ObjectiveTo study on the different therapeutic effects and potential mechanisms of Rhei Radix et Rhizoma(RH) before and after steaming with wine on constipation model mice. MethodsFifty-four male ICR mice were randomly divided into control group, model group, lactulose group(1.5 mg·kg-1), high, medium and low dose groups of RH and RH steaming with wine(PRH)(8, 4, 1 g·kg-1). Except for the control group, the constipation model was replicated by gavage of loperamide hydrochloride(6 mg·kg-1) in the other groups. After 2 weeks of modeling, each administration group was gavaged with the corresponding dose of drug solution, and the control and model groups were given an equal volume of normal saline, 1 time/d for 2 consecutive weeks. After administration, the feces were collected for 16S rRNA sequencing, the levels of gastrin(GAS), motilin(MTL), interleukin-6(IL-6), γ-interferon(IFN-γ) in the colonic tissue were detected by enzyme-linked immunosorbent assay(ELISA), the histopathological changes of colon were observed by hematoxylin-eosin(HE) staining, flow cytometry was used to detect the proportion changes of CD4+, CD8+ and regulatory T cell(Treg) in peripheral blood. ResultsCompared with the control group, the model group showed significantly decrease in fecal number in 24 h, fecal quality and fecal water rate(P<0.01), the colon was seen to have necrotic shedding of mucosal epithelium, localized intestinal glands in the lamina propria were degenerated, necrotic and atrophied, a few lymphocytes were seen to infiltrate in the necrotic area in a scattered manner, the contents of GAS and MTL, the proportions of CD4+, CD8+ and Treg were significantly reduced(P<0.01), the contents of IL-6 and IFN-γ were significantly elevated(P<0.05, P<0.01). Compared with the model group, the fecal number in 24 h, fecal quality and fecal water rate of high-dose groups of RH and PRH were significantly increased(P<0.05, P<0.01), the pathological damage of the colon was alleviated to varying degrees, the contents of GAS, MTL, IL-6 and IFN-γ were significantly regressed(P<0.05, P<0.01), and the proportions of CD4+ and CD8+ were significantly increased(P<0.01), although the proportion of Treg showed an upward trend, there was no significant difference. In addition, the results of intestinal flora showed that the number of amplicon sequence variant(ASV) and Alpha diversity were decreased in the model group compared with the control group, and there was a significant difference in Beta diversity, with a decrease in the relative abundance of Lactobacillus and an increase in the relative abundances of Bacillus and Helicobacter. Compared with the model group, the ASV number and Alpha diversity were increased in the high-dose groups of RH and PRH, and there was a trend of regression of Beta diversity to the control group, the relative abundance of Lactobacillus increased, and the relative abundances of Bacillus and Helicobacter decreased. ConclusionRH and PRH can improve dysbacteriosis, promote immune system activation, inhibit the release of inflammatory factors for enhancing the gastrointestinal function, which may be one of the potential mechanisms of their therapeutic effect on constipation.

Objective:To establish the normal reference value of the left ventricular remodeling index(RI)in healthy adults, and to evaluate the phenotypes of RI in various left ventricular geometries and its relationship with left ventricular systolic and diastolic functions in asymptomatic hypertensive patients. Methods:A retrospective study design was employed,906 healthy Han Chinese volunteers were selected as the control group from the multicenter study "Echocardiographic Measurements in Normal Chinese Adults"(EMINCA),which was conducted in 2012. The normal reference range of left ventricular RI was established based on the 95% CI( xˉ ± 1.96 s). Statistical analyses were conducted to evaluate differences in RI across age groups and between genders. A total of 340 asymptomatic hypertensive patients in the Department of Health Management,Sichuan Provincial People's Hospital from July 2023 to March 2024 were prospectively included as the hypertension group. Conventional echocardiography and two-dimensional speckle tracking imaging were used to assess left ventricular structure and function. Parameters such as left ventricular mass index(LVMI),RI,relative wall thickness(RWT),the ratio of early diastolic mitral inflow velocity to the average early diastolic mitral annular velocity(E/e'),and the ratio of early diastolic to late diastolic mitral inflow velocities(E/A)were calculated. Differences of these parameters between the hypertension group and the healthy control group were compared. The hypertension group was stratified into 3 subgroups based on left ventricular posterior wall thickness at end-diastole(LVPWd)and RI:the non-LV hypertrophy group, the left ventricular hypertrophy with normal RI group, and the left ventricular hypertrophy with low RI group. Relevant parameters were compared among these subgroups, and the statistical significance of the differences was analyzed. Pearson correlation analysis and multiple linear regression analysis were performed to explore the relationships between left ventricular RI and GLS,LVEF,the average mitral annular peak systolic velocity(s'),e',E/e',E/A. Results:In the healthy control group,RI showed a declining trend with increasing age in both sexes. Furthermore, the RI was significantly higher in the female group compared to the male group( P<0.001). Compared with the healthy control group,LVMI and E/e' increased while RI,e' and s' decreased in the hypertension group(all P < 0.001). Among the 3 subgroups of the hypertension group, compared with the other two groups,LVMI and E/e' increased while absolute GLS and s' reduced in the left ventricular hypertrophy with low RI subgroup(all P <0.001). Pearson correlation analysis revealed that RI was negatively correlated with GLS and E/e'( r=-0.457,-0.281;all P < 0.001),and positively correlated with LVEF,e',s' and E/A( r=0.229,0.394,0.150,0.172;all P < 0.05). Multivariate linear regression analysis demonstrated that left ventricular RI was independently associated with GLS,LVEF,e',s',E/e' and E/A. Conclusions:The normal reference range of left ventricular RI tends to decrease with age and is typically higher in females than in males. In asymptomatic patients with primary hypertension,RI,systolic and diastolic functions of the left ventricular are lower, while LVMI is higher. Among these patients, functional impairment is more pronounced in patients with left ventricular hypertrophy and reduced RI. This indicates that left ventricular RI may offer an imaging basis for further classification and stratification of structural and functional abnormalities in the left ventricle of asymptomatic hypertensive patients.

Schizophrenia (SZ) stands as a severe psychiatric disorder. This study applied diffusion tensor imaging (DTI) data in conjunction with graph neural networks to distinguish SZ patients from normal controls (NCs) and showcases the superior performance of a graph neural network integrating combined fractional anisotropy and fiber number brain network features, achieving an accuracy of 73.79% in distinguishing SZ patients from NCs. Beyond mere discrimination, our study delved deeper into the advantages of utilizing white matter brain network features for identifying SZ patients through interpretable model analysis and gene expression analysis. These analyses uncovered intricate interrelationships between brain imaging markers and genetic biomarkers, providing novel insights into the neuropathological basis of SZ. In summary, our findings underscore the potential of graph neural networks applied to multimodal DTI data for enhancing SZ detection through an integrated analysis of neuroimaging and genetic features.

Chinese Journal of Postgraduates of Medicine 2025;48(6):548-554
Objective:To investigate the relationship between soluble vascular cell adhesion molecule-1 (sVCAM-1), high mobility group protein B1 (HMGB1), and heart failure after percutaneous coronary intervention (PCI) in patients with unstable angina (UA).Methods:This study was a retrospective study. The data of 196 UA patients who underwent PCI treatment in Xi′an Daxing Hospital from January 2019 to December 2023 were collected through the hospital′s electronic medical record system. The patients were followed up for 6 months after PCI, and the follow-up data were completed. According to the occurrence of heart failure during the follow-up period, they were divided into the occurrence group (35 cases) and the non occurrence group (161 cases). The general data, serum sVCAM-1, HMGB1 and other laboratory indicators before PCI were compared between the two groups, and the relationship between serum sVCAM-1, HMGB1 and the occurrence of heart failure in UA patients after PCI was analyzed. Results:Compared with the non occurrence group, the occurrence group had higher proportion of diabetes mellitus and multi-vessel disease : 28.57% (10/35) vs. 13.66% (22/161), 62.86% (22/35) vs. 32.30% (52/161); larger left ventricular diameter : (4.73 ± 0.54) cm vs (4.52 ± 0.51) cm; higher levels of serum cardiac troponin I (cTnI), creatine kinase MB isoenzyme (CK-MB), sVCAM-1 and HMGB1: 0.07 (0.05, 0.08) μg/L vs. 0.05 (0.04, 0.06) μg/L, (8.16 ± 1.58) μg/L vs. (7.08 ± 1.21) μg/L, (458.26 ± 41.23) μg/L vs. (422.19 ± 37.41) μg/L, (6.39 ± 1.44) μg/L vs. (5.24 ± 1.45) μg/L, there were statistical differences ( P<0.05). Logistic regression analysis showed that the occurrence of heart failure in UA patients after PCI may be related to a larger left ventricular diameter, abnormal levels of serum cTnI, sVCAM-1, and HMGB1 ( P<0.05). Restrictive cubic spline (RCS) analysis revealed a non-linear dose-response relationship between serum sVCAM-1, HMGB1, and the occurrence of heart failure in UA patients after PCI ( P<0.05). When serum sVCAM-1 ≥ 422.37 μg/L and HMGB1 ≥ 5.26 μg/L, the risk of heart failure in patients after PCI increased with the level of both. Serum sVCAM-1 and HMGB1 had a positive interactive effect on the occurrence of heart failure in UA patients after PCI. Decision curves and column charts were drawn to measure the model, it was found that the predictive model constructed with serum sVCAM-1 and HMGB1 assisted serum cTnI had certain predictive efficacy for the occurrence of heart failure in UA patients after PCI. Conclusions:The occurrence of heart failure in UA patients after PCI may be related to abnormally high levels of sVCAM-1 and HMGB1. As the levels of both increase, the risk of heart failure in patients increases. Using both to assist in predicting the risk of heart failure in patients has certain value.
