1.Study of Single-cell Adhesion Kinetics by Fluidic Force Microscopy

ObjectiveCell adhesion is a critical process that regulates cellular physiological functions. Quantitative characterization of adhesion dynamics is essential for elucidating the intrinsic mechanical mechanisms underlying cellular activities. Although atomic force microscopy-based single-cell force spectroscopy is widely used for single-cell adhesion measurements, it requires complex chemical modifications for preparation of live-cell probes, leading to limitations such as cumbersome operation, low throughput, and potential impacts on cell viability. Fluidic force microscopy, which combines atomic force microscopy with microfluidic probes, is a technique allowing the operation of force-controlled nanopipettes in aqueous environments. By applying negative or positive pressure via a pressure controller, a single living cell can be captured onto or released from the cantilever under physiological conditions. This procedure offers a simple workflow and high assay throughput for single-cell adhesion measurements without the need for chemical functionalization. In this study, fluidic force microscopy-based single-cell force spectroscopy was adopted to achieve long-term quantitative characterization of single-cell adhesion dynamics in a simpler and more efficient manner, comparing the dynamic differences in adhesion establishment between two cell lines with different differentiation levels. MethodsHEK 293T and hTERT RPE-1 cells were non-invasively captured on the cantilever of a fluidic force microscope via its integrated microfluidic system during 40 h of adhesion culture. Cell-substrate detachment assays were performed, and force-distance curves were recorded to extract key mechanical adhesion parameters, including adhesion force, adhesion energy, and maximum detachment distance. These measurements were combined with real-time monitoring of cell spreading area to systematically characterize the dynamic evolution of single-cell adhesion. ResultshTERT RPE-1 cells rapidly entered a stable adhesion phase within 1 h after seeding, with both area-normalized adhesion force and area-normalized adhesion energy reaching peak values. In contrast, HEK 293T cells required 4 h to achieve stable adhesion. Subsequently, the adhesion force, adhesion energy and maximum detachment distance of hTERT RPE-1 and HEK 293T cells stabilized at approximately 240 nN vs. 30 nN, 2.2 pJ vs. 0.12 pJ and 6 μm vs. 4 μm, respectively. hTERT RPE-1 cells reached the peak of area-normalized adhesion parameters earlier than HEK 293T cells, with their peak area-normalized adhesion force and area-normalized adhesion energy being substantially elevated relative to HEK 293T cells. HEK 293T cells presented stronger linear correlations among adhesion energy, maximum detachment distance and adhesion force compared with hTERT RPE-1 cells. For both cell lines, cell spreading area exhibited a weak correlation with adhesion force. Whereas the area-normalized adhesion parameters of HEK 293T cells remained relatively constant throughout the adhesion process, hTERT RPE-1 cells exhibited elevated values in the early phase, followed by a gradual decline. These results indicated distinct dynamic adhesion patterns between the two cell types, with hTERT RPE-1 cells exhibiting stronger adhesion strength and higher adhesion efficiency. ConclusionIn this study, fluidic force microscopy-based single-cell force spectroscopy was successfully applied to perform long-term in situ quantitative measurement of the adhesion dynamics in single adherent cells. The approach revealed divergent adhesion patterns between HEK 293T and hTERT RPE-1 cells, suggesting a close association between cell differentiation and adhesion behaviors. These findings provide quantitative mechanical evidence for further understanding the underlying mechanisms of cell adhesion.
2.Analysis of depressive symptoms and associated factors among primary and secondary school students in the in depth monitoring counties Rural Nutrition Improvement Program

Objective:
To understand the prevalence and related factors of depressive symptoms among primary and secondary school students in the in depth monitoring counties of China s Rural Compulsory Education Nutrition Improvement Program, so as to provide a basis for prevention and psychological intervention of depressive symptoms among children and adolescents in rural areas.
Methods:
In November 2022, a stratified random sampling method was adopted to collect height and weight data, basic personal and family information of 7 949 primary and secondary school students from grade three to grade nine through physical measurements and questionnaires in 56 key monitoring schools implementing the Student Nutrition Improvement Program in 7 in depth monitoring counties (Jalaid Banner in Inner Mongolia, Jinzhai County in Anhui, Mao Xian in Sichuan, Tiandeng County in Guangxi, Mian County in Shaanxi, Zhaozhou County in Heilongjiang and Youxi County in Fujian), and to obtain the information related to their depressive symptoms through the self assessment questionnaire on depression. Multivariate Logistic regression analysis was conducted to analyze the prevalence of depressive symptoms among primary and secondary school students, as well as their related factors.
Results:
The detection rate of depressive symptoms among primary and secondary school students in the in depth monitored counties was 23.5%. Logistic regression analysis showed that the probability of detecting depressive symptoms was higher among female students, middle school students, students whose video screen duration per day was >2 h, and students whose parents marital status was divorced or widowed ( OR =1.40, 1.64, 1.60, 1.24), and students whose sleep duration reached the recommended standard, whose parents usually accompanied them daily for time was 60-<120 min and ≥120 min, and students whose mothers literacy level was middle school graduation had lower probability of detecting depressive symptoms ( OR =0.85, 0.84, 0.71, 0.76) ( P < 0.05 ).
Conclusion
The detection rate of depressive symptoms among students in the in depth monitoring area is high, and targeted interventions need to be developed for students to reduce the risk of mental health problems.
3.Progress in experimental models of viral myocarditis

Viral myocarditis is a myocardial disease resulting from various viral infections.Due to the com-plexity of its pathogenesis,effective prevention and treatment options are currently lacking.Establishing appropriate ex-perimental models is crucial for studying the pathogenic mechanisms,disease progression,early diagnosis,and the devel-opment of new drugs and therapies for viral myocarditis.This article reviews the construction methods,research advance-ments,and applications of common experimental models of viral myocarditis,which range from cell models to small ani-mal models,including mice,hamsters,and rabbits,as well as larger animals such as pigs and non-human primates.Ad-ditionally,we summarize and discuss future research directions,providing a theoretical foundation and technological guid-ance for the prevention and treatment of viral myocarditis in clinical settings.
4.Research progress on cuproptosis inducers overcoming tumor multidrug resistance

In recent years,cancer incidence has been rising.Chemotherapy is a key clinical treatment,yet long-term use of antitumor drugs can lead to multidrug resistance(MDR).This review introduces the causes and mechanisms of MDR,mainly including genomic mutations,cellular autophagy,tumor micro-environment,and apoptosis.Subsequently,the concept and mo-lecular mechanisms of Cuproptosis are introduced,pointing out that the abnormal accumulation of Cu2+in cells and the chain reaction triggered by the binding of excess Cu2+with mitochon-drial enzymes ultimately lead to cell death.Finally,copper ionophores,copper complexes as drugs,and strategies for indu-cing cuproptosis to reverse drug resistance are introduced.The prospects for exploring antitumor drugs that can reverse MDR are briefly discussed,bringing new ideas to overcoming tumor resist-ance.
5.Renal metabolomics study of Huangqi preparations for alleviating nephrotoxicity of cisplatin

Aim To investigate the effect of Huangqi injection(HI)and Huangqi oral solution(HO)on cisplatin-induced nephrotoxicity(CIN)based on un-targeted metabolomics technology and the underlying mechanisms.Methods Sprague Dawley(SD)rats were randomly divided into the blank group,cisplatin(CDDP)model group,HI treatment group,and HO treatment group,then the CIN model was built with low dose multiple intraperitoneal injections of CDDP.Pre-liminary evaluation of the renal protective efficacy of HI and HO was performed by measuring serum creatinine(Scr),blood urea nitrogen(BUN),and organ indi-ces.Further screening and identification of potential biomarkers(PBs)related to CIN and HI/HO pharma-cological effects were attained through metabolomics studies of renal tissues,and pathway enrichment analy-sis was conducted.Results HI and HO significantly restored the abnormal increase in renal function indica-tors and abnormal decrease in organ indices caused by CDDP,as well as significantly improved the abnormal renal metabolic profile induced by CDDP,indicating that both HI and HO had good alleviating effects on CIN.HI significantly reversed 47 out of 54 CIN related PBs,mainly involving metabolic pathways such as glycerophospholipid metabolism,tryptophan metabo-lism,pantothenate and CoA biosynthesis;HO signifi-cantly reversed 18 out of 54 CIN related PBs,mainly involving metabolic pathways such as taurine and hypo-taurine metabolism,ascorbate and aldarate metabo-lism,pentose and glucuronate interconversions.Con-clusions Both HI and HO have significant alleviating effects on CIN.In the short term,HI salleviating effect is superior to that of HO.Overall,the mechanisms by which both alleviate CIN are mainly related to regula-ting lipid metabolism,amino acid metabolism.
6.Analysis on Characteristics of Isokinetic Muscle Strength of the Healthy Lower Limb and Muscle Activity During Single-Leg Jumping Movements in Patients with Acute Non-Contact Anterior Cruciate Ligament Injury

Objective The isokinetic muscle strength indicators and surface electromyography(sEMG)signals of the healthy lower limb side of patients with non-contact anterior cruciate ligament(ACL)injury in the acute phase were compared with those of healthy individuals,so to provide a theoretical basis for the prevention of secondary ACL rupture in the healthy lower limb side of patients with ACL injury.Methods 21 patients with ACL injury in the acute phase(ACL injury group)and 21 healthy individuals(control group)were subjected to isokinetic muscle strength tests for hip flexion,extension,adduction,abduction,and knee flexion,and sEMG signals of the vastus medialis,vastus lateralis,semitendinosus,biceps femoris,and gluteus maximus were collected during the single-leg jumping movements.The differences between the two groups in relative peak torque,antagonist muscle ratio,root mean square amplitude,and co-contraction index were analyzed by independent sample t-test.Results The extension of the knee(P=0.040)and hip flexion strength(P=0.041)were lower in the healthy lower limb side of patients with ACL injury.In the single-leg landing action,there were lower activation of the vastus medialis muscle in the pre-activation phase(P=0.014),lower activation of the vastus medialis(P=0.030)and gluteus maximus(P=0.002)in the reaction phase,and also lower co-activation levels of the biceps femoris-semitendinosus(P=0.020).Conclusions At the early stage of injury,the function of the healthy lower limb side of patients with ACL injury changes,which may indicate why these patients face a greater risk of contralateral lower limb injury.
7.Model establishment for quantitative analysis of saponins of Paris polyphylla by near-infrared spectroscopy

AIM To establish a rapid quantitative analysis model for saponins in Paris polyphylla var.yunnanensis(PPY)by near infrared spectroscopy.METHODS The contents of polyphyllins Ⅰ,Ⅱ,Ⅶ and there total content in PPY were determined by HPLC,while spectral data within the range of 10 000 to 4 000 cm-1 were collected.A quantitative analysis model was established by combining these data with partial least squares regression(PLSR).Multivariate scatter correction(MSC)and vector normalization(SNV)were applied prior to further preprocessing the spectra with original,first-order derivative(1stD),or second-order derivative(2ndD)treatments.Lastly,the model was optimized through non-smoothing(NS),Norris Derivative filtering(Nd),and Savitzky-Golay filtering(S-G)method.Model stability was evaluated based on correlation coefficients and variance.The predicted contents of each saponin component in the validation set samples were calculated.RESULTS The contents of polyphyllins Ⅰ,Ⅱ,Ⅶ were 0.42-17.98,0.46-10.44,0.23-3.86 mg/g,respectively.The total content ranged from 2.91 to 22.1 mg/g.The optimal parameters of three saponins were achieved when selecting the MSC+2ndD+S-G pretreatment method.The corresponding ratio of line segment length to segment gap was 13∶5,15∶5,11∶5,with correlation coefficients of 0.982,0.930,0.958,respectively.The root mean square errors of calibration(RMSEC)were 0.702,0.797,0.238,and the root mean square errors of prediction(RMSEP)were 1.120,0.835,0.304,respectively.The optimal parameters for the total content were obtained when selecting the MSC+2ndD+NS pretreatment method,with a correlation coefficient of 0.970,a RMSEC of 1.090,and a RMSEP of 1.740.CONCLUSION This accurate and rapid method can be used for detection of saponin contents in P.Polyphylla.

Objective This study investigated feasible cases and their significance in promoting the"generalized"development of inspection through digital-intelligent equipment.Methods A qualitative research approach was used,involving interviews conducted between February 2025 and March 2025 with experts in traditional Chinese medicine diagnostics,clinical research methodology,medical engineering integration,and related disciplines,using both online and offline methods.In accordance with the Consolidated Criteria for Reporting Qualitative Research,feasible cases involving the specific application of digital equipment in various parts of observation were collected through item enrichment.The significance of extending observation capabilities via these cases was analyzed,along with the overall implications of integrating digital technologies with traditional inspection method.Results Interviews were completed with 11 experts from domestic universities and research institutes in the fields of traditional Chinese medicine diagnosis,medical engineering integration,and related disciplines.A total of 78 feasible cases of digital-intelligent inspection were identified,along with 69 insights regarding the significance of enhancing the inspection capabilities.These insights were synthesized into two dimensions and 23 holistic meanings.The first dimension is to expand the scope of inspection,including obtaining internal environmental characteristics,observing external environmental characteristics,expanding thermodynamic characteristic data,and crossing time and space.The second dimension is to improve the quality of observation and diagnosis information collection and analysis,including 19 specific meanings,such as standardized collection environment,objective quantification,and refined observation.Conclusion Digital-intelligent equipment plays a significant role in expanding the scope of inspection content and achieving high-quality acquisition and analysis of extensive inspection information.These advancements extend and enrich the capabilities of traditional inspection method in traditional Chinese medicine.
10.Analysis of clinical characteristics of multiple myeloma complicated with second primary malignancies

Objective:This study aims to explore the clinical characteristics of patients with multiple myeloma concurrently presenting with a second primary malignancy.By analyzing the causes based on literature reports,this study aims to deepen the understanding of multiple primary cancers in multiple myeloma patients,and provide some assistance for auxiliary examinations and risk assessments of multiple myeloma patients.Methods:A retrospective analysis was conducted on the clinical data of four patients with multiple myeloma who also had a second primary malignancy admitted to The Second Affiliated Hospital,Chongqing Medical University.Each patient was diagnosed with multiple myeloma through bone marrow aspiration and had a second primary malignancy confirmed through pathological biopsy.Results:The onset age of the 4 patients ranged from 42 to 81 years old;three were males and one was female.One patient had smoldering myeloma with a second primary hematological malignancy(follicular lymphoma),while the other 3 patients had concurrent solid malignancies,including bladder cancer,esophageal cancer,and breast cancer,respectively.Conclusion:Multiple myeloma accompanied by a second primary malignancy is rare,and early pathological biopsy is necessary for diagnosis to avoid missed diagnosis.
