1.Application of Raman spectroscopy in the quality control and in vitro permeation studies of topical drug formulations

Transdermal drug delivery offers advantages such as safety, convenience, and high patient compliance. However, the complex structure of the skin and significant individual variability, particularly the barrier function of the stratum corneum, result in generally low bioavailability for topical formulations. The formulation of topical drug products is complex, with numerous factors influencing quality, which requires strict control of product quality. Raman spectroscopy, as a non-destructive vibrational technique, combined with chemometric methods, imaging technology, and other spectroscopic techniques, can be applied to study the key quality attributes of topical drug formulations. The applications of Raman spectroscopy in studies of the crystal form, particle size distribution, excipient research, and in vitro transdermal experiments of topical drug formulations were summarized. In particular, it focused on the use of Raman spectroscopy in the selection of skin samples for in vitro permeation tests, the study of drug spatial distribution in the skin, and the interactions between drugs and the skin.
2.Clinical study on the reconstruction of webbed toes with advanced kite-flap after syndactyl split in children

Objective:To investigate the clinical effect of the proximal interphalangeal kite flap in repairing the wound of the posterior web of the split toe.Methods:A retrospective case series study was used to analyze the clinical data of children with syndactyly after toe web wounds were repaired with proximal kite flap between toe webs in the Yidu Central Hospital of Weifang from May 2010 to March 2019. The satisfaction of flap repair effect (the total score is 5-10) and the operation effect according to toe function, toe web slope, and depth were evaluated. The measurement data of normal distribution were expressed in Mean±SD and analyzed by paired sample t-test. Results:A total of 10 children with syndactyly were included, including four males and six females, aged from 6 months to 9 years. Seven cases were on the right side, and there were on the left. All were soft tissue connexions, incomplete syndactyly 6 feet, complete syndactyly 4 feet. All flaps survived and were followed up for an average of 13 months (range, 6-24 months). The score of satisfaction was 8.6±2.7. The appearance, color, elasticity, and texture of the reconstructed web are similar to those of the uninjured side. At the last follow-up, there was no significant difference between the abduction of the separated toe and that of the healthy side (43.57 ± 3.82)° and (44.39 ± 4.25)°( P=0.64). There was no significant difference between the two groups ( P=0.66). The reconstruction depth of the toe web was (1.19 ± 0.23) cm, which was not significantly different from that of the healthy side (1.21 ± 0.27) cm ( P=0.85). Conclusions:The proximal interphalangeal kite flap is one of the ideal methods to repair the wounds of the posterior web of the congenital syndactyly. However, the number of cases in this study is small, and the follow-up time is short. The curative effect of the children with the growth and development process needs further research.

4.The epidemiological characteristics and clinical diagnosis and treatment of 43 cases of novel coronavirus pneumonia

Objective:To retrospectively analyze the epidemiological and clinical characteristics of novel coronavirus pneumonia (COVID-19), and to provide evidence for its further prevention and control.Methods:The epidemiological history, clinical symptoms, pulmonary CT changes, treatment plan (hormone intervention or not), and negative conversion time of nucleic acid were summarized and analyzed in 43 COVID-19 patients.Results:All the 43 patients had clear contact history, among which the number of patients transmitted from conference microphone was the largest (13 cases). There were 2 cases of critical disease, 4 cases of severe disease and 37 cases of common type. The main symptoms were fever (30 cases, 69.8%) and cough (40 cases, 72.1%). Lung CT mainly showed single or multiple ground glass shadows (40 cases), which were all absorbed by the treated lesions, among which 28 cases had a transient aggravation of the lesions during the treatment. All the 43 cases were treated with integrated traditional Chinese and western medicine, 2 cases in critical condition were treated with non-invasive ventilator, and 19 cases were treated with low-dose hormone. The average nucleic acid negative conversion time was 13.28 days, and the use of hormones had no significant effect on the negative conversion time ( P>0.05). Conclusions:COVID-19 of common type is the commonest. Early respiratory support therapy is recommended for critical cases. Hormone intervention makes no obvious influence on the negative conversion time of nucleic acid. Close contact in a closed environment greatly increases the risk of transmission.
5.Design of a dual wavelength multi-channel laser acupuncture instrument

Objective To design a laser acupuncture instrument with dual wavelength and multi-channel,under the guidance of traditional Chinese medicine acupuncture theory and according to low-level laser induced effect in bio-tissues and compatibility characteristics of acupuncture points.Methods Using SCM and semiconductor laser,hardware circuitry was designed to achieve the basic functions of the instrument,and the corresponding PC software was developed.Results The laser acupuncture treatment for several acupuncture points could be carried out at the same time with the same or different laser parameters (wavelength,power density,continuous/pulse),and each channel could output 635 nm and 808 nm laser,with continuous adjustable power.Conclusions The dual wavelength multi-channel laser acupuncture instrument can meet the needs of compatibility during clinical acupuncture treatment and is easy to operate.
6.Evaluation of Removing Pyrogen Effect of Activated Carbon in Traditional Chinese Medicine Injection Technics

Objective To establish the pyrogen removing effect of activated carbon in the technics of traditional Chinese medicine (TCM) injections. Methods The content of bacterial endotoxin concentration was detected by kinetic turdimetric assay to evaluate the effect of removing pyrogen before and after using activated carbon in concentrated solution of TCM injection. Results The activated carbon adsorption rate of Shuanghuanglian concentrated solution≥70%and the activated carbon adsorption rate of Danshen concentrated solution>95%. Conclusion Pyrogen manufacturing process is scientific and rational by adding activated carbon adsorption in powder injection of TCM. The bacterial endotoxin of large doses can't be removed fully by activated carbon adsorption.
7.Investigating the killing effect of photodynamic therapy induced by different photosensitizers on human leukemia cell in vitro

Objective To investigate and compare the killing effect of photodynamic therapy (PDT)induced by hematoporphyrin derivative (HpD),hematoporphyrin monomethyl ether (HMME) and photocarcinorin (PsD007) on human leukemia cells K562 in vitro.Methods Human leukemia cells were cultured with serial concentrations of photosensitizers followed by irradiation of different dosage of laser light,then MTT colorimetric assay was applied to measure the relative survival rate of PDT for the cells.Results Significant difference in the inhibitory between the PDT group and control group was observed (P<0.05).The survival rate of PDT for the cells elevated along with the increase in the concentration of sensitizer and dose of laser light.When the photosensitizer concentration was bigger (25 μg/ml) or the energy density was bigger (7.2 J/cm2),the effect of PsD007 was better than HMME,and they were significantly better than HpD (P<0.05).Conclusion PDT has significant killing effect on human leukemia cells K562,and its relative inhibitory rate appears to be correlated with the dose of sensitizer and laser light irritation.The effect of PDT is related to the photosensitizers.The effect of HpD-PDT is not as effective as PsD007 and HMME.On the conditions of higher energy density and larger photosensitizer concentration,the effect of PsD007-PDT is better than HMME-PDT.
8.Apoptosis induced by HMME mediated photodynamic therapy in murine hepatoma cell line MM45T-Li

Objective To investigate the effects of HMME mediated photodynamic therapy (PDT) on apoptosis of murine hepatoma cell line MM45T-Li.MethodsThe photodynamic treatment on MM45T-Li cells was performed in vitro using HMME as photosensitizer and 630 nm laser as light source.The ceils were incubated with HMME of 2.5,5,10 and 20 μg/ml for 4 h respectively,and then irradiated with laser at different dosages.The dark toxicity of HMME and the activity of cells after PDT were evaluated by MTT assay.The effect of PDT on apoptosis were observed by Hoechst fluorescent staining analysis.ResultsMTT assay showed that there were no significant effects on cell growth in each concentration group of HMME when the cells were not exposed to light.The inhibition ratio of cells increased with laser dosage at HMME concentration of 10 and 20 μg/ml.Chromatin condensation,nuclear pyknosis and karyorrhexis were observed after PDT by Hoechst fluorescent staining analysis.The apoptosis rate increased in groups of 5.4 J/cm2 and 7.2 J/cm2 compared with that of control (P<0.05).Conclusion HMME mediated PDT can efficiently induce the apoptosis of MM45T-Li cells.
