1.Zuogui Wan Improve Ovarian Inflammatory Microenvironment and Stemness of Ovarian Germline Stem Cells in Ovarian Aging via cGAS/STING Signaling Pathway

ObjectiveTo investigate the mechanism of Zuogui Wan (ZGW) in improving ovarian inflammatory microenvironment and stemness of ovarian germline stem cells (OSCs) for treating ovarian aging via the cyclic guanosine monophosphate/adenosine monophosphate synthase (cGAS)/stimulator of interferon genes (STING) signaling pathway. MethodsForty C57BL/6 female mice were randomly divided into a blank group, a model group, a low-dose ZGW group (2.7 g·kg-1), a high-dose ZGW group (5.4 g·kg-1), and an estradiol valerate group (0.15 mg·kg-1), with 8 mice in each group. Except the blank group, all other groups received a single intraperitoneal injection of cyclophosphamide at 120 mg·kg-1 to establish an ovarian aging mouse model. After successful modeling, each group was continuously administered for 4 weeks, once daily. The physiological status of the mice was observed, and the ovarian index was calculated. The estrus cycle of the mice was monitored. Hematoxylin-eosin (HE) staining was used to observe pathological changes in ovarian tissue. Enzyme-linked immunosorbent assay (ELISA) was used to detect serum sex hormone levels. Serum inflammatory factors interleukin-1β (IL-1β), tumor necrosis factor-α (TNF-α), and mouse interleukin-6 (IL-6) levels were detected using kits. Western blot was used to detect the protein expression of ovarian cGAS, STING, p-STING, TANK-binding kinase 1 (TBK1), p-TBK1, interferon-induced transmembrane protein 3 (Fragilis), and Vasa homolog protein (MVH). Quantitative real-time polymerase chain reaction (Real-time PCR) was used to detect the mRNA expression of inflammatory factors in ovarian tissue. Immunofluorescence double labeling was performed to locate OSCs in ovarian tissues, and fluorescence intensities of OSCs markers MVH and octamer binding transcription factor 4 (Oct4) were calculated. ResultsCompared with the blank group, the model group showed reduced body weight, ovarian wet weight, and ovarian index (P<0.01) and a disordered estrus cycle (P<0.01). In addition, the levels of serum follicle-stimulating hormone (FSH), TNF-α, IL-6, and IL-1β were increased (P<0.01), while anti-Müllerian hormone (AMH) and estradiol (E2) levels were decreased (P<0.01). The protein expression of cGAS, p-STING/STING, and p-TBK1/TBK1 in ovarian tissue was increased (P<0.05, P<0.01), while that of OSCs stemness factors MVH and Fragilis was reduced (P<0.01). Immunofluorescence indicated a reduction in MVH and Oct4 expression in OSCs (P<0.01). The mRNA expression of inflammatory factors TNF-α, IL-6, and IL-1β in ovarian tissue was increased (P<0.05, P<0.01). Compared with the model group, the treatment groups exhibited improved body weight, ovarian wet weight, and ovarian index (P<0.05) and a reduced rate of estrus cycle disorder (P<0.05, P<0.01). The levels of serum FSH, TNF-α, IL-6, and IL-1β were decreased (P<0.05, P<0.01), while AMH and E2 levels were increased (P<0.01). The protein expression levels of cGAS, p-STING/STING, and p-TBK1/TBK1 in ovarian tissue were decreased (P<0.05), while the protein expression of MVH and Fragilis was increased (P<0.05), and the fluorescence intensities of MVH and Oct4 were increased (P<0.05, P<0.01). The mRNA expression of inflammatory factors in ovarian tissue was decreased (P<0.05). ConclusionZGW alleviate ovarian inflammatory response, regulate ovarian microenvironment homeostasis, and maintain stemness of OSCs in ovarian aging mice probably by modulating the cGAS-STING signaling pathway, thereby improving ovarian function and delaying ovarian aging.
2.Research on Chinese Medical Classics Compiled in Taoist Canon

[Objective]To determine the medical classics of Taoist Canon for laying a foundation for further research through arrangement and summary of the traditional Chinese medicine literature of Zhengtong Taoist Canon written in the Ming Dynasty.[Methods]Firstly,the literature review method was used to summarize and sort out the medical classics described by the predecessors.Then,combined with the interdisciplinary methods of traditional Chinese medicine,bibliography and Taoist medicine,the medical classics Zhengtong Taoist Canon were put forward,sorted and analyzed.[Results]A total of 152 medical classics of Zhengtong Taoist Canon were sorted out,and they were divided into four categories,including 20"basic theory of traditional Chinese medicine",8"Herbalism",14"Medical formula"and 110"Medical Taoist Classics".And the"Medical Taoist Classics"included 24"taking medicine books",52"inner danology books",10"coition medicine books"and 24"comprehensive books".[Conclusion]Taoist Canon contains many medical classics related to traditional Chinese medicine.These medical classics provide literature basis and theoretical support for scholars to deeply study the Taoist medical thoughts from the perspective of philology.
3.Meta-Analysis of the Effect of Resistance Exercise Prescription on Glucose and Lipid Metabolism in Overweight and Obese Adults

Objective To explore the effect of resistance exercise on glycolipid metabolism in over-weight or obese people,with the aim of providing reference for the clinical development of appropriate resist-ance exercise programs.Methods PubMed,Cochrane Library,EmBase,Web of Science,CNKI,Wanfang Data Knowledge Service Platform and VIP database were systematically searched to obtain the relevant litera-ture on the effects of resistance exercise on glucose and lipid metabolism from the establishment of the library to December 31,2023 in overweight and obese populations.The type of literature was limited to randomized controlled studies in which the exercise intervention included resistance exercise in the test group but did not include resistance exercise in the control group.The literature was screened by 2 researchers according to in-clusion and exclusion criteria and relevant data were extracted.Meta-analysis of included literature was per-formed using RevMan 5.3 software,and publication bias was assessed using funnel plots.Results A total of 17 papers and 700 study subjects were included,with 351 cases in the test group,and 349 cases in the control group.Meta-analysis results showed that compared with the control group,the glycosylated hemoglobin of the test group with resistance exercise[SMD(95%CI)=-0.30(-0.53--0.07),P=0.010],fasting blood glucose[SMD(95%CI)=-0.58(-0.90--0.26),P<0.001],insulin re-sistance index[SMD(95%CI)=-0.90(-1.42--0.38),P<0.001],and low-density lipoprotein cholesterol(LDL-C)[SMD(95%CI)=-0.28(-0.53--0.04),P=0.020]levels were reduced.Subgroup analysis showed that the glucose metabolism levels of the test group with only resistance exercise and a combination of resistance-anaerobic exercise were significantly lower than those of the control group(both P<0.05).The heterogeneity of the effect of resistance exercise on glucose metabolism in Asian popu-lations(I2=57%)was smaller than that in Caucasion(I2=76%)and other populations(I2=94%),and the level of decrease in glucose metabolism indexes was significant[SMD(95%CI)=-1.15(-1.73--0.57),P<0.001].Conclusion Resistance exercise can significantly reduce the level of glycolipid me-tabolism indexes in overweight or obese people.

5.The effects of maxillary protraction on soft and hard tissue in patients with cleft lip and palate in the mixed dentition period

Objective To study the three-dimensional changes of soft and hard tissue in male patients with unilateral non-syndromic cleft lip and palate in the mixed dentition period before and after maxillary protraction.Methods Twenty male patients with unilateral non-syndromic cleft lip and palate in the mixed dentition period treated by maxillary anterior traction in the Department of Orthodontics of Affiliated Stomatological Hospital of Nanjing Medical University were selected(average age(10.6±1.23)years old).Cone beam CT was taken before and after treatment.Dolphin 3D 11.95 software was used for three-dimensional measurement and analysis.SPSS 25.0 software package was used for statistical analysis.The self-controlled paired t test was used to compare the changes in soft and hard tis-sues of male patients with unilateral cleft lip and palate before and after treatment.The changes in the anterior displacement of the ANS point,the anterior displacement of point A,and the posterior displacement of point B were tested using the one-sample t test.Results The sagittal skeletal changes were significantly increased in ∠ SNA(P<0.01),∠ANB(P<0.01),Y axis(P<0.05),the forward displacement of ANS point(P<0.01)and A point(P<0.01)and the backward displacement of B point(P<0.01),but ∠SNB(P<0.05)was decreased significantly.The vertical skeletal changes showed that ∠MP-FH(P<0.01),∠MP-SN(P<0.05)and the dis-tance of ANS-Me(P<0.05)were increased significantly,but ∠SN-PP(P<0.01)was decreased significantly.The dental changes inclu-ding ∠U1-NA(P<0.01),the distance of U1-NA(P<0.01),∠U1-SN(P<0.01),overjet and the Wits were increased significantly,but ∠ L1-NB(P<0.01),the distance of L1-NB(P<0.01)and ∠L1-MP(P<0.01)were decreased significantly.The changes of soft tis-sue including ∠ S-Ns-Sn(P<0.01),∠ Sn-Ns-Bs(P<0.01),the distance of UL-EP(P<0.01)and LL-UL(P<0.01)were increased sig-nificantly.Conclusion After the treatment of maxillary protraction,the forward growth of maxilla will be possibly promoted on patients with cleft lip and palate in the peak of growth timing,as well as the intermaxillary relationship and soft tissue profile,but the side effects should be paid attention to.
6.Development of the Chinese Premature Ovarian Insufficiency Quality of Life Scale

Objective To develop a quality of life(QoL)scale for Premature Ovarian Insufficiency(POI)with high reliability and validity in accordance with China's national conditions.Methods The theoretical framework of the scale was constructed under the guidance of traditional Chinese medicine theory with reference to the development method of the QoL scale.Literature analysis method,medical record review method,patient interview method,Delphi expert consultation method and clinical investigation were used to establish,improve and screen the scale items to form the final scale,and the characteristics and differentiation in terms of reliability and validity and feasibility of the final scale were evaluated.Results After screening the entries by the Delphi method and clinical surveys,the Chinese Premature Ovarian Insufficiency Quality of Life Scale was formed,which included three domains of physiology,psychology,and society,and eleven aspects such as menstruation,fertility,and physical symptoms,with 30 entries and two overall evaluations.The Cronbach's coefficient of this scale was 0.910,the fold-half coefficient was 0.956,and the re-test correlation coefficient was 0.868,which showed good internal consistency,fold-half and re-test reliabilities;the scale extracted 9 factors with characteristic root>1,and the cumulative variance contribution rate was 71.063%,the comparative fit index was 0.959,and the goodness-of-fit index was 0.919,and the degree of correlation was tested has showed that the content validity,structural validity and calibration correlation validity were good;the difference in the total scores of the scale between patients with POI and the healthy population was statistically significant,and the scale had a good degree of differentiation.Conclusion As confirmed by this study,the Chinese Premature Ovarian Insufficiency Quality of Life Scale has good reliability,validity,differentiation and feasibility,and can be used as a tool for evaluating the quality of life of POI patients in China as well as enriching the efficacy evaluation system of TCM in treating POI,and it is suitable for clinical popularization and application.
7.Effect of monocular form deprivation during the pre-critical period on dendritic spine density and morphology in V1 neurons

Objective:To investigate the effect of monocular form deprivation (MD) during the pre-critical period of visual development on the density and morphology of dendritic spines in mouse primary visual cortex (V1) neurons.Methods:Twenty SPF male C57BL/6J mice with eyes opened on postnatal day 14 (P14) were selected and divided into MD and control groups using a random number table, with 10 mice in each group.The MD group was fed to P18 after 4 days of MD in the right eye, and the control group was raised to P18 under the same feeding conditions.All mice were decapitated after cardiac perfusion, and the sections were stained with the cell membrane fluorescent probe 1, 1′-dioctadecyl-3, 3′, 3′-tetramethylindocarbocyanine perchlorate, and imaged by laser scanning confocal microscopy to observe and compare the differences in density and morphology of dendritic spines in bilateral V1 neurons between the control group and the MD group.This study was approved by the Animal Ethics Committee of Tianjin Medical University (No.TMUaMEC2022004).Results:The total density of dendritic spines in the V1 area on the left side of the control group, the right side of the control group, the left side of the MD group, and the right side of the MD group were (7.57±0.25), (7.42±0.25), (6.54±0.18), and (7.51±0.29)spines/10 μm, respectively, with a statistically significant overall difference ( F=3.818, P<0.05).The total density of dendritic spines in the left V1 area of mice in the MD group was significantly lower than that in the left side of the control group and the right side of the MD group, and the differences were statistically significant (both P<0.05).There was a significant difference in the proportion of the four types of dendritic spines in V1 neurons on both sides between the two groups ( χ2=26.295, P=0.002).There was a significant difference in the proportion of the four types of dendritic spines between the left V1 of the MD group and the left and right V1 of the control group (both P<0.008 3).There was a significant difference in the filopodia-type dendritic spine density in bilateral V1 neurons between the two groups ( F=3.253, P<0.05).Compared with the left V1 area of the control group, the density of filopodia-type dendritic spines in the left V1 area of the MD group decreased significantly, with a statistical significance ( P<0.05).There was no significant difference in the density of thin-type, mushroom-type, and stubby-type dendritic spines in bilateral V1 area neurons between the two groups ( F=1.760, 2.618, 1.749; all P>0.05). Conclusions:MD during the pre-critical period of visual development can cause a decrease in the total density of dendritic spines and significant changes in the compositional proportions in the V1 contralateral to the deprived eye, and is mainly manifested by a decrease in the number of filopodia, suggesting that abnormal visual experience can cause plastic changes in the number and structure of synapses in the visual cortex during the pre-critical period of visual development.

Energy metabolism disorders are a critical factor contributing to cognitive dysfunction.Astrocytes,as key suppliers of energy substrates to neurons,help to sustain normal cognitive function via their extensive energy metabolism activities in the brain,coordinated by various energy supply mechanisms.Imbalances in astrocyte-neuron energy metabolism coupling lead to abnormal neuronal activity,thereby driving the pathogenesis and progression of Alzheimer's disease(AD).It is therefore essential to investigate the pathological mechanisms and therapeutic targets associated with AD from the perspective of energy metabolism to support the future development of AD treatments and interventions.Traditional Chinese medicine(TCM),characterized by multi-component and multi-target effects,has shown potential efficacy for regulating cerebral energy metabolism,suggesting that modulating brain energy metabolism may represent a significant pathway by which TCM might ameliorate AD.This review systematically elucidates the mechanistic role of the dysregulation of astrocyte-neuron bioenergetic coupling in the pathogenesis and progression of AD,and critically evaluates recent advancements in therapeutic interventions mediated by TCM.

10.Research advances in treatment of cerebral ischemia-reperfusion injury by traditional Chinese medicine via modulating NLRP3 inflammasome-related signaling pathways

Cerebral ischemia-reperfusion injury(CIRI)is a pathological condition characterized by the aggra-vation of the secondary injury in the brain region previously deprived of blood supply upon the restoration of blood flow.nu-cleotide-binding oligomerization domain-like receptor protein 3(NLRP3),a pivotal inflammatory complex,plays an inte-gral role in the pathophysiology of CIRI.Its activation can instigate a robust inflammatory cascade,culminating in nerve cell death.In light of these insights,this paper reviews the structural characteristics,signaling transduction pathways,and mechanistic involvement of NLRP3 in CIRI.Additionally,the drug intervention strategies targeting inflammatory cor-puscles are discussed,with the aim of providing reference materials for the prevention and treatment of CIRI by traditional Chinese medicine.
