1.Efficiency and safety of haematopoietic stem cell collection in healthy donors

[Objective] To explore the key factors affecting the efficiency and safety of hematopoietic stem cell apheresis. [Methods] The clinical data of 59 healthy donors who underwent allogeneic hematopoietic stem cell donation in the First Affiliated Hospital of Anhui Medical University from January 2021 to June 2024 were retrospectively analyzed. The number of CD34+ cells was used to evaluate the eligibility of stem cell collection. The effects of donor gender, age, patient weight, as well as the number of WBC, MNC, RBC, Hb, HCT, PLT, CD34+ cells, CD34+ percentage and instrument operating parameters on collection efficiency were analyzed. [Results] A total of 59 donors were enrolled, and 68 occasions of stem cell apheresis were performed, with a qualified collection rate of 56%. Donor gender, age, patient weight, total blood circulation volume, anticoagulant dosage, collection time, calcium gluconate dosage and RBC, Hb, HCT levels were not significantly correlated with the collection effect (P>0.05). Multivariate logistic regression analysis showed that the number of MNC cells, CD34+ cells and stem cell product volume were the key factors affecting the efficiency and safety. A total of 12 donors had mild adverse reactions during the collection process, and all of them were improved after treatment. [Conclusion] Optimizing apheresis strategy based on the three factors of MNC, WBC count and stem cell product volume on the day of collection will help to achieve high-quality collection and improve the success rate of transplantation.

With the rapid advancement of artificial intelligence technology, chatbots have shown great potential in the healthcare sector. From personalized health advice to chronic disease management and psychological support, chatbots have demonstrated significant advantages in improving the efficiency and quality of healthcare services. As the scope of their applications expands, the relationship between technological complexity and practical application scenarios has become increasingly intertwined, necessitating a more comprehensive evaluation of both aspects. This paper, from the perspective of he althcare applications, systematically reviews the technological pathways and development of chatbots in the medical field, providing an in-depth analysis of their performance across various medical scenarios. It thoroughly examines the advantages and limitations of chatbots, aiming to offer theoretical support for future research and propose feasible recommendations for the broader adoption of chatbot technologies in healthcare.
3.The epidemiological survey of depressive and anxiety disorder among people aged 18 and above in Henan Province in 2021

Objective:This survey aims to investigate the prevalence, distribution, and medical treatment of depressive and anxiety disorder among people aged 18 and above in Henan province.Methods:An epidemiological survey was conducted in 30 counties (cities and districts) of Henan province from November 2020 to March 2021. A total of 10 800 permanent urban and rural residents aged 18 and above were selected by the stratified multi-stage random sampling method. The Patient Health Questionnaire-9 (PHQ-9) and Generalized Anxiety Disorder Scale (GAD-7) were used for primary screening of depressive and anxiety disorders respectively. Then all participants were divided into high-, medium-, and low-risk groups based on the primary screening results. All participants in high-risk groups, 40% participants randomly selected in medium-risk groups, and 10% randomly selected in low-risk groups underwent a diagnostic procedure for depressive and anxiety disorder conducted by psychiatrists according to the 10th revision of the International Statistical Classification of Diseases and Related Health Problems (ICD-10). The adjusted prevalence, standardized prevalence, and 95 %CI were calculated based on the number of patients diagnosed with depressive or anxiety disorders in the three risk groups in 12 months. The distribution of the prevalence of two mental disorders were compared using the chi-square test and Fisher′s exact probability test. Results:Among the 10 057 participants who completed the survey, 229 were diagnosed with depressive disorder and 412 with anxiety disorder. The 12-month standardized prevalence of depressive and anxiety disorder were 1.96% (95 %CI=1.51%-2.32%) and 3.90% (95 %CI=3.57%-4.23%), respectively. The prevalence rate was higher in populations living in rural areas, aged ≥60 years old, having a lower education level, having a lower monthly income, performing less regular exercise, having chronic disease, having poor physical health, and lack of mental health literacy. The treatment rates of patients with depressive and anxiety disorders were 32.8% (75/229) and 25.5% (105/412), respectively. The main reasons for non-seeking medical services were the unawareness that poor mental health status could be a disease and the consideration that mental health problems were not serious. Conclusion:The prevalence of depressive and anxiety disorder in urban and rural residents of Henan province is lower than that in other provinces of China and the corresponding national data. These prevalence rates vary greatly among populations with different demographic characteristics. The treatment rate is still low in the current studied population.

OBJECTIVE:
To evaluate the effect of rosmarinic acid (RA) on mitophagy and hypertrophy of cardiomyocytes exposed to high glucose (HG).
METHODS:
Rat cardiomyocytes (H9c2) exposed to HG (25 mmol/L) were treated with 50 μmol/L RA or with both RA treatment and Parkin siRNA transfection, with the cells cultured in normal glucose (5.5 mmol/L) and HG as the controls. The expressions of PINK1, Parkin and LC3II/LC3I in the cells were detected by Western blotting. The formation of mitochondrial autophagosomes was observed by transmission electron microscope. Excess activation of cardiac fibroblasts inevitably induces cardiac fibrosis. Emodin has been used as a natural medicine against several chronic diseases. The objective of this study is to determine the effects of emodin on cardiac fibrosis and the underlying molecular mechanisms. Intragastric administration of emodin markedly decreased left ventricular wall thickness in a mouse model of pathological cardiac hypertrophy with excess fibrosis induced by transaortic constriction (TAC) and suppressed activation of cardiac fibroblasts induced by angiotensin II (AngII). Emodin upregulated expression of metastasis associated protein 3 (MTA3) and restored the MTA3 expression in the setting of cardiac fibrosis. Moreover, overexpression of MTA3 promoted cardiac fibrosis; in contrast, silence of MTA3 abrogated the inhibitory effect of emodin on fibroblast activation. Our findings unraveled the potential of emodin to alleviate cardiac fibrosis upregulating MTA3 and highlight the regulatory role of MTA3 in the development of cardiac fibrosis.
