1.Genome-wide Characterization and Prokaryotic Expression of UGT Gene Family in Dipsacus asper Wall.ex Henry

Objective To explore the biosynthesis of Dipsacus asper Wall.ex Henry triterpenoid saponin,and the UGT gene in Dipsacus asper Wall.ex Henry has been analyzed by the identification of whole genome,genome and prokaryotic expression.Methods The laboratory self-tested sequenced protein sequence files of the Dipsacus asper Wall.ex Henry genome were used.To validate the conserved domains of the sequence of the Dipsacus asper Wall.ex Henry UGT gene,BLASTP and hmmsearch were utilized.Prot-Param,SOMPA,MAGA7.0,Tbtools and other tools were used to investigate the protein physicochemical properties,protein structure,and covariance analysis of the Dipsacus asper Wall.ex Henry UGT gene family,and using the joint analysis of transcriptomic data and metabolomics data,two glycosyltransferases that might be related to triterpene saponin biosynthesis were screened,and expression vectors were constructed for prokaryotic expression.Results 44 Dipsacus asper Wall.ex Henry UGT genes were identified from the Dipsacus asper Wall.ex Henry genome.The length of Dipsacus asper Wall.ex Henry UGT proteins ranged from 49 to 1083 amino acids,with an average molecular weight of 24.86 kDa and an isoelectric point of 4.31-8.59.Dipsacus asper Wall.ex Henry UGT gene family was distributed on eight chromosomes.The phylogenetic tree constructed from the sequences of Dipsacus asper Wall.ex Henry,Arabidopsis thaliana and identified UGTs showed that glycosyltransferase gene families in Dipsacus asper Wall.ex Henry were mainly in the UGT73,UGT81,UGT85,and UGT80 families.Cis-acting element analysis showed that light-responsive elements were the most prevalent elements in the promoter regions of UGT gene family members.Two glycosyltransferases potentially related to triterpene saponin biosynthesis were screened using combined transcriptomics and metabolomics analysis,and were successfully expressed in prokaryotic form.Conclusion In this study,two candidate genes related to the biosynthesis of Dipsacus asper Wall.ex Henry triterpenoid saponins were jointly screened for prokaryotic expression using multi-omics,and were subjected to prokaryotic expression for further validation of the function of the genes.
2.A qualitative study on the awareness and management strategy improvement of childhood hypertension

Objective:
To summarize strategies for improving childhood hypertension, so as to provide evidencebased recommendations for poliymaking and practice childhood hypertension management in China.
Methods:
From March to April 2024, child health stakeholders from five districts in Hangzhou were selected using a combination of stratified and convenience sampling methods. Data were analyzed using a groundedtheory approach. During the indepth interview phase, six policymakers were interviewed. Focus group discussions were conducted with school administrators, healthcare providers, and parents, comprising a total of 62 participants.
Results:
Through threelevel coding, 116 initial categories were identified(e.g., "trend of younger age" "difficulty in behavior change"), 35 main categories (e.g., "higher incidence compared to the past" "caused by comprehensive influencing factors"), and 12 core categories (e.g., "epidemic status" "influencing factors"). Finally, the cognitive status, problem analysis, and management strategies of children hypertension were constructed.
Conclusion
Effective prevention and control of childhood hypertension requires coordinated efforts among governments, schools, families, and society to establish a comprehensive management system, with dynamic monitoring and evaluation to optimize policy implementation.
3.Role of senescent cells in bone-related diseases:An updated review

Bone-related diseases are a group of inflammatory diseases characterized by abnormal bone resorption and formation,with a wide variety of diseases involving age,inflammation,obesity,genetics,radiation,and other pathogenic factors.In recent years,se-nescent cells have become an important mechanism for age-related diseases,and their role in bone-related diseases has attracted the at-tention of researchers.Senescent cells are gradually being used as therapeutic targets for bone-related diseases.In this paper,we pres-ent a review of the mechanism of senescent cells in various types of bone-related diseases,which will provide new strategies for the pre-vention and treatment of bone diseases.

6.Application Regularity and Mechanism of Chinese Marine Drug Pair Haliotidis Concha-Oystreae Concha Based on Literature Mining Technology

ObjectiveTo analyze the clinical application hotspots, development trends, compatibility characteristics, application rules, and formulation mechanisms of the Chinese marine drug pair Haliotidis Concha-Oystreae Concha in order to provide references for its clinical medication and further research. MethodBy means of various modern literature databases such as China National Knowledge Infrastructure (CNKI), modern clinical prescriptions containing Haliotidis Concha-Oystreae Concha, as well as the clinical application hotspots, were retrieved, followed by visualized analysis of hotspots and development trends of their clinical applications using Citespace. The drug composition, efficacy and indications, and drug dosages in the prescriptions were statistically analyzed. Additionally, various statistical software including SPSS Modeler 18.0 were employed to analyze the indications, syndromes, and formulation rules of Haliotidis Concha-Oystreae Concha. ResultThe visualized analysis included 90 articles, revealing a gradual decrease in publications in this field in recent years. Key clinical application keywords were identified as hypertension, collateral deficiency producing wind, insomnia, etc. Eighty clinical prescriptions were retrieved, involving 121 drugs. Frequency analysis of compatibility demonstrated that the top 10 drugs were Uncariae Ramulus cum Uncis, Gastrodiae Rhizoma, Os Draconis, Achyranthis Bidentatae Radix, Paeoniae Radix Alba, Chrysanthemi Flos, Scutellariae Radix, Gardeniae Frucuts, Glycyrrhizae Radix et Rhizoma, and Polygoni Multiflori Caulis. Association rule analysis showed that core combinations included "Uncariae Ramulus cum Uncis-Achyranthis Bidentatae Radix" and "Os Draconis-Pheretima-Chuanxiong Rhizoma". Through factor reliability analysis, new drug combinations were derived, such as "Gastrodiae Rhizoma-Polygoni Multiflori Caulis-Eucommiae Cortex-Taxilli Herba-Leonuri Herba", "Achyranthis Bidentatae Radix-Uncariae Ramulus cum Uncis", "Scutellariae Radix-Glycyrrhizae Radix et Rhizoma-Margarita-Prunellae Spica", "Os Draconis-Pheretima-Bombyx Batryticatus", "Chrysanthemi Flos-Chuanxiong Rhizoma", "Poria-Acori Tatarinowii Rhizoma", and "Paeoniae Radix Alba-Gardeniae Fructus-Sclerotium Poriae Pararadicis". The Haliotidis Concha-Oystreae Concha drug pair was mainly used to treat diseases with liver Yang hyperactivity syndrome, with hypertension accounting for 40.00%, migraines for 30.00%, and dizziness for 15.00%. In the treatment of liver Yang hyperactivity syndrome, the main categories of compatible drugs were liver-pacifying and wind-extinguishing ones (19.86%), blood-activating and stasis-resolving ones (12.13%), and spirit-calming ones (10.08%). High-frequency drugs in the prescriptions function to reduce blood pressure through multiple pathways, such as increasing nitric oxide (NO) levels, downregulating angiotensin Ⅱ (Ang Ⅱ), and inhibiting angiotensin-converting enzyme (ACE). ConclusionThrough comprehensive analysis of the results, the Haliotidis Concha-Oystreae Concha drug pair is commonly used for hypertension with liver Yang hyperactivity syndrome, and is often combined with deficiency-tonifying, liver-pacifying and wind-extinguishing, heat-clearing, and spirit-calming drugs, aiming to simultaneously extinguish wind, relieve spasms, and pacify the liver to subdue Yang, while also clearing heat to relax bowels, stabilizing the mind, and enhancing the liver-pacifying and Yang-subduing effects of this drug pair.
7.Predictive value of ALBI score and PALBI score in assessing the risk of liver insufficiency or liver failure after hepatectomy

Objective:To explore the predictive value of preoperative albumin-bilirubin (ALBI) score and platelet-albumin-bilirubin (PALBI) score in evaluating the risk of post-hepatectomy liver dysfunction (PLD) or post-hepatectomy liver failure (PHLF) of primary hepatocellular carcinoma (HCC).Methods:The clinical data of 106 primary HCC patients who received partial liver resection in the First Hospital of Shanxi Medical University from April 2018 to January 2023 were retrospectively analyzed. According to the presence of PLD or PHLF complication or not based on 50-50 criteria, all recipients were divided into PLD or PHLF group and non-PLD or non-PHLF group. The differences of preoperative serological detection indexes were compared between the two groups. Multivariate logistic regression was used to analyze the influencing factors of PLD or PHLF in primary HCC patients. The 50-50 criteria was taken as the gold standard, and the receiver operating characteristic (ROC) curve was used to assess the efficacy of ALBI score and PALBI score in judging of PLD or PHLF of primary HCC patients.Results:Among the 106 HCC patients, 13 cases (12.26%) of them suffered PLD or PHLF, while 93 cases (87.74%) did not. The age of patients in PLD or PHLF group and non-PLD or non-PHLF group was (60±7) years, (58±10) years, respectively; and there were 10 cases (76.92%) males, 69 cases (74.19%) males, respectively in both groups. There were statistically significant differences in aspartate aminotransferase, γ glutamyl transpeptidase, total bilirubin, albumin, tumor diameter, Child-Pugh score, ALBI score and PALBI score before operation between the two groups (all P < 0.05). The results of multivariate regression analysis showed that Child-Pugh score ( HR = 2.250, 95% CI 1.097-4.615, P = 0.027), ALBI score ( HR = 10.374, 95% CI 2.767-38.890, P = 0.001) and PALBI score ( HR = 33.074, 95% CI 4.677-233.894, P < 0.001) were independent influencing factors of PLD or PHLF in primary HCC patients ( P < 0.05). ROC curve result showed that the area under the curve of Child-Pugh score for predicting PLD or PHLF in HCC patients was 0.647 (95% CI 0.548-0.738), the area under the curve of ALBI score was 0.791 (95% CI 0.701-0.864), and the area under the curve of PALBI score was 0.794 (95% CI 0.704-0.866). The optimal cut-off value of Child-Pugh score for predicting PLD or PHLF in primary HCC patients was 6.500, the sensitivity was 38.5%, and the specificity was 92.5%. The optimal cut-off value of ALBI score for predicting PLD or PHLF in primary HCC patients was -2.345, the sensitivity was 76.9%, and the specificity was 77.4%. The optimal cut-off value of PALBI score for predicting PLD or PHLF in primary HCC patients was -2.050, the sensitivity was 69.2%, and the specificity was 92.5%. Conclusions:Both PALBI score and ALBI score is better than Child-Pugh score in the predictive efficacy of PLD or PHLF for primary HCC patients after hepatectomy.
8.Pharmacological Effect of Glycyrrizae Radix et Rhizoma Compatibility and Its Mechanism

Glycyrrizae Radix et Rhizoma has high medicinal value and is widely used in compatibility. It is used most frequently in the compatibility of Chinese medicine prescriptions,and is known as ''Guolao''(national medicine) and "master of all medicines". The characteristic active ingredients are mainly liquitin,glycyrrizic acid,glycyrrizin,and licochalcone. In different compatibilities,based on traditional and modern pharmacological theories,the corresponding effect of Glycyrrizae Radix et Rhizoma are brought into play through different mechanisms. Based on the traditional pharmacology of Glycyrrizae Radix et Rhizoma for tonifying spleen,replenishing Qi,clearing heat,removing toxin,dispelling phlegm,relieving cough and pain,and harmonizing various medicines,this paper used herbal authentication to analyze its compatibility application and mechanism. It was found that Glycyrrizae Radix et Rhizoma played corresponding effect in compatibilities through "tonification","harmonization",and "regulation". For example,Glycyrrizae Radix et Rhizoma was combined with tonics including Ginseng Radix et Rhizoma and Atractylodis Macrocephalae Rhizoma to tonify the five Zang-organs through its strong tonifying effect,combined with Paeoniae Radix Alba and Aconiti Lateralis Radix Praeparata to relieve emergencies and pains through harmonizing medicine power and properties,and combined with Rhei Radix et Rhizoma and Natrii Sulfas to reduce medicine intensity through regulating medicine properties and body characteristics. The application law and mechanism of the modern pharmacological compatibility of Glycyrrizae Radix et Rhizoma were analyzed by data mining and network pharmacology. It was found that the modern clinical formula was often compatible with Glycyrrizae Radix et Rhizoma for anti-inflammation,cardiovascular and cerebrovascular protection,anti-virus,and anti-tumor,Ephedrae Herba and Scutellariae Radix for anti-inflammation,Bambusae Caulis in Taenias,Salviae Miltiorrhizae Radix et Rhizoma,and Aurantii Fructus Immaturus for cardiovascular and cerebrovascular protection,and Ophiopogonis Radix and Chuanxiong Rhizoma for nerves protection. Meanwhile,the important targets of the characteristic ingredients were protein kinase B1 (Akt1),interleukin-6 (IL-6),tumor necrosis factor (TNF),and epidermal growth factor receptor (EGFR). The important characteristic pathways such as tyrosine kinase inhibitor resistance pathway and cyclic guanosine monophosphate (cGMP)/protein kinase G (PKG) signal pathway played the role of cardiovascular and cerebrovascular protection,and proteoglycan pathway in cancer played a neuroprotective role. This study is expected to provide references for the rational compatibility and application of Glycyrrizae Radix et Rhizoma,as well as the compatibility application of Chinese medicine prescriptions.
9.Decreased vitamin D-binding protein level portends poor outcome in acute-on-chronic liver failure caused by hepatitis B virus

Background/Aims:
Acute-on-chronic liver failure (ACLF) is a catastrophic illness. Few studies investigated the prognostic value of vitamin D-binding protein (VDBP) for hepatitis B virus (HBV)-related ACLF (HBV-ACLF) resulted in conflicting results.
Methods:
Two prospective HBV-ACLF cohorts (n=287 and n=119) were enrolled to assess and validate the prognostic performance of VDBP.
Results:
VDBP levels in the non-survivors were significantly lower than in the survivors (P<0.001). Multivariate Cox regression demonstrated that VDBP was an independent prognostic factor for HBV-ACLF. The VDBP level at admission gradually decreased as the number of failed organs increased (P<0.001), and it was closely related to coagulation failure. The areas under the receiver operating characteristic curve (AUCs) of the Child-Pugh-VDBP and chronic liver failuresequential organ failure assessment (CLIF–SOFA)-VDBP scores were significantly higher than those of Child-Pugh (P<0.001) and CLIF-SOFA (P=0.0013). The AUCs of model for end-stage liver disease (MELD)-VDBP were significantly higher than those of MELD (P= 0.0384) only in the case of cirrhotic HBV-ACLF patients. Similar results were validated using an external multicenter HBV-ACLF cohort. By longitudinal observation, the VDBP levels gradually increased in survivors (P=0.026) and gradually decreased in non-survivors (P<0.001). Additionally, the VDBP levels were found to be significantly decreased in the deterioration group (P=0.012) and tended to be decreased in the fluctuation group (P=0.055). In contrast, they showed a significant increase in the improvement group (P=0.036).
Conclusions
The VDBP was a promising prognostic biomarker for HBV-ACLF. Sequential measurement of circulating VDBP shows value for the monitoring of ACLF progression.
10. Analysis on internal mechanism of zedoary turmeric in treatment of liver cancer based on pharmacodynamic substances and pharmacodynamic groups

Zedoary tumeric (Curcumae Rhizoma, Ezhu in Chinese) has a long history of application and has great potential in the treatment of liver cancer. The antiliver cancer effect of zedoary tumeric depends on the combined action of multiple pharmacodynamic substances. In order to clarify the specific mechanism of zedoary tumeric against liver cancer, this paper first analyzes the mechanism of its single pharmacodynamic substance against liver cancer, and then verifies the joint anti liver cancer mechanism of its “pharmacodynamic group”. By searching the research on the antihepatoma effect of active components of zedoary tumeric in recent years, we found that pharmacodynamic substances, including curcumol, zedoarondiol, curcumenol, curzerenone, curdione, curcumin, germacrone, β-elemene, can act on multi-target and multi-channel to play an antihepatoma role. For example, curcumin can regulate miR, GLO1, CD133, VEGF, YAP, LIN28B, GPR81, HCAR-1, P53 and PI3K/Akt/mTOR, HSP70/TLR4 and NF-κB. Wnt/TGF/EMT, Nrf2/Keap1, JAK/STAT and other pathways play an antihepatoma role. Network pharmacological analysis showed that the core targets of the “pharmacodynamic group” for anti-life cancer are AKT1, EGFR, MAPK8, etc, and the core pathways are neuroactive live receiver interaction, nitrogen metabolism, HIF-1 signaling pathway, etc. At the same time, by comparing and analyzing the relationship between the specific mechanisms of pharmacodynamic substance and “pharmacodynamic group”, it is found that they have great reference significance in target, pathway, biological function, determination of core pharmacodynamic components, formation of core target protein interaction, in-depth research of single pharmacodynamic substance, increasing curative effect and so on. By analyzing the internal mechanism of zedoary tumeric pharmacodynamic substance and “pharmacodynamic group” in the treatment of liver cancer, this paper intends to provide some ideas and references for the deeper pharmacological research of zedoary tumeric and the relationship between pharmacodynamic substance and “pharmacodynamic group”.
