1.Role of sphingolipid metabolism signaling in a novel mouse model of renal osteodystrophy based on transcriptomic approach.

BACKGROUND:
Renal osteodystrophy (ROD) is a skeletal pathology associated with chronic kidney disease-mineral and bone disorder (CKD-MBD) that is characterized by aberrant bone mineralization and remodeling. ROD increases the risk of fracture and mortality in CKD patients. The underlying mechanisms of ROD remain elusive, partially due to the absence of an appropriate animal model. To address this gap, we established a stable mouse model of ROD using an optimized adenine-enriched diet and conducted exploratory analyses through ribonucleic acid sequencing (RNA-seq).
METHODS:
Eight-week-old male C57BL/6J mice were randomly allocated into three groups: control group ( n = 5), adenine and high-phosphate (HP) diet group ( n = 20), and the optimized adenine-containing diet group ( n = 20) for 12 weeks. We assessed the skeletal characteristics of model mice through blood biochemistry, microcomputed tomography (micro-CT), and bone histomorphometry. RNA-seq was utilized to profile gene expression changes of ROD. We elucidated the functions of differentially expressed genes (DEGs) using gene ontology (GO) analysis, Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis, and gene set enrichment analysis (GSEA). DEGs were validated via quantitative real-time polymerase chain reaction (qRT-PCR).
RESULTS:
By the fifth week, adenine followed by an HP diet induced rapid weight loss and high mortality rates in the mouse group, precluding further model development. Mice with optimized adenine diet-induced ROD displayed significant abnormalities in serum creatinine and blood urea nitrogen levels, accompanied by pronounced hyperparathyroidism and hyperphosphatemia. The femur bone mineral density (BMD) of the model mice was lower than that of control mice, with substantial bone loss and cortical porosity. ROD mice exhibited substantial bone turnover with an increase in osteoblast and osteoclast markers. Transcriptomic profiling revealed 1907 genes with upregulated expression and 723 genes with downregulated expression in the femurs of ROD mice relative to those of control mice. Pathway analyses indicated significant enrichment of upregulated genes in the sphingolipid metabolism pathway. The significant upregulation of alkaline ceramidase 1 ( Acer1 ), alkaline ceramidase 2 ( Acer2 ), prosaposin-like 1 ( Psapl1 ), adenosine A1 receptor ( Adora1 ), and sphingosine-1-phosphate receptor 5 ( S1pr5 ) were successfully validated in mouse femurs by qRT-PCR.
CONCLUSIONS
Optimized adenine diet mouse model may be a valuable proxy for studying ROD. RNA-seq analysis revealed that the sphingolipid metabolism pathway is likely a key player in ROD pathogenesis, thereby providing new avenues for therapeutic intervention.

2.Feasibility analysis of bilateral uterine artery embolization via distal radial artery access

Objective To explore the feasibility and safety of bilateral uterine artery embolization(UAE)via distal radial artery access.Methods Thirty patients who underwent bilateral UAE were selected.They were divided into distal radial artery group(14 cases)and femoral artery group(16 cases).The clinical signs,puncture times,operation time,compression hemostasis time,discomfort scores,microcatheter non-use rates,and complication rates of the two groups were analyzed,the feasibility and safety of bilateral UAE via distal radial artery access were evaluated.Results The mean number of puncture times in the distal radial artery group was 1.6 times that of the femoral artery group,and the puncture pain score was 1.5 times that of the femoral artery group(P<0.05).The operation time and puncture point compression hemostasis time in the distal radial artery group were shorter than those in the femoral artery group,and the discomfort score of compression hemostasis in the distal radial artery group was lower than that in the femoral artery group(P<0.01).The proportions who did not use microcatheters in the two groups accounted for 28.6%and 6.3%,respectively,the difference was not statistically significant(P>0.05).Four patients with poor access vessels were found in the distal radial artery group(P<0.05).Conclusion Bilateral UAE via distal radial artery access is safe and feasible.

4.Effect of miR-222-3p in endothelial progenitor cell-derived exosomes on skin wound healing in diabetic mice

Objective To explore the effect of miRNA-222-3p in endothelial progenitor cell-derived exosomes(EPCs-Exo)on skin wound healing in diabetic mice.Methods Endothelial progenitor cell(EPCs)derived from C57BL/6 mouse bone marrow were identified by fluorescence staining.Subsequently,EPCs-Exo isolated from the media of EPCs were identified,and high-throughput sequencing of EPCs-Exo miRNA was completed.The skin injury model of diabetic mice was established.According to different groups,the wounds were treated with externally ap-plied phosphate buffer solution(PBS)and EPCs-Exo;Subcutaneous injection of PBS,agomiR-222-3p,and an-tagomiR-222-3p at the edge of the wound was performed continuously for 14 days,and the wound healing rate was observed.Meanwhile,immunofluorescence methods were used to investigate the changes in the expression levels of ROS,CD31,and VEGF in the wound margin tissue before and after treatment.Results EPCs-Exo significantly ac-celerated skin wound healing in diabetic mice,reduced the level of ROS,and increased the expression level of VEGF and CD31 in the wound margin tissue(P<0.05).High-throughput sequencing showed that miRNA-222-3p was highly expressed in EPCs-Exo.Local subcutaneous injection of miRNA-222-3p into wound margin tissue signifi-cantly promoted the skin wound healing of diabetic mice,reduced the level of ROS,and increased the expression level of VEGF and CD31 in the wound margin tissue(P<0.05).Conclusion MiRNA-222-3p promotes wound healing in diabetic mice and plays an important role in EPCs-Exo promoting wound healing.
5.Analysis of exercise density and exercise load in different physical classes for second year junior high school students from Beijing City

Objective:
To explore the exercise intensity and effective exercise load of different physical fitness classes for second year junior high school students in Beijing, so as to provide a reference for optimizing the teaching content of physical fitness classes, reasonably regulating exercise load and the scientific development of physical fitness.
Methods:
From October to November 2022, 30 second year junior high school students from a middle school in Xicheng District, Beijing were selected to have their heart rate monitored during physical fitness classes using Polar heart rate monitors. Heart rate intervals and training impulse (TRIMP) were used to quantify exercise intensity and load. Independent sample t-test was used to statistically analyze differences in heart rate across different dimensions.
Results:
The average heart rate of students in physical fitness classes was (140.62±9.41) counts/min, with effective exercise load (heart rates ≥120 counts/min) time accounting for 77.2%, and heart rate load of moderate to vigorous physical activity (MVPA) time accounting for 51.9%. The average heart rates for speed, speed endurance, and endurance physical fitness classes were (137.89±8.82) (137.67±11.27) and (145.35±8.98) counts/min, respectively. The average heart rates of male and female students in physical education class were (144.22±24.95) and (136.31±28.78) counts/min, and the difference was not statistically significant ( t=4.04, P >0.05). The cumulative durations of effective exercise load among male and female students were (34.08± 7.52 ) and ( 28.43 ±5.39) min, respectively. The average TRIMP value for physical fitness classes were (100.46±16.56), with a exercise density of 72.06%.
Conclusions
The exercise intensity of physical fitness classes for secondyear junior high school students in Beijing is appropriate, mainly with moderate to vigorous intensity, and the accumulated time of MVPA is sufficient. The average heart rate and exercise density of students meet the requirements. Scientific setting of physical fitness class exercise load should be based on individual differences in students physical fitness, in order to improve the physical health level of middle school students.
