Aim To examine the pathogenesis of disul-fide death gene in vascular dementia(VD)by bioin-formatics analysis of disulfide death differentially ex-pressed genes(DEGs)combined with experimental verification. Methods The death DEGs of disulfide were screened and their correlation was analyzed. The VD patients data in the data set were analyzed by clus-tering and typing and gene set variation. The clustering risk of DEGs was tested with a nomogram model, and the optimal learning model was predicted. After the es-tablishment of VD rat model, water maze test,HE stai-ning and RT-qPCR detection were performed to verify the results of health information. Results Four DEGs including SLC7A11 were obtained, which had antago-nistic or synergistic interaction with each other. The genetic data could be divided into two subtypes with significant differences. After typing,VD disulfide DEGs were mainly concentrated in GnRH signaling pathways. The accuracy of the nomogram prediction model was high.Generalized linear was the best ma-chine learning model.Compared with the sham opera-tion group, the escape latency of rats in the model group was prolonged, the number of crossing platforms decreased, the relative mRNA expression levels of Slc3a2 and Slc7a11 decreased, and LRPPRC in-creased.Conclusions SLC7A11 and other disulfide death DEGs and its related GnRH signaling pathway may be an important part of the pathogenesis of VD di-sulfide death.SLC3A2,LRPPRC and SLC7A11 can be used as characteristic genes in the regulation of VD by disulfide death, which may affect VD progression through the regulation of disulfide death.

Objective:To explore the role of nuclear receptor-binding SET-domain protein 1(NSD1)in the pathogenesis of nonobstructive azoospermia(NOA)by regulating the expressions of relevant genes. Methods:We detected the expression of NSD1 in the testis tissue of 7 male patients with obstructive azoospermia(OA)and 18 with NOA by qPCR and immunofluorescence assay, and determined the modification level of H3K36me2 in the testes of two groups of patients by immunofluorescence staining,Western blot and immunoprecipitation(IP).We examined the difference in the enrichment of H3K36me2 in the testis tissue by chromatin IP-based sequencing(ChIP-Seq),analyzed the genomic distribution and target genes using bioinformatics, and verified the expression levels of the target genes in the testes of the two groups of patients by qPCR.Results:Compared with the patients with OA,those with NOA showed dramatically decreased mRNA and protein expressions of NSD1(P=0.000 8).The binding of NSD1 to H3K36me2 was observed in the testis tissue of both the two groups of patients, while the modification level of H3K36me2 was evidently reduced in the NOA males.H3K36me2 was distributed mainly in the intergenic region in the testes of the two groups of patients, but the enrich-ment of H3K36me2 was obviously decreased in the NOA group. The differentially H3K36me2-enriched genes were involved in various biological processes, including tissue development, and cell morphogenesis. Results of ChIP-Seq and qPCR showed significantly down-regulated expressions of the target genes KIT,SPO11 and ACRV1 in the testis tissue of the NOA males compared with those in the OA patients(P<0.01).Conclusion:The levels of NSD1 and H3K36me2 are decreased in testis tissue of the NOA patient,H3K36me2 is highly enriched in the spermatogenesis-related key genes KIT,SPO11 and ACRV1,and the down-regulated expression of NSD1 impairs spermatogenesis.

Journal of Regional Anatomy and Operative Surgery 2025;34(7):563-569
Objective To determine the knee joint cartilage thickness using different methods and explore the feasibility of mathematical statistical models of dataset for the prediction of cartilage thickness. Methods A total of 304 patients diagnosed as knee osteoarthritis(OA)combined with varus deformity and undergoing unilateral total knee arthroplasty at the Second Affiliated Hospital of Army Medical University from March 2023 to March 2024 were selected for the study. All patients had complete preoperative and postoperative clinical data. The healthy cartilage at four anatomical sites of patients, including the distal femur lateral condyle, lateral tibial plateau, posterior medial femoral condyle, and posterior lateral femoral condyle were selected, and the knee joint cartilage thickness was determined based on preoperative MRI analysis, robotic navigation system tracing, tissue section of surgical specimen and digital vernier caliper. The baseline indicators of demographics, disease and imaging ffor patients were collected to construct a dataset, and four models of linear regression analysis, principal component analysis,Least Absolute Shrinkage and Selection Operator(LASSO)regression analysis, and K-nearest neighbors(KNN)analysis were established for predicting the accuracy, determination coefficient(R2)and root mean square error(RMSE),and the regression equation for predicting cartilage thickness was established. Results The knee joint cartilage thicknesses determined by preoperative MRI analysis, robotic navigation system tracing, tissue section of surgical specimen had no statistically significant difference with that by digital vernier caliper(P>0.05).The predictive efficiencies of models of linear regression analysis, principal component analysis, and LASSO regression analysis for the knee joint cartilage thickness all failed to meet the expectations(R2<0.3,RMSE>0.03).The predictive effect of KNN model on the cartilage thickness of the distal femur lateral condyle and lateral tibial plateau was not ideal(R2=0.23,RMSE=0.29),while it had potential predictive value(accuracy=0.21,accuracy=0.15).Conclusion The prediction model of knee joint cartilage thickness based on individual parameters has certain scientificity, and the feasibility of KNN model is relatively high. However, due to insufficient sample size and unclear individual parameter weight, the efficiencies of the four established prediction models are not ideal, which fails to provide definite prediction equations. Therefore, the construction scheme of the prediction model still needs to be further optimized.
