1.Qishen Granule protects against myocardial ischemia by promoting angiogenesis through BMP2-Dll4-Notch1 pathway.

OBJECTIVE:
Therapeutic angiogenesis has become a promising approach for treating ischemic heart disease (IHD). The present study aims to investigate the effects of Qishen Granule (QSG) on angiogenesis in myocardial ischemia (MI) and the potential mechanism.
METHODS:
In vivo study was conducted on rat model of myocardial infarction. QSG was performed daily at a dose of 2.352 g/kg for four weeks. Cardiac function was assessed by echocardiogram and pro-angiogenic effects were evaluated by Laser Doppler and CD31 expression. Oxygen-glucose deprivation (OGD) was applied in cultured human umbilical vein endothelial cells (HUVECs). Cell viability, wound healing and tube formation assay were used to test functions of HUVECs. ELISA and Western blots were used to assess protein expressions of bone morphogenetic protein 2-delta-like 4-notch homolog 1 (BMP2-Dll4-Notch1) signaling pathway.
RESULTS:
The results showed that QSG improved heart function, cardiac blood flow and microvessel density in myocardial ischemic rats. In vitro, QSG protected HUVECs by promoting the cell viability and tube formation. QSG upregulated bone morphogenetic protein-2 (BMP2) and downregulated delta-like 4 (Dll4) and notch homolog 1 (Notch1) expressions both in rats and HUVECs.
CONCLUSION
QSG protected against MI by promoting angiogenesis through BMP2-Dll4-Notch1 pathway. BMP2 might be a promising therapeutic target for IHD.
2.Relation of weight change after twenty on subsequent diabetes risk

Objective:To explore the association between weight change from age 20 to middle age and diabetes risk.Methods:A total of 10 104 residents over 40 years old were recruited in Guangzhou. The final analysis data set is a cohort of 6 272 patients from the REACTION study. Quartile of weight changes from age 20 to middle-age was used as an independent variable to explore its association with newly diagnosed diabetes. Simple and multivariable logistic regression analysis were used to calculate OR and 95% CI. Results:The prevalence of newly diagnosed diabetes in study cohort was 15.7%. Weight changes from age 20 to middle-age was significantly higher in diabetes group( P<0.001). Logistic regression analysis found in general population and overweight people [24.0≤body mass index(BMI)<28.0 kg/m 2], after adjusting for gender, age, educational level, family history of diabetes, drinking, SBP, waist circumference, cholesterol, high-density lipoprotein-cholesterol(HDL-C), TG, ALT, AST, the risk of diabetes increased in line with the magnitude of weight change( P=0.081). While in group of lean, normal and obese adult, no significant association has been found. Conclusion:The risk of diabetes increased with weight change in general population and overweight people, indicating that weight change in adults after age of 20 was positively associated with diabetes. It is suggested the importance of weight management in the prevention of dialetes.
3.Study on associated protein markers in urine of patients with chronic renal failure

Objective To investigate the associated protein markers in urine of patients with chronic renal failure (CRF) based on surface enhanced laser desorption and ionization time of flight mass spectrometry (SELDI-TOF MS)technique.Methods Urine samples were taken from 150 CRF patients and 50 healthy people,and investigated by proteomic techniques with H4 gene chip.Results Compared to healthy group,141 different protein peaks were identified within the range of 1000 to 20 000 M/Z in the protein map of CRF group,whose differences were all significant (all P<0.01).The decision tree model for CRF urine was constructed after bioinformation analysis to significantly differentiate between CRF and healthy group.The accuracy rate,sensitivity and specificity of the decision tree model were 96.0%,100.0% and 94.7% respectively.In CRF group and healthy group,the different protein peaks in urine were identified to probably be 17 proteins with reference to SwissProt database.Conclusions Candidate protein markers in urine are screened and prediction model of CRF urine is established.The markers are identified with the database which provides a more accurate prediction and solid evidence for early diagnosis of CRF.
4.Research of the EEMD method to pulse analysis of traditional Chinese medicine based on different amplitudes of the added white noise.

The ensemble empirical mode decomposition (EEMD) can be used to overcome the mode mixing problem of empirical mode decomposition (EMD) effectively. The EEMD method and Hilbert-Huang Transform (HHT) can be used to analyze pulse signals of Traditional Chinese Medicine (TCM). The amplitudes of the added white noise were about 0.1 and 0.2 time standard deviation of the investigated signal respectively. The difference of average frequency and average energy of every mode between normal pulse, slippery pulse, wiry pulse and wiry-slippery pulse were demonstrated based on different amplitudes of the added white noise. The results showed that it is more in line with clinical practice when the amplitude of the added white noise is about 0.2 time standard deviation of the investigated signal.

5.Investigation in status quo of evidence awareness of internship nursing students and countermeasures

Objective In order to grasp the status quo of the internship nursing students' evidence awareness in our province, to improve the teaching measures, and to promote nursing safety. Methods We delivered questionnaires to 480 nursing students who were exercising practice in some major hospitals in our province about their contemporary consciousness of evidence, the results underwent analysis. Results The results showed that the awareness of nursing evidence was very poor in the new period. Conclusions It's imperative to enhance the normative education about evidence awareness among the nursing students.
6.The effect of short hairpin RNA and DNAzyme gene therapy on latent membrane protein-1 expression in nasopharyngeal carcinoma.

OBJECTIVE:
To study the inhibitory effects of short hairpin RNA (pshLMP1) combined with DNAzyme or alone on latent membrane protein-1 (LMP1)expression in HNE1 cell lines.
METHOD:
The pshLMP1 and DNAzymes were co-transfected with pEGFP-N1-1158 (a expression vector encoding LMP1 and EGFP fusion protein) into HNE1 cells, divided into 4 groups as positive control, RNA interference, combining and DNAzyme groups. The green fluorescence were analyzed and then the mRNA and protein of LMP1 gene were detected.
RESULT:
The HNE1 cells expressing green fluorescence in combining group were significantly less than that in RNA interference group (P < 0.01); The inhibition efficiency of green fluorescence in combining group were 86. 31% Wp 88.88% ,which were higher than that in RNA interference group with 75.15%. The detection of LMP1 mRNA and proteins showed that combining group had a higher inhibition ability.
CONCLUSION
Combining gene therapy with pshLMP1 and DNAzyme is superior to pshLMP1 alone to inhibit LMP1 gene expression.

7.Analysis of EBV-LMP1 gene mutation in plasma of patients with nasopharyngeal carcinoma in Chongqing.

OBJECTIVE:
To detect and analyze the EBV-LMP1 gene N terminus XhoI-site mutation and C terminus 30 bp deletion in plasma of patients with nasopharyngeal carcinoma (NPC) in Chongqing of China.
METHOD:
DNA extraction and PCR amplification was used in plasma of 48 NPC patients and 40 control non-NPC cases from Chongqing of China. All the PCR production of N terminus was digested by enzyme XhoI, then segregated in 8% PAGE. All- the PCR production of C terminus was segregated in 8% PAGE too. Compared with B95-8 cell, some of the PCR production were sequenced and analyzed with software.
RESULT:
LMP1 was amplified successfully from 48 of 48 NPC cases (100%) and from 38 of 40 non-NPC cases (95%) by PCR, but none of LMP1 XhoI-site mutation and none of 30 bp deletion was found by digesting, segregating and Sequencing.
CONCLUSION
EBV-LMP1 gene N terminus XhoI-site and C terminus 30 bp deletion have no mutation in plasma of NPC or non-NPC cases in Chongqing of China. The precise relationship of EBV-LMP1 gene mutation or deletion with NPC pathogenesis needs further investigation.

Objective:To explore the relationship of optimal pulse-taking pressure among cun,guan,chi pulse of of healthy persons.Methods:264 Healthy undergraduate were included and pulse signals were collected from cun,guan,chi in cunkou area.The relationship of optimal pulse-taking pressure among three-region pulse,ratios and body mass index(BMI)were researched.At the same time,to calculate relationship of optimal pulse-taking pressure among cun,guan,chi pulse.Results:The optimal pulse-taking pressure of cun,guan,chi pulse was positively correlated with BMI.The optimal pulse-taking pressure of guan and cun pulse was significantly smaller than that of chi pulse.The optimal pulse-taking pressure of cun pulse is similar to that of guan pulse,which was 90 percents of the chi pulse.Conclusion:There was significant difference in optimal pulse-taking pressure between cun,guan,chi pulse.The result was favourable to the optimizing the program of pulse device,consummating of three-probe pulse sensor and studying pulse thoroughly.
