1.Mechanism by which Tongan Decoction regulates synovial macrophage polarization in rats with knee osteoarthritis

BACKGROUND:Developed by the esteemed Chinese medicine master Wei Guikang,Tongan Decoction has proven highly effective in treating knee osteoarthritis.However,the mechanism of action is yet unclear.OBJECTIVE:To elucidate how Tongan Decoction modulates synovial macrophage polarization as a therapeutic strategy for knee osteoarthritis in a rat model.METHODS:(1)We employed high-throughput microRNA sequencing and polymerase chain reaction to analyze the differentially expressed miRNA in synovial macrophages of normal and knee osteoarthritis rats.Predicted target genes of miR-27a were identified using bioinformatics databases,with subsequent validation through luciferase assays.A total of 68 Sprague-Dawley rats were randomly divided into normal control group(n=16),model group(n=16),miR-27a overexpression group(n=12),Tongan Decoction group(n=12),and Tongan Decoction+miR-27a inhibition group(n=12).The miR-27a overexpression group and the Tongan Decoction+miR-27a inhibition group were injected with miR-27a mimic and miR-27a inhibitor in the right knee joint cavity,respectively,once daily for 5 continuous days.On the 15th day after modeling,the Tongan Decoction group and the Tongan Decoction+miR-27a inhibition group were given Tongan Decoction by gastric lavage,and the other three groups were given saline by gastric lavage,once daily for 14 continuous days.After administration,behavioral tests,X-rays,hematoxylin-eosin staining of synovial and cartilage tissues of the knee joint and immunofluorescence staining of synovial tissues of the knee joint,RT-PCR,and Western blot were performed.RESULTS AND CONCLUSION:High-throughput sequencing of miRNA showed low expression of miR-27a in synovial tissues of rats with knee osteoarthritis.The target gene of miR-27a was nuclear factor κB,and luciferase assay showed that the two could bind to each other.Behavioral assays showed that miR-27a overexpression or Tongan Decoction alleviated joint dysfunction in rats with knee osteoarthritis,and miR-27a inhibition antagonized the effect of Tongan Decoction.X-ray films and hematoxylin-eosin staining showed that miR-27a overexpression or Tongan Decoction reduced the degree of knee osteoarthritis and miR-27a inhibition weakened the therapeutic effect of Tongan Decoction.The results of RT-PCR and western blot assay showed that compared with the normal control group,the expression of interleukin 10 was reduced in the model group(P<0.05),and the expression of matrix metalloproteinase 13,interleukin 1β,and nuclear factor κB was elevated in the model group(P<0.05).miR-27a overexpression or Tongan Decoction could differently reverse the changes in the above-mentioned indexes,while miR-27a inhibition weakened the effect of Tongan Decoction.Immunofluorescence staining results showed that CD86 protein expression in the model group was higher than that in the normal control group(P<0.05),and CD206 protein expression was lower than that in the normal control group(P<0.05);miR-27a overexpression group and Tongan Decoction had lower CD86 protein expression than that in the model group(P<0.05),and higher CD206 protein expression than that in the model group(P<0.05);in the Tongan Decoction+miR-27a inhibition group,CD86 protein expression was higher(P<0.05)and CD206 protein expression was lower than that in the Tongan Decoction group(P<0.05).Tongan Decoction mitigates knee osteoarthritis by upregulating miR-27a expression and suppressing nuclear factor κB activity,which improves knee joint function and further treats knee osteoarthritis.
2.Application of solid collagen-based materials in medical devices

BACKGROUND:Collagen is the main component of extracellular matrix.With its stable triple helix structure,good biological characteristics and machinability,collagen is an ideal biomedical material and has been widely used in the field of medical devices.OBJECTIVE:To analyze the main product morphology,technical characteristics,preparation process,and clinical application of solid collagen-based materials.METHODS:The research on solid collagen-based materials in the field of medical devices included in PubMed and CNKI was searched by computer.The English and Chinese search terms were"collagen,collagen sponge,collagen membrane,collagen powder,collagen gel,decellularized extracellular martix,medical device,clinical application."After preliminary screening of all articles according to inclusion and exclusion criteria,92 articles with higher quality and relevance were retained for review.RESULTS AND CONCLUSION:Solid collagen-based materials from animal sources have been used as materials for the preparation of medical devices,and remarkable progress has been made in raw material extraction,preparation technology,and clinical application.Marine biological collagen and recombinant collagen have increased the selectivity of collagen sources.There are various types of solid collagen-based medical devices and various preparation methods,among which crosslinking modification is an important way to improve material properties,which is worthy of further study.In terms of product development ideas,research on solid collagen-based medical device products is developing in the direction of trivialization and biochemistry,and the research on compounding with other biological materials and drugs has also shown great potential.Although many collagen-based medical devices have been used clinically in China,product development in the fields of artificial skin,cartilage repair,and nerve injury repair is still insufficient,and further breakthroughs and innovations in technology and industrialization are needed.
3.Cognitive reserve and social cohesion in influence on fall-related self-awareness among the elderly in community

Objective To study the level of fall-related self-awareness among the elderly in community and to explore the influence of cognitive reserve and social cohesion on fall-related self-awareness,thereby providing a reference for developing targeted interventions.Methods Toally 500 old people in communities were recruited by convenience sampling in Hengyang between August and September 2024.Data were collected with the general information questionnaire,neighbourhood cohesion scale,self-awareness of falls in elderly scale,and cognitive reserve index questionnaire.Hierarchical multiple linear regression analysis was employed to explore the effects of cognitive reserve and social cohesion on fall-related self-awareness while controlling for demographic variables.Results A total of 467 valid questionnaires were returned.The score of fall-related self-awareness was 54.60±6.64(moderate level).Social cohesion scores was 29.84±5.52(moderate level),and cognitive reserve score was 93.27±10.83(moderate level).The cognitive reserve and social cohesion were both positively correlated with fall-related self-awareness(all P<0.01).Community integration accounted for 9.10%of its total variability,and cognitive reserve accounted for 5.70%of the total variability of fall vigilance,while controlling for variables such as body mass index(BMI),sleep quality,sedentary habits,fear of falling and financial resources.Conclusion The elderly in communities exhibit a low level of fall-related self-awareness.More attention to risk of falling is required along with developing targeted and individualised training programs aiming at improvement of cognitive reserve and social cohesion,thereby reducing the incidence of falls.

7.The influence of ultra-shortwave irradiation on Ca2+-ATPase expression after cerebral ischemia and reperfusion

Objective To observe the influence of ultra-shortwave (USW) irradiation on infarct volume and Ca2+-ATPase (SPCA) secretion after brain ischemia and reperfusion.Methods Eighty Sprague-Dawley rats were randomly divided into a sham operation group (n=8),a model group (n=36) and a USW group (n=36).The animal model of middle cerebral artery ischemia and reperfusion (MCAO/R) was established using the suture method in the rats of the model and USW groups,while the sham operation group was given the same operation but without inserting the thread plug.One day,3 days and 7 days after the intervention,12 rats were sacrificed and the infarct volumes and SPCA1 protein expression were measured using 2,3,5-triphenyltetrazolium chloride staining and western blotting.Results No white infarcted tissue was found in the sham operation group.In the model and USW groups the volume of infarcted tissue decreased with time.Significantly less infarcted volume was observed in the USW group compared to the model group at each time point.The SPCA1 levels in the brain tissue were lower than in the sham operation group after one and 3 days of USW treatment,but they were significantly lower in the model group as well.As time went by,the average SPCA1 level increased significantly in the model and USW groups.A slightly higher SPCA1 level was observed in the USW group compared to the model group after one day of treatment,but with no significance.However,significant differences were found between them after 3 and 7 days of intervention.Conclusion Ultra-shortwave irradiation can protect against MCAO/R injury by decreasing the infarcted volume,which may be related to down-regulation of SPCA1,minimizing nerve cell apoptosis and promoting neural functional recovery,at least in rats.
8.Detection of food-borne rotavirus by molecular motor biosensor.

To develop a specific, rapid and convenient method based on molecular motor biosensor to detect food-borne rotavirus. A specific probe was encompassed the conservative region of rotavirus's VP7 segment, and a molecular motor detect device was constructed by connecting probes to F0F1-ATPase molecular motor through biotin-streptavidin system. This biosensor's sensitivity was 0.005 ng/mL for rotavirus RNA. Extracted virus RNA was conjugated with the biosensor separately, at the same time ATP was synthesized. By comparing fluorescence intensity, we can detect rotavirus RNA in samples. This method possessed specificity for rotavirus, without any cross-reaction with Hepatitis A virus and noroviris, and it could be accomplished within 1 h. We detected 15 samples using this method and the results were compared with RT-PCR results. This method is sensitive and specific for rotavirus, and it can be used to detect food-borne rotavirus.

9.Value of diffusion weighted imaging in diagnosis and treatment of gastric cancer.

With the capability of measuring the water molecular diffusion qualitatively and quantitatively, diffusion weighted imaging (DWI) is currently the optimal and the only magnetic resonance method to observe water molecular diffusion motion in living tissues. Along with the advances in magnetic resonance technology, particularly the development of fast imaging sequences, DWI has been widely used in the imaging of liver, pancreas, kidneys and other solid organs. In recent years, DWI has been applied in the stomach. This article summarizes recent research on the application of DWI for the diagnosis and treatment of gastric cancer.

10.The gene characterization of VP1 and 2A of Enterovirus type 71

Objective To detect VP1 and 2A genes of Enterovirus type 71 (EV71) isolated from clinical specimens of patients with light or heavy symptoms and analyze the homogeneity and phylogenetic tree. Methods Fifty clinical specimens of children with hand-foot-and-mouth disease ( HFMD) were dealed with, which were tested by RT-PCR assay with specific primer pairs for EV71. EV71 isolates from patients with light or heavy clinical symptoms were tested by RT-PCR assay with two specific primer pairs for VP1 and 2A genes of EV71 respectively. All of the PCR products were sequenced and compared with that of previously isolated EV71 isolates available from GenBank by homogeneity and phylogenetic tree analyses. Results The RT-PCR results indicated that 30 isolates were EV71, 13 of 30 isolates were from clinical specimens of patients with light symptoms of hand-foot and mouth, the other were from clinical specimens of patients with heavy symptoms of complications. VP1 genes and 2A genes of 10 EV71 isolated strains including 5 light strains and 5 heavy strains were sequenced and compared with that of previously isolated 5 EV71 Chinese isolates available from GenBank (fuyangEU703814.1, xi_anHM003207. 1, shandongEU753418.1, shenzhenFJ607337.1, henanGU366191. 1) by homogeneity and phylogenetic tree analyses. The homogeneity of VP1 and 2A genes of the 10 EV71 isolated strains and 5 previously isolated strains were between 94.7% -99.4% and 93.6% -99.3% respectively, with the representative isolates of A and B genotypes was between 81.0%-84. 6% and 78. 4%-82. 2% respectively. The data suggested that all of the 10 Chinese isolates belong to EV71 genotype C. There were only 87.8% -90.2% homology among these 10 strains and the representative strains of C1, C2, C3 sub-genotypes of EV71 but 96. 8% -99.6% homology among these 10 strains and the representative strains of C4 sub-genotypes of EV71, this suggested that these 10 Chinese isolates composed the C4 sub-genotype, of the C genotype, that formed a single branch in the phylogenetic tree. Conclusion EV71 of sub-genotype C4 distributed in Mainland China, and VP1 genes have close genetic relationship between isolated strains. There is no obvious difference in 2A genes between clinical specimens of patients with light or heavy symptoms by homogeneity and phylogenetic tree analyses.
