Objective:To analyze incidence trends and age distribution of cutaneous melanoma in Jiangsu province from 2010 to 2019.Methods:In this cross-sectional study, data on the reported incidence of cutaneous melanoma from 2009 to 2019 were obtained from 16 cancer registries in Jiangsu Province, which had complete and continuous records after quality control. The Jointpoint log-linear regression model was used, and the average annual percentage change (AAPC) was calculated to analyze incidence trends of cutaneous melanoma in different genders and areas (urban or rural). Data from the Chinese population census in 2000 were used for calculation of age-standardized incidence rates by Chinese standard population. The Jointpoint regression model was used to evaluate the trends in the average age at onset over time. If there were no inflection points, a t test was used to compare changes in the average age at onset and age-specific proportions among different years. The age-period-cohort model was used to analyze changes of the incidence in birth cohorts in different years. Results:The age-standardized incidence rate of cutaneous melanoma in Jiangsu province was 0.26/100 000 in 2009 and 0.49/100 000 in 2019. Overall, the provincial incidence rates showed an increasing trend (AAPC in age-standardized incidence rates: 6.17%, P < 0.001). In the subgroup analyses, increasing trends were observed in different genders and areas (AAPC in age-standardized incidence rates: 4.24% for males, 8.01% for females, 5.17% for urban areas, 7.62% for rural areas, all P < 0.001). The standardized average age at onset of cutaneous melanoma was 59.95 years in 2009 and 58.84 years in 2019, with no significant trend in average age at onset over time ( P = 0.196). People aged 60 years and above had an AAPC of 1.20% ( P = 0.020) in the actual incidence proportion of cutaneous melanoma in 2019 compared with 2009. The age-period-cohort model showed that the incidence progressively increased with the year of birth in age groups among patients of different genders and in different areas (all P < 0.05) . Conclusions:The overall incidence of cutaneous melanoma was relatively low in Jiangsu province from 2009 to 2019, however, an upward trend was observed. Patients in both rural and urban areas, as well as female and male populations, all exhibited increasing incidence rates. The incidence rates of cutaneous melanoma increased with age.

Objective To construct N6-methyladenosine related long non-coding RNA(LncRNA)pairing model for renal cell carcinoma based on bioinformatics analysis of the cancer ganome atlas(TCGA)database and to explore its prognosis value.Methods Transcriptome data of RNA-sep for renal cell carcinoma and its related clinical information were downloaded from the TCGA database.Perl software was used to organize and separate LncRNA and messenger RNA(mRNA)from the transcriptome data.A total of 564 tissues from renal cell carcinoma cases and 72 normal tissues were obtained,and thus 540 renal cancer patients were eventually included.Random data table method was used to divide 540 patients with renal cancer into a training group(n=275)and a validation group(n=265)by caret.M6A related LncRNA pairing models were established based on the single factor and multivariate COX regression analysis.The risk assessment equation was obtained using the LASSO regression algorithm.The risk scores were calculated based on this equation,and the optimal critical point of the median risk value was applied to divide all patients into high-risk and low-risk groups.Kaplan-Meier survival analysis was used to make a survival curve for the differences between high and low risk groups in the overall sample.The gene ontology(GO)and Kyoto encyclopedia of genes and genomes(KEGG)pathway enrichment analyses were conducted using the Cluster Profiler software package.The relationship between N6-methyladenosine related LncRNA pairing model and immune cell infiltration was analyzed by R software.Results Kaplan-Meier survival analysis showed the total survival time of patients in the low-risk group was significantly higher than that of patients in the high-risk group of the training group(P<0.05).Compared with high risk group,the overall survival time of patients(G1～2,G3～4,Ⅰ～Ⅱ,or Ⅲ～Ⅳ,age≤65 years,or patients>65 years old)in low risk group was higher(P<0.05).Differential gene enrichment analysis was obtained for high and low risk groups,which mainly enriched with many differential genes such as muscle contraction,rhabdomytic cell differentiation,myofibril,receptor activation activity,and vascular smooth muscle contraction.The highest driver genes in high risk group and low risk group exhibited mutation frequency and mutation information,and their risk score was positively correlated with the degree of T cell and plasma cell infiltration(r=0.638,P=0.001).Conclusion Bioinformatics-based analysis of the N6-methyladenosine related LncRNA pairing models can be helpful to predict the prognosis of patients with renal cancer.It provides new ideas for the prognosis evaluation and optimal treatment strategy of renal cancer,and contributes to further analyzing the molecular mechanism of the occurrence and development of gastric cancer in the future.

This quick guideline aims to standardize the diagnosis and treatment of novel coronavirus Omicron infection, and to improve the disease management abilities of clinicians.

Objective To investigate the effect of effective fractions from Radix Vitis Davidii Foex. on fracture healing. Methods SD rats were randomly divided into model control group,Yunnan Baiyao group,and effective fraction group (n= 36 each group). Fracture models were established. Rats in the Yunnan Baiyao group and effective fraction group were given 3.0 g?kg-1?d-1 of Yunnan Baiyao and effective fraction from Radix Vitis Davidii Foex.,respectively,and rats in the model control group was treated with the same volume of pure water. Alkaline phosphatase and Ca concentration in serum were determined.For radius fracture structure,digital radiography and RT-PCR detection of VEGF mRNA expression were performed. Results The peak concentration of the alkaline phosphatase and Ca concentration in serum were significantly higher in the effective fraction group than in the Yunnan Baiyao group,and peak value appeared earlier,fracture healing score was higher, fracture line disappeared earlier,and VEGF mRNA expression was significantly higher in three weeks after the fracture (all P<0. 01) . Conclusion The effect of the effective fraction from RadixVitis Davidii Foex.on the fracture healing is strong.

OBJECTIVETo study the signaling pathways associated with lipopolysaccharide (LPS)-induced inflammation in islet micro-endothelial cells (IMECs) and the mechanism of pravastatin intervention.

METHODSIMECs exposed to LPS, SB203580, pravastatin, or SB203580+pravastatin were examined for cell apoptosis with Hoechst staining and flow cytometry and for expression levels of total-p38, photophosphorylation-p38 (p-p38) and iNOS with Western blotting.

RESULTSThe apoptosis rate and expression levels of total-p38, p-p38, iNOS in IMECs all increased after LPS exposure. Pravastatin, SB203580, and their combination significantly attenuated LPS-induced enhancement of cell apoptosis and total-p38, p-p38, and iNOS expressions in IMECs.

CONCLUSIONLPS-induced inflammatory toxicity in IMECs is associated with the activation of P38MAPK and iNOS/NO signaling pathways. Pravastatin can inhibit these pathways and suppress the apoptosis and necrosis of IMECs to relieve the cell inflammatory injuries.
