1.Optimization of the Molding Process of Classical Prescription Xiebai Granules Based on Box-Behnken Design-Response Sur-face Method and BP Neural Network Method

OBJECTIVE To optimize the molding process of Xiebai Granules(XG)using the Box-Behnken design-response surface method combined with the BP neural network method,and to evaluate the consistency of particle quality between different bat-ches by establishing physical fingerprint.METHODS Dry paste powder was used as the main drug,dry granulation was adopted,and the forming rate,dissolution rate,moisture absorption rate and angle of repose of the granules were used as evaluation indexes,the ex-cipients dextrin,maltodextrin and lactose of the particles,were screened by single factor test combined with simplex-lattice design and entropy weight method,and the optimal excipient ratio was selected.The entropy weight method combined with the Box-Behnken de-sign-response surface method and the BP neural network algorithm were used to optimize the process parameters,and the process veri-fication was carried out.The physical fingerprint was used to comprehensively characterize the bulk density(Da),hygroscopicity(H),moisture(HR),tap density(Dc),angle of repose(α),Hausner ratio(IH),relative uniformity index(Iθ),Carr index(IC),and in-terparticle pore number(Ie),and the consistency of particle quality in different batches was evaluated.RESULTS The optimal ratio of excipients was dextrin 15%,maltodextrin 48%,and lactose 37%.The optimal process parameters were conveying speed 95 r·min-1,pressure wheel speed 4 r·min-1 and hydraulic pressure 7 MPa.The similarity of the physical fingerprints of the five bat-ches of XG was greater than 0.98.CONCLUSION The optimized molding process of XG is stable and feasible,and the quality of different batches of XG is stable,which can provide a reference for the development and industrial scale-up production of XG.

3.Traceability of syphilis antibody detection in preserved samples

【Objective】 To investigate whether the current retention methods in blood stations can fully meet the traceability requirements of blood test results by analyzing the traceability of retained samples for syphilis antibody testing. 【Methods】 Thirty-four one-assay-positive deep-well plate retention samples, 68 double-assay-positive deep-well plate retention samples and 263 negative retention blood braids and corresponding deep-well plate retention samples that expired retention period for syphilis antibody testing from 2014 to 2020 in our center were collected. The TP-ELISA assays of two manufacturers were used for retesting, and the results were recorded and compared with the original results statistically. 【Results】 The concordance rate of the double-assay-positive and single-assay-positive samples with their corresponding deep-well plate samples was 98.53%(67/68) and 67.65%(23/34), respectively(P<0.05). 4.Effect of Chinese herbal medicine immunomodulator on T lymphocyte immune function in immature rats with obstructive jaundice

Objective To evaluate the effect of Chinese herbal medicine immunomodulator on T lymphocyte immune function in peripheral blood and intestinal mucosa of immature rats with obstructive jaundice. Methods Three-weeks Wistar rats were randomly divided into four groups. (n= 12, in each): normal control group, sham operation group, obstructive jaundice (OJ) group, OJ + Chinese herbal medicine immunomodulator (OJ+ZY) group. Except for the normal control group, the others' bile duct stones were ligatured to establish rat models with obstructive jaundice. The percentage of CD4+ and CD8+ tlymphocytes and the ratio of CD4+/CD8+ in peripheral blood and intestinal mucosa of immature rats was detected by flow cytometry. Results The percentage of CD4+ cell [(36.2 ±4.2)%, (28.8±1.8)% respectively] and the ratio of CD4+/CD8+ [(1.14±0.39), (1.37±0.34)respectively] in OJ group were lower than those in normal control group [peripheral blood: CD4+(41.5±5.3)%,CD4+/CD8+(1.37±0.19); intestinal mucosa: CD4+(32.3± 2.4)%, CD4+/CD8+ (1.84+0.28) and sham operation group (peripheral blood: CD4+ (41.2±5.5)%, CD4+/CD8+ (1.45±0.27); intestinal mucosa: CD4+(31.5 ± 2.7)%, CD4+/CD8+ (1.63±0.58)] . The difference was statistical significant(P<0.05). The percentage of CD4+ cell [(42.7±6.3)%, (33.6±2.4)% respectively] and the ratio of CD4+/CD8+ [(1.56±0.46), (1.84±0.56)respectively] in OJ+ZY group, were higher than those in OJ group(P<0.05). The difference was statistical significant(P<0.05). Conclusion Chinese herbal medicine immunomodulator can increase T lymphocyte immune function in immature rats with obstructive jaundice, but has no significance in normal control group as well as sham operation group.
5.Effect of Astragalus on T Lymphocyte Immune Function in Immature Rats with Obstructive Jaundice

Objective To evaluate the effect of Astragalus on T lymphocyte immune function in peripheral blood and intestinal mucosa of immature rats with obstructive jaundice. Methods Three-week-old Wistar rats were randomly divided into four groups(n=12,in each):control group, sham operation group, obstructive jaundice(OJ)group,OJ+Astragalus (OJ+A)group.The percentage of CD4 and CD8 T lymphocytes, the ratio of CD4 to CD8 in peripheral blood and intestinal mucosa of immature rats was detected by flow cytometry. Result The percentage of CD4+ cell and the ratio of CD4 to CD8 in the OJ group were lower than those in the control group and the sham operation group,with significant difference(P<0.05).The percentage of CD4 cell and the ratio of CD4 to CD8 in the OJ+A group were higher than those in the OJ group(P<0.05).Conclusion Astragalus can increase T lymphocyte immune function in OJ immature rats.
