1.Mechanism of Dingkun Dan in Promoting Ovarian Granulosa Cell Proliferation in Polycystic Ovary Syndrome via microRNA-30d-5p-Mediated Targeting of Smad2

Objective To investigate the therapeutic mechanism of Dingkun Dan(DKD)in polycystic ovary syndrome(PCOS)by examining the effects of microRNA-30d-5p on ovarian granulosa cells(GCs).Methods A PCOS rat model was established using dehydroepiandrosterone(DHEA).Normal rat GCs and PCOS rat GCs were cultured in vitro and divided into four groups:blank control group,model group,low-dose DKD group,and high-dose DKD group.After grouping,GCs viability was assessed using the methyl thiazolyl tetrazolium(MTT)assay,GCs apoptosis was analyzed by flow cytometry,and the gene expression of transforming growth factor β1(TGF-β1),Smad2,Smad3,and microRNA-30d-5p in GCs was measured by real-time polymerase chain reaction(RT-PCR),protein expression of TGF-β1,Smad2,and Smad3 in GCs was detected by Western Blot.Results Compared with the blank control group,the model group exhibited significantly decreased GCs viability,increased GCs apoptosis,upregulated mRNA and protein expression of TGF-β1,Smad2,and Smad3,and downregulated microRNA-30d-5p expression,the differences were statistically significant(P<0.01).Compared with the model group,both low-and high-dose DKD groups showed increased GCs viability,reduced GCs apoptosis,downregulated mRNA and protein levels of TGF-β1 Smad2 and Smad3,elevated microRNA-30d-5p expression,and the differences were statistically significant(P<0.05 or P<0.01).Conclusion DKD promotes GCs proliferation by targeting Smad2 via microRNA-30d-5p,suggesting a potential therapeutic role in PCOS-related ovulatory dysfunction.
2.Obstructive Ileocolitis in Patients With Obstructed Colorectal Cancer – A Matched Case Control Study.

PURPOSE: Obstructive ileocolitis is an ulcero-inflammatory condition which typically occurs in the ileum or colon proximal to an obstructing colorectal lesion. If left unresolved, it often leads to intestinal perforation. We present a matched case control study of patients with obstructive ileocolitis caused by colorectal cancer to determine if any factors can predict this condition. METHODS: This is a retrospective review of 21 patients with obstructive colorectal cancer and histologically proven obstructive ileocolitis from 2005 to 2015 matched for age and sex with 21 controls with obstructing colorectal cancer without obstructive ileocolitis. RESULTS: The 21 patients with obstructive ileocolitis had a median age of 71 years (range, 52–86 years). The most common presenting symptom was abdominal pain (n = 16, 76.2%), followed by vomiting/nausea (n = 14, 66.7%) and abdominal distension (n = 12, 57.1%). Interestingly, the radiological feature of pneumatosis intestinalis was noted in only 1 case. No significant differences were observed in baseline comorbidities, clinical presentations, or tumor characteristics between the 2 groups. Patients with obstructive ileocolitis were found to have a significantly higher total leucocyte count (17.1 ± 9.4×109/L vs. 12.0 ± 6.8×109/L, P = 0.016), lower pCO2 (32.3 ± 8.2 mmHg vs. 34.8 ± 4.9 mmHg, P = 0.013), lower HCO3 (18.8 ± 4.5 mmol/L vs. 23.6 ± 2.7 mmol/L, P < 0.001), lower base excess (-6.53 ± 5.32 mmol/L vs. -0.57 ± 2.99 mmol/L, P < 0.001) and higher serum lactate levels (3.14 ± 2.19 mmol/L vs. 1.19 ± 0.91 mmol/L, P = 0.007) compared to controls. No radiological features were predictive of obstructive ileocolitis. CONCLUSION: Patients with obstructive ileocolitis tend to present with metabolic acidosis with respiratory compensation, raised lactate, and worse leucocytosis. Radiological features are not useful for predicting this condition.

3.Application of real-time fluorescence isothermal RNA amplification assay in the determination of Mycobacterium tuberculosis in pleural effusion

Objective To evaluate clinical value of utilization of real‐time fluorescence isothermal RNA amplification assay (SAT)for detecting Mycobacterium tuberculosis(MTB) in pleural effusion samples of patients with tuberculous pleurisy .Methods 90 hydrothorax specimens of patients with tuberculous pleurisy and 52 hydrothorax specimens of patients with non‐tuberculous pleurisy were selected ,and MTB was detected by using Roche culturing method ,SAT and TB DNA fluorescence quantitative expan‐sion method respectively .The sensitivity ,specificity and clinical value of SAT for detecting MTB were analysed .Results Taking Roche culturing method as the standard ,the sensitivity ,specificity and Youden index of SAT for detecting MTB were 90 .9% , 72 .1% and 0 .630 respectively .In patients clinically diagnosed with tuberculous pleurisy ,the positive detection rate of hydrothorax specimens detected by using Roche culturing method and SAT were 24 .4% and 43 .3% respectively ,and had statistically significant differences(χ2 =7 .166 3 ,P=0 .001 4) .Conclusion SAT is a rapid ,sensitive and specific method for the detection of MTB in pleu‐ral effusion samples ,which could improve the detection rate of MTB and decrease incidence of missed diagnosis and misdiagnosis .
4.Progress of research on mathematical model for transmission of schistosomiasis

Mathematical model has been increasingly applied in the description of the transmission of schistosomiasis, the prediction and selection of schistosomiasis prevention measures, and it has being developed constantly. This paper briefly introduces several typical mathematical models for transmission of schistosomiasis and summarizes their advancements.
