1.Application of subjective gravity detection in vestibular system diseases

Subjective visual gravity detection includes subjective visual vertical(SVV)detection and subjec-tive visual horizontal(SVH)detection,which refers to the detection of the angle between patients' perceived gravity vertical line and gravity horizontal line and the actual gravity vertical line and horizontal line when people exclude vis-ual reference and in dark environment.The results are used to evaluate the function of the utricle of the subjects,and then provide the basis for diagnosing vestibular system diseases.With the continuous improvement of SVV &SVH examination technology and the understanding of the mechanism,SVV & SVH examination has been widely applied to clinical work.At present,there are some progresses of srvesvit SVV & SVH in the diagnosis,evaluation and treatment of diseases of the vestibular system such as vestibular neuronitis,Meniere disease and benign paroxys-mal positional vertigo(BPPV),and this article reviews these progresses.

3.Analysis of pathogen distribution and epidemic characteristics of 233 influenza virus negative influenza-like cases in Yantai

Objective:To understand the spectrum of pathogens and epidemic characteristics of respiratory infectious diseases in influenza virus-negative influenza-like cases in Yantai, and provide reference for disease prevention and control and clinical diagnosis and treatment.Methods:From March 2020 to February 2021, nasopharyngeal swab samples of 233 influenza virus-negative influenza like cases were collected in all sentinel hospitals monitored by Yantai National Influenza Network Laboratory, and 22 respiratory pathogens were detected by multiple fluorescence quantitative polymerase chain reaction to analyze epidemiological characteristics.Results:The total pathogen detection rate of 233 samples was 69.96% (163/233). A total of 17 respiratory pathogens were detected. The top three pathogens were human coronavirus (HCoV, 32.62%), rhinovirus/enterovirus (RhV/EV, 17.17%) and Legionella pneumophila (LP, 16.74%). The detection rates in different age groups were 80.28% (57/71) in the 0-15 years old group, 62.65% (52/83) in the 16-30 years old group, 68.18% (30/44) in the 31-45 years old group, 64.28% (9/14) in the 46-60 years old group, and 71.43% (15/21) in the >60 years old group. There was no significant difference among the groups. Respiratory pathogens were detected throughout the year, mainly in a single pathogen carrying mode (44.21%), and there was no significant difference in the physical examination rate of respiratory pathogens in different seasons. The seasonal prevalence of various pathogens was different, and the detection rate of HCoV 229E was the highest in spring (68.75%); the detection rate of rhinovirus/enterovirus was higher in autumn (26.98%) and winter (23.08%); the detection rate of LP was high in spring (19.05%) and summer (27.27%); the detection rate of human parainfluenza virus (HPIV) in spring (22.22%) was significantly higher than that in summer (3.64%). The number of HPIV and Bordetella pertussis (Bp) detected in the 0-15 year old group was the highest, and the detection rate was statistically significant among different age groups. Conclusions:The continuous monitoring of respiratory pathogens such as HCoV, RhV, EV, LP, HPIV should be strengthened to understand their epidemiologic characteristics and the standardization of pathogenicity, which provides data support and reference for epidemiological investigation of outbreaks that may be caused by other pathogens.
4.Analysis of the serum antibody test results and clinical diagnostic value in patients with novel coronavirus pneumonia

Objective:To provide reference for clinical case diagnosis and treatment applications using descriptive analysis of patient-specific IgM and IgG test result of novel coronavirus pneumonia (COVID-19).Methods:Chemical luminescence was used to detect the levels of 423 confirmed or suspected cases IgM and IgG antibody, and the test result of patients with different clinical characteristics were compared and analyzed.Results:The positive rates of confirmed cases of COVID-19 were 80.4% (314/388) and 98.2% (381/388), for IgM and IgG respectively, and the positive rates of COVID-19 suspected cases specific IgM and IgG were 0.0% (0/24) and 45.8% (11/24), respectively. In patients at 6 weeks or more after the onset of the disease, the positive rate of IgG antibody was 100%. The level of IgG titer was generally higher in cases of 5~8 weeks after onset (mean: 112.70 AU/ml) than in cases of 1-4 weeks after onset (mean: 85.01 AU/ml) (U=8 531, P<0.0001). The level of IgG titer was higher in severe type cases than that of patients with ordinary type illness, with an average of 137.61 AU/ml. Conclusions:Specific IgM and IgG antibodies have strong application value in COVID-19 case diagnosis, and it is recommended to strengthen the tracking and monitoring of IgM and IgG titer levels in patients.
5.Value of combined utilization of urinary CysC and NAG in the diagnosis of renal tubules damage

Objective To discuss the diagnostic value of combined utilization of urinary cystatin C (CysC) and β‐N‐acetyl‐glu‐cosaminidase (NAG) in renal tubules injury .Methods Urine samples were collected from 130 cases of patients with renal tubular damage (case group) and 100 cases of health individuals (normal control group) ,respectively .The concentration of CysC and the activity of NAG in urine were detected .Results The sensitivity ,specificity ,and positive predictive value of combined utilization of CysC and NAG in diagnosis of renal tubules damage were 85 .4% ,89 .6% and 82 .1% ,respectively .The concentration of CysC and the activity of NAG in case group after treatment were both significantly different from those before treatment (P<0 .05) .Conclu‐sion Combined utilization of urinary CysC and NAG has a significant value for the diagnosis and the therapeutic monitoring in re‐nal tubules injury .
6.Preparation of PEI-RGD/~(125)I-(α_v)ASODN and its inhibitory effect on invasive ability of HepG2 cells

Objective:To study the effects of ~(125)I-(α_v)ASODN on the in vitro invasive ability of heptocellular carcino-ma cell line(HepG2) through PEI-RGD-mediated receptor process. Methods: Intergrin α_v-specific antisense oligonucle-otide was labeled with ~(125)I, and PEI-RGD/~(125)I-(α_v)ASODN complex was prepared by combining ~(125)I-(α_v)ASODN with polyethyleneimine derivative PEI-RGD. PEI-RGD/~(125)I-(α_v)ASODN complex was transferred into HepG2 cells through the receptor-mediated process. The effect of PEI-RGD/~(125)I-(α_v)ASODN complex on the invasive ability of HepG2 cells was examined by Boyden chamber invasive assay. Results: (1) The labeling yield and radiochemical purity of ~(125)I-(α_v) ASODN were(73.78±4.09)% and(96.68±1.38)%, respectively, and the labeled compound had a good stability in vitro after 48 h at 37℃; (2) The ability of HepG2 cells to uptake PEI-RGD/~(125)I-(α_v)ASODN reached its peek ([12.77±0.85] % ) when PEI-RGD/~(125)I-(α_v)ASODN was at 4 μl/2 μg ([12.77±0.85] %), and then gredually decreased thereafter. So the dosage of PEI-RGD/~(125)I-(α_v)ASODN for the following experiment was chosen as 2 μl/1 μg; (3) The invasive capacity of HepG2 cells was significantly reduced in PEI-RGD/~(125)I-(α_v)ASODN group compared with those in other experiment and control groups (P <0.01 ). Conclusion: ~(125)I-(α_v)ASODN mediated by PEI-RGD can effectively inhibit the invasive capacity of HepG2 cells.
