1.Relationship between GH/IGF-1 axis and type 2 diabetic kidney disease in adults

Objective This study examined changes in serum growth hormone(GH)and insulin-like growth factor 1(IGF-1)levels,as well as insulin resistance,in patients with type 2 diabetes mellitus(T2DM)across different stages of diabetic kidney disease(DKD).It explored the relationship between these factors and the occurrence and progression of DKD,with the aim of identifying new targets for the prevention and treatment of DKD.Methods In this cross-sectional study,T2DM patients aged 18-80 years were selected from the Department of Endocrinology,The Second Affiliated Hospital of Xi'an Jiaotong University,from November 30,2019 to March 30,2021.Their serum GH and IGF-1 levels were tested.Results The serum GH level increased gradually in the groups,and the difference was statistically significant(P<0.05).The level of serum IGF-1 decreased first and then increased among the groups,but with no significant difference(P>0.05).HOMA-IR showed no significant correlation among the groups(P>0.05),but HOMA-β gradually decreased among the groups,with statistically significant difference(P<0.05).Multivariate Logistic regression analysis showed that the increase of serum GH level was closely related to the progression of T2DM to early DKD,and that the increase of serum IGF-1 level might be related to the deterioration of renal function in patients with T2DM at DKD stage.HOMA-IR and HOMA-β were not found to have correlation with the occurrence or development of DKD in T2DM patients.Conclusion In patients with T2DM,elevated serum GH level is closely associated with the occurrence and progression of early DKD.The increase of serum GH level may promote the occurrence of early DKD in T2DM,and the increase of serum IGF-1 level may be related to the deterioration of renal function in clinical DKD stage.In this study,we did not find correlation of HOMA-IR and HOMA-β with the occurrence or development of DKD in T2DM patients.

3.Clinical observation of goserelin combined with estradiol valerate in the treatment of endometriosis in patients of childbearing age after laparoscopic surgery

OBJECTIVE To observe the efficacy and safety of goserelin combined with estradiol valerate in the treatment of endometriosis (EMs) in patients of childbearing age after laparoscopic surgery. METHODS The clinical data of 144 patients with EMs of childbearing age who underwent laparoscopic surgery in our hospital from January 2020 to December 2022 were collected retrospectively, and those patients were divided into control group (70 cases) and combination group (74 cases) based on different postoperative medication regimens. Control group was given Acetate goserelin sustained-release implant on the 1st to 5th day of their first menstrual cycle after laparoscopic surgery, while combination group was additionally given Estradiol valerate tablets on the basis of control group. The short-term and long-term efficacy of 2 groups were observed; preoperative and postoperative pain scores, ovarian reserve function, and the incidence of adverse reactions were all recorded in 2 groups. RESULTS There was no statistically significant difference in short-term total effective rate, 1-year postoperative recurrence rate and the incidence of decreased libido, vaginal dryness, gastrointestinal reactions, rash, or arrhythmia between two groups (P>0.05). The natural pregnancy rate of combination group was significantly higher than control group at 1 year after surgery, while the incidence E-mail:wanghuiling0518@126.com of early miscarriage, hot flashes and sweating, emotional fluctuations, insomnia and fatigue, joint muscle pain, dizziness and headache were significantly lower than control group (P<0.05). However, at 6 months after surgery, pain scores and the levels of anti-Müllerian hormone, follicle stimulating hormone, luteinizing hormone, estradiol of both groups were significantly lower than before surgery in the same group, while the number of antral follicle count was significantly higher than before surgery in the same group; the levels of anti-Müllerian hormone, luteinizing hormone, estradiol, and the number of antral follicles in the combination group were significantly higher than control group, while pain score and follicle stimulating hormone level were significantly lower than control group (P<0.05). CONCLUSIONS The combination of goserelin and estradiol valerate after laparoscopic surgery has a significant therapeutic effect and good safety in the treatment of EMs of childbearing age.
4.Progress and analysis of 2019 novel coronavirus vaccine research and development

To compare the representative novel coronavirus vaccine in the world at present and to provide a new and more comprehensive reference for the research and development of novel coronavirus vaccine. This review summarized the virological and immunological basis of novel coronavirus vaccine and made a detailed comparison and analysis of some representative vaccines based on different technologies and clinical trials in phase 2a/3. The immune effect of many kinds of novel coronavirus vaccines in phase 3 clinical trials around the world is encouraging, but the relationship between vaccine immunogenicity and safety and the reliability of clinical trial data still need to be paid close attention to. Designing new vaccine targets, inducing more extensive immune protection, improving immunogenicity, strict clinical trials and establishing large-scale cohort studies are higher requirements for novel coronavirus vaccine research and development.
5.Construction of a lentivirus vector for Trail gene in rats

BACKGROUND:Adenovirus, expressing within a limited period, can limit the expression time and amount of target genes that is not conducive to ongoing experiments. Here, we select adenovirus as vectors for genetic recombination with tumor necrosis factor (TNF)-related apoptosis-inducing ligand (Trail) gene fragment.
OBJECTIVE:To explore the construction of recombinant lentiviral vector carrying Trail in rats
METHODS:The Trail gene was obtained:according to GenBank in rat Trail gene sequence (NM_145681.1), we designed the gene specific primers of Trail-Age I-F and Trail-AgeI-R, and used AgeI as enzyme cutting site. PCR was applied to amplify Trail gene from rat cDNA Library and construct recombinant plasmids after cutting Trail gene to be cloned into expression vector GV218 by AgeI. Recombinant plasmids were transfected into 293T cells by Lipofeetamine2000 encapsulated recombinant plasmid and auxiliary packaging carrier. The Trail protein of lentiviral plasmids was expressed. Fol owing virus col ection, we identified virus titer and extracted protein from cells to detect Trail expression by western blot assay.
RESULTS AND CONCLUSION:Screened positive Escherichia coli DH5a competent cells were sequenced with 861 bp, which was consistent with Trail nucleotide sequence in GenBank. After transfection 2 days, virus liquid was col ected and confirmed as recombinant plasmid including Trail gene by PCR and Trail proteins expressed in 293T cells by western blot assay. Hole dilution method and real-time fluorescent quantitative PCR determination showed that the virus titer was 2×109 TU/mL. In this study, recombinant lentiviral vector carrying Trail is successful y constructed by homologous recombination in Escherichia coli.
6.Clinical Study on Irbesartan and Aspirin Combined with Tetramethylpyrazine in the Treatment of Early Diabetic Nephropathy

OBJECTIVE: To explore the clinical efficacy of irbesartan, enteric-coated aspirin combined with tetramethylpyrazine in the treatment of diabetic nephropathy. METHODS: 75 patients with diabetic nephropathy were randomly divided into treatment group(n=40) and control group(n=35). Both groups were administered with 150 mg irbesartan(p.o.) and 100 mg enteric-coated aspirin (p.o.) once a day. Treatment group were additionally treated with 0.9% normal saline containing 200 mg tetramethylpyrazine once a day (i.v.gtt). In this study, 5'-CMP was sulfonated, and then the modified 5'-CMP was connected to a protein carrier as an immunogen to immunize BALB/c mice. After cell hybridization, screening and recloning , a McAb (B10) with high sensitivity and specificity was selected. In a dot Hot using the McAb B10, less than 0.05 pg of sulfonated DNA could be detected while 10 ng of DNA was not coloured The result showed that the sensitivity of McAb B10 was higher than that of the McAb from "Chemiprobe" kit
