1.Quality consistency evaluation of Tongmai preparations

AIM To evaluate the quality consistency of Tongmai Granules,Tongmai Tablets,Tongmai Capsules and Tongmai Oral Liquid.METHODS The HPLC fingerprints were established,after which the contents of danshensu,protocatechuic aldehyde,3'-hydroxy puerarin,puerarin,puerarin apioside,daidzin,ferulic acid,salvianolic acid B and salvianolic acid A were determined,and cluster analysis and principal component analysis were adopted in the quality analysis from the perspective of daily intake.RESULTS There were 21 common peaks in the fingerprints for 39 batches of samples with the similarities of 0.765-0.997.Various batches of samples were clustered into 5 categories,2 principal components demonstrated the accumulative variance contribution rate of 83.53% .The daily intakes of various constituents in different dosage forms exhibited obvious differences,especially for that of salvianolic acid B,which were low in tablets and capsules,and their heterogeneities existed among the same dosage forms.CONCLUSION This simple and accurate method can provide a reference for the quality evaluation of Tongmai preparations from different manufacturers.
2.FRMD4A promotes autophagy in placental trophoblast cells in preeclampsia

Aim To investigate the role of FRMD4A in autophagy of placental trophoblast cells in preeclampsia(PE).Methods The placental tissues and clinical data of normal pregnancy and PE were obtained,and the histopathological changes were observed by HE staining.An in vitro model of hypoxia-induced HTR-8/SVneo trophoblast cells was established.The expres-sions of LC3B Ⅱ/Ⅰ and p62 in placental tissues and hypoxic cell models were analyzed by Western blot.The expression of FRMD4A was detected by qRT-PCR,Western blot and immunofluorescence,and the correlation between the expression level of FRMD4A and the clinical characteristics of the subjects was ana-lyzed by Pearson correlation analysis.Hypoxia induced trophoblast cells were transfected with si-FRMD4A,and the expression of LC3 B Ⅱ/Ⅰ and p62 was analyzed by Western blot.Results Compared with the normal group,the expression of LC3B Ⅱ/Ⅰ in PE placental tissues and hypoxia-induced trophoblast models was significantly upregulated,while the expression of p62 was significantly downregulated.Meanwhile,the ex-pression of FRMD4A increased significantly.Moreo-ver,its expression was positively correlated with the maternal systolic blood pressure,diastolic blood pres-sure,and platelet count,but negatively correlated with the neonatal weight(P<0.01).In addition,hypoxia-induced trophoblast cells transfected with si-FRMD4A showed a significant decrease in LC3B Ⅱ/Ⅰ and an increase in p62 expression.Conclusions The expres-sion of FRMD4A is upregulated in PE placenta and hy-poxia-induced trophoblast cell model.Interfering with it can significantly hinder the autophagy process of trophoblast cells,suggesting that it may serve as a po-tential molecular target to participate in the pathologi-cal process of PE.
3.Progress in role and intervention of M2 microglia in ischemic stroke

Stroke is one of the main diseases that threaten human health,including ischemic stroke and hemorrhagic stroke,with the former being the main cause.The important pathogenesis of ischemic stroke includes neuroinflammation,oxidative stress,and excitatory toxic damage,and neuroinflammation plays an impor-tant role in the pathogenesis and rehabilitation process of ische-mic stroke.Microglia are inherent immune cells in the central nervous system,which monitor the site of injury and respond to the immune response as soon as a stroke occurs.The activated microglia are mainly polarized into pro-inflammatory M1 type and anti-inflammatory M2 type.The latter improves neurological dys-function by inhibiting neuroinflammation,promoting neuronal re-generation and myelin repair,and maintaining the integrity of the blood-brain barrier.It suggests that it may be a potential target for treating ischemic stroke by combating acute phase injury and promoting chronic phase rehabilitation.Precise regulation of M1/M2 activation has important therapeutic value in cerebral protection in ischemic stroke.This article focuses on the role of M2 microglia in ischemic stroke and the mechanism of various drugs or acupuncture and moxibustion therapy regulating the transformation of microglia into M2 type,in order to provide theo-retical basis for clinical treatment of stroke and new drug devel-opment.
4.Construction and In Vitro Testing of Genipin Cross-linked Hemerythrin Nanoparticles

Objective:To explore the feasibility of modifying hemerythrin molecules with natural cross-linker genipin,and evaluate its efficacy and safety.Methods:Hemerythrin was isolated and purified from sipunculid worms using tangential flow ultrafiltration.Subsequently,genipin cross-linked hemerythrin nanoparticles(GHrNPs)were constructed by adding 20%w/w genipin under mildly acidic conditions,and glutaraldehyde cross-linked hemerythrin nanoparticles(GAHrNPs)were constructed by adding 10%w/w glutaraldehyde under mildly alkaline conditions.The diameter,dispersity index,zeta potential,functional group structure,P50,and Hill coefficient of the two nanoparticle groups were measured.The two nanoparticle groups at different concentrations were co-cultured with vascular endothelial cells for 24 hours,then the cell viability and NO concentration in the culture medium were measured.Results:After glutaraldehyde/genipin molecular cross-linking,infrared spectra showed the continuous presence of amide bands Ⅰ and Ⅱ.The hydrated particle sizes of hemerythrin,GHrNP and GAHrNP were(93.14±2.11),(109.53±3.54),and(115.65±2.65)nm,dispersity indexes were 0.30±0.06,0.27±0.05,and 0.25±0.03,zeta potentials were(-24.00±1.54),(-19.52±1.31),and(-18.90±1.25)mV,P50 values were(9.28±0.22),(8.50±0.54),and(5.75±0.90)mmHg,and Hill coefficients were 1.61±0.14,1.58±0.17,and 1.41±0.22,respectively.The average hydrated particle size increased after cross-linking with hemerythrin,the negative value of the zeta potential decreased(both P<0.05).The P50 value of GAHrNP was significantly decreased than that of hemerythrin and GHrNP(P<0.05).The viability of vascular endothelial cells in the GHrNP group was higher than that in the GAHrNP group at different mass concentrations(P<0.05).The NO concentration in the culture medium of vascular endothelial cells in the GHrNP group was higher than that in the GAHrNP group only at 2.0 mg/ml(P<0.05).Conclusion:Hemerythrin molecules cross-linked by genipin can form stable nanoparticles with good oxygen-carrying activity and lower cytotoxicity compared to glutaraldehyde.
5.Clinical Application of Multimodal MRI in Cognitive Impairment Associated with Cerebral Small Vessel Disease

Objective:To explore the clinical application value of multimodal Magnetic Resonance Imaging(MRI)in cognitive impairment related to Cerebral Small Vessel Disease(CSVD).Methods:This study was a retrospective analysis of 60 CSVD patients,who were divided into cognitive impairment group(n=32)and normal group(n=28)based on Montreal Cognitive Assessment scale(MoCA).Compare the general information and MRI parameters of two groups of patients,use correlation analysis and logistic regression analysis to investigate the relationship between MRI parameters and cognitive dysfunction,and use receiver operating characteristic curve(ROC)to explore the value of MRI parameters in diagnosing cognitive dysfunction in cerebral small vessel disease.Results:Compared with the cognitively normal group,the cognitive impairment group had older age,higher apparent diffusion coefficient(ADC),lower anisotropy score(FA),and an overall increase in microbleeds(P<0.05).Age(r=-0.510),ADC value(r=-0.591),and overall number of microbleeds(r=-0.369)were significantly negatively correlated with MoCA score(P<0.05),while FA value(r=0.262)was significantly positively correlated with MoCA score(P<0.05).The logistic regression results indicate that age,overall microbleeds,ADC value,and FA value are influencing factors for cognitive impairment in patients with cerebral small vessel disease.ROC curve analysis showed that the combination of age,ADC,FA,and microbleeds significantly improved diagnostic efficacy(AUC=0.990,95%CI=0.974-1.000).Conclusion:The joint analysis of multimodal MRI parameters can provide important imaging evidence for the early identification of cognitive dysfunction in CSVD.
6.Dahuang Zhechong pill regulates stress fiber remodeling mediated by mechanistic signaling pathway to alleviate liver cirrhosis

Aim To explore the effect of the classical famous prescription Dahuang Zhechong pill(DHZCP)on relieving liver cirrhosis by regulating the stress fiber remodeling mediated by mechanistic signaling pathway and to explore the underlying mechanism.Methods Mice were randomly divided into the control group,model group,DHZCP low-dose group,DHZCP high-dose group,and Colchicine-positive control group.The liver cirrhosis mouse model was constructed by intrap-eritoneal injection of olive oil-solubilized CCl4.HE staining and serologic markers were used to reflect liver injury.Masson staining was used to evaluate collagen deposition in liver tissue.ELISA was applied to detect vasoactive molecules and cancer indicators.Atomic force microscopy was employed to detect liver tissue stiffness.Color Doppler diagnostic instrument was used to assess portal blood flow velocity.Western blot was utilized to detect ROCK2 expression and phosphoryla-tion of YAP,Cofilin,and MLC.Results The liver tis-sues in the model group had obvious inflammatory cell infiltration and collagen deposition,accompanied by significant elevation of serum transaminases and fibrosis indexes.Similarly,vasoactive molecules and cancer in-dicators were elevated,and the mechanoregulatory pro-tein ROCK2 expression and phosphorylation of Cofilin and MLC were elevated,with YAP being strongly de-phosphorylated.Both low and high doses of DHZCP re-versed the pathological changes,serological indices,and inhibited the activation of the stress fiber(SF)re-modeling mechanistic signaling pathway.Conclusion DHZCP effectively ameliorates liver tissue lesions in mice with liver cirrhosis,and its mechanism may be re-lated to the inhibition of SF remodeling mechanistic signaling pathway.

10.Research progress of spinal-pelvic characteristics in adolescent patients with idiopathic scoliosis.

Adolescent idiopathic scoliosis (AIS) is a common spinal deformity in adolescents, with potential causes etiologies associated with mesenchymal stem cells, genetic factors, histological features, and biomechanical aspects. Biomechanically, the pelvis, serving as the central and majort load-bearing structure, exhibits morphological and alignment abnormalities highly correlated with the development of AIS. Recent studies have extensively explored three-dimensional pelvic parameters and kinematics, demonstrating that abnormal pelvic characteristics may contribute to AIS onset and progression and are increasingly incorporated into clinical interventions. This review summarizes sagittal and coronal features of the spine-pelvis, as well as the influence of three-dimensional kinematic features on the pathogenesis of AIS, providing insights for advancing the study of spine-pelvis features related to AIS.
