The aim of this paper was to investigate the mechanism of the active peptide DP17 of Eupolyphaga steleophaga in the treatment of hyperlipidemia rats. HPLC and MADIL-TOF/TOF-MS were used for the amino acid sequence analysis and solid-phase synthesis on the active peptide of E. steleophaga which were obtained by biomimetic enzymatic hydrolysis, separation and purification. The hyperlipidemia model was established by feeding with high-fat diet.Twenty days later, the rats in the blank group and the model group were given the saline and the rats in remaining groups were given the corresponding drugs by oral administration. After administration for 4 weeks, the levels of triglyceride(TG), total cholesterol(TC) and low density lipoprotein(LDL) in serum, the levels of TG, TC, adenosine monophosphate(AMP), adenosine triphosphate(ATP) in liver tissues and TG in feces were detected, respectively. Hematoxylin-eosin(HE) staining was used to observe the pathological changes of liver tissues. The Real-time fluorescence quantitative PCR method was used to detect the expression of acetyl coa carboxylase(ACC) and hydroxymethylglutaryl-coa reductase(HMGCR) mRNA in liver tissues. The expression of mammalian target of rapamycin(mTORC1) protein and adenosine 5'-monophosphate-activated protein kinase(AMPK) in liver tissues were detected by Western blot. The analysis showed that the amino acid sequence of active peptide from E. steleophaga was DAVPGAGPAGCHPGAGP(DP17). The results of pharmacological experiments showed that after oral administration of DP17 in rats, the levels of TG, TC and LDL in serum as well as TG and TC levels in liver tissues were significantly decreased(P<0.05), while the levels of AMP, ATP in liver tissues and TG content in feces were significantly increased(P<0.05); the liver steatosis of rats was significantly relieved; the expression of ACC, HMGCR mRNA and mTORC1 protein in liver tissues were significantly reduced, while the expression of AMPK phosphorylated protein was significantly increased(P<0.05). DP17, the active peptide of E. steleophag can significantly reduce lipid accumulation in liver tissues, and it may play a role in reducing blood lipids by regulating the energy metabolism balance in the body and activating AMPK/mTOR signaling pathway.

Intestine, the body's largest digestive and immune organ, always affects the occurrence and development of cardiovascular disease process.Especially the trimethylamine N-oxide as one of the metabolic derivatives produced by intestinal microbiota, can increase the risk of atherosclerosis and promote cardiovascular diseases such as chronic heart failure. Therefore, changing the level of trimethylamine N-oxide in the circulation by taking different measures to intervene the structure, composition and metabolic activity of intestinal flora can affect the occurrence and development of the disease. Thus, the intestinal flora is recognized as a new target for the treatment of cardiovascular diseases.

Objeetive As to the high incidence of arteriovenous fistula(AVF) stenosis, surgical operation will result in the exhaustion of vascular resources in patients, while percutaneous transluminal angioplasty(PTA) can maintain vascular resources for ostomy. However, there is still no clear definition between the choices of PTA and surgical resection. The aim of this study was to compare the efficacy of PTA and surgical resection followed by reconstruction for the treatment of arteriovenous fistula stenosis in order to find appropriate treatment. Methods Retrospective analysis had been done on 46 hemodialysis patients with arteriovenous fistula stenosis in Nanjing BenQ hospital from January 2015 to March 2017,which included 22 cases treated with PTA (PTA group) and 24 cases treated with surgical operation (operation group).Comparison was made in general clinical situation, patency rate at six months after surgery, over patency time and adverse reactions to surgery between the two groups. Results The number of stenoses in PTA group was bigger than that in operation group and the difference was of statistic significance (2.78±1.43 vs 1.67±0.71,P<0.05).There was no significant difference in patency rate between the two groups (P =0.828).There were 57 venous stenoses in PTA group, among which 12 stenoses were anastomotic (21.05%) with 79.3% average stenosis degree and 43 stenoses were at venous outflow tract of fistula (75.44%) with 84.26 average stenosis degree. In PTA group,3 patients had hematoma brachial puncture position and recovered by self-absorption without special treatment. In operation group,1 patient had mild blood oozing and recovered after treatment;4 patients recovered gradually from mild swelling on the back of the hand of the operation side.No difference was found in adverse reactions between two groups (P>0.05).Conclusion PTA treatment is preferred for multiple stenoses(n ≥ 3),which ensures better preservation of vascular resources at a comparable patency rate.

OBJECTIVE To study the mechanism of loganin, the effective componet of Cornus officinalis,on protecting the glomerulus mesangial cell (GMCs) injured by advanced glycation end products (AGEs).METHODS GMCs were divided into control, model (AGEs: 200mg/L),loganin (0.1,1,10μmol/L) groups. The cell proliferation,FN and COL-Ⅳ secretion, microstructure lesions,RAGE,SphK1,S1P and TGF-β protein expressions were measured.RESULTS AGEs induced cell proliferation,FN and COL-Ⅳ secretions, microstructure lesions,RAGE,SphK1,S1P and TGF-β protein expressions were suppressed by loganin.CONCLUSION Loganin can improve the AGEs induced GMCs injury by regulating the AGEs/RAGE/SphK1 signal pathway, and then ameliorate the diabetic nephropathy.

Objective To explore the regularity of time-dependent changes in morphology and biomechanical properties of brain tissues in pigs, and value the feasibility of deducing the postmortem interval (PMI).Methods Brain tissues were taken from 42 pigs and kept in an artificial climate chamber with the temperature of 25 ℃ and humidity of 75%.The samples were collected from telencephalon at sequential time intervals (0,12,24,36,48,60 h;n =6) according to the principle of predefined time, position, direction, ratio, quantity and shape. The samples fixed with formaldehyde were then immediately tested by mechanical testing machine to obtain their biomechanical parameters and the histological sections were prepared. Results With the extension of PMI (0-60 h),brain tissues gradually became discolored, weak, mudding and liquefied under the influence of autolysis and putrefaction. Both clearance area of the white matter and its integrated optical density (IOD) significantly increased during 0-48 h.Biomechanical properties of brain tissues including the limit load, average force, elastic modulus and fracture energy all presented a declining tendency at the interval of 12-60 h.The limit load was considered highly statistically significant, and statistical differences were found in average force, elastic modulus and fracture energy. Conclusions There exists a significantly negative structure-activity relationship between the morphology of brain tissues and biomechanical properties. The limit load of postmortem brain tissues in 60 h is the optimum in the window period, which can be used as a new method for estimating PMI.
