Objective:This study aims to explore the independent and interactive effects of childhood trauma (CT) and major depressive disorder (MDD) on amygdala subregion volumes and to examine whether volumetric changes in these subregions mediate the relationship between CT and depressive severity. Methods:A total of 129 MDD patients and 127 age- and sex-matched healthy controls were recruited from Nanjing Brain Hospital between October 2022 and November 2024. All participants underwent 3D-T 1 weighted MRI scans, and amygdala subregions were segmented using the FreeSurfer software. Depressive and anxiety symptoms were assessed with the 17-item Hamilton Depression Rating Scale (HAMD 17) and the Hamilton Anxiety Scale (HAMA),respectively. Childhood trauma exposure was evaluated via the Childhood Trauma Questionnaire (CTQ). Generalized linear models (GLM) were applied to analyze the main and interactive effects of MDD diagnosis (depression/healthy controls) and CT (presence/absence),adjusting for age, estimated intracranial volume, sex, medication history, and education years. Partial correlation and mediation analyses were conducted to explore associations between amygdala subregion volumes and clinical measures in MDD patients. Results:MDD diagnosis was independently associated with increased volumes in the right central nucleus ( Wald χ2=9.09, P=0.026) and medial nucleus ( Wald χ2=10.08, P=0.026). CT exposure was independently associated with reduced volumes in the right central nucleus ( Wald χ2=7.99, P=0.047) and medial nucleus ( Wald χ2=9.20, P=0.047). No significant interaction effects between MDD and CT were observed in any amygdala subregion. Mediation analysis revealed that reduced right medial nucleus volume partially mediated the relationship between total CTQ scores and depressive severity (proportion mediated: 26.69%,95% CI=0.002-0.060) and mediated the association between emotional neglect and depressive severity (proportion mediated: 26.75%,95% CI=0.006-0.150). Such mediating effects were not found for the right central nucleus. Conclusion:CT and MDD exhibit divergent patterns of influence on amygdala subregions. CT is linked to volumetric reductions, whereas MDD is associated with volumetric enlargement. Reduced volume of the right medial nucleus mediates the relationship between CT and depression severity.

Tendons play a crucial role in the motor system. However, tendon injuries can result in pain and function decline, a condition known as tendinopathy. Pain is often the earliest symptom, significantly impacting the lives and work of patients. There are many methods for addressing tendinopathy-associated pain, but the treatment is lengthy and the outcomes are not satisfactory. The main cause is that the mechanism of tendinopathy-associated pain is not completely clear yet. Therefore, it is of great value to identify the mechanism of tendinopathy-associated pain in order to discover new therapeutic strategies. To this end, the authors reviewed the research progress in the mechanisms of tendinopathy-associated pain, including changes in tissue structure, pain mediators, central regulation, etc, to provide a reference for researches on the mechanisms and clinical treatment of tendinopathy-associated pain.

Objective:To investigate the expression of macrophage migration inhibitory factor (MIF) in bone marrow fluid and peripheral blood of patients with acute myeloid leukemia (AML) and its effect on the expression of interleukin-8 (IL-8) in bone marrow mesenchymal stem cells (BM-MSC).Methods:Fifty bone marrow fluid samples and 50 peripheral blood samples were collected from 50 patients with AML diagnosed in the First People's Hospital of Yunnan Province from October 2017 to January 2019, of which 17 patients were newly diagnosed, 26 patients were complete remission (CR), and 7 patients were partial remission (PR) or non-remission (NR). Fifty plasma samples from 50 healthy subjects and 50 bone marrow fluid samples from 50 patients with iron deficiency anemia were used as the controls. Enzyme-linked immunosorbent assay (ELISA) was used to detect the level of MIF protein in the samples, and the relationship between MIF expression level and clinicopathological characteristics of AML patients was analyzed. BM-MSC was successfully isolated and cultured from 42 bone marrow fluid samples of AML patients, the suitable samples for experiment were chosen and divided into BM-MSC control group (untreated BM-MSC), recombinant human macrophage migration inhibitory factor (rhMIF) group and rhMIF+ISO-1 group. ELISA and real-time fluorescence quantitative polymerase chain reaction were used to detect the expression level of IL-8 protein and mRNA in each BM-MSC group. Results:The expression levels of MIF protein in bone marrow fluid and plasma in AML group were (24.9±7.7) ng/ml and (60.5±12.1) ng/ml, the difference was statistically significant ( P < 0.01), and those in control group were (5.3±2.6) ng/ml and (2.0±1.1) ng/ml, respectively, and there were statistical differences between the two groups (t values were 136.71, 33.97 and 17.58, all P < 0.01). MIF protein expression levels in bone marrow fluid and plasma of AML patients in newly diagnosed group and PR+NR group were higher than those in CR group, and the differences were statistically significant (all P < 0.01). MIF protein expression levels were higher in bone marrow fluid and plasma of patients with ≥60 years of age, peripheral blood white blood cell count ≥30×10 9/L and bone marrow myeloblast ratio > 0.50 (all P < 0.05), but the differences were not statistically significant between patients with different gender (both P > 0.05). The detection results of each BM-MSC group showed that rhMIF promoted the IL-8 expression in BM-MSC at the gene and protein levels, which could be inhibited by the MIF inhibitor ISO-1 (all P < 0.01). Conclusion:The increased expression levels of MIF in bone marrow fluid and plasma of patients with AML are associated with the disease progression, and rhMIF can promote the expression of IL-8 in BM-MSC.

Methods Literatures from CNKI, WANFANG MED ONLINE and CQVIP were organized with Excel, we searched literatures on status quo of extended care for patients with hypertension in China from lst January 2007 to 31st December 2016,and counted with the bibliometric method. And the number, publication year, author, source journal, fund, research type, implementation of extended care, evaluation method and research subjects of these literatures were analyzed. Results Totally 187 literatures were included in this study. The number of literatures increased year by year since 2011. Guangdong, Jiangsu and He'nan were the top 3 provinces where these literatures came, which accounted for 61 literatures (32.62%). 64.70% of the literatures came from general hospitals. A total of 11 journals published more than 4 literatures. There were 15 funded papers among the literatures (8.02%). The research type mainly was randomized controlled trial (136 literatures), but only 43 of them explained their specific random method (31.62%). There was no standard for intervention personnel, and the intervention time varied from 1 to 12 months. The control state, quality of life, treatment compliance and health behavior were research highlights in extended care. Conclusions In recent years, the number of literatures on extended care for patients with hypertension has increased year by year, but the quality of these literatures needs to be improved. Nursing staffs shall carry out evidence-based nursing, apply for more research fund, and establish an extended care model which fits the patients with hypertension in China as soon as possible.
