Objective To explore the mechanism of acupuncture's effect on hippocampal neuron apoptosis in a post-traumatic stress disorder(PTSD)rat model via the reactive oxygen species(ROS)/c-Jun N-terminal kinase(JNK)/p53 pathway.Methods Thirty rats were randomly divided into normal control group(NC group),PTSD rat model group(PTSD group)and electroacupuncture intervention group(EA group),with 10 rats in each group.The results of behavioral test(open field experiment and elevated cross maze experiment)were compared.The terminal deoxynucleotidyl trans-ferase dUTP nick-end labeling(TUNEL)staining method was used to detect hippocampal neuron apoptosis in each group.DHE fluorescence staining was employed to measure ROS levels in the brain tissue of each group.The content of Ros index[malondialdehyde(MDA),superoxide dismutase(SOD)and glutathione(GSH)]in brain tissue of each group was detected.Western blot analysis was per-formed to determine the protein expression levels of JNK,p-JNK,p53 and PUMA in hippocampal tissue.Results The total movement distance of rats in the PTSD group was significantly shorter,and the total number of uprightand central cell entry times were significantly shorter than those in the NC group(P＜0.05).The total distance moved by rats in the EA group was significantly longer,and the total number of upright and central lattice entry times were significantly higher than those in the PTSD group(P＜0.05).The percentage of times and time of entering the open arm in the PTSD group was significantly lower,and the anxiety index was significantly higher than that in the NC group(P＜0.05).The percentage of times and time of entering the open arm in the EA group was significantly higher,and the anxiety index was significantly lower than that in the PTSD group(P＜0.05).The neuronal apoptosis rate of the PTSD group was significantly higher than that of the NC group(P＜0.05);the neuronal apoptosis rate in the EA group was significantly lower than that in the PTSD group(P＜0.05).The mean fluorescence intensity of ROS in brain tissue of the PTSD group was significantly higher than that of the NC group(P＜0.05);the mean fluorescence intensi-ty of ROS in the EA group was significantly lower than that in the PTSD group(P＜0.05).The MDA content in brain tissue of the PTSD group was significantly higher,and SOD activity and GSH content were significantly lower than those of the NC group(P＜0.05);the MDA content in the brain tissue of the EA group was significantly lower,and the SOD activity and GSH content were sig-nificantly higher than those of the PTSD group(P＜0.05).The p-JNK/JNK and the expression levels of p53 and PUMA in brain tissue of the PTSD group were significantly higher than those of the NC group(P＜0.05);the p-JNK/JNK and the expression levels of p53 and PUMA in the brain tissue of the EA group were significantly lower than those of the PTSD group(P＜0.05).Conclusion Acu-puncture can inhibit hippocampal neuron apoptosis and relieve fear symptoms in PTSD rats,possibly through the activation of the ROS/JNK/p53 pathway.

Objective:To explore the clinical effects of Wenjing Decoction in treating endometriosis syndrome with cold coagulation and blood stasis.Methods:A randomized controlled trial was performed. A total of 90 cases were collected and with distributed random number method of drugs were divided into control group and the treatment group with 45 cases each. The treatment group was given Wenjing Decoction granules, while control group was given placebo (10% Wenjing Decoction). Both groups were taken for 14 consecutive days from the first day of menstruation as a course of 1, a total of 3 courses. Before and after treatment, main observation indexes including dysmenorrhea, Traditional Chinese Medicine (TCM) syndromes and the secondary outcome including scores of menstruation, short form McGill pain questionnaire (SF-MPQ) were detected by subjective evaluation, interleukin-8 (IL-8), tumor necrosis factor-α (TNF-α), and prostaglandin F2α (PGF2α) level in human serum tissue were evaluated by enzyme linked immunosorbent assay and the clinical and TCM syndromes specific effects were determined.Results:Totally 7 patients were lost, including 1 case in the treatment group and 6 cases in control group. The number of actual observation cases was 83 (44 cases in the treatment group and 39 cases in control group). The clinical effective rate in the treatment group [75.00% (33/44)] was statistically higher than that in control group [23.08% (9/39), P<0.001]. As for therapeutic effect of TCM syndromes, the effective rate in the treatment group [70.45% (31/44)] was obviously higher than that in control group [20.51% (8/39), P<0.001]. After treatment, compared with the results of control group, menstruation [2.00 (1.00,4.00)], dysmenorrhea [7.25 (5.00,9.88)], SF-MPQ [10.00 (4.50,18.00)], TCM syndromes [13.50 (9.25, 20.00)] and the data of IL-8 [9.80 (5.73,27.53) ng/L], TNF-α [5.05 (4.10,6.20) ng/L] and PGF2α [(147.16±22.24) ng/L] in the treatment group were decreased obviously [4.00 (2.00,6.00), P=0.022; 8.00 (6.50,12.00), P=0.030; 13.00 (4.00,24.00), P<0.001; 17.00 (9.00, 26.00), P<0.001; 18.20 (10.00,30.90) ng/L, P=0.013; 5.80 (4.50,10.70) ng/L, P=0.035;(165.17±30.33) ng/L, P=0.001]. Conclusion:Wenjing Decoction can treat TCM syndromes of endometriosis patients with cold coagulation and blood stasis, and the clinical curative effect was significant. The mechanism may involve enhancing immunity and relieving uterine smooth muscle contraction.

OBJECTIVE: To explore the protective and therapeutic effects of Faecalibacterium prausnitzii (Fp) supernatant on ulcerative colitis (UC) in mice induced by dextran sulfate sodium (DSS) and the underlying mechanisms.
 METHODS: Forty male mice were randomly divided into a control group, a model group, a treatment group and a prevention group (n=10 in each group). The colorectal histopathologic damage score (HDS) were calculated; the proportion of helper T cell (Th17) in mononuclear cells (MNC) in spleen, the levels of IL-17A and IL-6 in plasma were detected; the mRNA levels of transcription factor retinoic acid-related orphan receptor-γt (ROR-γt), interleukin (IL)-17A and IL-6 in colon mucosa tissues were also determined.
 RESULTS: Compared with the model group, the colon HDS in the treatment group and the prevention group were significantly decreased (both P<0.05), but there was no significant difference in the treatment group and the prevention group (P>0.05). The proportion of Th17 cells in spleen in the treatment group and the prevention group was also remarkably lower than that in the model group (both P<0.01). The levels of IL-17A and IL-6 in plasma in the treatment group were significantly lower than those in the model group (both P<0.05). The mRNA expression of ROR-γt, IL-17A and IL-6 in the colon mucosa tissues in the treatment group were remarkably lower than those in the model group (all P<0.05). But there was no statistic difference in the level of IL-6 in the plasma and the colon mucosa tissues between the prevention group and the model group (P>0.05).
 CONCLUSION Fp supernatant has protective and therapeutic effects on ulcerative colitis in mice induced by DSS, which might be mediated by decrease of Th17 and IL-17A levels in the plasma and the colon mucosa tissues. Fp supernatant also can decrease mice colitis by reducing IL-6 levels.

AIM: To establish the method of determining procyanidin B2 content in grape seed extract by HPLC. METHODS: The determination was carried out with ZORBAX SB-C_ 18 column. The mobile phase consisted of acetonitrile-2% acetate buffer under the gradient elution condition, detection wavelength was at 280nm. RESULTS: There was a good linear relationship between the peak area and concentration in the range of 1~30 ?g/mL for procyanidin B2. The average of procyanidin B2 (n=5) was 99.29% and RSD=1.64%, respectively. CONCLUSION: The method is simple, accurate, reproducible and can be used for assay of procyanidin B2.
