1.Cervical spine range of motion measurements in flight trainees

Objective To measure the range of cervical spine mobility in flight trainees and analyze its correlation with flight task performance.Methods A total of 55 participants were enrolled(30 in the experimental group,25 in the control group).An independently developed AI-based digital cervical spine function assessment system was used to measure rotational ranges in yaw,pitch,and roll directions,with comparative analysis performed against contralateral rotation.A subjective scoring scale was designed,and flight instructors of the experimental group were invited to provide performance ratings to explore the relationship between cervical mobility and flight task performance.Results Significant differences were observed between the experimental and control groups in left rotation(P<0.01),right rotation(P<0.05),and flexion(P<0.05).Within the experimental group,significant differences were found between left and right rotation(P<0.01)and between flexion and extension(P<0.01).Correlation analysis between flight performance scores and cervical mobility revealed a significant association between situational awareness and yaw range(r=-0.415,P<0.05).Conclusion The experimental group exhibited significant differences in certain cervical mobility parameters compared to the control group,suggesting that occupational characteristics may influence cervical mobility.The correlation between yaw range and situational awareness implies a potential link between head-neck yaw capability and vestibular-visual system functions,though further validation is required.

Objective:To investigate the effect of Jaceosidin on immune escape of colorectal cancer(CRC)cells by regulating cyclic guanosine monophosphate-adenylate synthase(cGAS)-stimulator of interferon gene(STING)signal pathway.Methods:Human CRC cells HCT116 were cultured in vitro and grouped into control group,Jaceosidin-L group(25 μmol/L),Jaceosidin-M group(50 μmol/L),Jaceosidin-H group(100 μmol/L),activator group[100 μmol/L Jaceosidin+10 μmol/L cGAS activator manganese chloride(MnCl2·4H2O)],and inhibitor group(100 μmol/L Jaceosidin+1 μmol/L cGAS inhibitor RU.521);CCK-8 method was applied to de-tect the proliferation of HCT116 cells;flow cytometry was applied to detect apoptosis of HCT116 cells;HCT116 cells were co-cultured with NK cell to detect NK cell killing activity;ELISA was applied to detect the levels of IFN-γ and Granzyme B in the supernatant of co cultured cells;Western blot and qRT-PCR were applied to detect the expression of cGAS-STING signaling pathway and apoptosis related factors.Results:Compared with the control group,the A450 value and Bcl-2 expression of HCT116 cells in the Jaceosidin-L,Ja-ceosidin-M,and Jaceosidin-H groups were obviously reduced,the apoptosis rate,and the expression of cGAS,STING and Bax were obviously increased,and were dose-dependent(P<0.05);compared with the control co culture group,the levels of IFN-γ and Gran-zyme B,and NK cell killing activity in the supernatant of the Jaceosidin-L,Jaceosidin-M,and Jaceosidin-H co culture groups were significantly increased,in a dose-dependent manner(P<0.05);cGAS activator MnCl2·4H2O enhanced the inhibitory effects of high-dose Jaceosidin on HCT116 cell proliferation,immune escape,and the promoting effect on cell apoptosis,cGAS inhibitor RU.521 weakened the inhibitory effects of high-dose Jaceosidin on HCT116 cell proliferation,immune escape,and the promoting effect on cell apoptosis.Conclusion:Jaceosidin inhibits HCT116 cell proliferation,immune escape,and promotes cell apoptosis by activating cGAS-STING signaling pathway.
4.Synthesis and anti-breast cancer activity of novel cyclic mono-carbonyl curcumin analogues

OBJECTIVE To design and synthesize mono-carbonyl curcumin analogues(MCACs)and investigate the activities of them against breast cancer.METHODS The analogues F1,F2,and F3 were obtained by aldol condensation reaction,and their antitumor activities(including the activities of human breast cancer cell MCF-7 and human lung cancer cell A549)were detected by MTT assay[evaluated with half inhibitory concentration(IC50)].The results of MTT assay were compared with those of curcumin.Bioinformatics methods were used to collect the core targets of analogues F1,F2 and F3 acting on breast cancer,and then molecular docking verification was carried out.The cell experiments were conducted to investigate the effects of high,medium and low concentrations(16,8,4 μmol/L)of F1,F2 and F3 on the expression of the first core target protein as well as the effects of medium concentration of F1,F2 and F3 on the expression of cleaved-caspase-3.RESULTS Compared with curcumin,IC50 of analogues F1,F2 and F3 to A549 and MCF-7 cells(except for IC50 of analogue F2 to A549 cells)were decreased significantly(P<0.05 or P<0.01);among them,IC50of analogue F2 to MCF-7 cell was the lowest,being(9.67±1.27)μmol/L.Bioinformatics analysis showed that index of affinity of analogues F1,F2 and F3 with the first core target epidermal growth factor receptor(EGFR),protein kinase B(AKT)and AKT were 5.909 2,8.402 5 and 6.486 6,respectively;high concentration of F1 could significantly reduce the phosphorylation level of EGFR protein in MCF-7 cells(P<0.01),while low,medium,and high concentrations of F2 and high concentration of F3 could significantly reduce the phosphorylation level of AKT protein in MCF-7 cells(P<0.05 or P<0.01).Medium concentration of F1,F2,and F3 could significantly increase the expression level of cleaved-caspase-3 protein in MCF-7 cells(P<0.01).CONCLUSIONS Designed and synthesized MCACs F1,F2 and F3 all have good anti-breast cancer activity,and F2 has better anti-breast cancer activity.

6.Research progress of injection fear related assessment tools

Injection fear is widespread in the population, which can cause patients to tolerate or avoid injection, reduce treatment compliance, and increase the burden of healthcare. Choosing appropriate injection fear assessment tools in clinical practice is helpful to understand the degree, psychological characteristics and influencing factors of individual injection fear. In this paper, the contents, characteristics and application methods of fear of injection assessment tools at home and abroad are reviewed, in order to provide reference for the application and development of fear of injection assessment tools for medical staff.
7.Effects of web standardized patients in online clinical nursing practice of undergraduate nursing students in Psychiatry Department

Objective:To evaluate the teaching effect of online clinical nursing practice in Psychiatry Department for undergraduate nursing students by using Web standardized patients (Web-SP) .Methods:In June 2021, 219 undergraduate nursing students from a university in Beijing were selected as research subjects by convenient sampling. We organized a week of online nursing practice in Psychiatry Department, set up an online practice teaching and research group, recruited and trained 15 standardized patients (SP) among hospital nurses, and designed 12 roles including schizophrenia patients, depression patients, and bipolar disorder patients with typical symptoms according to the actual clinical cases. According to the practice plan, an online practice class was opened on Tencent conference platform. The clinical teachers broadcastedand interviewed SP and SP's family members on the spot. Studentsinteracted with the online teachers and SP through video, voice or text. After the practice, 219 undergraduate nursing students were issued SP Simulated Quality of Standardized Patient Scale (SQSPS) and self-made Web-SP Use Effect Evaluation Questionnaire in Psychiatry Department online. A total of 219 questionnaires were distributed online, and 219 valid questionnaires were recovered, with a valid recovery rate of 100%.Results:The total score of SQSPS of 219 nursing students was (92.08±10.89) . The overall evaluation score of nursing students on the application of Web-SP in online practice was (46.52±4.60) , and the highest score was "deepened empathy for patients", which was (4.79±0.47) . The second was "deepened the understanding of psychiatric nursing", which was (4.75±0.68) . The third was "understanding the impact of symptoms on patients", which was (4.73±0.52) .Conclusions:Applying Web-SP to online clinical nursing practice in Psychiatry Department can meet the practice needs of nursing students for Psychiatric Department and nursing courses, and Web-SP after systematic training can replace real patients to achieve teaching goals.
8.Study on Clear Optic Evaluation Method of Intraocular Lenses.

The study explored the methods of determining the diameter of clear optic of intraocular lenses by analyzing the design principles and influencing factors of clear optic. Measure and compare the clear optic diameters of intraocular lenses from -10.0 D to 36.0 D respectively. The measurement results of the two methods have good consistency and can be used in the measurement of clear optic. But considering the accuracy and convenience of actual measurement, profile projector measurement principle is more suitable for conventional mass production testing. The Schlieren technique method can provide more useful information in product development and feature description stage, and can be used as an auxiliary verification method for clear optic diameter measurement.

10.Anterior Gradient 3 Promotes Breast Cancer Development and Chemotherapy Response

Purpose:
Anterior gradient 3 (AGR3) belongs to human anterior gradient (AGR) family. The function of AGR3 on cancer remains unknown. This research aimed to investigate if AGR3 had prognostic values in invasive ductal carcinoma (IDC) of breast cancer and could promote tumor progression.
Materials and Methods:
AGR3 expression was detected in breast benign lesions, ductal carcinoma in situ and IDC by immunohistochemistry analysis. AGR3’s correlations with clinicopathological features and prognosis of IDC patients were analyzed. By cell function experiments, collagen gel droplet-embedded culture drug sensitivity test and cytotoxic analysis, AGR3’s impacts on proliferation, invasion ability, and chemotherapeutic drug sensitivity of breast cancer cells were also detected.
Results:
AGR3 was up-regulated in luminal subtype of histological grade I-II of IDC patients and positively correlated with high risks of recurrence and distant metastasis. AGR3 high expression could lead to bone or liver metastasis and predict poor prognosis of luminal B. In cell lines, AGR3 could promote proliferation and invasion ability of breast cancer cells which were consistent with clinical analysis. Besides, AGR3 could indicate poor prognosis of breast cancer patients treated with taxane but a favorable prognosis with 5-fluoropyrimidines. And breast cancer cells with AGR3 high expression were resistant to taxane but sensitive to 5-fluoropyrimidines.
Conclusion
AGR3 might be a potential prognostic indicator in luminal B subtype of IDC patients of histological grade I-II. And patients with AGR3 high expression should be treated with chemotherapy regimens consisting of 5-fluoropyrimidines but no taxane.
