Objective:To explore the differences in reproductive tract microbiota between patients with moder-ate and severe intrauterine adhesions(IUA)and infertile patients with normal uterine cavities and the clinical sig-nificance.Methods:Thirty patients were enrolled in the study from June 1,2022 to September 12,2022.Among them,15 cases were confirmed as moderate to severe IUA by hysteroscopy(group A),and 15 cases of primary infertility were excluded from intrauterine diseases through outpatient hysteroscopy(group B).Perform high-throughput sequencing of secretions from the vagina,cervix,and uterine cavity separately.Results:There is no difference in the α diversity of intrauterine microbiota between the two groups,but the α diversity of intrauterine mi-crobiota in group A is significantly higher than that of cervical microbiota(P=0.036).At the genus level,the top 10 relative abundances are the same for both groups.At the species level,the abundance of lactobacillus iners in group A is significantly lower than that in group B(P=0.001).There is no statistically significant difference in the abundance of microorganisms in the vagina,cervix and uterine cavity of group A.Conclusions:The types of en-dometrial microbiota in IUA patients and infertile women are roughly the same,but the composition is different.There are differences in the diversity of reproductive tract microbiota in patients with IUA,with a higher diversity of microbiota in the uterine cavity.Which suggests that the pathogenesis of IUA may be related to the changes of endometrial microbiota,increased microbial diversity,complex microbial species,especially the imbalance of the microecology of lactobacillus,which is not conducive to the repair of endometrial damage.

Objective To assess the effect of low‐dose aspirin on endometrium after transcervical resection for severe intrau‐terine .Methods Eighty women after transcervical resection and foley balloon for severe intrauterine adhesions were divided into 4 groups randomly .Twenty cases in each group .Group A :fill jiale 3 mg ,3 times/day ,1 consecutive month ;Group B :fill jiale 3 mg ,3 times/day plus aspirin 50 mg ,1 time/day ,1 consecutive month ;Group C :fill jiale 3 mg ,3 times/day ,75 mg of aspirin ,1 time/day ,1 consecutive month ;Group D :fill jiale 3 mg ,3 times/day ,100 mg of aspirin ,1 time/day ,1 month in a row .Endometrial thickness , pattern of endometrium ,the resistance index(RI) and pulsed index (PI) were observed and recorded the menstrual blood volume . The endometrial cavity were performed through hysteroscope after one month .Results First ,all endometrial thickness in group B , C ,D on HCG day were thicker than group A ,there were significant difference(P< 0 .05) ;A‐type endometrium rate of group B ,C , D were higher than that of group A ,however ,RI and PI were lower than that of group A ,there had significant difference compared with group A(P< 0 .05) ;Second ,the menstrual blood volume have been great changed in group B ,C ,D ,there had significant differ‐ence compared with group A(P< 0 .05) ,there were no significant changes between the three group(P> 0 .05) ;Third ,the endome‐trial cavity in group B ,C ,D were recovery well(P< 0 .05) ,there were no significant changes between the three group(P > 0 .05) . Conclusion low‐dose aspirin combined fill jiale can significantly improve the growth of endometrial caused by intrauterine adhe‐sions cutting and recovery .

Objective:To study the biological effects of grape seed extract(GSE) on the human ovary cancer cells and explore its molecular mechanism. Methods: Human ovary cancer cells line SKOV3 were cultured.They were put into 96-well plate and added with solutions of GSE of the terminal concentrations of 25,50,100,200,400 ?g/ml respectively,and 24,48,72 hours later cell growth curve was drawn. MTT assay was used for other SKOV3 cells co-incubated with GSE of the terminal concentration of 25,50,100,200,400 ?g/ml respectively so as to calculate the proliferation inhibition rate. SKOV3 cells were treated with GSE of the concentration of 25 ?g/ml for 24,48 hours respectively. Flowcytometry(FCM)was used to analyze the DNA cycle and TUNEL was used to calculate the apoptotic rate. Annexin-V labeling method was used to detect the positive rate of apoptotic cells. SKOV3 cells were treated with GSE of the concentration of 25 ?g/ml for 24,48,72 hours.Western blotting was used to detect the protein expression of caspase-3. Results:GSE dose-dependently inhibited the proliferation of the SKOV3 cells in a dose-dependent manner.Treated by GSE,the progress of cells at stage into G2/M stage was inhibited.Tunel showed that treated by GSE (25 ?g/ml)for 24、48 h the apoptotic rates of the cells were 31.98%and 45.78%, respectively. Annexin-V showed that after incubation with GSE (25 ?g/ml)for 12,24,48 h,the apoptotic rate were 6.71%,19.05%, 36.55%,respectively. Western blotting showed that the caspase-3 protein expression were up-regulated.Conlusion:GSE inhibits the proliferation of malignant human ovary cancer cells and induces their apoptosis. Expression of caspase-3 protein may be related to cell growth inhibition and the apoptosis mediated by GSE in vitro.

Methods Human ovary cancer cell line SKOV3 were co-cultured with GSPE solution of the terminal concentrations of 25,50,100,200,400 ?g/ml respectively in 96-well plate. At 24,48,72 h after coculture,the following parameters were detected: cell growth curve,the inhibition rate of SKOV3 cells by MTT assay,DNA cycle by FCM,the apoptosis of SKOV3 by TUNEL and Annexin-V labeling method,the mRNA and protein expressions of survivin by semi-quantitative RT-PCR and Western blotting,respectively. Results GSPE dose-dependently inhibited the proliferation of the SKOV3 cells. Treated by GSPE,the progress of SKOV3 cells at S stage into G/M stage was inhibited. TUNEL showed that treated by 25 ?g/ml GSPE for 24,48 h,the apoptotic rates of SKOV3 cells were 31.98%,45.78% respectively. Annexin-V showed that after incubation with 25 ?g/ml GSPE for 24 h,the apoptotic rate was 14.68%. The survivin mRNA and protein expressions were both down-regulated. Conclusion GSPE inhibits the proliferation of malignant human ovary cancer cells and induces their apoptosis. Expression of survivin mRNA and protein may be related to cell growth inhibition and to the apoptosis mediated by GSPE in vitro.
