The Janus kinase/signal transducers and activators of transcription (JAK-STAT) control natural killer (NK) cells development and cytotoxic functions, however, whether long non-coding RNAs (lncRNAs) are involved in this pathway remains unknown. We found that miR155HG was elevated in activated NK cells and promoted their proliferation and effector functions in both NK92 and induced-pluripotent stem cells (iPSCs)-derived NK (iPSC-NK) cells, without reliance on its derived miR-155 and micropeptide P155. Mechanistically, miR155HG bound to miR-6756 and relieved its repression of JAK3 expression, thereby promoting the JAK-STAT pathway and enhancing NK cell proliferation and function. Further investigations disclosed that upon cytokine stimulation, STAT3 directly interacts with miR155HG promoter and induces miR155HG transcription. Collectively, we identify a miR155HG-mediated positive feedback loop of the JAK-STAT signaling. Our study will also provide a power target regarding miR155HG for improving NK cell generation and effector function in the field of NK cell adoptive transfer therapy against cancer, especially iPSC-derived NK cells.

A 26-year-old male presented with tremor of bilateral shoulders and hands as the major symptom and also had cognitive and emotional abnormalities for more than 1 year, who was diagnosed as cerebral hepatolenticular degeneration (HLD) in Tongji Hospital of Huazhong University of Science and Technology in October 2021. The serum ceruloplasmin and urine copper levels of the patient were 0.023 g/L and 3760.00 μg/24 h, respectively, and the Kayser-Fleischer (K-F) ring was seen in the cornea. Genetic testing revealed a homozygous mutation of ATP7B gene c.2975C>T (p.Pro992Leu), while transcranial sonography (TCS) showed lenticular nucleus hyper-echogenicity. The literature was searched using hepatolenticular degeneration and transcranial sonography as key words; and 9 articles involving 150 HLD cases were obtained. The lenticular nucleus hyper-echogenicity was presented in 76.9% HLD patients (150/195), while only in 12.7% healthy subjects (17/134) ( P<0.001), suggesting that advanced transcranial sonography can detect the metal deposition and may be used for diagnosis of cerebral HLD.

Objective:To design and compile quality evaluation questionnaire on problem-based learning (PBL) of Neurology and to test its reliability and validity.Methods:Referring to the framework of the evaluation system and according to the characteristics of the discipline, we designed the questionnaire on PBL of Neurology. The clinical medical students who participated in neurologic PBL course were taken as the objects of investigation. The reliability of the questionnaire and the reliability of internal consistency were tested. Correlation analysis and exploratory factor analysis were used to evaluate the validity of the questionnaire.Results:The questionnaire on PBL of Neurology included three first-level indicators: teaching plan quality evaluation, teaching method evaluation and teaching quality evaluation, containing 6 second-level indicators and 25 third-level indicators. The correlation coefficient ρ=0.990 ( P<0.01) showed that the retest reliability was good. The cronbach's α coefficient is 0.901, meeting the internal consistency reliability requirement. And the content validity correlation coefficient was 0.307-0.7 ( P<0.05), indicating that each third-level index had a significant correlation with the total score, and the questionnaire item design was good. The three first-level indicators were used as common factors to evaluate the structural validity by exploratory factor analysis and identify the direction of optimization. Conclusion:The questionnaire has good reliability and validity, which can be used as a tool for scientific evaluation on PBL of neurology, and also provide an optimization direction for further research in the future.

[Objective] To detect and genotype HPV in cutaneous wart tissue.[Methods] This study recruited 132 outpatients with cutaneous warts in the Affiliated Hospital of Zunyi Medical College.Of the 132outpatients,46 had verruca vulgaris,38 had verruca plantaris,and 48 had verruca plana.Ten healthy human and 15 patients with condyloma acuminatum served as the control.Tissue specimens were obtained from the lesions of these patients as well as from the normal skin of the healthy controls.DNA was extracted from the specimens followed by PCR amplification with general and type-specific primers.[Results] Of the 132 wart specimens,128 (97.0％) were positive for HPV,111 (84.1％) for HPV2,89 (67.4％) for multiple types of HPV,55 (41.7％) for two types of HPV,28 (21.2％) for three types of HPV,and 6 (4.6％) for four types of HPV.The most prevalent HPV types were HPV2 followed by HPV1 in verruca vulgaris and verruca plantaris,HPV2 followed by HPV10,HPV3 and HPVl in verruca plana.Multiple types of HPV were identified in 50.0％(23/46) of the verruca vulgaris specimens,76.3％ (29/38) of the verruca plantaris specimens,and 77.1％(37/48) of the verruea plana specimens.Among patients infected with more than one type of HPV,the combination of HPV1 and 2 was the most common (35,39.3％),followed the combination of HPV1,2 and 10(12,13.5％).[Conclusion]s The most prevalent HPV type appears to be HPV2,followed by HPV1,in patients with cutaneous warts in Zunyi area of Guizhou province.HPV2 infection is associated with various types of cutaneous warts.There is a high incidence of coinfection with multiple types of HPV in patients with cutaneous warts,especially in those with verruca plana.Moreover,HPV3 and 10 are newly identified in verruca vulgaris,HPV 10 in verruca plantaris,HPV 1 and HPV7 in veruca plana.
