1.Association between systemic lupus erythematosus and hypothyroidism:a two-sample Mendelian randomization study

Objective To investigate the relationship between systemic lupus erythematosus(SLE)and hypothyroidism using bidirectional two-sample Mendelian randomization(MR)method.Methods The Genome-Wide Association Study data of SLE and hypothyroidism were obtained online.Independent single nucleotide polymorphisms closely related to SLE were screened as instrumental variable(Ⅳ),and outlier values were tested and eliminated by MR-PRESSO tool of R 4.3.1 software.The inverse variance weighted(IVW),MR-Egger,weighted mode(WM),weighted median(WME)and simple mode(SM)were used for MR analysis,and the values of odds ratio(OR)and 95%confidence interval(95%CI)were used to evaluate whether there was an association between SLE and hypothyroidism.The Cochran's Q heterogeneity test was performed for the results of IVW and MR-Egger,the pleiotropy test was performed by Egger-intercept method,and the sensitivity analysis was performed by elimination test one by one.F value was calculated to evaluate whether there was a weak Ⅳ bias.Results MR analysis results showed that there was a positive causal relationship between SLE and hypothyroidism in the overall population,and the results calculated by IVW,MR-Egger,WM and WME were statistically significant,with OR(95%CI)being 1.004(1.002-1.005),1.004(1.001-1.008),1.004(1.002-1.007),and 1.004(1.002-1.006),respectively.The heterogeneity test results for IVW and MR-Egger were P=0.086 and P=0.098,respectively,indicating no heterogeneity;the Egger-intercept result was P=0.295,indicating no pleiotropy;sensitivity analysis showed MR results were stable;and all F values were greater than 10,indicating no weak Ⅳ bias.Conclusion Compared with healthy people,the risk of hypothyroidism in patients with SLE is significantly higher.
2.DDX3X/NF-κB pathway mediates early neurons apoptosis in mice with subarachnoid hemorrhage

Objective:To study the role of DDX3X/NF-κB pathway in early neuronal apoptosis in subarachnoid hemorrhage(SAH)mice.Methods:The mouse model of SAH was established by internal carotid artery puncture,and the neurological function score of the mice was evaluated.The DDX3X expression was knocked down using recombinant lentivirus expressing DDX3X targeted shRNA(Lv-shDDX3X),or the NF-κB pathway was inhibited by NF-κB-IN-1(IN-1).Western Blot was used to detect the expression of DDX3X and NF-κB(p65)in mouse cortex.TUNEL/NeuN staining was used to detect the apoptosis of cerebral cortex neurons.Results:Twenty-four hours after SAH operation,the neurological function of mice was significantly impaired(P<0.05).While the expression of DDX3X was signifi-cantly increased and the expression of NF-κB(p65)was significantly decreased in the cortex(P<0.05).When the DDX3X expression is knocked down firstly,then SAH surgery is performed.The neurological function of mice was sig-nificantly recovered,and the expression of NF-κB(p65)protein was significantly higher than that in SAH group(P<0.05);If the NF-κB activity was inhibited by IN-1 while DDX3X knockdown,there is no significant recovery of neuro-logical function in SAH mice.TUNEL/NeuN staining showed that the number of TUNEL-positive neurons in the brain tissue after DDX3X knockdown was less than that in the SAH group(P<0.05),while the number of TUNEL-positive neurons was not significantly reduced when IN-1 was used to inhibit NF-κB activity at the same time of DDX3X knock-down.Conclusion:DDX3X/NF-κB mediated cell death in mice with early brain injury after SAH.
3.The combination of YAP/TAZ predicts the clinical prognosis in patients with cholangiocarcinoma after radical resection

Objective:Cholangiocarcinoma (CCA) is a malignant tumor derived from bile duct epithelial cells with extremely poor prognosis. The Hippo-Yes-associated protein (YAP)/transcription activator with PDZ binding motif (TAZ) signaling plays a critical role in cancer stem cell biology. Previous studies have shown that the positive expression of YAP/TAZ in CCA predicts larger tumor size and unfavorable clinical outcomes. We aim to evaluate the prognostic value of YAP/TAZ detection in CCA patients.Methods:CCA patients who underwent radical resection were retrospectively analyzed at our institution from January 2011 to June 2016. Postoperative pathological specimens were scored by YAP/TAZ immunohistochemical staining. The prognostic value of YAP/TAZ was analyzed by multivariate Cox-proportional hazards model.Results:A total of 91 CCA patients were enrolled. During a median follow-up time of 11.0 months, 69.2% patients relapsed and 45.1% died. The median OS and DFS were 10.7 months and 8.8 months respectively. The YAP/TAZ dual positive patients owned a worse TNM stage ( P=0.015), poorer tissue differentiation ( P=0.007), and a higher CA199 than those in negative patients. Multivariate Cox analysis identified that YAP/TAZ dual positivity as a significant factor predicted poorer OS ( P=0.010) and DFS ( P=0.028) in CCA patients after radical resection. In subgroup analysis, YAP/TAZ combination also significantly predicted OS ( P=0.044) and DFS ( P=0.043) in CCA patients with positive lymphatic metastasis and/or surgical margin who required adjuvant therapy. Conclusions:YAP/TAZ positivity is an independent predictive factor for survival in CCA patients after radical resectiony.
