1.Exploration on the mechanism of Xuanfei Dahe Decoction in the treatment of asthmatic mice based on network pharmacology and experimental validation

Objective:To explore the mechanism of Xuanfei Dahe Decoction in treating asthmatic mice through network pharmacology and animal experiments.Methods:The active components and their targets of Xuanfei Dahe Decoction were retrieved from TCMSP, and the asthma targets were obtained by searching the GeneCards, DisGeNet, OMIM, TTD and DrugBank databases. PPI network of intersection targets was constructed using string database and Cytoscape 3.9.0 software. Go function and KEGG pathway enrichment were analyzed by metascape database. The mice were divided into a blank group of 6 mice and a model group of 30 mice according to the random number table method. The asthma model was prepared in the model group. Totally 30 successfully modeled mice were divided into the model group, the dexamethasone group, and Xuanfei Dahe Decoction low-, medium- and high-dosage groups according to the random number table method, with 6 mice in each group. On the 5th day, gavage was initiated. Xuanfei Dahe Decoction low-, medium- and high-dosage groups were respectively gavaged with Xuanfei Dahe Decoction liquid at concentrations of 6.84, 13.68 and 27.36 g/kg. The dexamethasone group was gavaged with dexamethasone acetate tablets at concentrations of 2.73 mg/kg. The blank group and the model group were gavaged with the same volume of sterile normal saline once a day for 14 consecutive days. The pathological changes of lung tissue were observed by HE staining, the level of serum IL-17 was detected by ELISA, and the expression of IL-17 in lung tissue was detected by immunohistochemistry.Results:120 asthma targets and 13 key targets were obtained from Xuanfei Dahe Decoction. Pathway enrichment analysis suggested that IL-17 signaling pathway was one of the key pathways. Compared with the model group, the levels of IL-17 in the serum of the low-dose, medium-dose and high-dose Xuanfei Dahe Decoction groups and the expression of IL-17A in the lung tissue of the medium-dose Xuanfei Dahe Decoction group decreased ( P<0.05). Conclusion:Xuanfei Dahe Decoction may treat asthma by restrain airway inflammation mediated by Th17/IL-17.
2.Anti-inflammatory and antioxidant mechanism of berberine for sepsis rats

Objective To investigate the anti-inflammatory and antioxidant effects of berberine for sepsis rats based on kelch-like ECH-associated protein 1(Keap1)/nuclear factor erythroid 2-relat-ed factor 2(Nrf2)/antioxidant response element(ARE)signaling pathway.Methods Adult health-y rats were randomly divided into normal group(20 rats)and sepsis group(80 rats).The sepsis model in rats of the sepsis group was established by cecal ligation and puncture.According to differ-ent doses of berberine administered by gavage,the sepsis group was further divided into model group(0 mg/kg,without berberine administration),low-dose group(25 mg/kg),medium-dose group(50 mg/kg)and high-dose group(100 mg/kg),with 20 rats in each group.Both the model group and the normal group were given an equal volume of pure water by gavage.Ten rats were randomly selected from each group for a survival test,and they were continuously observed for 5 days to com-pare the survival rates among the groups.Lung,kidney,and liver tissues of rats in each group were collected for hematoxylin-eosin(HE)staining,enzyme-linked immunosorbent assay(ELISA),Western blot,and real-time fluorescence quantitative polymerase chain reaction(qRT-PCR)detection.The pathological tissue changes,serum inflammatory cytokine levels,serum biochemical indicator levels,and the relative expression levels of pathway-related proteins and their mRNAs in each group were observed and compared.Results The survival rates of rats in the medium-dose and high-dose groupsat various time points were all higher than those in the model group,and the differences were statistically significant(P<0.05).Compared with the normal group,the model group showed in-creased levels of serum alanine aminotransferase(ALT),aspartate aminotransferase(AST),serum creatinine(SCr),blood urea nitrogen(BUN),interleukin-6(IL-6),interleukin-1 β(IL-β)and tumor necrosis factor-α(TNF-α).There were obvious pathological injuries in lung,liver,and kid-ney tissues.The relative expression levels of Nrf2 and heme oxygenase-1(HO-1)proteins and their mRNAs in lung tissues were decreased,while the relative expression levels of Keap1 mRNA and Keap1 protein were increased,and the differences were statistically significant(P<0.05).Com-pared with the model group,the serum SCr and BUN levels in the medium-dose group,and the ser-um ALT,AST,SCr and BUN levels in the high-dose group were all decreased,with statistically sig-nificant differences(P<0.05).Compared with the model group,the pathological injuries in lung,liver,and kidney tissues of rats in the low-dose,medium-dose,and high-dose groups were all alle-viated.The relative expression levels of Nrf2 mRNA and HO-1 mRNA as well as Nrf2 and HO-1 proteins in the lung tissues showed a dose-dependent increase,while the relative expression levels of Keap1 mRNA and Keap1 protein exhibited a dose-dependent decrease.The serum levels of IL-6 and TNF-α in the low-dose group,as well as the serum levels of IL-6,IL-1 β and TNF-α in the medium-dose and high-dose groups were all reduced(P<0.05).Conclusion Berberine can alleviate the inflammatory response and activate the antioxidant response in sepsis rats by regulating the Keap1/Nrf2/ARE signaling pathway.

Objective To study the application effect of quality control circle(QCC)in reducing the dissatisfaction rate of physical examination clients in health management center.Methods To establish QCC,selected the health check-up popula-tion in our hospital in September-2019 and March-2020,through the questionnaire investigation and analysis,compare the dis-satisfaction of the clients before and after the quality control circle.Results After carrying out QCC activities,the dissatisfaction of physical examination clients was significantly lower than that before QCC,and the difference was statistically significant(P<0.05).Conclusion The activities of QCC in the health management center can effectively improve the quality of the physical examination work and reduce the dissatisfaction of the customers in the physical examination.It is of great significance to the health management.
4.The clinical application of high-sensitivity cardiac troponin T in diagnosis of non-ST-segment elevation acute coronary syndrome

Objective To investigate the clinical application of high-sensitivity cardiac troponin T (hs-cTnT) in the diagnosis of patients with non-ST-segment elevation acute coronary syndrome (NSTE-ACS). Methods The clinical data of 126 patients with NSTE-ACS admitted to the Department of Emergency of the Third People's Hospital in Yunnan Province from July 2016 to June 2017 were retrospectively analyzed, including 76 patients in non-ST-segment elevation myocardial infarction (NSTEMI) group and 50 patients in unstable angina (UA) group. Moreover, the clinical data of venous blood hs-cTnT and creatine kinase MB (CK-MB) were collected at immediate admission, 4 hours and 12 hours after admission to the hospital in the two groups. At last, the differences in levels of hs-cTnT and CK-MB were compared between the two groups. Results The hs-cTnT and CK-MB in NSTEMI group were gradually increased with the prolongation of therapeutic time, the elevation ratios of hs-cTnT were 88.2% (67 cases) and 100.0% (76 cases), 100.0% (76 cases) at admission and 4 hours, 12 hours after admission respectively; and the elevation ratios of CK-MB were 60.0% (30 cases), 80% (40 cases) and 96% (48 cases) respectively at the above time points; there were no significant differences in UA group. The levels of hs-cTnT and CK-MB in NSTEMI group were significantly higher compared to those of UA group at admission and 4 hours, 12 hours after admission [hs-cTnT (μg/L): the levels were respectively 0.182±0.052 vs. 0.010±0.001, 2.421±0.084 vs. 0.011±0.012, 5.647±0.012 vs. 0.11±0.012, CK-MB (U/L): the levels were respectively 36.3±12.1 vs. 8.1±0.5, 179.6±26.3 vs. 8.5±0.5, 286.0±23.2 vs. 7.9±0.7, all P < 0.05]. Conclusion The blood level of hs-cTnT in acute myocardial infarction (AMI) patients is obviously increased, which can be used as the basis to distinguish NSTEMI from UA.
5.Determination of Dracorhodin and Curcumin in Dieda Pills by UPLC

Objective:To determine the content of dracorhodin and curcumin in Dieda pills by UPLC .Methods:A UPLC method was adopted.The determination was performed on a Waters Acquity UPLC BEH C 18 column(100 mm ×2.1 mm,1.7 μm) with the mobile phase consisting of acetonitrile-0.05 mol· L-1 NaH2PO4(50 ∶50).The detection wavelength was 440 nm for dracorhodin and 431 nm for curcumin, the column temperature was 30℃and the flow rate was 0.1 ml· min-1 .Results:There was a good linear range of 0.001 8-0.036 4 μg(r=0.999 9)for dracorhodin and 0.000 8-0.015 6 μg(r=0.999 9) for curcumin.The average recovery for dracorhodin was 97.94%(RSD=0.89%) and that for curcumin was 98.45%(RSD=0.91%).Conclusion:The method is simple, rapid and reproducible ,which can be used for the determination of dracorhodin and curcumin in Dieda pills .
6.Pharmacodynamics study of Yuyang Powder

AIM: To study the action of Yuyang Powder(Herba Swertiae, Corium Elephantis, Rhizoma Bletillae, etc.) on the experimental gastric ulcer. METHODS: The rat gastric ulcer models were induced by burned acetic acid, irritabillity stimulus, ethanol injure gastric mucosa and pyloric ligation, respectively. The ulcer was obsered and the content of gastric juice, gastric acidity and pepsin were measured. RESULTS: Yuyang Powder could significantly reduce the areas of ulcers, restrain gastric ulcer, promote gastric ulcer heal, reduce gastric juice volume, restrain gastric secretion (in pyloric ligation). CONCLUSION: Yuyang Powder has the preventive and therapeutic in gastric ulcer.
7.Chemical constituents of Isodon. excia (Maxin.) Hara in Changbai Mountain

AIM: To study the chemical constituents of Isodon.excia(Maxin.) Hara in Changbai Mountain. METHODS: Chromatograms column was used to separate and purify the chemical constituents.The structures were elucidated on the basis of physic-chemical properties and spectrum data. RESULTS: Two diterpenoids compounds exciain(Ⅰ) and exciain(Ⅱ),were separated and identified. CONCLUSION: The compound(Ⅰ) is obtained from this plant for the first time.
