1.A primary study on the establishment of a low-titer group O whole blood donor bank

Objective: To investigate and analyze the IgM/IgG antibody titer levels and population characteristics of local type O blood donors, and to provide data support for the establishment of a low-titer group O blood donor bank. Methods: Whole blood samples were collected from 527 type O blood donors. The agglutination of IgM and IgG anti-A/anti-B antibodies at titers 64 and 128 was assessed using an enzyme immunoassay reader. The distribution of antibody agglutination was displayed using GraphPad Prism 9.5. Statistical analysis was performed to compare antibody agglutination differences among donors of different genders, age groups, and donation frequencies. Results: At a titer of 64, the non-agglutination rate of IgM anti-A/anti-B was 71.35%, and that of IgG anti-A/anti-B was 54.46%. At a titer of 128, the non-agglutination rate of IgM anti-A/anti-B was 83.68%, and that of IgG anti-A/anti-B was 70.21%. At a titer of 64, the agglutination rate of IgM anti-B was significantly higher in female donors than in male donors (23.08% vs 13.71%, P<0.05). The agglutination rates of IgM anti-A/anti-B at a titer of 64 decreased with age in different age groups (anti-A: 26.22% vs 18.28% vs 8.49%; anti-B: 19.82% vs 11.83% vs 5.66%, P<0.05). The agglutination rates of IgM anti-A/anti-B at a titer of 64 were both higher in first-time donors than in repeat donors (anti-A: 24.00% vs 15.82%; anti-B: 18.00% vs 10.73%, P<0.05). The agglutination rate of IgG anti-A at a titer of 128 was higher in first-time donors than in repeat donors (26.57% vs 6.21%, P<0.05). Conclusion: The establishment of a low-titer type O whole blood donor bank should primarily target males, donors aged>30 years and repeat donors, with both IgM and IgG antibodies included in the antibody testing scope.
2.Drug resistant of multidrug-resistant organisms and prevention and control effectiveness of multidisciplinary collaboration mode

OBJECTIVE To retrospectively analyze the drug resistance characteristics of the patients with multidrug-resistant organisms(MDROs)infections who were hospitalized from 2022 to 2023 and observe the effect of multi-disciplinary teamwork(MDT)mode so as to provide scientific bases for prevention and control of MDROs infec-tions and hospital-associated infections.METHODS A total of 639 patients with MDROs infection who were hospi-talized in Jianyang People's Hospital from Jan.2022 to Dec.2023 were recruited as the research subjects.The clinical data were collected from the patients,the drug resistance characteristics of bacteria were analyzed.The effects of MDT and pharmacological supervision on treatment of the patients with MDROs infection were observed and compared.RESULTS The methicillin-resistant Staphylococcus aureus(MRSA)(359 strains,56.18%)was dominant among the pathogens isolated from the 639 patients with MDROs infections,followed by the carbapen-em-resistant Klebsiella pneumoniae(CRKP)(96 strains,15.02%)and carbapenem-resistant Acinetobacter bau-mannii(82 strains,12.83%).Of the patients with MDROs infection,150(23.47%)were from critical care medicine department,94(14.71%)from pediatrics department,and 82(12.83%)from general surgery de-partment.The result of drug susceptibility test showed that the S.aureus strains were susceptible to linezolid,daptomycin,vancomycin and tigecycline;most of the gram-negative bacteria were susceptible to carbapenems,while the A.baumannii strains were highly resistant to the commonly used antibiotics.The total isolation rate of MDROs and the case-time infection rate of MDROs infections were 14.32%and 0.05%,respectively,after MDT and pharmacological supervision were carried out,lower than those before carried out;the effective treatment rate of the patients with MDROs was 76.47%after MDT and pharmacological supervision were carried out,higher than that before they were carried out,and there were significant differences(all P<0.05).CONCLUSION MDT and pharmacological supervision may improve the curative effect of the patients with MDROs infection and reduce the isolation rate of MDROs as well as the incidence of hospital-associated infections.

4.Analysis on the Expression of Basement Membrane Protein in Endometrial Carcinoma and Its Relationship with Immune Invasion

Objective Immune infiltration,as well as their implications across various cancers,and to establish a clinical prognostic model based on the findings.Methods The expression profile and clinical information of endometrial cancer tissue and adjacent tissues in TCGA endometrial cancer database were used,the differential expression genes were screened out for analysis,and the mRNA co-expressed by LncRNA was analyzed by GO enrichment and KEGG signaling pathway.The differential genes between endometrial cancer and adjacent tissues were intersected with the basement membrane protein genes.The selected differentially expressed genes were combined with survival status and survival time to screen out Hub genes by Lasso-cox regression analysis.Multivariate Cox regression analysis was used to establish a prognostic model,and pan-cancer analysis and immunoinfiltration correlation analysis were further performed.Results Six basal membrane protein Hub genes,ADAMTS5,EVA1C,THBS4,CTSD,ITGAV and LAMA1,were identified as associated with the prognosis of patients with endometrial cancer,and found that the survival rate of patients decreased significantly with the increase of risk score.Pan-cancer analysis found that these 6 genes were significantly different in most cancer types,and high expression in GBMLGG(glioma),LGG(low-grade glioma of the brain),LAML(acute myeloid leukemia),UVM(Uveal melanoma),ACC(adrenal cortical cancer)and other cancers had poor prognosis.The potential role of these genes in tumor immunotherapy was also explored,and significant negative correlation was found with Th 17 cells,Th2 cells,NK CD56 bright cells and other immune cells(P<0.01),and significant positive correlation was found with Tcm,iDC,Eosinophils,aDC and other immune cells(P<0.01).Conclusion Basal membrane protein gene has high clinical value in the diagnosis and prognosis of endometrial cancer,and can be used as a prognostic marker and potential therapeutic target for patients with endometrial cancer.
5.Study on the Influence of Yunnan Rice Products on Blood Sugar

Objective To explore the effect of Yunnan specialty rice products on blood sugar by measuring the glycemic index of 5 Yunnan special rice foods:rice noodles,rice cakes,rice rolls,sour rice noodles and dry rice noodles.Methods Following the national standard method to determine the carbohydrate content of 5 Yunnan specialty rice products,and the target amount of the test substance was calculated.Food Glycemic Index Determination Method was used to determine the glycemic index of 5 Yunnan specialty rice products and observe their impact on blood sugar.Results The GI value of Yunnan specialty food rice noodle is 63,rice cake is 64,rice roll is 46,sour rice noodles is 38,and dry rice noodles is 33.Conclusion Yunnan specialty foods rice noodle and rice cake belong to medium GI foods,and diabetes patients should reduce consumption;rice roll,sour rice noodles,and dry rice noodles belong to low GI foods and can be a better staple food source for diabetes patients.
6.Research progress on CAR T-cell neurologic toxicity

Chimeric antigen receptor (CAR) T-cell therapy is an effective new treatment for hematologic malignancies. Currently, two CAR T-cell products have been approved for clinical use by the U. S. FDA. A barrier to widespread use of CAR T-cell therapy is post-infusion toxicity, including primarily cytokine release syndrome and neurologic toxicity, in which neurotoxicity is the main factor for incidence rate and mortality rate.As there is still a lack of pathophysiological research on this symptom, this review describes existing neurologic toxicity and insights into the pathophysiology of this syndrome, which provides new opportunities for targeted therapeutic interventions to modulate CAR T-cell therapy toxicities.
7.Liver injury induced by Congrong Tongbian oral liquid (苁蓉通便口服液)

A 25-year-old female patient took Congrong Tongbian oral liquid 20 ml once daily by herself for constipation. After 3 months of intermittent medication, the patient gradually developed dark urine and yellowish skin and sclera, accompanied by nausea, vomiting, and decreased appetite; 4 months later, laboratory tests showed alanine aminotransferase (ALT) 1 359 U/L, aspartate aminotransferase (AST) 859 U/L, alkaline phosphatase (ALP) 160 U/L, total bilirubin (TBil) 131.5 μmol/L, and direct bilirubin (DBil) 99.3 μmol/L. Liver injury caused by Congrong Tongbian oral liquid was considered, then the drug was discontinued, and IV infusions of magnesium isoglycyrrhizinate injection 200 mg dissolved in 5% glucose injection 250 ml once daily and polyene phosphatidylcholine injection 20 ml dissolved in 5% glucose injection 250 ml once daily were given. After the above treatments, the patient′s symptoms were gradually improved. Two weeks later, the patient′s urine color was basically normal, gastrointestinal symptoms disappeared, and liver function showed ALT 137 U/L, AST 87 U/L, ALP 126 U/L, and TBil 34.9 μmol/L. At a 2-month follow-up, the patient′s liver function returned to normal. The patient′s liver injury was most likely related to the Polygonum multiflorum in Congrong Tongbian oral liquid.

9. Simultaneous determination of 14 chlorinated hydrocarbons in urine by headspace gas chromatography-mass spectrometry

OBJECTIVE: To establish a headspace gas chromatography-mass spectrometry method for the determination of 14 chlorinated hydrocarbons in urine. METHODS: The urine sample 4.00 mL and anhydrous sodium sulfate 3.00 g were added into a 10.00 mL headspace bottle, then the headspace bottle was maintained at 70 ℃ for 40.0 min. After headspace pretreatment, 14 chlorinated hydrocarbons in headspace air were separated in the DB-5 MS capillary column of the gas chromatography and detected by mass spectrometer. RESULTS: There was a good linear relationship of 14 chlorinated hydrocarbons in urine in the range of 0.62-1 630.00 μg/L. The linear correlation coefficient was greater than 0.999 0.The minimum detectable concentration was 0.19-0.43 μg/L and the minimum quantitative concentration was 0.62-1.44 μg/L. The average recovery rate was 89.8%-107.1%. The within-run relative standard deviation(RSD) was 4.0%-8.5% and the between-run RSD was 6.3%-9.1%. Urine samples can be stored at 4 ℃ or-8 ℃ for 3 days and below-20 ℃ for 7 days. CONCLUSION: This method is rapid, simple, sensitive, accurate and has little interference,which can be used as a method for detecting 14 kinds of chlorinated hydrocarbons in urine samples of patients with occupational poisoning.
10. Detecting human urinary thiocyanate by gas chromatography with pre-column derivatization

OBJECTIVE: To establish a method for detecting human urinary thiocyanate by gas chromatographic and pre-column derivatization with 2,3,4,5,6-pentafluorobenzyl bromide( PFB-Br). METHODS: A total of 20. 0 μL of urine was taken and 1. 0 m L of acetonitrile and 100. 0 μL of PFB-Br were added for derivative reaction. The gas chromatography was directly used for measurement. RESULTS: The urinary thiocyanate concentration showed a good linear range of 1. 000-10. 000 mg/L. The linear correlation coefficient was 0. 999 6. The minimum detection concentration was 0. 112 mg/L,and the minimum quantitative concentration was 0. 411 mg/L( 20. 0 μL urine sample). The standard recovery rate was 97. 22%-102. 04%.The within-run relative standard deviation( RSD) of this method was 1. 56%-5. 35%. The between-run RSD was 1. 46%-5. 10%. Hydrocyanic acid ions interfered with the measurement. Other common inorganic ions such as chloride,sulfate,and nitrate ions did not interfere with the measurement results. The samples can be stored at 4 ℃ for at least 15 days. CONCLUSION: This method is suitable for detecting human urinary thiocyanate.
