Objective:To construct a matching model between difficulty grading of peripheral venipuncture in infants and nurse's venipuncture competence, and to evaluate its effectiveness. Methods:Convenience sampling was used to select pediatric patients who were treated with peripheral venous infusion in the infusion room of the Affiliated Xuzhou Children's Hospital of Xuzhou Medical University from March to June 2024, and to establish a model for matching the difficulty grading of peripheral venipuncture in infants and the nurse's venipuncture competence. Pediatric patients before the implementation of the matching model (March to April 2024) were set up as control group, and pediatric patients after the implementation of the matching model (May to June 2024) were set up as observation group. The success rate of first peripheral venipuncture was counted and compared between the two groups. Results:The per capita first puncture success rate of the nurses in control group was (93.94±4.67) %, and the per capita first puncture success rate of the nurses in observation group was (96.10±2.65) %, and the difference was statistically significant ( P<0.05) . Conclusions:The establishment of a matching model between difficulty grading of peripheral venipuncture in infants and nurse's venipuncture competence can improve the success rate of first pediatric peripheral venipuncture and promote a steady improvement in the technical competence of pediatric venipuncture nurses.

Objective:To establish a rapid detection method for human astrovirus based on TaqMan-probe real-time fluorescent quantitative reverse transcription-polymerase chain reaction (RT-PCR).Methods:According to the conservative sequence of human astrovirus ORF1 b gene, we designed the amplification primers and specific fluorescent probe to establish the human astrovirus TaqMan real-time fluorescent quantitative RT-PCR rapid detection method. The specificity, sensitivity and stability of the method were evaluated. We also used this method to detect human astrovirus in clinical samples. Results:The established human astrovirus TaqMan real-time fluorescent quantitative RT-PCR detection method has good specificity and repeatability for human astrovirus, and the sensitivity can reach 10 2 copies/μl. After testing the clinical samples, the detection rate of human astrovirus by our method was 100%. Conclusions:The human astrovirus TaqMan real-time fluorescent quantitative RT-PCR detection method established in this study is simple, rapid, sensitive, specific and stable. It can be used for clinical human astrovirus detection and epidemiological investigation.

Objective To study the Chlamydia trachomatis(CT)and Ureaplasma urealyticum (UU)infection in Guangzhou area, and analyze the consistency of simultaneous amplification and testing (SAT)and conventional methods(CT was detected by latex immunochromatography, UU was detected by liquid culture method). Methods A total of 12 120 samples of urogenital secretions or urine samples were collected from Sun Yat-Sen Memorial Hospital of Sun Yat-Sen University from January 2015 to December 2016.CT-RNA and UU-RNA were detected by the SAT technique, a part of samples were tested by conventional methods at the same time. The positive rates of CT and UU by SAT and the conventional methods between different gender and age groups were analyzed by χ2test, the consistencies between different detection methods were analyzed by Kappa test. Results The positive rate of CT was 4.05%(356/8 781), UU 33.69%(1 125/3 339)in Guangzhou from 2015 to 2016.The positive rate of UU was significantly higher than that of CT(χ2=1 981,P<0.01).Of 145 specimens for CT test, the coincidence rate between SAT and latex immunochromatographic method was 96.55%(140/145), which showed good consistency(Kappa=0.65).Of 186 specimens for UU test, the coincidence rate of the results between the SAT method and liquid culture was 92.47%(172/186),which showed strong consistency(Kappa=0.81). Conclusions The positive rate of UU was significantly higher than that of CT in Guangzhou. The SAT method and conventional methods to detect CT and UU show high consistency, which can provide the evidence for clinical diagnosis of CT and UU infection.
