1.Analysis of the Therapeutic Effect of Triple Antibiotic Paste Combined with Pulp Revascularization on Pulp Necrosis of Permanent Teeth in Children

Objective:Exploring the therapeutic effect of triple antibiotic paste combined with pulp revascularization on pediatric permanent tooth pulp necrosis.Methods:A retrospective study was conducted on 90 young children with permanent tooth pulp necrosis treated from October 2019 to October 2022.Based on different treatment methods,the children were divided into two groups,namely the traditional group and the experimental group,with 45 cases in each group.All patients were treated with pulp revascularization.The traditional group used conventional calcium hydroxide paste for root canal disinfection during the operation,while the experimental group used triple antibiotic paste.Two groups of patients were followed up for 2 years.After 6 months of treatment,the clinical efficacy,changes in gingival crevicular fluid growth factors and inflammatory factors before and after treatment were compared between the two groups.The growth and development of tooth roots were also compared between the two groups after 2 years of treatment,as well as the pulp activity at 6 months,1 year,and 2 years after treatment.The incidence of complications during the follow-up period was also compared between the two groups.Results:The total effective rate of the combined group was 82.22%,which was higher than the traditional group's 66.67%(P<0.05);There was no significant difference in the levels of growth factor and inflammatory factor in the gingival crevicular fluid between the two groups before treatment(P>0.05).After a 6-month follow-up,the levels of VEGF and bFGF in the gingival crevicular fluid of both groups increased,with the combined group being higher than the traditional group(P<0.05).The inflammatory factors interleukin-8 and IL-6 in the gingival crevicular fluid of both groups decreased,while the combined group was lower than the traditional group(P<0.05);Before treatment,there was no significant difference in the thickness of the root canal wall,crown to root ratio,and apical foramen diameter between the two groups(P>0.05).After a 2-year follow-up,the levels of root canal wall thickness and crown to root ratio increased in both groups,with the combined group being higher than the traditional group(P<0.05).The apical foramen diameter decreased in both groups,with the combined group being lower than the traditional group(P<0.05).After treatment,the deposition rate of dental pulp bone like tissue in the combined group was lower than that in the traditional group(P<0.05);Follow up for 6 months,1 year,and 2 years revealed that the positive rate of pulp vitality in the combined group was higher than that in the traditional group(P<0.05);There was no significant difference in the incidence of complications between the two groups(P>0.05).Conclusion:The application of triple antibiotic paste in pulp revascularization surgery can improve the clinical efficacy of pediatric permanent tooth pulp necrosis,promote root growth and development,enhance pulp activity,and may be related to improving periodontal tissue related growth factors and stronger antibacterial and anti-inflammatory effects in children.Moreover,this therapy has high safety.
2.Application Effect of Concentrated Growth Factor Combined with Dual Antibacterial Drug Paste in Young Permanent Teeth Children with Pulp Necrosis

Objective:To observe the application effect of concentrated growth factor combined with dual antibacterial drug paste in young permanent teeth children with pulp necrosis.Methods:This study was a prospective study,79 young permanent teeth children with pulp necrosis who were admitted to our hospital from May 2022 to May 2023 were divided into control group(treated with conventional pulp revascularization combined with dual antibacterial drug paste,n=39 cases)and study group(treated with concentrated growth factor pulp revascularization combined with dual antibacterial drug paste,n=40 cases)according to the random number table method.Follow up for 12 months,the clinical efficacy,pain visual analogue scale(VAS),cementoid tissue deposition rate,root canal wall thickness,root length,bite force,chewing efficiency and satisfaction of the children's families were compared.Result:The total clinical effective rate in the study group was higher than that in the control group(P<0.05).After 6 months and 12 months of treatment,the pain VAS scores decreased and cementoid tissue deposition rate increased between two groups,and the pain VAS scores in study group were lower than those in control group,the cementoid tissue deposition rate was higher than that in the control group(P<0.05).After 6 months and 12 months of treatment,the root canal wall thickness and root length increased between two groups,and the root canal wall thickness and root length in the study group were higher than those in the control group(P<0.05).After 6 months and 12 months of treatment,the bite force and chewing efficiency increased between two groups,and the inbite force and chewing efficiency in the study group were higher than those in control group(P<0.05).The total satisfaction rate in study group was higher than that in control group(P<0.05).Conclusion:Concentrated growth factor combined with dual antibacterial drug paste in young permanent teeth children with pulp necrosis can increase the total clinical efficacy,significantly increase cementoid tissue deposition rate and chewing ability,promote the regeneration of dental tissue,relieve pain,and the satisfaction of the children's families is relatively high.

5.Protective effects of pueraria isoflavones on myocardial injury in ovariectomized rats via LKB1/AMPK/PGC-1α signaling pathway

AIM To study the protective effects and mechanism of pueraria isoflavones on myocardial injury in ovariectomized rats.METHODS Thirty-six rats were randomly divided into the sham operation group,the model group,the estradiol valerate group(0.1 mg/kg)and the low,medium and high dose pueraria isoflavones groups(55,110,220 mg/kg).In contrast to the rats of the sham operation group having their small pieces of adipose tissue removal around the ovaries,rats of the other groups had their bilateral ovaries excised,followed by the 16-week corresponding oral drug administration 2 weeks later at a once daily frequency for,6 days a week.At the end of the 16th week,the rats had their hemodynamics[systolic pressure(SBP),diastolic pressure(DBP),mean pressure(MBP),left ventricular systolic pressure(LVSP),left ventricular diastolic pressure(LVMP),and the maximum rate of increase and decrease of left ventricular pressure during isovolumic contraction(±dp/dtmax)]detected by PowerLab;their cardiac pathological changes observed by HE staining;their levels of creatine kinase(CK),lactate dehydrogenase(LDH),total cholesterol(TC),triglyceride(TG),high density lipoprotein cholesterol(HDL-C),low density lipoprotein cholesterol(LDL-C)and glucose(Glu)in plasma detected by biochemical analyzer;their myocardial level of adenosine triphosphate(ATP)detected by colorimetry;their mRNA expressions of glucose transporter 4(GLUT4),lactate dehydrogenase A(LDHA),carnitine palmitoyl transferase-1(CPT-1α),acyl coenzyme A carboxylase(ACC),liver kinase B1(LKB1),adenylate-activated protein kinase(AMPK)and peroxisome proliferator-activated receptor γ coactivator factor 1α(PGC-1α)detected by RT-qPCR;and their myocardial expressions of energy metabolism related proteins LKB1,p-AMPK/AMPK and PGC-1α detected by Western blot.RESULTS Compared with the model group,the pueraria isoflavones groups displayed decreased levels of SBP,DBP,MBP,LVSP,LVMP(P<0.05,P<0.01);increased-dp/dtmax(P<0.05,P<0.01);improved myocardial fibrinolysis,gap widening and inflammatory infiltration caused by ovariectomy;decreased activities of LDH and CK(P<0.05);increased myocardial ATP level(P<0.05,P<0.01);decreased levels of TC,TG,LDL-C and Glu(P<0.05,P<0.01);increased HDL-C level(P<0.05,P<0.01);increased myocardial mRNA expressions of GLUT4,LDHA,CPT-1α,ACC,LKB1,AMPK and PGC-1α(P<0.05,P<0.01);and increased protein expressions of myocardial LKB1,p-AMPK/AMPK and PGC-1α(P<0.05,P<0.01).CONCLUSION Pueraria isoflavones are protective to myocardial injury in ovariectomized rats,and the mechanism may lie in the improvement of energy metabolism-related myocardial proteins via LKB1/AMPK/PGC-1α signaling pathway.
6.Effects of Dianxianqing Granules on Tau protein in a mouse model of Alzheimer' s disease via NLRP3/Caspase-1 pathway

AIM To study the effects of Dianxianqing Granules on Tau protein in a mouse model of Alzheimer's disease (AD).METHODS The mice expressing P301S mutant Tau variant were randomly divided into the model group,the MCC950 group (NLRP3 inhibitor,10 mg/kg),the Dianxianqing Granules group (12.48 g/kg),the MCC950+Dianxianqing Granules group,in contrast to the C57BL/6 mice of the control group.After 5 months of administration,the mice had their learning and memory ability tested by Y maze test and Morris water maze test;their cerebral morphological changes observed by HE staining;their cerebral expressions of Caspase-1 and GSDMD proteins detected by immunohistochemical method;their expression of cerebral Tau protein detected by immunofluorescence;and their cerebral expressions of Tau,p-Tau (ser202),p-Tau (thr205),NLRP3,Caspase-1,IL-1β and IL-18 detected by Western blot.RESULTS Compared with the control group,the model group displayed decreased rate of spontaneous alternate reaction and times of crossing platform (P<0.05,P<0.01);abnormal hippocampal morphology,decreased number of neurons,increased cerebral positive expressions of Caspase-1 and GSDMD (P<0.05);deposition of a large number of brown granules in cytoplasm,and increased protein expressions of Tau,p-Tau (ser202),p-Tau (thr205),NLRP3,Caspase-1,IL-1βand IL-18 in the hippocampus and the cortex (P<0.05,P<0.01).Compared with the model group,the group intervened with Dianxianqing Granules demonstrated both increased rate of spontaneous alternate reaction and times of crossing platform (P<0.05);complete and normal morphology of the brain,a diversity of fine neurons,reduced cerebral positive expressions of Caspase-1 and GSDMD (P<0.05);and decreased protein expressions of Tau,p-Tau (ser202),p-Tau (thr205),NLRP3,Caspase-1,IL-1β and IL-18 in the hippocampus and the cortex (P<0.05,P<0.01).CONCLUSION Dianxianqing Granules may inhibit Tau protein expression in the mouse model of AD via NLRP3/Caspase-1 pathway.

The rare-earth-elements-doped upconversion nanoparticles NaYF4:Yb3+,Er3+were synthesized by solvothermal method,and NaYF4:Yb3+,Er3+@SiO2 were prepared by coating SiO2 on the surface of NaYF4:Yb3+,Er3+by inverse microemulsion method in this work.Based on the fluorescence quenching principle between NaYF4∶Yb3+,Er3+@SiO2 and SQA-Fe3+,a NaYF4∶Yb3+,Er3+@SiO2-SQA-Fe3+fluorescence nanosensor was constructed for detection of trace hydrogen peroxide(H2O2).Under optimal conditions,the linear range of this method for detecting H2O2 was 1.8?84.0 μmol/L,with detection limit(3σ)of 0.47 μmol/L.The recoveries of H2O2 spiked in milk were 98.4%?99.7%.This method could be used for detection of H2O2 residue in milk samples,with advantages such as low detection limit,good stability and strong anti-interference ability.
8.Development and validation of a stromal-immune signature to predict prognosis in intrahepatic cholangiocarcinoma

Background:
Intrahepatic cholangiocarcinoma (ICC) is a highly desmoplastic tumor with poor prognosis even after curative resection. We investigated the associations between the composition of the ICC stroma and immune cell infiltration and aimed to develop a stromal-immune signature to predict prognosis in surgically treated ICC.
Patients and methods:
We recruited 359 ICC patients and performed immunohistochemistry to detect α-smooth muscle actin (α-SMA), CD3, CD4, CD8, Foxp3, CD68, and CD66b. Aniline was used to stain collagen deposition. Survival analyses were performed to detect prognostic values of these markers. Recursive partitioning for a discrete-time survival tree was applied to define a stromal-immune signature with distinct prognostic value. We delineated an integrated stromal-immune signature based on immune cell subpopulations and stromal composition to distinguish subgroups with different recurrence-free survival (RFS) and overall survival (OS) time.
Results:
We defined four major patterns of ICC stroma composition according to the distributions of α-SMA and collagen: dormant (α-SMAlow/collagenhigh), fibrogenic (α-SMAhigh/collagenhigh), inert (α-SMAlow/collagenlow), and fibrolytic (α-SMAhigh/collagenlow). The stroma types were characterized by distinct patterns of infiltration by immune cells. We divided patients into six classes. Class I, characterized by high CD8 expression and dormant stroma, displayed the longest RFS and OS, whereas Class VI, characterized by low CD8 expression and high CD66b expression, displayed the shortest RFS and OS. The integrated stromal-immune signature was consolidated in a validation cohort.
Conclusion
We developed and validated a stromal-immune signature to predict prognosis in surgically treated ICC. These findings provide new insights into the stromal-immune response to ICC.
