1.Research on the resistance of hepatitis B virus to core protein allosteric modulators

Hepatitis B virus (HBV) infection represents a significant global health challenge. Current therapeutic options, such as interferon and nucleoside analogues (NAs), are limited by issues including long-term medication toxicity, the virus's propensity to develop drug resistance, and the inability to eradicate covalently closed circular DNA (cccDNA). Core protein allosteric modulators (CpAMs), as an emerging class of anti-HBV drugs, target HBcAg to interfere with capsid assembly. This not only blocks viral nucleocapsid formation and inhibits viral replication but also indirectly destabilizes the cccDNA pool, while exhibiting a relatively high genetic barrier to resistance. This article systematically evaluates HBV drug resistance to CpAMs and proposes anti-resistance strategies, including the combination of nucleoside analogues with CpAMs, enhancing protein-drug interactions, targeting HBcAg degradation, designing multi-target inhibitors, and employing combination therapy. Looking ahead, the integration of structural biology, computational chemistry, and immunotherapy will offer innovative approaches for developing novel, highly effective, and low-resistance inhibitors, thereby advancing the functional cure of hepatitis B.
2.Feasibility study of the post process of CTA on head and neck based on AI deep learning on stenosis assessment

Objective:To explore the feasibility of the post process of computed tomography angiography(CTA)on head and neck based on artificial intelligence(AI)deep learning on stenosis assessment.Methods:A total of 108 patients who underwent CTA on head and neck at Shunyi Hospital of Beijing Traditional Chinese Medicine Hospital from January 2022 to June 2023 were selected,and they were divided into an AI group(54 cases)and a manual group(54 cases)based on different diagnostic methods.Dual source CT was used to conduct examination,and data was transmitted to"Shenrui Dr.Wise Head and Neck CTA Intelligent Assistance System"and"Siemens Syngo View Post Process Workstation".The image evaluation was conducted on a 5-point scale.The time-consuming of post process and diagnosis of CTA images of the artery of head and neck between two groups were observed and compared,and the quality of CTA images of the artery of head and neck,and the diagnostic results of the degree of the artery stenosis of head and neck also were observed and compared between two groups.Results:The differences of the time-consuming of post process of images and the time-consuming of diagnosis between two groups were all statistically significant(t=52.315,24.509,P<0.05),respectively.The intra-group correlation coefficient(ICC)of each branch of the artery of head and neck was 0.996 between two physicians.In AI group,the score of common carotid artery was(4.77±0.12),and the score of internal carotid artery was(4.56±0.13),and the score of vertebral artery was(4.55±0.16),and the score of middle cerebral artery was(4.78±0.16).In manual group,the score of common carotid artery was(3.02±0.12),and the score of internal carotid artery was(3.02±0.12),and the score of vertebral artery was(3.02±1.14),and the score of middle cerebral artery was(3.11±1.09).The differences of the image scores of each main branch of the artery of head and neck between two groups were significant(t=107.165,94.590,13.812,15.753,P<0.05),respectively,and scores of AI group all higher than these of manual group.The result of digital subtraction angiography(DSA)was used as the"gold standard".A total of 44 stenosis arteries were included for comparison,and the DSA results indicated that there were 12 cases with mild stenosis,12 cases with moderate stenosis and 10 cases with severe stenosis in AI group,and there were 12 cases with mild stenosis,8 cases with moderate stenosis and 7 cases with severe stenosis in manual group.There was a favorable diagnostic consistency in AI group(Kappa=0.898,P<0.05).The compliance rate of each stenosis of AI group was higher than that of manual group.The sensitivity(96.80%)and specificity(66.52%)of AI group were all higher than those(68.36 and 14.75%)of manual group,and the difference of specificity was significant.Conclusion:The time-consuming of post process,the time-consuming of diagnosis and the quality of CTA image of the artery of head and neck in AI mode are better than those in manual mode,and the consistency of diagnostic efficacy between two modes is favorable.
3.Epidemiological features and trends of endometrial cancer: 10 081 cases analysis of 62 hospitals in Guangdong province

Objective To explore the epidemiology characteristics of endometrial cancer (EC).Methods We retrospectively analyzed 10081 patients diagnosed with EC from 62 hospitals between 2000 and 2010 in Guangdong province.Results The mean age at diagnosis was 52.8 ± 9.3.The proportion were 19.3%,64.2%,16.6% in patients with ages ≤45,> 45-60,> 60 respectively.From 2000 to 2010,the mean ages at each year were no statistic significance.The number of cases of EC were positively correlated with years(r =0.964,P < 0.001).The number of cases of patients with ≤ 30 years old (r =0.857,P =0.001),≤35 years old (r =0.866,P =0.001),≤40 years old (r =0.952,P < 0.001),≤ 45 years old (r =0.952,P <0.001) were positively correlated with years.The ratios of patients with ≤30 years old (x2 =10.390,P =0.407),≤35 years old (x2 =11.651,P =0.309),≤ 40 years old (x2 =17.329,P =0.067),≤45 years old (x2 =5.154,P =0.881) during these eleven years were no statistic significance.The ratios of type Ⅰ EC at 2000-2010 were no statistic significance.Conclusions EC often present in patients aged from > 45-60 years old.The case number of EC showed an increasing trend.However,the proportion of young patients was stable.The endometroid adenocarcinoma was the main histological type of EC.
4.A Comparative Study on Quality of Different Populations of Wild Herbs ofPsammosilene tunicoides

This article was aimed to carry a comparative study on the quality of 16 populations of wild herbs of rare and endangered plant ofPsammosilene tunicoidesin order to provide a basis for its domestication, GAP and selective breeding. Methods for determination of moisture content, total ash and extract content were taken in this comparative study. The results showed that different populations had different moisture content, total ash and extract content. The moisture content and the total ash ofXiang-Shan population were the lowest; and the extract content was the highest. The total ash ofLiu-De population was the highest; and the extract content was the lowest. The moisture content ofShun-Zhou population was the highest. It was concluded that different populations had different quality among 16 populations of wild herbs. Therefore, there was a probability to choose the best population.
5.Incidence of pocket hematoma after electrophysiological device placement:dual antiplatelet therapy versus low-molecular-weight heparin regimen

Background Given the increasing number of patients who require dual antiplatelet (DAP) therapy and electrophysiological device (EPD) placement, perioperative antiplatelet management is a current challenge. In this study, we investigated the incidence of pocket hema-toma formation after EPD placement in patients undergoing DAP therapy or an alternative low-molecular-weight heparin (LMWH) regimen. Methods This clinical observational study was performed from July 2010 to July 2012. In total, 171 patients were enrolled in the analysis after meeting the inclusion criteria. These patients were divided into two groups: 86 patients were treated with DAP therapy at the time of device implantation, and the DAP therapy was discontinued for 5 to 7 days and replaced with enoxaparin before device implantation in the other 85 patients. Adenosine phosphate (ADP)-mediated platelet aggregation and arachidonic acid-induced platelet aggregation were tested preoperatively. We compared the incidence of pocket hematoma between the two groups and the association of pocket hematoma develop-ment with ADP-mediated platelet aggregation and arachidonic acid-induced platelet aggregation.Results The incidence of pocket hema-toma in the patients who continued DAP was lower than that in the patients who replaced the dual antiplatelet regimen with LMWH (3.49%vs. 16.47%, respectively;X2 = 6.66,P < 0.01). Among the patients who continued DAP therapies, the rate of ADP-mediated platelet aggre-gation inhibition in patients with pocket hematomas was higher than that in patients without pocket hematomas. None of the patients under-going DAP or enoxaparin therapy developed pocket infection, thromboembolic events, or other serious complications. Multiple logistic re-gression analysis revealed that LMWH therapy was an independent risk factor for the development of pocket hematoma (RR = 0.054, 95%CI = 0.012-0.251). Furthermore, patients undergoing LMWH therapy were 5.1-fold more likely to develop pocket hematomas than were DAP-treated individuals.Conclusion Continuance of DAP therapy does not increase the risk of pocket hematoma formation after EPD placement.
