1.Scoping review of factors influencing social alienation in stroke survivors

Objective:To conduct a scoping review of the current state of research on social alienation in stroke survivors, its influencing factors, and assessment tools, and to provide a theoretical basis as well as a reference for clinical prevention of social alienation in stroke survivors.Methods:A systematic search was conducted for relevant literatures on the factors affecting social alienation of stroke survivors in domestic and foreign databases such as PubMed, Cochrane Library, Wanfang Database, etc, and the search period was established until April 29, 2024. After a rigorous assessment of quality, the pertinent literature was condensed and scrutinized.Results:A total of 11 literatures were included, with 9 Chinese literatures and 2 English literatures, the research instrument was based on the General Detachment Scale, involved 22 influencing factors of social alienation among stroke survivors.Conclusions:Stroke survivors lack specific assessment tools for social alienation. In the future, localized and specific assessment tools should be developed, and targeted intervention measures should be formulated based on the influencing factors related to the social alienation of stroke survivors to improve their social alienation.

3.Establishment and application of LAMP-Taqman detection method of porcine pseudorabies virus

A LAMP-Taqman rapid detection system for porcine pseudorabies virus(PRV)was de-veloped based on LAMP and quantitative PCR.LAMP primers were designed for PRV conserved sequences,and the loop primer modified by the fluorescent quenching group was used as the Taq-man probe.The composition optimization,specificity,sensitivity and repeatability of the LAMP-Taqman system were tested using positive samples and recombinant plasmid as templates.Thirty-eight samples of pork swabs were tested with the commercial LAMP detection kit in parallel to verify the actual detection effect of the LAMP-Taqman detection system.The results showed that the optimal final concentration of each component was as follows:PRV-FIP/BIP 0.8 μmol/L,Bst DNA polymerase 0.7 U/μL,Taq DNA polymerase 0.24 U/μL,dNTPs 1.6 mmol/L,MgSO47.2 mmol/L.This system had good specificity and did not cross-react with other virus samples.The linear correlation coefficient of gradient samples was 0.995,the coefficient of variation of repeatable tests was less than 3.000%,and the minimum detection limit could reach 2.81 ×102 copies/μL.The test results of the actual swab samples were consistent with the commercial isothermal fluores-cence detection reagents.In conclusion,the PRV detection system established by LAMP-Taqman method in this study is specific,sensitive,stable and accurate,and is a reliable technical method suitable for the accurate detection of porcine PRV.

This article analyzed the necessity of multi-component exercise, the insufficiency and prospect of multi-component exercise in elderly patients with sarcopenia, and provides certain reference significance for medical personnel to carry out multi-component exercise intervention in elderly patients with sarcopenia in China.
5.Early outcome of transapical transcatheter aortic valve replacement for aortic insufficiency

Objective To assess the early outcome of transapical transcatheter aortic valve replacement (TAVR) for patients with aortic insufficiency. Methods The patients with aortic valvular disease who underwent transapical TAVR from October 2020 to October 2022 in the Department of Cardiac and Vascular Surgery, the First Affiliated Hospital of Anhui Medical University were enrolled in the current retrospective study. The patients with aortic stenosis were assembled in a group A, and the patients with aortic insufficiency were assembled in a group B. The improvements of heart function and complications were assessed for the two groups. Results A total of 56 patients were enrolled, including 32 males and 24 females with an average age of 73.34±5.10 years. There were 31 patients in the group A and 25 patients in the group B. There was no statistical difference between the two groups in the age, gender, height, weight, hypertension, coronary artery disease, peripheral vascular disease, chronic obstructive pulmonary disease, renal disorder or classification of heart function (P>0.05). Also, there was still no statistical difference in the rate of permanent peacemaker implants, emergent open surgery, valve re-implants, or perivalvular leakage (P>0.05). After TAVR, the left ventricular diastolic diameter, left ventricular ejection fraction, complicated moderated mitral and tricuspid regurgitation were significantly improved in both groups compared with preoperative findings (P<0.05); however, there was no statistical difference in these parameters between groups (P>0.05). Conclusion Interventional valve (J-Valve) in the treatment of patients with aortic insufficiency through transapical TAVR significantly improves cardiac function and reduces functional valve regurgitation.
6.Advances in liquid chromatography-mass spectrometry analysis of several important secondary metabolites in plant metabolomics.

Metabolomics, which mainly studies the metabolite components of organisms, tissues, cells and their dynamic changes, is an emerging omics technology following genomics and proteomics. Metabolites are the final products of cellular regulation, and the concentration of metabolites is considered to be the ultimate response of a biological system to genetic or environmental changes. Secondary metabolites with chemical diversity are widely present in living organisms, thus accurate quantification of secondary metabolites through appropriate analytical platforms is an important task of metabolomics. Liquid chromatography-tandem mass spectrometry (LC-MS/MS) is the most commonly used method for the detection of metabolites, providing a basis for the wide application of plant secondary metabolites. This review summarizes the advances of using LC-MS/MS techniques for the detection of phytohormone, folic acid, flavonoids and other secondary metabolites.

7.Investigation and analysis of the correlation between TCM constitution and fatigue of the seafarers undertaking long-term maritime work

Objective:To analyze the incidence of fatigue in the seafarers undertaking long-term maritime work and the correlation between Traditional Chinese Medicine(TCM)constitution and fatigue,so as to provide basic data support for formulating constitution conditioning programs.Methods:An epidemiological survey of fatigue and TCM constitution was conducted in 425 males who had been working at sea for 50 days by cross-sectional sampling method with TCM constitution scale and modified Fatigue Impact Scale.Results:The number of people with balanced constitution was the largest,accounting for 49.18%;the top three imbalanced constitutions were qi deficiency constitution(11.29%), yang deficiency constitution(9.41%),and phlegm dampness constitution(9.18%). The overall fatigue rate was 16.71%. The fatigue rate of people with balanced constitution was the lowest,accounting for 0.96%;among the people with imbalanced constitutions,the fatigue rate of people with qi stagnation was the highest(58.33%),followed by the people with qi deficiency(45.83%). Statistical analysis based on the average rank of fatigue scores showed that the fatigue score of the people with balanced constitution was significantly lower than those of the people with imbalanced constitutions( P<0.05). Conclusion:Balanced constitution, qi deficiency constitution,and yang deficiency constitution are the common TCM constitution types in the seafarers undertaking long-term maritime work. There is a certain correlation between TCM constitution and fatigue. The people with qi stagnation and qi deficiency constitutions are more prone to fatigue.

9.Preventive effect and mechanism of hyperbaric oxygen on chemotherapy-induced peripheral neuropathic pain

Objective:To investigate the effect of hyperbaric oxygen (HBO) on the prevention of chemotherapy-induced peripheral neuropathic pain (CIPNP), and to observe its mechanism by targeting spinal cannabinoid receptors (CBRs).Methods:A total of 75 male Sprague-Dawley(SD) rats were randomly divided into 5 groups (15 rats in each group), i. e. blank control group, CIPNP control group, CIPNP+ HBO group, CIPNP+ HBO+ AM630 group, and CIPNP+ HBO+ AM251 group. The model rats with CIPNP were established by injecting paclitaxel (i.p.). Each group with HBO intervention received the HBO treatment on the second day after each of the 5 times of paclitaxel injection. The CIPNP+ HBO+ AM630 and CIPNP+ HBO+ AM251 groups were administered with AM630 (an antagonist of CBR2) and AM251 (an antagonist of CBR1) respectively before each HBO treatment. The behavioral test was carried out to measure the mechanical withdrawal threshold (MWT) of rats by von fery filaments before the experiment and every 7 days during the experiment. The expressions of CBR1 and CBR2 were tested by Western blotting. The expression of glial fibrillary acidic protein (GFAP) was tested by immunohistochemistry (ICH) and Western blotting. And the expressions of inflammatory cytokines in the spinal cord, i. e. Interleukin-1β (IL-1β) and tumor necrosis factor-α (TNF-α), were detected by enzyme-linked immunosorbent assay (ELISA).Results:Compared with that of the blank control group, the MWT of the CIPNP control group was significantly decreased ( P<0.01), and the difference was most significant [(15.46±2.83) g vs. (4.33±3.53) g; P<0.01] especially on the 21st day of the experiment. The expressions of spinal GFAP, IL-1β, and TNF-α were significantly increased, and the differences were statistically significant ( P<0.05). Compared with those of the CIPNP control group, the MWT and spinal CBR2 of the CIPNP+ HBO group were significantly increased ( P<0.05), the GFAP, IL-1β, and TNF-α in the spinal cord were significantly decreased ( P<0.05), and the above-mentioned effects could be blocked by intraperitoneal injection of AM630, while there was no such reverse effect after intraperitoneal injection of AM251. Conclusion:HBO can prevent paclitaxel-induced CIPNP, and its mechanism may be related to the activation of spinal CBR2 and then the blocking of the activation of GFAP and the expression of inflammatory cytokines in the spinal cord.
