1.Analysis of immunogenicity of African swine fever virus p37 recombinant protein in mice

The aim of this study is to explore the immunogenicity of African swine fever virus p37 recombinant protein in mice.C57BL/6J mice were immunized subcutaneously in the abdomen using p37 recombinant protein as antigen.The second immunization was performed 21 d after the first immunization.Serum-specific antibody levels were detected by ELISA;serum cytokine levels were detected using a multifactor assay technique;mice splenic lymphocytes were isolated 7 d after sec-ondary immunization,and the number of splenic lymphocytes secreting IFN-γ after recombinant protein stimulation was detected by ELISpot;and the ratio of CD4+T cells to CD8+T cells was detected by flow cytometry.The results of indirect ELISA showed that p37 recombinant protein could stimulate mice to produce high levels of specific antibodies;ELISpot showed that p37 recom-binant protein could significantly stimulate splenic lymphocytes to produce IFN-γ(P<0.001)and activate cellular immune responses;the results of flow cytometry showed that it could signifi-cantly stimulate the differentiation of T-lymphocytes to CD4+T-lymphocytes(P<0.001).In ad-dition,serum levels of IL-2,IL-4,IFN-γ,and TNF-α immune-related cytokines were significantly higher after the second immunization.Immunization of mice with p37 recombinant protein induced strong humoral and cellular immune responses with good immunogenicity,providing reference for the subsequent epitope identification and functional study of p37 protein and the antigen screening of ASF mRNA vaccine.
2.Therapeutic effect of occlusive dressing therapy on skin lesions in synovitis-acne-pustulosis-hyperostosis-osteomyelitis syndrome

Objective To observe the efficacy of occlusive dressing therapy combined with infra-red lamp irradiation on skin lesions in patients with synovitis-acne-pustulosis-hyperostosis-osteomyeli-tis(SAPHO)syndrome.Methods A total of 52 patients with SAPHO syndrome were selected as study subjects.They were randomly divided into control group and observation group using random number table method,with 26 patients in each group.The control group received conventional treat-ment,while the observation group received occlusive dressing therapy combined with infrared lamp ir-radiation on the basis of the control group.The skin lesion scores,Dermatology Life Quality Index(DLQI)scores,Self-rating Anxiety Scale(SAS)scores,and Self-rating Depression Scale(SDS)scores of the two groups were compared.The clinical efficacy and adverse reactions of the two groups were also statistically analyzed.Results One and two months after treatment,the skin lesion scores of both groups were lower than those before treatment,and the scores in the observation group were lower than those in the control group(P<0.05).After treatment,the DLQI,SAS,and SDS scores of both groups were lower than those before treatment,and the scores in the observation group were lower than those in the control group(P<0.05).The total effective rate in the observation group was 96.15%,which was higher than 80.77% in the control group(P<0.05).There was no statistically significant difference in the incidence of adverse reactions between the two groups(P>0.05).Con-clusion Occlusive dressing therapy combined with infrared lamp irradiation can significantly improve the symptoms of skin lesions and the psychological state of patients with SAPHO syndrome,as well as enhance their quality of life and the therapeutic effect of the disease.

African swine fever(ASF)is a highly contagious disease caused by the African swine fe-ver virus(ASFV).To evaluate the immunogenicity of the pp62(CP530R)protein of ASFV,the recombinant CP530R protein was expressed in HEK 293F cells transfected with the plasmid pMAL-Fc-CP530R.Six-week-old female C57BL/6J mice were immunized with 10 μg of purified pp62 protein via subcutaneous injection,followed by a booster immunization with the same dosage at 21 days(enhanced).The humoral and cellular immune responses in the mice were then assessed using ELISA,flow cytometry,and ELISpot assays.Western blot analysis confirmed that the pp62 protein was successfully expressed,with a molecular weight of approximately 118.5 kDa.ELISA results indicated that a high level of specific antibodies was detected in the immunized mice,with antibody titers reaching up to 1∶1 638 400 at 7 days after the secondary immunization.The pro-portion of CD8+T lymphocytes in the immunized mice increased compared to the control group(P<0.05).Results from the Q-PlexTM Mouse Cytokine Screen demonstrated that the secretion levels of IFN-γ,IL-2,IL-4,and IL-10 in serum were significantly upregulated in the immunized mice following secondary immunization(P<0.001).In summary,these findings indicate that the pp62 protein can significantly stimulate both humoral and cellular immunity in mice,laying the groundwork for further studies on the function of the ASFV pp62 protein and the identification of novel vaccine antigens for ASF.
5.The characteristics of retinal neovascularization and tube diameter and the levels of miR-126 and miR-141-3p in peripheral blood of patients with diabetic retinopathy

Objective:To analyze the characteristics of retinal neovascularization and tube diameter and the levels of miR-126 and miR-141-3p in peripheral blood of patients with diabetic retinopathy (DR).Methods:A total of 130 patients with DR Admitted to the Xi′an First Hospital from January 2020 to December 2023 were selected as the study objects (DR group), and 139 patients with type 2 diabetes who did not develop retinopathy during the same period (non-DR group) were selected. The characteristics of retinal neovascularization and tube diameter and the levels of miR-126 and miR-141-3p in peripheral blood were compared between the two groups. The patients with DR were divided into two groups according to whether retinal neovascularization and vascular diameter abnormalities occurred. The levels of miR-126 and miR-141-3p were compared between the two groups. Logistic regression analysis was used to evaluate the relationship between miR-126 and miR-141-3p levels in peripheral blood and retinal neovascularization and vascular diameter abnormalities in DR patients. Receiver operating characteristic (ROC) curve was drawn to examine the predictive efficacy of peripheral blood miR-126 and miR-141-3p levels on retinal neovascularization and tube diameter abnormalities in patients with DR.Results:There were significant differences in retinal neovascularization and vascular diameter between DR And non-DR patients. The retinal features of DR patients were significant, showing that a large number of new blood vessels form in and around the optic disc, spread to the entire retina, and form a fragile network structure, which was easy to be damaged and adhere to the vitreous cortex, and wound not disappear by itself and might invade the vitreous. The vascular diameter varied greatly, partial dilation showed abnormal blood flow, and partial stenosis reflected vascular damage. There was no significant retinal neovascularization in the non-DR group. The levels of miR-126 and miR-141-3p in the DR group were significantly lower than those in the non-DR group (all P<0.05). Among 130 patients with DR, 62 of them had abnormal retinal neovascularization and tube diameter, accounting for 47.69%. The levels of miR-126 and miR-141-3p in peripheral blood of DR patients with retinal neovascularization and vascular diameter abnormalities were lower than those in the non-DR group (all P<0.05). Logistic regression analysis showed that the levels of miR-126 and miR-141-3p in peripheral blood were associated with abnormal retinal neovascularization and tube diameter in DR patients (all P<0.05). ROC curve analysis showed that miR-126 and miR-141-3p had good predictive value for the occurrence of retinal neovascularization and tube diameter abnormalities in patients with DR. Conclusions:Some patients with DR have abnormal characteristics of retinal neovascularization and tube diameter, and peripheral blood miR-126 and miR-141-3p are related to the neovascularization and tube diameter abnormalities in patients with DR.

8.Clinical characteristics and treatment of central nervous system aspergillosis: an analysis of 37 cases

Objective:To summarize the clinical characteristics and therapeutic efficacy of central nervous system (CNS) aspergillosis.Methods:The clinical manifestations, laboratory examination, neuroimaging features, treatment and prognosis of 37 cases of CNS aspergillosis diagnosed and treated in the First Medical Center of People′s Liberation Army General Hospital from January 2000 to January 2021 were retrospectively analyzed. According to the correlation between intracranial lesions and paranasal sinus lesions, they were divided into two groups: rhino-cerebral aspergillosis (RA, n=21) group and cerebral aspergillosis (CA, n=16) group. Results:Only 16.2% (6/37) of CNS aspergillosis patients had a clear background of immunosuppression, but 35.1% (13/37) were complicated with diabetes. The most common clinical manifestations were headache (73.0%, 27/37), cranial nerve involvement (59.5%, 22/37) and fever (37.8%, 14/37). Cerebrospinal fluid characteristics included increased pressure (53.8%, 14/26), increased white blood cell count (46.7%, 14/30), decreased glucose (30.0%, 9/30), increased protein (70.0%, 21/30), and high positive results of the metagenomic next-generation sequencing (mNGS) of pathogenic microorganism (7/10). Cranial magnetic resonance imaging showed that commonly involved sites were sinus, orbital apex, posterior orbit, cavernous sinus (43.2%, 16/37) and cerebral lobes (27.0%, 10/37). Treatment options included antifungal drugs alone (64.9%, 24/37), combination of drugs and surgery (27.0%, 10/37) and surgery alone (8.1%, 3/37). Compared with the CA group, RA group had fewer males [47.6% (10/21) vs 14/16, χ2=6.34, P=0.012] and older age [(54.2±19.4) years vs (38.4±18.4) years, t=2.50, P=0.017], and was more prone to headache [85.7% (18/21) vs 9/16, χ2=4.00, P=0.046) and cranial nerve involvement [81.0% (17/21) vs 5/16, χ 2=9.31, P=0.006]. The misdiagnosis rate of these patients in the early stage was 73.0% (27/37). A total of 29 patients (85.3%, 29/34) were treated with voriconazole successively, and the course of treatment was 3.0 (0.5, 10.4) months. Compared with salvage therapy, the mortality of primary therapy was lower (4/17 vs 9/12, χ2=7.54, P=0.006). All patients were followed up to December 2021, and 17 patients died, with a mortality rate of 45.9% (17/37). Conclusions:CNS aspergillosis may have no definite immunosuppressive background. Some of CNS aspergillosis patients are complicated with diabetes, and the clinical manifestations of the disease lack specificity, with high misdiagnosis rate in the early stage, no inflammatory changes in cerebrospinal fluid, and high positive rate of mNGS for pathogenic microorganism. Early and long-term application of voriconazole can significantly reduce the mortality rate.
9.Research on magneto-acoustic-electrical tomography method based on liquid metal contrast agent and M sequence coded excitation.

Magneto-acoustic-electric tomography (MAET) boasts high resolution in ultrasound imaging and high contrast in electrical impedance imaging, making it of significant research value in the fields of early tumor diagnosis and bioelectrical monitoring. In this study, a method was proposed that combined high conductivity liquid metal and maximum length sequence (M sequence) coded excitation to improve the signal-to-noise ratio. It was shown that, under rotational scanning, the liquid metal significantly improved the signal-to-noise ratio of the inter-tissue magneto-acoustic-electric signal and enhanced the quality of the reconstructed image. The signal-to-noise ratio of the signal was increased by 5.6, 11.1, 21.7, and 45.7 times under the excitation of 7-, 15-, 31-, and 63-bit M sequence code, respectively. The total usage time of 31-bit M sequence coded excitation imaging was shortened by 75.6% compared with single-pulse excitation when the same signal-to-noise ratio was improved. In conclusion, the imaging method combining liquid metal and M-sequence coding excitation has positive significance for improving MAET image quality.

10.Application of methoxyamine combined with target guided fluid in elderly patients undergoing pancreaticoduodenectomy

Objective:To investigate the effect of methoxyamine combined with target guided fluid in elderly patients undergoing pancreaticoduodenectomy.Methods:90 elderly patients undergoing pancreaticoduodenectomy were randomly divided into methoxyamine group and control group, with 45 cases in each group. The patients in both groups were treated with intravenous inhalation combined anesthesia. The stroke volume variation (SVV) was maintained at 7%-10% and the central venous pressure (CVP) was 4-8 cmH 2O. In methoxyamine group, 3 μg/(kg·min) methoxyamine was continuously pumped, while the control group was pumped with the same amount of normal saline at the same speed. The intraoperative infusion volume, urine volume, bleeding volume, blood transfusion cases, intraoperative mean arterial pressure, heart rate, blood gas analysis results, B-type natriuretic peptide (BNP), creatinine, urea nitrogen level and postoperative exhaust time were compared between the two groups. Results:Compared with the control group, the patients in methoxyamine group had less infusion volume, urine volume, lower postoperative BNP level and heart rate, shorter postoperative exhaust time (all P<0.05), and higher mean arterial pressure ( P<0.05). There was no significant difference in blood loss, blood transfusion cases, PaO 2, PaCO 2, pH, creatinine and urea nitrogen between the two groups (all P>0.05). In addition, the number of patients in the methoxyamine group who used pressor drugs was less than that in the control group ( P<0.05), and the frequency of bradycardia was more than that in the control group ( P<0.05). The proportion of tachycardia and urapidil was similar in the two groups (all P>0.05). Conclusions:Methoxyamine combined with target guided fluid therapy can reduce the intraoperative infusion volume of pancreaticoduodenectomy in elderly patients, stabilize circulation, shorten postoperative exhaust time, and contribute to the recovery of gastrointestinal function.
