1.Analysis of distribution characteristics and drug resistance of mucous and non-mucous Streptococcus pneumoniae infections

Objective To compare the distribution characteristics and drug resistance of mucous and non-mucous Streptococcus pneumoniae(SP).Methods SP isolated from clinical specimens of the Second Affiliated Hospital of Xiamen Medical College from January 2023 to June 2024 were collected.They were isolated,cultured and identified,and their susceptibility to antibacterial drugs was determined.Results A total of 291 SP strains were isolated,of which 20 strains were mucous SP and 271 strains were non-mucous SP.Children played a dominant role of non-mucous SP infection,but mucous SP mainly infected adults and the children above 5 years old.Both phenotypes of SP were mainly characterized by pulmonary infection.Non-mucous SP had a relatively high sensitivity rate to vancomycin,linezolid,penicillin,ertapenem,chloramphenicol,levofloxacin,moxifloxacin and ofloxacin.It had a high resistance rate to compound sulfamethoxazole,clindamycin,erythromycin and tetracycline.Mucous SP was completely sensitive to penicillin,amoxicillin,cefotaxime,ceftriaxone,ertapenem,vancomycin,linezolid,levofloxacin and moxifloxacin,and had a relatively high resistance rate to clindamycin,erythromycin and tetracycline.Conclusion The laboratory should enhance the detection capacity of mucous SP.Mucous SP is highly sensitive to common antibiotics.Clinicians should select antibiotics based on drug sensitivity results to delay the occurrence of drug resistance.
2.Establishment and identification of organoid derived from patients with pancreatic cancer based on suspension culture

Objective:To explore the application of the suspension culture method in pancreatic cancer organoid construction.Methods:Cell suspensions obtained from 8 pancreatic cancer tissue samples at the Second Hospital of Lanzhou University between July 2023 and March 2024, were prepared by digested pancreatic cancer tumor tissues using mixed enzymes, inoculated into ultra-low adsorption culture plates for suspension culture, and when the organoids were cultured to a certain size, passaging and freezing were initiated, and their structural morphology was observed by inverted microscope. Hematoxylin-eosin (HE) staining showed that pancreatic cancer organoids were lumpy or irregularly tubular, with obvious nuclear atypia, and were remarkably similar in tissue structure to pancreatic cancer tissue.Results:Among the 8 pancreatic cancer tissue samples, pancreatic cancer organoids were successfully constructed in three patients, and HE staining showed that pancreatic cancer organoids had a high degree of structural similarity with tumor tissues. Immunohistochemistry suggested that CK7, CK19, P53, and Ki-67 were expressed in the pancreatic cancer organoids and tumor tissues of case origin in more or less the same way.Conclusion:The suspension culture method is able to construct pancreatic cancer-like organs that are approximately the same as the originating tumor tissues at the tissue level.

5.Bioinformatics analysis of differential gene expression in HEp-2 cells infected with human respiratory syncytial virus

Objective:To find clues potentially valuable for fighting against infection with human respiratory syncytial virus (HRSV), the differentially expressed genes in HEp-2 cells infected with HRSV were analyzed.Methods:Gene expression profiles of HEp-2 cells infected with HRSV were collected from the public gene expression omnibus (GEO) database. The differentially expressed genes following HRSV infection at each time point of 4, 8, 12, and 15 hours were found using R language. The differentially expressed genes were analyzed by gene ontology (GO), KEGG pathway and protein-protein interaction network (PPI). Genes with relatively high protein interaction in PPI were randomly selected for quantitative reverse transcription-polymerase chain reaction (qRT-PCR) verification at the transcription level from HEp-2 cells after HRSV infection at 4 hours.Results:A total of 101 differentially expressed genes were determined, including 92 upregulated genes and 9 downregulated genes. Function enrichment analysis revealed that HRSV infection could cause significant changes in multiple signaling pathways such as immune response in HEp-2 cells. The results of qRT-PCR were consistent with the trend of transcriptome data.Conclusions:The differentially expressed genes and the change of signaling pathways in HRSV-infected HEp-2 cells is of great significance to the studies on pathogenic mechanism and prevention of HRSV infection.
