1.Differences and related factors of preschool children s evaluation by parents and teachers using the Strengths and Difficulties Questionnaire

Objective:
To explore differences in the factors influencing parents and teachers assessments of preschool children s mental health using the Strengths and Difficulties Questionnaire (SDQ), so as to provide reference for promoting children s mental health.
Methods:
A retrospective analysis was conducted on the SDQ survey data of 14 763 middle and senior kindergarten children in Nanshan District, Shenzhen, from March to June 2023. Chi square χ 2 tests were used to analyze differences in mental health assessments between parents and teachers. Multivariate Logistic regression was employed to examine the factors influencing parental assessments, and Kappa coefficients were used to evaluate the consistency between parent and teacher evaluations.
Results:
The positive rate of mental health problems reported by parents (7.2%) was significantly higher than that reported by teachers (6.2%) ( χ 2=254.27, P <0.01). Gender differences revealed that parents reported a lower positive rate for boys (7.9%) compared to teachers (8.5%), whereas for girls, the parental positive rate (6.4%) was higher than that reported by teachers (3.8%) ( χ 2=163.59, 81.26, all P <0.01). Age related differences showed that parental positive rates for 4, 5, and 6 year olds (8.5%, 7.4%, 5.8%) were consistently higher than teachers assessments (6.3%, 6.7%, 5.4%) ( χ 2=41.23, 157.53, 63.67, all P <0.05). Univariate analysis of parental assessments indicated higher positive rates among boys (7.9%), 4 year olds (8.5%), mothers aged 20-35 ( 6.6 %), mothers with high school education or below (9.8%), fathers aged 23-40 (6.4%), fathers with high school education or below (10.3%), and children exposed to secondhand smoke (7.9%) ( χ 2=23.56-235.24, all P <0.01). Multivariate Logistic regression identified lower parental education levels and exposure to secondhand smoke as significant risk factors for abnormal SDQ assessments by parents ( χ 2=2.05, 1.62, 3.15, all P <0.05). The Kappa coefficients for parent-teacher agreement across SDQ subscales and total difficulties ranged from 0.04 to 0.12 (all P <0.01).
Conclusions
Parental education level and exposure to secondhand smoke are significant factors influencing preschool children s mental health. Differences exist between parental and teacher assessments of children s mental health, and incorporating teacher evaluations can provide a more comprehensive understanding of preschoolers psychological well being.
2.Establishing N-ELISA-based method for rapid detection of neutralizing antibodies against respiratory syncytial virus

Objective:To prepare rabbit polyclonal antibodies against respiratory syncytial virus (RSV) N protein and use them as the detection antibodies to establish a N-ELISA-based method for rapid detection of neutralizing antibodies.Methods:A plasmid of pET30a-N for the expression of RSV N protein was constructed. After purification, the protein was immunized into New Zealand rabbits to prepare polyclonal antibodies, which were used as the detection antibodies. Positive serum samples were diluted and used to neutralize RSV (100 TCID 50/well). Hep-2 cells were inoculated and cultured, and then the cells were fixed with 80% acetone. ELISA was performed to detect RSV N protein in infected cells. When the absorbance value of a well was below the cut-off value, it was regarded as the positive well in the neutralization test. The highest dilution of a positive well serum was the neutralizing antibody titer. After optimizting the antibody dilution, detection time, cell density and the duration of neutralization, the method for neutralizing antibody detection was established based on N-ELISA. The established method was verified by analyzing the influences of different cell generations and edge effects, and calculating the accuracy, repeatability and precision. The correlation between the established method and microneutralization method was analyzed by detecting human RSV IgG-positive serum. Results:The plasmid pET30a-N was successfully constructed, and the expressed N protein showed high purity and good specificity. After the third immunization, the antibody titer in rabbit serum was 1∶51 200, and the antibodies could specifically bind to RSV. The prepared rabbit anti-RSV N polyclonal antibodies had a titer of 1∶51 200, and showed good specificity. The neutralizing antibodies could be detected on day 4 with the established method, and the duration of neutralization was shortened to 30 min. Cell generations and the position of wells in the 96-well plate (edge well and non-edge well) had no significant effect on the method, and the repeatability, precision and accuracy of the method were good. In the detection of 64 RSV IgG-positive human serum samples by the established method and microneutralization method, the correlation coefficient was 0.929 6, indicating a good positive correlation between the two methods.Conclusions:A N-ELISA-based method for rapid neutralizing antibody detection is successfully established, which can be used to evaluate the serum antibody level after RSV vaccination.
3.Analysis of chest imaging features of novel coronavirus pneumonia, bacterial pneumonia and viral pneumonia

Objective:To investigate and summarize the chest CT imaging features of patients with novel coronavirus pneumonia (COVID-19), bacterial pneumonia and other viral pneumonia.Methods:Chest CT data of 102 patients with pulmonary infection due to different etiologies were retrospectively analyzed, including 36 patients with COVID-19 admitted to Hainan Provincial People's Hospital and the Second Affiliated Hospital of Hainan Medical University from December 2019 to March 2020, 16 patients with other viral pneumonia admitted to Hainan Provincial People's Hospital from January 2018 to February 2020, and 50 patients with bacterial pneumonia admitted to Haikou Affiliated Hospital of Central South University Xiangya School of Medicine from April 2018 to May 2020. Two senior radiologists and two senior intensive care physicians were participated to evaluated the extent of lesions involvement and imaging features of the first chest CT after the onset of the disease.Results:Bilateral pulmonary lesions were more common in patients with COVID-19 and other viral pneumonia, and the incidence was significantly higher than that of bacterial pneumonia (91.6%, 75.0% vs. 26.0%, P < 0.05). Compared with other viral pneumonia and COVID-19, bacterial pneumonia was mainly characterized by single-lung and multi-lobed lesion (62.0% vs. 18.8%, 5.6%, P < 0.05), accompanied by pleural effusion and lymph node enlargement. The proportion of ground-glass opacity in the lung tissues of patients with COVID-19 was 97.2%, that of patients with other viral pneumonia was 56.2%, and that of patients with bacterial pneumonia was only 2.0% ( P < 0.05). The incidence rate of lung tissue consolidation (25.0%, 12.5%), air bronchial sign (13.9%, 6.2%) and pleural effusion (16.7%, 37.5%) in patients with COVID-19 and other viral pneumonia were significantly lower than those in patients with bacterial pneumonia (62.0%, 32.0%, 60.0%, all P < 0.05), paving stone sign (22.2%, 37.5%), fine mesh sign (38.9%, 31.2%), halo sign(11.1%, 25.0%), ground-glass opacity with interlobular septal thickening (30.6%, 37.5%), bilateral patchy pattern/rope shadow (80.6%, 50.0%) etc. were significantly higher than those of bacterial pneumonia (2.0%, 4.0%, 2.0%, 0%, 22.0%, all P < 0.05). The incidence of local patchy shadow in patients with COVID-19 was only 8.3%, significantly lower than that in patients with other viral pneumonia and bacterial pneumonia (8.3% vs. 68.8%, 50.0%, P < 0.05). There was no significant difference in the incidence of peripheral vascular shadow thickening in patients with COVID-19, other viral pneumonia and bacterial pneumonia (27.8%, 12.5%, 30.0%, P > 0.05). Conclusions:The probability of ground-glass opacity, paving stone and grid shadow in chest CT of patients with COVID-19 was significantly higher than those of bacterial pneumonia, and it was more common in the lower lungs and lateral dorsal segment. In other patients with viral pneumonia, ground-glass opacity was distributed in both upper and lower lungs. Bacterial pneumonia is usually characterized by single lung consolidation, distributed in lobules or large lobes and accompanied by pleural effusion.
4.Development of Urban empty-nesters Home Safety Risk Factor Assessment Scale

Objective:To test the reliability and validity of Urban empty-nesters Home Safety Risk Factor Assessment Scale (Uen-HAS), in order to provide an instrument for assessing the safety risk factors of urban empty-nesters.Methods:Totally 530 urban empty-nesters from Fuzhou City, Fujian Province were employed by convenient sampling method for household survey, from August to October 2018. Item analysis and validity and reliability test were conducted.Results:The final version of urban empty nest elderly home safety risk factor assessment scale (Uen-HAS) contains 5 scales, 12 sub-scales and 55 items. The Cronbach α coefficient of the scale was 0.896. The content validity index (S-CVI) of the total scale was 0.881. The correlations of the scores between each item and the total scale ranged from 0.509 to 0.812 with statistical significance ( P<0.05). Conclusion:The scale has good reliability and validity. It could be an instrument to access safety risk factors of urban empty nesters.
5.Effect of Increasing Efficacy and Decreasing Toxicity of Limax Extract on Cyclophosphamide in the Treatment of Hepatocellular Carcinoma

OBJECTIVE:To investigate the effect of increasing efficacy and decreasing toxicity of Limax extract (LE)on cyclophosphamide(CTX)in the treatment of hepatocellular carcinoma. METHODS :The mice were randomly divided into normal group,model group ,CTX group (0.02 g/kg),LE low-dose ,medium-dose and high-dose groups (LEL,LEM,LEH group ,0.6,1.2,2.4 g/kg),CTX+LE low-dose ,medium-dose and high-dose combination groups (CTX+LEL,CTX+LEM,CTX+ LEH group ,the same dose as single drug group ),with 10 huangrenbin518@163.com mice in each group. Except for normal group ,other groups were inoculated with hepatoma cells H 22 in the left ar mpit to establish tumor bearing models. After 24 h of inoculation ,normal group and model group were intragastrically given normal saline , and administration groups were intragastrically given corresponding drugs ,once a day ,for 10 days. On the second day after the last administration ,the general conditions of mice in each group were observed ;the body mass ,thymus index (LI),spleen index (SI)were measured ;the tumor inhibition rate was detected. The effect (q)of combination therapy was evaluated by King ’s formula . The counts of WBC ,RBC and PLT ,serum contents of ALT ,ALT,Scr and BUN were detected in model group ,CTX group and combination groups ,and the contents of MDA,SOD and GSH ,the levels of VEGF ,TNF-α and IL-6 in the tumor tissue were detected by colorimetry and ELISA in above groups. The protein expression of oncogenes (p53,Bcl-2 and Bax )were detected by immunohistochemical method in model group,CTX group and CTX+LEM group. RESULTS :The mice in the model group were in poor spirit and had symptoms of excessive drinking and eating ;although the body weight ,TI and SI were not significantly abnormal compared with normal group (P>0.05),WBC count and AST content were significantly increased ,ALT and BUN contents were significantly decreased (P< 0.05 or P<0.01). Compared with model group ,above symptoms of mice were all improved in administration groups. The tumor weight of administration groups ,TI and SI of CTX group and TI of combination groups were decreased significantly ,but tumor weight of LEL group and LEH group ,TI and SI of LE single groups and combination groups were significantly higher than CTX group;tumor weight of combination groups were significantly lower than CTX group (P<0.05 or P<0.01). The tumor inhibition rates of administration groups were 29.58%-72.08%. The q values of CTX+LEL group ,CTX+LEM group and CTX+LEH group were 1.03,0.97 and 0.86,respectively. Compared with model group ,WBC count ,AST and BUN contents of CTX group ,MDA contents of combination groups ,VEGF,TNF-α and IL-6 levels of administration groups ,the protein expression of Bcl- 2 in CTX group and CTX+LEM group were decreased significantly ;the activities of SOD and GSH of administration groups ,the protein expression of p 53 in CTX+LEM group and Bax in CTX group ,CTX+LEM group were increased significantly (P<0.05 or P< 0.01);WBC counts and AST contents of administration groups ,ALT content of CTX+LEM group ,SOD activity of CTX+LEH group and GSH activity of CTX+LEM group were all significantly higher than those of CTX group ;MDA content of CTX+LEH group,VEGF and TNF-α levels of CTX+LEM group and CTX+LEH group,IL-6 levels of administration groups were all significantly lower than CTX group (P<0.05 or P<0.01). CONCLUSIONS :LE combined with CTX can increase the anti-tumor effect,and LE can reduce the toxicity of CTX induced immunosuppression and bone marrow suppression in mice ,with effect of increasing efficacy and decreasing toxicity. The effect may be related to antioxidant stress ,inhibition of angiogenesis and secretion of inflammatory factors ,and regulation of apoptosis protein expression.

AIM: To observe the anesthetic effect of nalbuphine used in ultrasound-guided transvaginal oocyte retrieval and its effect on embryo quality and pregnancy outcome. METHODS: Four-hundred patients who underwent ultrasound-guided transvaginal oocyte retrieval were randomly divided into two groups (n=200): nalbuphine group (N group) and control group (C group). The patients were in the bladder lithotomy position. Patients in N group were given nalbuphine 0.1 mg/kg intravenously 2 minutes before induction of anesthesia, patients in C group were given normal saline intravenously, and patients in both groups were induced with propofol 1.5 mg/kg. 7.Effects of Hymagic-4D on sensitive skin: a randomized, parallel, self-controlled clinical study

Objective:To evaluate the efficacy and safety of Hymagic-4D multi-essence (miniHA, acetylated sodium hyaluronate, sodium hyaluronate, sodium hyaluronate cross-linked polymer) on sensitive skin.Methods:The clinical trial was conducted from April 2, 2019 to April 30, 2019 in the Cosmetic Dermatology Laboratory of the Third Affiliated Hospital of Sun Yat-Sen University. A total of 33 subjects (average age of 43±9.5) completed the trial. Subjects diagnosed as sensitive skin according to the criteria were enrolled. Hymagic-4D were randomly applied on one side of the face while macromolecular hyaluronic acid was applied on opposite side for 28 days. Skin biophysical properties, lactic test, clinical efficacy and safety were evaluated by the investigators at day 0, day 7, day 14 and day 28. Skin water content, TEWL and a*value were measured at the same time.Results:Dates showed that at day 7, day 14 and day 28, the a* values on Hymagy-4D using side were 8.54±3.08, 8.87 ±3.21 and 8.39±3.21, lower than that on the side of control 9.48±3.09, 9.51±3.30 and 9.03±2.95 ( P<0.01); skin roughness score, dryness score and erythema score on hymagy-4d using side were significantly lower than before ( P<0.01), and significantly lower than that on the control side ( P<0.01). Hydration, TEWL, pH value, L value and total score of lactic acid tingling test which were significantly improved on both facial skin compared to the baseline ( P<0.05) showed no statistical difference between two facial sides ( P>0.05) during 28-day treatment. Conclusions:This study demonstrates that Hymagic-4D has more effective in repairing skin barrier and inhibiting skin inflammation than single component hyaluronic acid on sensitive skin persons.
8.Optimization of the Extraction Technology of Total Flavonoids from Typhonium divaricatum

OBJECTIVE: To establish a method for the content determination of total flavonoids from Typhonium divaricatum, and to optimize its extraction technology. METHODS: The content of total flavonoids in T. divaricatum was determined by UV spectrophotometry. Using the extraction amount of total flavonoids from T. divaricatum as index, the volume fraction of ethanol, the ratio of material to liquid, the extraction time and times as factors, the extraction technology of total flavonoids from T. divaricatum was optimized by L9(34) orthogonal design, based on the single factor test. RESULTS: The linear range of rutin were 8-48 μg/mL (r=0.999 7); the quantification limit was 0.54 μg/mL, and the detection limit was 0.18 μg/mL; RSDs of precision, stability and repeatability tests were all lower than 2%. The recoveries were 99.61%-102.38%(RSD=1.15%, n=6). The optimal extraction technology was as follows: ethanol concentration of 70%, solid-liquid ratio of 1 ∶ 20 (g/mL), extraction time of 45 min, extracting for 2 times. Under this condition, the average content of total flavonoids from T. divaricatum was 2.74 mg/g. CONCLUSIONS: Established method is simple and accurate; the extraction process is stable and feasible.
9.Correlation between circulating tumor cells and clinicopathological features of early breast cancer.

To investigate the correlation between the number of peripheral blood circulating tumor cells (CTCs) and clinicopathological features of early breast cancer.
Methods: The clinical and pathological data from 100 patients with early breast cancer treated by a breast cancer treatment team in the Department of Breast Surgery, Second Xiangya Hospital, Central South University, were collected from January 2017 to December 2018. For these patients, their peripheral blood CTCs were detected, enumerated and typed by CanpatrolTM CTC assay.
Results: The positive rate of CTCs was 90% in peripheral blood of patients with early breast cancer, and the majority of molecular phenotypes was hybrid CTCs (55.6%). The positive rate of CTCs was only related to the pathological type of tumor (P<0.05), but not to other clinicopathological features. No correlation between clinicopathological features and the total number of CTCs, the number of epithelial CTCs or the number of hybrid CTCs was found. However, the number of mesenchymal CTCs was significantly correlated with the expression of hormone receptors and Ki-67 (r=0.200, P<0.05), and there was a significant correlation between the proportion of mesenchymal CTCs and the expression level of Ki-67 (r=0.213, P<0.05).
Conclusion: The number of CTCs is not correlated with all clinicopathological features, but patients with negative hormone receptor and high expression of Ki-67 probably have more hybrid CTCs.

10.Construction of evaluation index system of home safety of empty nest elderly in community based on Delphi method

Objective To construct the evaluation index system of home safety of empty nest elderly in community, and to provide a reference for the analysis of potential home safety hazards and the formulation of safety intervention measures for the empty nest elderly in the community. Methods From June 2018 to March 2019, based on literature review, Delphi expert consultation method was used to design the Expert Consultation Table for the Index System of home safety of the empty nest elderly in community, which included five first-level indicators (physiological status, home environment, psychological status, social support, safety knowledge, attitude and practice) and 52 second-level indicators. 15 experts in geriatric nursing, geriatrics and injury epidemiology were selected for two rounds of consultation. Results The positive coefficients of experts in the first and second rounds were 83.3% and 100.0%, respectively; the authoritative coefficient of experts was 0.91; Kendall W coefficients in the first and second rounds were 0.246 and 0.356, respectively (P<0.01); after two rounds of inquiry, an evaluation index system of the home safety of empty nest elderly in community was established, which consisted of 5 first-level indicators and 52 second-level indicators. Among them, the mean value of the importance assignment of the first level index was 4.64-4.91, the standard deviation was 0.30-0.67, and the coefficient of variation was 0.06-0.14;the mean value of the importance assignment of the second-level index was 3.55-4.91, the standard deviation was 0.30-0.92, and the coefficient of variation was 0.06-0.20. Conclusions Through Delphi expert consultation method, the evaluation index system of home safety of empty nest elderly in community is scientific and reliable. It plays a strong guiding role in the evaluation of home safety of empty nest elderly.
