1.Effectiveness of modified Morita therapy in the rehabilitation of long-term hospitalized patients with schizophrenia

Objective:To explore the effectiveness of modified Morita therapy in the long-term hospitalized patients of schizophrenia.Methods:A total of 84 long-term hospitalized patients of schizophrenia admitted to Minkang Hospital of Ningxia Civil Affairs Department from January 2020 to December 2022 were selected by convenience sampling method. Patients were randomly divided into a control group ( n=42) and an intervention group ( n=42) using a random number table method. The control group received psychotropic medication and routine psychiatric care, while the intervention group received additional modified Morita therapy for eight weeks. Before and after the intervention, the Positive and Negative Syndrome Scale (PANSS), Rosenberg Self-Esteem Scale, and Life Satisfaction Index A (LSIA) were used to assess clinical symptoms, life satisfaction, and self-esteem. Results:After the intervention, the intervention group showed significantly lower PANSS total scores [ (37.10±4.18) vs. (42.70±5.56), P<0.01] and general psychopathology symptom scores [ (19.98±2.63) vs. (22.59±3.04), P<0.01] compared to the control group. The Rosenberg Self-Esteem Scale and LSIA scores in the intervention group were significantly higher than those in the control group [ (28.89±4.17) vs. (26.61±5.74), P=0.02] and [ (11.39±1.88) vs. (9.33±2.57), P<0.01], respectively. Conclusions:Modified Morita therapy is effective in alleviating symptoms, improving life satisfaction, and enhancing self-esteem in long-term hospitalized patients in the remission stage of schizophrenia.

The mesencephalic astrocyte-derived neurotrophic factor (MANF) has been recently identified as a neurotrophic factor, but its role in hepatic fibrosis is unknown. Here, we found that MANF was upregulated in the fibrotic liver tissues of the patients with chronic liver diseases and of mice treated with CCl4. MANF deficiency in either hepatocytes or hepatic mono-macrophages, particularly in hepatic mono-macrophages, clearly exacerbated hepatic fibrosis. Myeloid-specific MANF knockout increased the population of hepatic Ly6Chigh macrophages and promoted HSCs activation. Furthermore, MANF-sufficient macrophages (from WT mice) transfusion ameliorated CCl4-induced hepatic fibrosis in myeloid cells-specific MANF knockout (MKO) mice. Mechanistically, MANF interacted with S100A8 to competitively block S100A8/A9 heterodimer formation and inhibited S100A8/A9-mediated TLR4-NF-κB signal activation. Pharmacologically, systemic administration of recombinant human MANF significantly alleviated CCl4-induced hepatic fibrosis in both WT and hepatocytes-specific MANF knockout (HKO) mice. This study reveals a mechanism by which MANF targets S100A8/A9-TLR4 as a "brake" on the upstream of NF-κB pathway, which exerts an impact on macrophage differentiation and shed light on hepatic fibrosis treatment.
4.Allosteric inhibition reveals SHP2-mediated tumor immunosuppression in colon cancer by single-cell transcriptomics.

Colorectal cancer (CRC), a malignant tumor worldwide consists of microsatellite instability (MSI) and stable (MSS) phenotypes. Although SHP2 is a hopeful target for cancer therapy, its relationship with innate immunosuppression remains elusive. To address that, single-cell RNA sequencing was performed to explore the role of SHP2 in all cell types of tumor microenvironment (TME) from murine MC38 xenografts. Intratumoral cells were found to be functionally heterogeneous and responded significantly to SHP099, a SHP2 allosteric inhibitor. The malignant evolution of tumor cells was remarkably arrested by SHP099. Mechanistically, STING-TBK1-IRF3-mediated type I interferon signaling was highly activated by SHP099 in infiltrated myeloid cells. Notably, CRC patients with MSS phenotype exhibited greater macrophage infiltration and more potent SHP2 phosphorylation in CD68+ macrophages than MSI-high phenotypes, suggesting the potential role of macrophagic SHP2 in TME. Collectively, our data reveals a mechanism of innate immunosuppression mediated by SHP2, suggesting that SHP2 is a promising target for colon cancer immunotherapy.
5.The feature of meibomian glands and tear film lipid layer in dry eye of children with long-term visual display terminals exposure

Objective To evaluate the feature of meibomian glands and tear film lipid layer thickness (LLT) in dry eye of children with long-term visual display terminals (VDTs) exposure.Methods Cross-sectional case control observation was performed.Eighty-four 6-14 years old children (149 eyes) with long or short time VDTs exposure from January to August 2017 in Tianjin Medical University Eye Hospital were enrolled,including 29 children 56 eyes for long time VDTs group and 55 children 93 eyes for short time VDTs group.Questionnaires were designed to detect disease history and ocular surface disease index (OSDI).Slit lamp microscopy,Keratograph 5 M,lipiview ocular surface interometry examination,corneal-conjunctival fluorescence staining,tear film break-up time (BUT) and Schirmer test (S I t) were performed on all the subjects.The subjective OSDI and clinical signs were compared between long time VDTs group and short time VDTs group and correlation between OSDI and clinical signs were analyzed in each group.This study followed the declaration of Helsinki.Informed consent was obtained from each subject or the guardian prior to any medical examination.Results Compared with the short time VDTs group,high OSDI value and long average noninvasive BUT (NIBUTav) were observed in the long time VDTs group,the differences were statistically significant (OSDI:t =-2.662,P =0.009;NIBUTav:Z =2.315,P =0.021).The meibomian gland dropout score was significantly different between the 2 group (x2 =11.779,P =0.008).In long time VDTs group,OSDI was negatively correlated with S I t (rs =-0.598,P =0.050) and positively correlated with meibomian gland dropout score and NIBUTav (rs =0.522,P =0.022;rs =0.597,P =0.011).In short time VDTs group,OSDI was positively correlated with first time noninvasive BUT (NIBUTf),NITBUTav and partial blink rate (rs =0.372,P =0.030;rs =0.408,P =0.008;rs =0.426,P =0.050).Conclusions Compared with short time VDTs exposure,the value of subjective OSDI was high and meibomian gland dropout was serious in long term VDTs exposure dry eye children.The OSDI value is associated with NIBUTf and partial blink ratio in short time VDTs dry eye children.
6.Identification of a novel COL2A1 variant in a pedigree affected with spondyloepiphyseal dysplasia congenita.

OBJECTIVE:
To explore the molecular basis for a pedigree affected with spondyloepiphyseal dysplasia congenita (SEDC).
METHODS:
The proband was subjected to whole exome sequencing. Suspected variant was verified by Sanger sequencing.
RESULTS:
All patients from the pedigree were found to carry a novel missense variant c.1394G>C (p.Gly465Ala) of the COL2A1 gene. The variant was not reported previously. Provean, Polyphen-2 and Mutation Taster software predicted that the variant is highly likely to be pathogenic.
CONCLUSION
The c.1394G>C (p.Gly465Ala) variant of the COL2A1 gene probably underlies the SEDC in this pedigree.

7.Evaluation of left atrial function in patients with severe preeclampsia by full-volume imaging technology

Objective To evaluate the left atrial(LA) function in patients with severe preeclampsia ( SPE) by full-volume imaging technology. Methods Forty-two patients of SPE were randomly chosen as SPE group,34 healthy pregnant women were selected as the control group. LA related function parameters including LA maximum volume( LAVmax),LA minimum volume( LAVmin),LA pre-atrium contraction volume(LAVpre),LA reservoir volume(LARV),LA passive emptying volume(LAPEV),LA contraction volume(LACV),LA expansion index(LAEI),LA passive emptying fraction(LAPEF),LA passive emptying fraction( LAAEF ) were separately required by biplane area-length method and full-volume imaging technology in antepartum and postpartum. Results Compared with control group before delivery,the parameters of SPE group before delivery such as LAVmax,LAVmin,LAVpre were higher,LAEI,LAPEF, LAAEF indexed to body surface area(BSA) were lower(all P <0.01).Compared with control group before delivery,the parameters of control group after delivery such as LAVmax,LAVmin,LAVpre were lower( all P <0.05),LAEI, LAAEF calculated as ratio to BSA were somewhat reduced,LAPEF calculated as ratio to BSA was relatively higher(all P >0.05).Compared with SPE group before delivery,the parameters of SPE group after delivery such as LAVmax,LAVmin,LAVpre were lower,LAEI,LAPEF,LAAEF indexed to BSA were higher( all P <0.01). Compared with control group after delivery,the parameters of SPE group after delivery such as LAVmax,LAVmin,LAVpre were still incresed ( all P < 0.05 ), LAEI,LAPEF, LAAEF indexed to BSA were relatively lower( all P >0.05). Conclusions Full-volume imaging technology can quantitatively evaluate the LA function. LA reservoir function,conduit function and booster pump function are reduced in SPE before delivery,LA rereservoir function,conduit function and booster pump function are basically recovery in SPE after delivery.
8.Effects of "one kind of high-quality nursing plan for one kind of disease" nursing model on self-efficacy and curative effect of depressive patients

Objective To explore the effects of"one kind of high-quality nursing plan for one kind of disease" nursing model on self-efficacy and curative effect of depressive patients with different severity. Methods Totally 120 depressive patients hospitalized in the department of mental health of our hospital from January 2014 to December 2015 were selected as the research object, and they were assigned to the observation group(n=60) and the control group(n=60). Patients in the control group were given routine nursing procedure, while patients in the observation group were given"one kind of high-quality nursing plan for one kind of disease" nursing model. Patients were evaluated by Hamilton depression scale ( HAMD) and general self-efficacy scale ( GSES) at the beginning of hospitalization, 15 d and 30 d after hospitalization. The evaluation results were compared between two groups. Patients in the observation group were divided into 3 groups based on the severity of depression, and the scores of HAMD and GSES at the beginning of hospitalization, 15 d and 30 d after hospitalization were compared among 3 groups ( mild group, moderate group and severe group).Results The GSES score in the observation group at 15 d (t =-2.79,P = 0.01) and 30 d ( t = 2. 60,P = 0.01) after hospitalization was higher than that in the control group; the HAMD score in the observation group at 15 d ( t = 2.19,P = 0.03) and 30 d ( t = 2.60, P = 0.01) after hospitalization was lower than that in the control group;the HAMD score-reducing rate of patients with mild depression ( P < 0. 01) and patients moderate depression ( P = 0.01) in the observation group was significantly higher than that of severe patients.Conclusions "One kind of high-quality nursing plan for one kind of disease" nursing model has significant effect in promoting the rehabilitation of depression and enhancing self-efficacy of depressive patients, especially the patients with mild depression and moderate depression. It also can make the patients more positively faced with the disease, so to obtain good curative effect.
9.Effects of electroacupuncture at Zusanli points on gut permeability in rats with intestinal ischemia/reperfusion injury

Objective To investigate the protective effects of elctroacupuncture(EA)at Zusanli(ST36) points on intestinal villas damage and mucosal permeability induced by small intestine pro-inflammatory factors in rats with intestinal ischemia/reperfusion(I/R). Methods 30 Sprague-Dawley(SD)rats were randomly divided into three groups(each,n=10):intestinal I/R group(model group),intestinal I/R+EA ST36 group(EA group)and intestinal I/R+sham EA group(SEA group). Rats were subjected to superior mesenteric artery(SMA)clamping at its root part to occlude the vessel for 30 minutes,followed by reperfusion for 60 minutes to form intestinal I/R models. Rats in EA group received EA at the bilateral ST36 points(2-3 mA,2-100 Hz)for 30 minutes immediately after ischemia,those in SEA group received EA at bilateral sham points(the point was located at 0.5 cm away from ST36 point in its lateral side)with the same frequency and intensity of stimulation as EA group for 30 minutes,and those in model group received no treatment. Animals were sacrificed 60 minutes after reperfusion and segments of distal part of ileum were harvested,then the levels of tumor necrosis factor-α(TNF-α)and interleukin-6(IL-6)in intestinal tissue were measured. Histopathologic changes were viewed and graded via light microscopy. A solution of fluorescein isothiocyanate(FITC)-dextran was injected into the lumen of the segment of intestine 30 minutes after reperfusion,systemic blood was drawn via abdominal aorta puncture at 60 minutes after reperfusion,and then the level of FITC-dextran in blood was measured to determine the changes in intestinal permeability. Results Compared to the model group and SEA group,EA ST36 significantly attenuated intestine TNF-α(pg/mg:3.01±0.50 vs. 8.65±1.02,8.42±1.41,both P<0.05)and IL-6 levels(pg/mg:2.51±0.15 vs. 6.34±0.86,6.13±1.12,both P<0.05),successfully maintained low gut injury scores(1.50±0.33 vs. 3.18±0.39,3.04±0.37,both P<0.05), and significantly reduced permeability of the distal ileum and the content of FITC-dextran(μg/L:282.42±73.92 vs. 856.22±229.47,844.22±239.47,both P<0.05). However,there were no significant differences in all above variables between SEA and model group(all P>0.05). Sections of distal ileum from animals in the model group and SEA group showed no obvious difference histologically,and the pathological manifestations were villous tip necrosis, blunt-shaped and collapse. Compared to the model group and SEA group,the intestinal villous injury in animals of EA group was much milder. Conclusion In rats with intestinal I/R injury,EA ST36 points has protective effect on the gut that is possibly due to the fact it may obviously lower the levels of the pro-inflammatory factors of small intestinal tissue,alleviate mucosal insult of gut and reduce the mucosal permeability.
