Objective:To evaluate the performance of our self-developed deep learning (DL) model which was designed to automatically classify the knee X-ray images into one non-surgical category and 4 surgical categories, including non-surgical knees (NSK), high tibial osteotomy (HTO), total knee arthroplasty (TKA), unicompartmental knee arthroplasty (UKA), and tibial plateau fracture fixation with an internal fixation plate (TPFF).Methods:The knee X-ray images were collected of the patients who had undergone knee joint surgery at Department of Orthopaedics, Fuzhou Second General Hospital from January 2017 to December 2022. On the Baidu EasyDL AI platform, a multi-class object recognition DL model was built using the You Only Look Once (YOLO) algorithm. The model was trained on a dataset of 1,281 knee anteroposterior X-ray images (including NSK, HTO, TKA, UKA, and TPFF) to generate a DL model which was able to automatically recognize and classify the knee X-ray images. The reliability of the model classification performance was evaluated by analyzing the 5 indicators [accuracy, sensitivity, specificity, positive predictive value (PPV), and negative predictive value (NPV)] using a test set of 450 knee anteroposterior X-ray images with the above 5 categories. The receiver operating characteristic curve was plotted and the area under the curve (AUC) was calculated to further quantify the classification performance of the model. Results:In the test set, on the whole, the model achieved an accuracy of 97.0%, a sensitivity of 92.4%, a specificity of 98.1%, a PPV of 92.4%, and a NPV of 98.1%, an AUC of 0.947, indicating a high reliability in classifying various categories. The model showed a best performance for TKA, with the 5 indicators being 99.1%, 99.0%, 99.1%, 97.1% and 99.7%. The model showed a slightly lower sensitivity for TPFF and HTO (87.0% and 86.0%, respectively). Conclusion:A successful DL model has been developed which can automatically classify the knee X-ray images into non-surgical and surgical categories due to its satisfactory performance, particularly in accuracy and AUC.

Objective Though HCV genotyping , liver ultrasound and liver function indicators were used to assess the relationship between HCV genotyping, viral RNA copy number and liver damage related indicators in patients with chronic hepatitis C. Methods A total of 105 Uyghur hepatitis C patients in Aksu, Xinjiang were recruited in our hospital. All patients were carried out a test of HCV RNA copy number. HCV genotyping was performed by fluorescence quantitative PCR method. Fully automatic biochemical analyzer was used for alanine aminotransferase (ALT), aspartate aminotransferase (AST), direct bilirubin (DIBL) and total bilirubin (TIBL) by commercial kits. Color doppler ultrasound system was used for liver ultrasound. Results The genotyping results of 105 Uyghur hepatitis C patients showed that type 1b, 2a, 3a, 3b and 6a were 46, 41, 8, 8 and 2, respectively. Type 1b was the main type of HCV virus. The proportion of RNA copy number, AST and ALT levels, and liver cirrhosis were higher in patients with type 1b. Statistical analysis showed that there was a significant correlation between different HCV virus types and HCV RNA copy number (P = 0.032). In terms of AST and ALT levels, there were significant differences between type 1b, type 2a and other genotypes (type 3b and type 3a and type 6a) (P < 0.01). In addition, patients with normal liver, enlarged liver spots, fatty liver and cirrhosis have significant differences among type 1b, type 2a and other genotypes (P < 0.01). Conclusion There are regional differences in HCV genotyping among Uygur people in Aksu, Xinjiang. HCV RNA copy number and degree of liver damage are correlated with different HCV genotypes, which is of great significance to guide the clinical diagnosis and treatment of HCV in local populations.

BACKGROUNDAllergic asthma is a chronic airway inflammatory disease partly characterised by high concentration of T help 2 (Th2) cytokines in bronchoalveolar lavage fluid (BALF). There is no report on the relation of peripherally circulating blood lymphocytes and asthma. We explored the balance of Th2/Th1 cytokines in asthmatic mice. Exogenous recombinant interleukin (IL) 33 acted on murine peripheral circulating blood lymphocytes, IL-5 cytokine was selected for assessing Th2 cytokines and interferon-gamma (IFN-γ) for Th1 cytokines.
METHODSFemale specific pathogen free BABL/c mice were sensitised by intraperitoneal injection of 20 µg of ovalbumin emulsified in 1 mg of aluminium hydroxide gel in a total volume of 200 µl, and challenged for 30 minutes in 7 consecutive days with an aerosol of 2 g ovalbumin in 100 ml of PBS. Then we collected BALF and isolated lymphocytes from the peripheral blood. The lymphocytes were divided into two groups: asthmatic group and normal group. Th1/Th2 cytokines was detected by enzyme-linked immunosorbent assay (ELISA) kits.
RESULTSIn the asthma group, we found numerous eosinophils and lymphocytes on the glass slides. We then confirmed that the optimal concentration of IL-33 was 10 ng/ml and time of IL-33 stimulating lymphocytes was 24 hours. In the asthma group, the production of IL-5 was significantly increased over normal group after stimulation with IL-33 (P < 0.05) and the production of IFNγ was supressed from IL-33 stimulated lymphocytes (P < 0.05).
CONCLUSIONIL-33 acts on lymphocytes of peripheral blood increasing secretion of Th2 cytokines and inhibiting secretion of Th1 cytokines.
