Objective:
To explore the network structure of middle school students basic psychological needs and psychological behavioral problems, and identify the core nodes within the network, as well as examine demographic subgroup differences, so as to provide support for targeted mental health interventions for adolescents.
Methods:
In September and October of 2023, a total of 2 000 junior and senior high school students were selected with multistage cluster random sampling from 8 schools in Jiaojiang District and Tiantai County, Taizhou City. An online self administered questionnaire was used to assess emotional and behavioral problems, perceived autonomy, self awareness, loneliness, and social support. The instruments included the Strengths and Difficulties Questionnaire (SDQ), Perceived Choice and Awareness of Self Scale (PCASS), Mental Health Literacy Questionnaire (MHLQ), University of California,Los Angeles Loneliness Scale (UCLA-LS), and the Multidimensional Scale of Perceived Social Support (MSPSS). A network analysis approach was employed to construct a network representing adolescents basic psychological needs and psychological behavioral problems, focusing on centrality measures and demographic subgroup differences.
Results:
A total of 418 students (20.9%) reported abnormal emotional and behavioral problems. Perceived autonomy and competence were negatively correlated with emotional problems (weights: 0.12, 0.14) and hyperactivity (weights: 0.10, 0.16). Social support showed negative correlation with peer relationship issues, hyperactivity, and conduct problems (weights: 0.16, 0.13, 0.10). Loneliness was positively correlated with emotional symptoms and peer relationship problems (weights: 0.28, 0.18). In the overall network, perceived relationships (social support and loneliness), emotional symptoms, and hyperactivity emerged as central nodes. Significant differences in network structure were observed between gender subgroups ( P =0.02). Girls internalizing issues were more influenced by loneliness and perceived autonomy frustration, while social support exhibited higher centrality in boys.
Conclusions
Perceived relationships, emotional problems, and hyperactivity are key nodes in the network of adolescents basic psychological needs and psychological behavioral problems. Loneliness demonstrates a prominent influence within the network, and the overall network exhibits gender differences.

Here we report the case of a CBF-AML patient with KIT p.D816V mutation who failed avapritinib induction therapy and subsequently underwent bridging allogeneic hematopoietic stem cell transplantation (allo-HSCT), along with a literature review. The patient was a 64-year-old male who did not achieve remission after induction therapy with the DA regimen (Daunorubicin + Cytarabine). After reinduction with avapritinib combined with the DCHG regimen (Decitabine + Homoharringtonine + Cytarabine + Granulocyte colony-stimulating factor), he attained complete remission (CR) and flow cytometry minimal residual disease (MRD) negativity, but the RUNX1::RUNX1T1 fusion gene remained positive. During consolidation therapy, flow MRD reappeared, and the fusion gene level progressively increased. The patient then underwent a 9/10 HLA-matched unrelated donor allo-HSCT. Post-transplant, the fusion gene persisted, prompting treatment with the "ITI" regimen (with dose adjustments, including sequential addition of interferon and interleukin-2, pomalidomide incorporation, and standard vs. escalated dosing of "ITI" regimen). Currently, MRD negativity has been maintained for over 5 months, with good treatment tolerance. This finding suggest that the "ITI" regimen may serve as a novel and well-tolerated therapeutic option for CBF-AML patients with persistent RUNX1::RUNX1T1 fusion gene positivity after allo-HSCT and KIT p.D816V mutation, particularly in cases of avapritinib treatment failure.

Senecavirus A (SVA) is a major viral pathogen causing disease in pigs, and effective monitoring of SVA infection is critical for disease control. In this study, we aimed to develop a reliable ELISA method for rapidly detecting neutralizing antibodies against SVA. We used HEK293F cells to express an SVA-specific porcine Fab antibody and verified the biological activity of the Fab antibody by indirect ELISA, immunofluorescence assay, virus neutralization test, and Western blotting. The Fab antibody was biotinylated and used as a competitive antibody to establish a competitive ELISA (C-ELISA) for detecting neutralizing antibodies against SVA. We then evaluated the C-ELISA in terms of sensitivity, specificity, repeatability, and result agreement rate with the VNT. The results showed that we successfully prepared an SVA-specific porcine Fab antibody, which showed high affinity for SVA. We named this antibody 1M33Fab and designated it as Bio-1M33Fab after biotin labeling. The assay conditions were optimized as follows: the coating concentration of SVA particles being 1 μg/mL, the working concentration of Bio-1M33Fab being 0.5 μg/mL, the optimal serum dilution of 1:10, and the optimal dilution of enzyme-labeled avidin being 1:30 000. At a percent inhibition (PI) of 47%, the assay demonstrated the highest sensitivity (96.88%) and specificity (100%), with no cross-reactivity observed with the positive sera of major porcine viral diseases. The intra-assay coefficient of variation ranged from 1.12% to 7.34%, while the inter-assay coefficient of variation ranged from 1.10% to 8.97%, indicating good repeatability. In the detection of 224 clinical pig serum samples, C-ELISA and VNT showed a result agreement rate of 93.75%. In conclusion, we successfully develop a C-ELISA method for detecting neutralizing antibodies against SVA by using a porcine-derived Fab antibody, which lays a foundation for the development of detection kits.

Objective To explore the relationship between the polymorphism of excision repair cross-complementation group 1 (ERCC1) C8092A locus and the efficacy and prognosis of platinum-based chemotherapy for lung cancer (LC), and to provide a theoretical basis for precision treatment of LC. Methods From January 2014 to October 2017, 120 patients from two tertiary hospitals in Haikou City, and with pathologically confirmed lung cancer treated with platinum-based chemotherapy were selected as the research objects. After informed consent was obtained, peripheral blood samples were collected for DNA extraction, and the genotype of ERCC1 C8092A locus was detected by mass spectrometry. WHO's Response Evaluation Criteria in Solid Tumours (RECIST) was used to judge patients' chemotherapy efficacy and patients' survival status was obtained by telephone follow-up and other means. Results Among the 120 LC patients, the genotype frequencies of ERCC1 C8092A locus were 67 cases of CC wildtype (55.8%), 45 cases of CA heterozygous type (37.5%), and 8 cases of AA rare mutation type (6.7%), which conformed to Hardy-Weinberg equilibrium (χ2=0.140, P>0.05). The total effective rate of chemotherapy was 32.5%, with the highest effective rate in patients with the CA genotype (42.2%) at the ERCC1 C8092A locus and the lowest in patients with the CC genotype (25.4%). The overall one-year survival rate was 68.3% and the three-year survival rate was 35.8%. The patients with ERCC1 C8092A AA genotype had the lowest survival rate, with a one-year survival rate of 50.0% and three-year survival rate of only 25.0%. However, there were no statistical differences in the overall survival rate among the three genotypes of carriers of ERCC1 C8092A (χ2=0.328, P=0.849). Conclusions The polymorphism of ERCC1 C8092A locus is associated with the efficacy of platinum-based chemotherapy for LC, and patients with CA genotype have the highest efficacy. The one-year and three-year survival rates of patients with CC genotype are significantly higher than those of CA and AA genotypes.

The inflammatory response is a systemic process produced by the body in response to stimulation such as infection or injury. The inflammatory response is closely related to the pathogenesis of various diseases, and plays an important role in the progression and outcome of diseases.Under normal conditions, the inflammatory response can maintain tissue homeostasis when the body is not seriously damaged. However,an uncontrolled inflammatory response that occurs when the body is subjected to more severe stimuli may often cause serious harm to the body. The termination of the ideal inflammatory response occurs after the elimination of the inflammatory stimulus. Therefore,it is particularly important to effectively modulate the extent and scope of the inflammatory response.β-arrestin 1(ARRB1)is a multifunctional regulatory protein that plays a key role in G protein-coupled receptor(GPCR)and non-GPCR-mediated signal transduction.Meanwhile, this protein also participates in the regulation of some immune cell development, differentiation and other functions.Although ARRB1 was originally thought to be a terminator of GPCR signal transduction and can inhibit the inflammatory response, recent studies suggest that its role in inflammatory responses is complex, with dual anti-inflammatory and pro-inflammatory effects.Based on this, this article reviews the relevant research on ARRB1 and the inflammatory response in recent years, focusing on the regulatory role of ARRB1 in signaling pathway transduction and immune cell development and differentiation function,as well as its mechanism of action in regulating the progression of inflammatory diseases,in order to provide new insights for the clinical precision treatment of inflammatory diseases and the screening of drug effect targets.

Objective:To analyze human papillomavirus (HPV) infection types and clinical characteristics in postmenopausal patients with high-grade squamous intraepithelial lesion or cervical cancer. Methods:The clinical data of 475 patients diagnosed with high-grade squamous intraepithelial lesions or cervical cancer, pathologically confirmed by colposcopic biopsy, were retrospectively analyzed. These patients received treatment at The Affiliated Suqian Hospital of Xuzhou Medical University (Nanjing Drum Tower Hospital Group Suqian Hospital) between September 2021 and September 2022. The patients were divided into two groups based on their menopausal status: the menopausal group and the non-menopausal group. The clinical data and the types of high-risk HPV infections were compared between the two groups. Results:Among the 475 patients, there were 142 in the menopausal group, with an average age of (57.63 ± 6.23) years, and 333 in the non-menopausal group, with an average age of (36.79 ± 6.85) years. The proportion of patients with no prior history of cervical cancer screening and those who sought medical attention due to clinical symptoms was higher in the menopausal group than in the non-menopausal group [23.2% (33/142) vs. 5.4% (18/333), χ2 = 33.03, P < 0.05]. Among the 475 patients, 347 underwent HPV typing tests, with the top five HPV types identified as 16, 52, 58, 33, and 18. The positive rate of HPV 16 was lower in the menopausal group compared with the non-menopausal group [43.8% (32/73) vs. 61.3% (168/274), χ2 = 7.53, P < 0.05], while the positive rate of HPV 52 was higher in the menopausal group [28.8% (21/73) vs. 13.5% (37/274), χ2 = 9.64, P < 0.05]. The proportion of multiple infections was also higher in the menopausal group compared with the non-menopausal group [47.9% (35/73) vs. 31.8% (87/274), χ2 = 6.63, P < 0.05]. The proportion of patients with type 3 transformation zone in the menopausal group was higher [63.4% (90/142) vs. 23.4% (78/333), χ2 = 69.52, P < 0.05], as was the proportion of cervical canal curettage [59.9% (85/142) vs. 33.6% (112/333), χ2 = 28.20, P < 0.05] compared with the non-menopausal group. The proportion of patients with cervical cancer in the menopausal group was higher than in the non-menopausal group [35.2% (50/142) vs. 8.7% (29/333), χ2 = 50.42, P < 0.05]. The proportion of patients with cervical cancer at stage ≥ IB in the menopausal group was 90% (45/50). Conclusion:In patients with high-grade squamous intraepithelial lesions or cervical cancer, HPV 16 is the most common type. In the menopausal group, there is a high proportion of patients who are positive for HPV 52, an elevated rate of multiple infections, a low cervical screening rate, and a high incidence of cancer, with many patients presenting at late stages.
