1.Association between gross motor development characteristics and child Chinese developmental dyslexia

Objective:To explore the association between the gross motor development of children and Chi-nese developmental dyslexia.Methods:A total of 54 children were enrolled,indadit 27 children with Chinese DD and 27 age-gender-matched normal children.The Test of Gross Motor Development-Third Edition(TGMD-3),and the balance tests from the Movement Assessment Battery for Children(M-ABC)were used to evaluate the children's gross motor development level.Children's reading ability was evaluated by primary school students'literacy test question bank,one-minute reading task,and the Reading Test on Pupils.Results:The overall analysis of motortests showed that the grossmotor total score,locomotor score,object control score,and balance score in the DD group were significantly lower than those in the normal control group(P<0.05).Logistic regression analysis revealed a negative association between balance scores,locomotor scores,object control scores,TGMD-3 total scores and indi-viduals with dyslexia(OR=3.08,1.35,1.16,1.13,Ps<0.05).Conclusion:The delayed gross motor development of children is associated with the occurrence of Chinese developmental dyslexia.
2.Analysis of tumor infiltrating lymphocytes changes after neoadjuvant chemotherapy with prognosis in high-grade serous ovarian cancer

Objective:To investigate expression changes of tumor infiltrating lymphocytes(TILs)and their subtypes CD4+TILs,CD8+TILs,CD20+TILs and Foxp3+TILs in paired tumor samples pre-and post-neoadjuvant chemotherapy(NACT)in high-grade serous ovarian cancer(HGSOC)patients and their association with chemotherapy sensitivity and prognosis.Methods:A total of 40 patients with HGSOC who underwent NACT and interval debulking surgery(IDS)at Fourth Hospital of Hebei Medical University from August 2017 to February 2023 were retrospectively analyzed.CD4+TILs,CD8+TILs,CD20+TILs and Foxp3+TILs expressions in paired tumor samples pre-and post-NACT were detected by immunohistochemical staining.Divided patients into two groups based on KELIM score and platinum sensitivity,expressions of TILs,CD4+TILs,CD8+TILs,CD20+TILs and Foxp3+TILs were compared be-tween two groups,and their correlation with prognosis was analyzed.Results:Density of TILs(P=0.003,P=0.01),CD8+TILs(P<0.001,P=0.014)and CD20+TILs(P<0.001,P=0.003)post-NACT in patients with KELIM scores≥1 and platinum sensitivity were in-creased compared with pre-NACT.In contrast,there was no significant change in density of TILs pre-and post-NACT in patients with KELIM scores<1 and platinum resistance.Patients with high expression of CD8+TILs pre-NACT had longer progression free survival(PFS)and overall survival(OS)than patients with lower expression(P<0.001,P=0.011);patients with high expression of TILs post-NACT showed prolonged median PFS and OS(P<0.001,P=0.009);patients with high expression of CD20+TILs post-NACT showed prolonged median PFS(P=0.005).In multivariate analysis,density of CD8+TILs pre-NACT(HR=0.309,P=0.032),density of TILs post-NACT(HR=0.192,P=0.014),age(HR=2.962,P=0.036)were independent prognostic factors for PFS.Conclusion:Patients with HGSOC who have increased TILs density after NACT are more likely to be platinum sensitive.Expression of TILs,CD8+TILs and CD20+TILs in tumor site can serve as effective prognostic factors in HGSOC patients.

5.Tracking observation of fine motor development in children aged 6-8 with attention deficit hyperactivity disorder

Objective:
To examine the developmental trajectory of fine motor ability in schoolage children with attention deficit hyperactivity disorder (ADHD) for two years, so as to provide scientific evidence to promote motor development in ADHD children.
Methods:
From April to June 2019, 31 children aged 6-8 years old were selected from a public elementary school. They were diagnosed with ADHD by two psychiatric professionals according to the Diagnostic and Statistical Manual of Mental Disorders, Fifth Edition (DSM-V) criteria. Additionally, 31 typical developmental children, matched for age, sex and IQ with the ADHD group, were recruited as the control group. Fine motor ability was assessed with tasks of hand manual dexterity in Movement Assessment Battery for Children-2 (MACB-2), and a followup assessment was conducted from April to June 2021. The development changes of fine motor ability between two groups of children were compared by using t test and repeated measures analysis of variance.
Results:
Between baseline and followup periods after two years, the total score of hand fine motor in the ADHD group did not show significant improvement (7.4±3.0, 8.0±3.4; t=-1.05, P>0.05), while there was a small effect size improvement in typically developing control group (9.5±2.1, 10.5±2.4; t=-2.12, effect size=0.38, P<0.05). Followup after two years, coin/peg throwing scores with dominant hand improved between ADHD group and control group (7.0±3.3, 9.5±3.2; 8.4±2.8, 11.6±1.6) (t=-3.74, -6.33, P<0.01; effect size=0.67, 1.14), with a smaller improvement in the ADHD group. The score for threading beads/threads decreased in between ADHD group and control group (7.9±2.4, 5.8±3.1; 9.2±1.1, 8.2±1.9) (t=3.89, 2.78, P<0.01; effect size=0.70, 0.50), with a greater decrease in the ADHD group.
Conclusions
The development speed of fine motor ability in children with ADHD aged 6-8 is slow and continues to lag behind normal developmental children. Fine motor development in children with ADHD should be closely monitored, and targeted interventions should be implemented when necessary.
6.AP2M1 inhibits proliferation and invasion of diffuse large B-cell lymphoma cells

Objective To evaluate the regulatory effect of the adaptor related protein complex 2 subunit μ1(AP2M1)on proliferation and invasion of diffuse large B-cell lymphoma(DLBCL).Methods Human diffuse large B-cell lymphoma cell line OCI-LY8 was aliquoted into control group,NC-shRNA group,AP2M1-shRNA group,NC-LV group,and AP2M1-LV group.Lipofectamine 2000 was used for cell transfection.Cell proliferation was detected by tetramethylazolium salt(MTT)method,apoptosis was detected by flow cytometry and cell migration and invasion were detected by Transwell assay.The protein expression of AP2M1,epidermal growth factor receptor(EGFR),p-phosphatidylinositol 3 kinase(PI3K),PI3K,p-protein kinase B(Akt)and AKT was detec-ted by Western blot.Results Compared with control group,the relative expression of AP2M1 mRNA and protein in the AP2M1-shRNA group was decreased(P<0.05).The relative cell viability was increased(P<0.05).The cell apoptosis rate was decreased(P<0.05).The counting number of migrating and invading cells was in-creased(P<0.05).The relative expression level of EGFR protein and the phosphorylation level of PI3K and AKT were increased(P<0.05).Compared with Control group,the expression of AP2M1 mRNA and protein relative ex-pression level in AP2M1-LV group was increased(P<0.05).The relative cell viability was decreased(P<0.05).The cell apoptosis rate was increased(P<0.05).The number of migrating and invading cells was decreased(P<0.05).The relative expression level of EGFR protein and the phosphorylation level of PI3K and AKT were all decreased(P<0.05).Conclusions The over-expression of AP2M1 partially inhibits the proliferation and invasion of DLBCL cells by inhibiting the EGFR/PI3K/AKT signaling pathway.
7.Bushenhuoxue Decoction Improves Hippocampal Synaptic Plasticity of Vascular Dementia Rat Model via PI3K-Akt-mTOR Signaling Pathway

Objective To observe the effect of Bushen Huoxue Decoction(BSHX)on PI3K/Akt/mTOR signaling pathway and explore its possible mechanism in improving synaptic plasticity in a vascular dementia(VD)rat model.Methods Sprague-Dawley rats were randomly divided into sham operation group(Sham group),model group(VD group),Bushenhuoxue decoction group(BSHXD group),nimodipine group(NMDP group),with 10 rats in each group.The VD model of rats was established by two-vessel(2-VO)occlusion method.Rats in BSHXD group were given BSHXD at a weight of 10.14 g·kg-1,while rats in the NMDP group were given nimodipine decoction at 11 mg·kg-1.The SHAM group and the VD group were given saline at a weight of 10 mL·kg-1 once a day for 4 weeks.Morris water maze was used to observe the spatial learning and memory ability of rats in each group.Nissl staining was used to observe the damage of Nissl bodies and neurons in CA1 area of hippocampus of rats.The expression of synaptophysin(SYN)and postsynaptic density protein 95(PSD-95)in hippocampal CA1 region was detected by immunohistochemistry.Golgi-Cox staining method was used to observe the number changes of dendritic branches and spines of hippocampal neurons.Transmission electron microscopy(TEM)observed the ultrastructural change of synapses.The protein and mRNA expressions of phosphatidylinositol 3-kinase(PI3K),serine-threonine kinase(AKT)and mammalian target of rapamycin(mTOR)in rat hippocampus were detected by Western blot and Reverse transcription quantitative PCR(RT-qPCR).Results Compared with the control group,the learning and memory ability of VD rats decreased.These rats showed abnormal synaptic ultrastructure of hippocampal neurons and neuronal cell damage,and this was accompanied by a decrease in the density of dendrite branches and dendritic spines of neurons.The expression of both SYN and PSD-95 proteins in the hippocampus decreased(P<0.05),and synaptic plasticity was damaged.Both mRNA and protein expression of PI3K,Akt,and mTOR in the hippocampus decreased in the VD rats(P<0.05).Also observed in VD rats was that administration of BHSX mitigated the learning and memory impairment observed in these animals,improved the morphology and synaptic ultrastructure of hippocampal neurons,increased the mRNA and protein expression levels of PI3K,Akt,mTOR,and increased the protein levels of SYN and PSD-95(P<0.05).Conclusion BSHX can alleviate the learning and memory impairment of VD rats and increase the protein expression levels of synapse-related proteins.These effects may be related to the promotion of synaptic plasticity by BSHX through activation of PI3K/Akt/mTOR signaling.
8.Effects of exercise intervention on reading abilities of children with Chinese developmental dyslexia

Objective To explore the impact of exercise intervention on the reading abilities of chil-dren with Chinese developmental dyslexia(CDD).Methods Twenty-seven CDD children were recruited from patients of Peking University Sixth Hospital,and randomly divided into a CDD exercise interven-tion group of 14 and a CDD control group of 13.Another 14 age-matched normal children were select-ed into a normal control group.All groups kept daily physical activities in school,while the CDD ex-ercise intervention group additionally underwent 8-week individualized gross and fine motor interven-tion.The literacy,reading fluency and reading comprehension of all groups were evaluated by using"Elementary School Students'Literacy Test and Evaluation Scale",one-minute word reading and ele-mentary school students'reading ability assessment.Results Before the intervention,there were no sig-nificant differences between the exercise intervention and control groups in the literacy level,reading fluency,and reading comprehension(P>0.05),which were all significantly lower than the normal con-trol group(P<0.05).However,after the intervention,significant improvement was observed in the aver-age literacy level,reading fluency,and reading comprehension of the CDD exercise intervention group,and the literacy and reading fluency of the CDD control group,as well as the average literacy level and reading comprehension of the normal control group.Moreover,there were no significant differ-ences between the CDD exercise intervention and control groups in the average literacy level and read-ing fluency score(P>0.05),both significantly lower than the normal control group(P<0.05).Mean-while,the reading comprehension score of the CDD exercise intervention group was significantly higher than the CDD control group(P<0.05),but significantly lower than the normal control group(P<0.05).Conclusion Eight-week targeted gross and fine motor intervention can effectively improve reading abili-ty of DD children,especially their reading comprehension.
9.The relationship between fine motor skills and executive function in boys with attention deficit hyperactivity disorder

Objective:
To investigate the relationship between fine motor skills and executive function in school aged boys with attention deficit hyperactivity disorder (ADHD) aged 6-10 years, so as to provide a reference for promoting executive function.
Methods:
From November 2022 to May 2021,65 boys with ADHD were recruited from a tertiary hospital in Beijing and a public elementary school in Beijing, and 65 boys with typical development according to age and sex were recruited as controls. Children s fine motor skills were assessed using the movement assessment battery for children, second version (MABC-2), and children s inhibitory control, working memory, and cognitive flexibility were assessed using the Stroop color word test (SCWT), Rey osterrich complex figure test (ROCFT) and trail making test (TMT), respectively. Independent samples t-tests were used to analyze the differences between the two groups, and Pearson correlation analysis and stratified regression analysis were used to explore the relationship between fine motor skills and executive functioning in boys with ADHD.
Results:
In the fine motor skills test, boys with ADHD scored significantly lower (8.68±3.62) than control boys (11.22±2.27)( t =4.80, P <0.05); in the executive function test, the results of SCWT, ROCFT, and TMT tests were significantly worse in boys with ADHD than in normal boys ( t =-4.53-4.42, P < 0.05). Correlation analysis showed that the fine motor skill scores of boys with ADHD were negatively correlated with the number of word sense errors ( r =-0.35) and color errors ( r =-0.42), and positively correlated with memory scores for delayed structure ( r =0.30) and detail ( r =0.25), which were negatively correlated with TMT-A ( r =-0.34),TMT-B reaction time ( r =-0.26), number of errors ( r =-0.43) ( P <0.05). Stratified regression analysis showed that fine motor skill scores of boys with ADHD were predictive of test results for inhibitory control, working memory and cognitive flexibility, with explanatory rates of 15%, 7%, and 19%, respectively.
Conclusion
Fine motor skills in boys with ADHD are correlated with executive functions, and fine motor skills could predict inhibitory control, working memory and cognitive flexibility to some extent. The development of fine motor skills in boy with ADHD could promote their executive functions.
10.Sex difference of fine motor skills of school-age children with attention deficit hyperactivity disorder

ObjectiveTo explore the characteristics of fine motor skills of school-age children with attention deficit hyperactivity disorder (ADHD) in different sexes. MethodsFrom April, 2021 to April, 2023, 66 ADHD children from grades one to four in Beijing Haidian Wanquan Primary School were screened by psychiatrists, and other 69 ADHD children were recruited from hospitals and patient groups of WeChat, accounting to 135 ADHD children. Meanwhile, a total of 135 typically developed (TD) children matched with sex and age (< 0.5 year) were recruited from the same primary school as controls. They were assessed with Movement Assessment Battery for Children (MABC-2) four subtests of manual dexterit. ResultsAfter controlling age, the subtest scores and the total score of fine motor of MABC-2 were less in ADHD boys and girls than in the TD boys and girls (P < 0.05). The main effect of sex was significant on the subtest scores and the total score of fine motor for ADHD children (F > 5.133, P < 0.05), and they were less in the boy than in the girls (P < 0.05). ConclusionThe development of fine motor delays in school-aged ADHD children, especially for the boys.
