1.Aquaporin 1 promotes proliferation and migration of tumor by up-regulating claudin-1 expression in colon cancer

With the rising incidence of colon cancer, several studies have indicated that aquaporin 1 (AQP1) expression is associated with the development of colon cancer. This study aims to elucidate the potential molecular mechanisms between them. Methods: We screened data from The Cancer Genome Atlas (TCGA) database and retrospectively examined AQP1 protein expression in 127 colon cancer patients to analyze the relationship between AQP1 expression and pathological stages, prognosis. We created stable colon cancer cell lines with differential AQP1 expression, the effect of AQP1 expression on the proliferation and migration of colon cancer cells was assessed by in vitro and in vivo studies, and explored potential molecular mechanisms through Western blotting. Results: High AQP1 expression was associated with poorer survival (overall survival [OS], p = .028) in colon cancer patients from the TCGA database. Similarly, retrospective clinical data indicated that high AQP1 expression was associated with reduced disease-free survival and OS (p = .036 and p = .017, respectively). The low-expressing AQP1 colon cancer cells exhibited a decrease in proliferation and migration ability of colon cancer cells compared to the overexpressing AQP1 group (p < .05) in vitro and in vivo. Immunohistochemistry and western blotting experiments validated heightened expression of N-cadherin, vimentin, and claudin- 1 in the tumor tissues of the overexpressing AQP1 group. Conversely, reduced AQP1 expression resulted in decreased expression of claudin- 1. Conclusions: AQP1 correlates with unfavorable prognosis in colon cancer and potentially enhances the proliferation and migration of colon cancer by up-regulating claudin-1 expression.
2.Predictive value of the early 18F-fluorodeoxyglucose positron emission tomography/computed tomography parameters percentage change in the maximum standardized uptake value and metabolic tumor volume for residual tumor lesions in patients with diffuse large B-cell lymphoma after chemotherapy

Objective To analyze the changes of early 18F-fluorodeoxyglucose positron emission tomography/computed tomography (18F-FDG PET-CT) metabolic parameters in patients with diffuse large B-cell lymphoma (DLBCL) before and after chemotherapy, and to systematically evaluate the early predictive efficacy of the above prarmeters for residual lesions. Methods Eighty patients with DLBCL admitted to our hospital from January 2021 to January 2025 were randomly selected as study subjects. All patients received 18F-FDG PET-CT examination. After 3 months of chemotherapy, patients were assessed according to the Lugano 2014 criteria. Patients without residual lesions were assigned to the complete remission group, while those with residual lesions were assigned to the residual lesion group. Clinical data and 18F-FDG PET-CT parameters were compared between the two groups. A logistic regression model was used to analyze the relationship between 18F-FDG PET-CT parameters and residual tumor lesions in DLBCL patients after chemotherapy. The receiver operating characteristic curve was used to analyze the predictive value of early 18F-FDG PET-CT parameters for residual tumor lesions in patients with DLBCL after chemotherapy. Results Compared with the complete remission group, the residual lesion group had a significantly higher proportion of patients with an international prognostic index score>3, as well as significantly higher maximum standardized uptake value (SUVmax), metabolic tumor volume (MTV), and total lesion glycolysis(TLG). The early percentage change in the maximum standardized uptake value (ΔSUVmax%) was significantly lower in the residual lesion group (P<0.05). There were no significant differences in age, sex, Ann Arbor stage, serum lactate dehydrogenase elevation, Eastern Cooperative Oncology Group performance status score ≥2, and molecular subtype between the two groups (P>0.05). Logistic regression analysis showed that an international prognostic index score >3 and a high MTV were independent risk factors for residual tumor lesions in DLBCL patients after chemotherapy, while a high ΔSUVmax% was a protective factor (P<0.05). Receiver operating characteristic curve analysis showed that the area under the curve for ΔSUVmax% and MTV alone in predicting residual tumor lesions after chemotherapy for DLBCL was 0.918 (95%CI=0.848-0.989) and 0.926 (95%CI=0.866-0.985), with cutoff values of 75.16% and 13.58 cm3, respectively. The combined prediction using both parameters yielded an area under the curve of 0.960 (95%CI=0.891-0.991), with a sensitivity of 91.38% and a specificity of 95.45%. Conclusion The early 18F-FDG PET-CT parameters ΔSUVmax% and MTV demonstrate high predictive value for residual tumor lesions in DLBCL patients after chemotherapy. Their combined use has improved predictive efficacy, which can provide a reference for clinical evaluation of chemotherapy efficacy in DLBCL.

This study explored the effects and underlying mechanism of Raf kinase inhibitory protein(RKIP)on apoptosis in mast cells sensitized by Echinococcus granulosus cyst fluid.Bone marrow-derived mast cells(BMMCs)were isolated and cultured from RKIP knockout(KO)and wild-type(WT)C57BL/6 mice.Cells were divided into control and sensitized groups.The sensitized group was incubated for 24 h in RPMI1640 medium containing 10%serum from mice infected with E.granulosus,then activated for 3 h or 6 h with E.granulosus cyst fluid.The control group was incubated for 24 h in RPMI1640 medium,and then received an equal vol-ume of PBS.Cells and supernatants were collected for analysis.Flow cytometry was used to detect the expression of CD117 and FcεRⅠα on BMMCs.The levels of β-hexosaminidase,IL-4,and TNF-α in the supernatant were quantified with ELISA.Western blot analy-sis was used to assess expression changes in RKIP,apoptosis-related proteins,and pathway proteins in BMMC before and after sensi-tization.Flow cytometry analysis revealed that after 4 weeks of induction,the CD117 and FcεRⅠα double-positivity rates on both WT and KO BMMC exceeded 90%.ELISA indicated that the E.granulosus cyst fluid resulted in significantly greater β-hexosaminidase re-lease(F=16.88,P<0.05),and levels of IL-4(F=16.51,P<0.05)and TNF-α(F=9.78,P<0.05)in the KO sensitized group than the WT sensitized group.With respect to the WT control group,the WT sensitized group showed significantly down-regulated pro-tein expression levels of RKIP(F=8.20,P<0.05)and Bcl-2(F=101.40,P<0.01)after 3 h,but significantly up-regulated levels of p-PI3K(F=8.04,P<0.05),p-Akt(F=32.52,P<0.01),p-P65(F=13.29,P<0.05),and cleaved-caspase-3(F=46.34,P<0.01).With respect to the WT sensitized group,the KO sensitized group showed significantly up-regulated protein expression of p-PI3K(F=8.45,P<0.05),p-Akt(F=8.58,P<0.05),p-P65(F=11.02,P<0.05),and Bcl-2(F=84.50,P<0.001)after 3 h,but significantly down-regulated expression of cleaved-caspase-3(F=15.66,P<0.05).In conclusion,RKIP may inhibit the PI3K/Akt/NF-κB pathway,thereby inducing apoptosis in mast cells sensitized by E.granulosus cyst fluid.This process may help ease aller-gic reactions caused by mast cells in echinococcosis,thus offering a promising new approach for preventing and treating such reactions.
4.Evaluation of joint end microfracture treatment combined with arthroscopy to improve the long-term effect of ankle joint fusion and influence on oxidative stress and complications during perioperative period

Objective To investigate the effect of joint end micro fracture treatment combined with arthroscopy on the long-term effect of ankle joint fusion and the effects on perioperative oxidative stress and complications.Methods 108 patients with end-stage ankle arthritis with internal and external inversion angles with ≤20° matching baseline data from February 2021 to April 2023 were selected.Among them,54 patients underwent arthroscopic ankle fusion as arthroscopic group,54 patients were treated with joint end microfracture combined with arthroscopic ankle joint fusion as improved group.The surgical indexes,perioperative oxidation indexes[malondialdehyde(MDA),total oxidation state(TOS),oxidative stress index(OSI)before surgery,1 d after surgery and 3 d after surgery]and antioxidant indexes[glutathione peroxidase(GSH-Px),superoxide dismutase(SOD),total antioxidant capacity(TAC)before surgery,1 d after surgery and 3 d after surgery]of the two groups were compared.Visual analogue scale(VAS score),American Orthopaedic Foot and Ankle Society(AOFAS score),before surgery,6 months after surgery and 1 year after surgery,excellent and good rate and complications were compared between the two groups,and fusion time and AOFAS score at 1 year after surgery of patients with different internal and external inversion angles were compared between the two groups.Results The intraoperative blood loss in the improved group was less than that in the arthroscopic group,postoperative hospital stay in the improved group was shorter than that in the arthroscopic group,the differences were statistically significant(P<0.05).The MDA,TOS and OSI levels in the improved group were lower than those in the arthroscopic group on day 1 and day 3 after operation,the differences were statistically significant(P<0.05).The GSH-Px,SOD and TAC levels in the improved group were significantly higher than those in the arthroscopic group at 1 and 3 days after surgery,the differences were statistically significant(P<0.05).The VAS score and AOFAS score at 6 months and 1 year after surgery in the improved group were not significantly different from those in the arthroscopic group(P>0.05).Comparison of the excellent rate of patients in the two groups,the difference was not statistically significant(P>0.05).The total complication rate of the improved group was significantly lower than that of the arthroscopic group,the difference was statistically significant(P<0.05).There was no significant difference in AOFAS scores between patients with 10°~20° and patients with<10° at 1 year after surgery(P>0.05).The fusion time of patients with internal and external inversion angles 10°~20° was longer than that of patients with<10°(P<0.05).Conclusion Joint end microfracture treatment combined with arthroscopy ankle fusion for end-stage ankle arthritis with ≤20° internal and external varus can avoid bone grafting and osteotomy with satisfactory efficacy,and has advantages in reducing intraoperative bleeding and complications,alleviating perioperative oxidative stress,and accelerating postoperative recovery.However,when internal and external varus malformations are obvious,the time of osseous fusion can be prolonged.
5.A study on the correlation between HPV DNA and IHC P16 expression in cervical lesions

Background and purpose:Human papilloma virus(HPV)infection status is crucial for diagnosing cervical precancerous lesions and classifying cervical cancer.High-risk(HR)HPV is often linked to P16 protein overexpression,so P16 detection via immunohistochemistry(IHC)is commonly used to assess HPV infection.However,the differences between HPV status and P16 expression remains unclear.An in-depth study of the correlation between HPV and P16 is essential for clinical guidance.Methods:We retrospectively collected clinical and pathological data of cervical lesions from 618 patients diagnosed at the Department of Pathology,Fudan University Shanghai Cancer Center from January 2020 to December 2023(Ethical number:050432-4-2307E).Polymerase chain reaction(PCR)reverse dot hybridization was used to detect HPV including HR and low-risk(LR)subtypes,and immunohistochemistry was used to detect P16 for comparative analysis.Based on different clinical and pathological diagnoses,the sensitivity and specificity of P16 expression in evaluating HPV infection were evaluated.Among the 618 cases of cervical lesions,there were 92 cases of cervical squamous cell carcinoma,257 cases of cervical adenocarcinoma,79 cases of high-grade squamous intraepithelial lesions(HSIL),105 cases of low-grade squamous intraepithelial lesions(LSIL),and 85 cases of chronic cervical inflammation.Results:According to clinical diagnosis,the HR-HPV positive rate in cervical squamous cell carcinoma was 88.0%(81/92),the P16 positive rate was 91.3%(84/92),and the overall consistency rate between P16 and HPV detection was 90.2%(88/92);for HR-HPV infection,the sensitivity and specificity of P16 were 96.3%and 45.5%.The positive rate of HR-HPV in adenocarcinoma was 54.5%(140/257),the positive rate of P16 was 58.8%(151/257),and the overall consistency rate between P16 and HPV detection was 82.5%(212/257);for HR-HPV infection,the sensitivity and specificity of P16 were 87.9%and 76.1%.In HSIL,the HR-HPV positive rate was 75.9%(60/79),the positive rate of P16 was 70.9%(56/79),and the overall consistency rate between P16 and HR-HPV detection was 82.2%(65/79);for HR-HPV infection,the sensitivity and specificity of P16 were 85.0%and 73.7%.In LSIL,the HR-HPV positive rate was 73.3%(77/105),the positive rate of P16 was 8.5%(9/105),and the overall consistency rate between P16 and HR-HPV detection was 33.3%(35/105);for HR-HPV infection,the sensitivity and specificity of P16 were 10.4%and 96.4%.In chronic cervical inflammation,the HR-HPV positive rate was 20%(17/85),the positive rate of P16 was 0.0%(0/85);for HR-HPV infection,the sensitivity and specificity of P16 were 0.0%and 100.0%.There was a significant positive correlation between P16 positivity and HPV16/18 in cervical squamous cell carcinoma,adenocarcinoma,and HSIL(P=0.000),while there was no significant correlation in LSIL and chronic cervical inflammation(P>0.05).Conclusion:In cervical squamous cell carcinoma and adenocarcinoma,the consistency of P16 expression and HPV DNA positivity are high,especially in HPV16/18 subtype.There is a good concordance between HR-HPV positivity and P16 protein overexpression.The positive expression of P16 in HSIL may initially reflect HPV infection status.However,in LSIL and chronic cervicitis,P16 expression may not accurately correlate with HPV infection.The inconsistency between P16 and HPV DNA testing could be influenced by multiple factors,including HPV subtypes,histopathological categories,specimen quality,and technical limitations.In clinical practice,it is recommended to conduct comprehensive analysis or employ multiple diagnostic methods to confirm HPV infection status for precise evaluation.

7.Clinical Study on the Treatment of Lung Cancer-Related Cough by Fire Needle Acupuncture Selection Based on CT Localization

Objective:To prospectively analyze and evaluate the clinical efficacy of fire needle acupuncture selection based on CT localization in the treatment of lung cancer-related cough.Methods:A total of 36 patients with lung cancer-related cough who were admitted to the Rehabilitation Hospital Affiliated to Fujian University of Traditional Chinese Medicine from January 2020 to December 2023 were prospectively included as research samples.According to the random number table method,they were divided into control group(18 cases)and observation group(18 cases).The control group was treated with routine etiological treatment and symptomatic treatment,while the observation group was treated with fire needle acupuncture selection based on CT localization on the basis of the control group.The course of treatment was 28 days.The efficacy improvement rate after 28 days of treatment,visual analogue scale(VAS),le icester cough questionnaire in mandarin-Chinese(LCQ-MC)and levels of peripheral serum cytokines interleukin-6(IL-6)and interleukin-8(IL-8)before and after 28 days of treatment were compared between the two groups.Results:After 28 days of treatment,the efficacy improvement rate of curative effect,VAS score and LCQ-MC score in the observation group were significantly higher than those in the control group(P<0.05).The levels of IL-6 and IL-8 in the two groups were significantly lower than those before treatment(P<0.05),but there was no significant difference between the two groups(P>0.05).Conclusion:The treatment of fire needle acupuncture selection guided by CT localization can significantly improve the therapeutic effect of patients with lung cancer-related cough,effectively reduce the cough symptoms of patients,improve the quality of life of patients,and alleviate the inflammatory response in the body.
