TCP family as plant specific transcription factor, plays an important role in different aspects of plant development. In order to screen TCP family members in tobacco, the homologous sequences of tobacco and Arabidopsis TCP family were identified by genome-wide homologous alignment. The physicochemical properties, phylogenetic relationships and cis-acting elements were analyzed by bioinformatics. The homologous genes of AtTCP3/AtTCP4 were screened, and RT-qPCR was used to detect the changes of gene expression upon 20% PEG6000 treatment. The results show that tobacco contains 63 TCP family members. Their amino acid sequence length ranged from 89 aa to 596 aa, and their protein hydropathicity grand average of hydropathicity (GRAVY) ranged from -1.147 to 0.125. The isoelectric point (pI) ranges from 4.42 to 9.94, the number of introns is 0 to 3, and the subcellular location is all located in the nucleus. The results of conserved domain and phylogenetic relationship analysis showed that the tobacco TCP family can be divided into PCF, CIN and CYC/TB1 subfamilies, and each subfamily has a stable sequence. The results of cis-acting elements in gene promoter region showed that TCP family genes contain low docile acting elements (LTR) and a variety of stress and metabolic regulation related elements (MYB, MYC). Analysis of gene expression patterns showed that AtTCP3/AtTCP4 homologous genes (NtTCP6, NtTCP28, NtTCP30, NtTCP33, NtTCP42, NtTCP57, NtTCP63) accounted for 20% PEG6000 treatment significantly up-regulated/down-regulated expression, and NtTCP30 and NtTCP57 genes were selected as candidate genes in response to drought. The results of this study analyzed the TCP family in the tobacco genome and provided candidate genes for the study of drought-resistance gene function and variety breeding in tobacco.

Objective To investigate the relationships of nutritional status and length of stay (LOS) with phase angle (PA) in patients with severe craniocerebral injury. Methods A total of 100 patients [Glasgow Coma Scale (GCS) score ≤8 points] admitted outside the department of neurology were selected as study objects. Body composition analysis and blood samples were used to determine the intracellular and extracellular water content, skeletal muscle, PA, prognostic nutritional index (PNI) and LOS. Patients were divided into low PA group (n=42) and normal PA group (n=58) based on PA values. Body composition related indexes, PNI and LOS were compared between the two groups. The correlation between PA and each index was analyzed. The influencing factors of malnutrition were analyzed. The influencing factors of malnutrition were analyzed. Results Intracellular water, total body water, body cell content and PNI in the low PA group were significantly lower, and LOS was significantly longer than that in the normal PA group (P < 0.05). PA was positively correlated with intracellular water, extracellular water, total body water, skeletal muscle, body cell content, bone mineral content, basal metabolic rate and PNI (P < 0.05), while PA was negatively correlated with LOS (P < 0.05). PA (OR=5.441, P=0.001, 95%CI, 2.011 to 14.719) and LOS (OR=8.373, P < 0.001, 95%CI, 3.079 to 22.765) were the influential factors in the occurrence of malnutrition. Conclusion PA is significantly correlated with nutritional status and LOS in patients with severe craniocerebral injury.

Objective To examine the clinical distribution of primary intracranial tumors and analyze the risk factors for postoperative complications.Methods From January 2018 to December 2022,the clinical data of 961 patients with primary intracranial tumor in the Department of Neurosurgery of Tiantan Hospital in Beijing were collected and analyzed retrospectively.To examine the clinical distribution of patients with primary intracranial tumor and present the incidence of postoperative complications.To compare the basic data of patients with and without postoperative complications,and analyze the risk factors leading to postoperative complications.Results There were 363 cases of glioma,231 cases of meningioma,158 cases of sellar tumors,142 cases of neurilemmoma and 67 cases of other types of tumors.There were 679 cases of supratentorial tumors and 282 cases of infratentorial tumors.Postoperative complications occurred in 279 patients,and the incidence of postoperative complications was 29.03％.The incidences of intracranial infection,pulmonary infection,hyponatremia,lower extremity venous thrombosis and central nervous system infection were 9.37％,5.41％,4.99％,4.47％and 4.27％,respectively.After surgery,D-dimer(D-D),fibrin degradation products(FPD),prothrombin time(PT)in patients with primary intracranial tumors were significantly higher than those in patients without primary intracranial tumors(P<0.05).Activation of partial thromboplastin time(APTT),and thrombin time(TT)levels were significantly higher than those before surgery(P<0.05).Fibrinogen(FIB)was significantly lower than that before surgery(P<0.05).There were significant differences in tumor location,intraoperative blood loss,operation time,anesthesia recovery time and postoperative coagulation function between patients with and without postoperative complications(P<0.05).Conclusions The common types of primary intracranial tumors include gliomas,meningiomas,sellar tumors and neurilemmoma,etc.Surgical treatment has a high risk of postoperative complications.Common postoperative complications include intracranial infection,pulmonary infection,hyponatremia,lower extremity venous thrombosis,and central nervous system infection.Tumor location,operation duration,intraoperative blood loss,anesthesia recovery time and postoperative coagulation dysfunction are all risk factors for postoperative complications.

OBJECTIVE: To explore the genetic basis for a child with metachromatic leukodystrophy (MLD). METHODS: Clinical data of the patient was collected. Genomic DNA was extracted from peripheral blood samples of the child and his family members. Potential variant was screened by whole exome sequencing (WES), and candidate variant was verified by Sanger sequencing. The pathogenicity the variant was analyzed by multiple sequence alignment of the amino acid sequence and three-dimensional model prediction of its protein product. RESULTS: The child was found to harbor compound heterozygous variants c.257G>A (p.R86Q) and c.467del (p.G156Afs*6) of the ARSA gene, among which the c.467del (p.G156Afs*6) frameshift variation was unreported previously. Multiple sequence alignment showed that the site of the c.257G>A (p.R86Q) missense variant is highly conserved. Three-dimensional structure modeling analysis showed that the partial deletion due to the p.G156Afs*6 variant may cause significant alteration of the structure of ARSA protein. CONCLUSION The discovery of novel variant in ARSA has enriched the mutational spectrum of MLD and may facilitate the understanding of the genotype-phenotype correlation of MLD.

Objective:To analyze the correlation between recessive obesity and lifestyle in physical examination population.Methods:A total of 1 026 people with body mass index (BMI) of 18.5<-24.9 kg/m 2 who completed physical examination in the Health Management Center of Affiliated Hospital of Qingdao University from March 1, 2019 to September 31, 2019 were included in this study. Inbody770 body composition analyzer was used to check the percent body fat, and the subjects were divided into recessive obesity group (405 cases) and control group (621 cases) with the results. The results of routine physical examination were compared between the two groups. The data of five dimensions including marital status, education level, income, occupation and dietary habits were obtained with lifestyle questionnaire. The correlation between recessive obesity and lifestyle was analyzed by binary logistic regression. Results:The proportion of women, BMI, total cholesterol, low density lipoprotein cholesterol and waist-hip ratio were significantly higher in the recessive obesity group than those in the control group [65.2% vs 27.1%, (21.83±1.63) vs (21.56±1.74) kg/m2, 4.68 (4.18, 4.22) vs 4.39 (4.13, 4.83) mmol/L, 2.54 (2.08, 3.00) vs 2.24 (2.13, 2.78) mmol/L, 0.87 (0.84, 0.90) vs 0.82 (0.80, 0.86)]; while the systolic blood pressure, albumin and aspartate aminotransferase were significantly lower [(114.99±11.49) vs (118.97±11.84) mmHg (1 mmHg=0.133 kPa), (45.13±2.83) vs (46.37±2.60) g/L, 15 (12, 18) vs 16 (14, 20) U/L] (all P<0.05). Unmarried ( OR=0.200, 95% CI: 0.123-0.325), eating less white meat [occasionally eating, OR=0.565, 95% CI: 0.304-1.053; rarely eating, OR=0.186, 95% CI: 0.094-0.368], eating less spicy food (occasionally eating, OR=0.298, 95% CI: 0.171-0.519; rarely eating, OR=0.828, 95% CI: 0.487-1.408), drinking more water (1 000-2 000 ml/d, OR=0.366, 95%CI: 0.218-0.615; ≥2 000 ml/d, OR=0.176, 95% CI: 0.087-0.356) were negatively correlated with the occurrence of recessive obesity (all P<0.05). Eating takeout food frequently ( OR=4.639, 95% CI: 2.412-8.923), eating too much edible oil ( OR=10.900, 95% CI: 4.376-27.148), drinking beer ( OR=3.702, 95% CI: 2.290-5.982) and infrequent physical exercise (occasional, OR=13.417, 95% CI: 6.907-26.066; rarely, OR=28.290, 95% CI: 13.532-59.142) were positively correlated with the occurrence of recessive obesity (all P<0.05). Conclusions:There is a correlation between recessive obesity and the lifestyle in physical examination population. Attention should be paid to control the intake of white meat, spicy food, edible oil and beer, ensure the amount of drinking water, reduce the frequency of takeout food, and increase physical exercise to prevent recessive obesity.

Objective:To investigate the short-term transplantation effect of different vitrification sollutions in order to optimize the cryopreservation protocol on human ovarian tissue.Methods:Ovarian tissues came from 4 patients who were treated surgically due to gynecological diseases in Peking University Shenzhen Hospital from March 2018 to December 2019. The size of 5-10 mm×10 mm×1 mm ovarian tissues were distributed to fresh control group and four experimental groups (G1, G2, G3 and G4). After equilibrium and thawing, the frozen ovarian tissues were grafted into chorioallantoic membrane (CAM). The transplanted ovarian tissues were collected 5 d after transplantation. The morphology of follicles was assessed by HE, the expression of CD105 protein was detected by immunohistochemistry and the microvascular density was recorded. The expression of Bcl-2 and Bax proteins was detected by Western blotting.Results:There was a significantly difference in the percentage of morphologically normal primordial follicles (MNPF) among G1 [63.2% (24/38)], G2 [68.3% (28/41)], G3 [61.9% (26/42)], G4 [69.6% (32/46)] and fresh control groups [82.8% (82/99), P=0.044]. Compared with fresh control group, the percentages of MNPF were statistically lower in the treatment groups (all P<0.05). The CD105-microvascular density (MVD) in fresh control group, G1, G2, G3 and G4 groups were 6.51±1.30/mm 2, 11.10±1.62/mm 2, 13.04±1.84/mm 2, 9.11±1.09/mm 2, 11.28±1.62/mm 2, respectively. The CD105-MVD in four treatment groups after transplantation was significantly higher than that of fresh control group ( P<0.001, P<0.001, P=0.022, P<0.001), and the CD105-MVD of G1, G2 and G4 groups was higher than that of G3 group ( P=0.024, P<0.001, P=0.034). The ratio of Bcl-2/Bax in G1, G2, G3, G4 groups and fresh control group were 0.71±0.37, 0.84±0.29, 0.45±0.18, 0.84±0.29 and 0.44±0.21, respectively. The ratio of Bcl-2/Bax of G2 group was statistically higher than that of fresh control group and G3 group ( P=0.025, P=0.038). Conclusion:After 5 d of transplantation into CAM, the percentage of normal primordial follicles is lower. Acorrding to the higher vascularization and anti-apoptotic ability, G2 and G4 are the optimal vitrification protocol for human ovarian tissue cryopreservation.

Objective To standardize an expert consensus on pulmonary rehabilitation nursing of chronic respiratory diseases. Methods On the basis of literature review, 5 experts were interviewed and 19 experts were consulted to establish the primary and secondary themes. 10 nursing experts in this field were invited to conduct peer review on the draft consensus to modify and improve the consensus content. Results After expert consultations，14 first-level themes and 40 second-level themes were established. The value of Ca (judgment coefficient),Cs (familiarity coefficient) and Cr (authority coefficient) were 0.895,0.839 and 0.867 ,which indicated the expert consultation have high authority and credibility. The Kendall coefficients of first-round expert consultation were 0.121 and 0.151, and they were 0.205 and 0.149 in the second round (P<0.05).In the first-round expert consultation, the coefficients of variation of the first and second themes were 0.197 and 0.200,and in the second round ,they were 0.202 and 0.237. They were all less than 0.25 , indicating that the expert's judgment results were relatively consistent. Conclusion As this consensus was developed based on 19 clinical nursing professionals from across the country, it is scientific and authoritative. This consensus can not only benefit to clinical nursing practice, but also lay the foundation for the development of guideline, and it still needs further theoretical and empirical research verification.

OBJECTIVE: To detect potential mutations of the PKHD1 gene in two pedigrees affected with infantile polycystic kidney disease. METHODS: Clinical data and peripheral venous blood samples were collected from the probands and their parents as well as fetal amniotic fluid cells. Genome DNA was extracted from the peripheral blood samples and amniotic fluid cells. Exons 32 and 61 of the PKHD1 gene were amplified with PCR and subjected to direct sequencing. RESULTS: The proband of pedigree 1 was found to carry c.4274T>G (p.Leu1425Arg) mutation in exon 32 and c.10445G>C (p.Arg3482Pro) mutation in exon 61 of the PKHD1 gene, which were inherited from her father and mother, respectively. The fetus has carried the c.4274T>G (p.Leu1425Arg) mutation. In pedigree 2, the wife and her husband had respectively carried a heterozygous c.5979_5981delTGG mutation and a c.9455delA mutation of the PKHD1 gene. No chromosomal aberration was found in the umbilical blood sample, but the genetic testing of their fetus was failed. Based on software prediction, all of the 4 mutations were predicted to be pathogenic. CONCLUSION PKHD1 c.4274T>G (p.Leu1425Arg), c.10445G>C (p.Arg3482Pro), c.5979_5981delTGG and c.9455delA were likely to be pathogenic mutations. The results have facilitated genetic counseling and prenatal diagnosis for the two pedigrees.

Objective? To explore the effects of family participation in the respiratory rehabilitation of the patients with chronic obstructive pulmonary disease (COPD) in terms of their treatment compliance and outcomes. Methods? Totally 80 COPD patients admitted in The First Affiliated Hospital of Guangzhou Medical University from March 2016 to August 2017 were selected and divided into the control group (n=40) and the observation group (n=40) according to the random number table. Patients in the control group received conventional respiratory rehabilitation intervention, while the patients had their family members participated in the rehabilitation process in the observation group. Treatment compliance and effects were compared between the two groups. Results? The compliance of the observation group totaled 97.50%, while that of the control group totaled 87.50% (χ2=7.207, P＜ 0.05). After 3 months' intervention, the walking distance within 6 min of the observation group was (278.72±45.89) m; the PEF pred% was (93.46±13.47)%; and FEV1% was (87.42±14.71)%, better than those of the control group (t=4.127,7.156,3.859;P＜ 0.05). Conclusions? Family participation in respiratory rehabilitation process can improve the compliance of COPD patients significantly, thus improve the treatment effects, which is worth promoting in clinical practice.
