S-methyl-L-cysteine sulfoxide (SMCO) is a non-protein sulfur-containing amino acid with a variety of functions. There are few reports on the enzymes catalyzing the biosynthesis of SMCO from S-methyl-L-cysteine (SMC). In this study, the flavin-containing monooxygenase gene derived from Schizosaccharomyces pombe (spfmo) was heterologously expressed in Escherichia coli BL21(DE3) and the enzymatic properties of the expressed protein were analyzed. The optimum catalytic conditions of the recombinant SpFMO were 30 ℃ and pH 8.0, under which the enzyme activity reached 72.77 U/g. An appropriate amount of Mg2+ improved the enzyme activity. The enzyme kinetic analysis showed that the Km and kcat/Km of SpFMO on the substrate SMC were 23.89 μmol/L and 61.71 L/(min·mmol), respectively. Under the optimal reaction conditions, the yield of SMCO synthesized from SMC catalyzed by SpFMO was 12.31% within 9 h. This study provides reference for the enzymatic synthesis of SMCO.

Background and Purpose:Previous studies have investigated the prognostic significance of skeletal muscle and adipose tissue composition and distribution in colorectal cancer patients, yet most have not differentiated between rectal and colon cancer patient cohorts. This study aimed to explore the relationship between body composition and long-term prognosis, and to develop a postoperative predictive model. Methods:Clinical data of rectal cancer patients who underwent surgical treatment at Qingdao University Affiliated Hospital from January 2018 to December 2021 were retrospectively collected.Inclusion criteria:①Age>18 years;② Preoperative colonoscopy and pathological diagnosis of colorectal cancer;③ Complete surgical resection;④Abdominal computed tomography(CT)scan 1 month before surgery.Exclusion criteria:① Clinical data is missing;② Multiple metastases of tumors;③ Tumor T stage 0 or carcinoma in situ;④ Severe artifacts lead to poor quality CT imaging, making it difficult to distinguish between fat and muscle;⑤ Inability to obtain follow-up results. This study has been approved by the Medical Ethics Committee of the Affiliated Hospital of Qingdao University(approval number:QYFYWZLL30313),and informed consent has been waived in the ethical approval process. The skeletal muscle index(SMI)and subcutaneous adipose tissue index(SATI)were calculated by dividing the areas of skeletal muscle and subcutaneous fat observed on CT scans by the square of the patient's height.Univariate and multivariate COX regression analyses were conducted to identify risk factors influencing recurrence-free survival(RFS)and overall survival(OS)in rectal cancer patients.Based on the results of the multivariate analysis,a nomogram prediction model was developed, its predictive power and accuracy were assessed using the receiver operating characteristic(ROC)curve, calibration plots and decision curve analysis(DCA),and internal validation was conducted. Results:A total of 696 patients were included in this study, with 96(13.8%)patients experiencing postoperative recurrence and 89(12.8%)patients dying.Multivariate COX regression analysis showed that SMI,SATI,tumor T stage and N stage were independent factors affecting the postoperative RFS and OS of patients.Nomogram prediction models for RFS and OS in rectal cancer patients were constructed based on the above independent predictors. The area under ROC curve(AUC)for 3-,4-and 5-year RFS was 0.862,0.846 and 0.824,respectively;the AUC for 3-,4-and 5-year OS was 0.886,0.898 and 0.875,respectively.The models were evaluated using calibration curves and decision curves, and internal validation was performed, which showed that the prediction accuracy of the models was good. Conclusion:CT body composition is an independent predictor of RFS and OS in rectal cancer patients, and the nomogram model developed based on these factors demonstrates good predictive value for patient prognosis.

Currently, the first-generation of proton pump inhibitors(PPIs)commonly utilized in clinical practice for the treatment of peptic ulcer include omeprazole, lansoprazole, and pantoprazole, and the second generation include rabeprazole and esomeprazole. The predominant mode of PPI administration is oral, which poses numerous challenges in clinical settings owing to their poor solubility, susceptibility to gastric acid degradation, short half-life,and limited stability. This article presents a comprehensive review of studies exploring the novel states of these five PPIs,include polymorphism,co-crystal drugs, and nanodrugs.Furthermore,it summarizes the advancements in research aimed at modifying the physicochemical properties of these drugs through innovative solid states, thereby enhancing their bioavailability and stability. This work offers a new perspective for researchers endeavoring to develop new states of pharmaceutical substances that incorporate PPIs.

Chinese Medical Journal 2023;136(3):253-268
Integrins are a family of transmembrane receptors that connect the extracellular matrix and actin skeleton, which mediate cell adhesion, migration, signal transduction, and gene transcription. As a bi-directional signaling molecule, integrins can modulate many aspects of tumorigenesis, including tumor growth, invasion, angiogenesis, metastasis, and therapeutic resistance. Therefore, integrins have a great potential as antitumor therapeutic targets. In this review, we summarize the recent reports of integrins in human hepatocellular carcinoma (HCC), focusing on the abnormal expression, activation, and signaling of integrins in cancer cells as well as their roles in other cells in the tumor microenvironment. We also discuss the regulation and functions of integrins in hepatitis B virus-related HCC. Finally, we update the clinical and preclinical studies of integrin-related drugs in the treatment of HCC.

Through the development of the guidance for case handling of radiological health law enforcement, health supervisors are instructed to accurately grasp the key points of law enforcement and case handling and standardize the process of collecting evidence and law application to ensure the correct implementation of administrative penalty. This article explains the structure and content of the guidance for case handling of radiological health law enforcement by taking the case of arranging radiation workers who have not undergone an occupational health examination to engage in the radiological occupational-disease-inductive operation in medical institutions as an example.

Objective:To collect the upper respiratory tract specimens from both domestically-spread and imported Coronavirus Disease 2019 (COVID-19) confirmed cases, sequence the whole genome of 2019 novel coronavirus (2019-nCoV) and analyze the genetic variation and the phylogenetic characteristics. Methods:The viral nucleic acids of all specimens were extracted, the sequencing library was constructed following the manufacturing instructions, and the sequencing data were analyzed by bioinformatics software. Results:The whole genomes of 2019-nCoV strains from 24 samples were all obtained and had high similarity with the genome of the reference strain Wuhan-Hu-1 (GenBank ID: NC_045512) and the similarity rates were all greater than 99.9%. Six 2019-nCoV strains from domestically-spread cases were classified into L-lineage and the others were S-lineage. The 2019-nCoV strains from the imported cases had 48 nucleotide mutation positions, but those from the domestically-spread cases only had 14 mutation positions. Most of 2019-nCoV strains from the imported cases (15/16) had the specific D614G mutation site in spike protein, while those from the domestically-spread cases had none. Conclusions:The 2019-nCoV strains from the imported cases in Tianjin city had more mutation sites than those from the domestically-spread cases, and most of 2019-nCoV strains from the imported cases had D614G mutations in spike protein which had more transmissive rates and were easier to spread among humans.
