1.Effects of hyperbaric oxygen combined with melatonin on migration of lung adenocarcinoma A549 cells and its possible molecular mechanism

Objective:To discuss the effect of hyperbaric oxygen (HBO) combined with melatonin (MLT) on the migration ability of lung adenocarcinoma A549 cells and its possible molecular mechanism.Methods:After the cell culture, lung adenocarcinoma A549 cells were divided into cell control group (control group), hyperbaric oxygen treatment group (HBO group), melatonin treatment group (MLT group), and hyperbaric oxygen combined with melatonin group (MLT+ HBO group). Wound healing assay was used to assess the migration ability of A549 cells, and Western blotting was used to detect the expression of migration-related protein matrix metalloproteinase 9 (MMP-9) in each group.Results:The migration distances of lung adenocarcinoma A549 cells in the MLT group and the MLT+ HBO group after 48 h were shorter than that in the control group, with statistically significant differences ( P<0.01); and the migration distance of lung adenocarcinoma A549 cells in the MLT+ HBO group was reduced even more obvious than that in the MLT group, and the difference was statistically significant ( P<0.01). The expression levels of MMP-9 in the HBO group and the MLT group were lower than that of the control group ( P<0.05); the expression level of MMP-9 in the MLT+ HBO group was even lower than those in the HBO group and the MLT group, and the difference was statistically significant ( P<0.05). Conclusion:Melatonin can inhibit the migration ability of lung adenocarcinoma A549 cells, and hyperbaric oxygen can enhance such inhibitory effect. The mechanism of action may be related to the reduced expression of migration-related protein MMP-9.

3.Effect of hyperbaric oxygen on the melatonin enhancement of the differentiation of gastric cancer cells

Objective To investigate the effects of hyperbaric oxygen (HBO) on the melatonin enhancement of the differentiation of gastric cancer cells SGC7901 and also to explore the possible mechanism involved.Methods The SGC7901 cells at logarithmic differentiation stage were divided into the control group,the melatonin treatment group and the HBO coupled with melatonin group.The rate of cell differentiation was detected by flow cytemctry,and endocan expression was measured by using Western blot assay.Results SGC7901 cells treated with melatonin could enhance the rate of the cells at differential stage (the rate of the melatonin treatment group was 74.35% ±0.88%,while that of the control group was 73.12% ± 1.01%).Melatonin treatment could also up-regulate the expression level of endocan cells (the expression level of the melatonin treatment group was 2.14 ± 0.12,while that of the control group was 1.99 ± 0.04).However,when treated with HBO + melatonin,both the rate of the cells at the differential stage (the rate of the cells of the HBO coupled with melatonin group was 76.86% ± 1.13%) and the expression of endocan cells (the rate of endocan cells of the HBO coupled with melatonin group was 2.57 ± 0.03) were further enhanced,and statistical significance could be noted,when compared with those of the control group (P < 0.05).Conclusions HBO treatment could obviously enhance the effect of melatonin on the differentiation of gastric cancer cells SGC7901,the mechanism of which might be associated with the up-regulation of endocan cells by melatonin.
