OBJECTIVES:
To explore the bioactive components in Jiawei Xiaoyao Pills (JWXYP) and their mechanisms for alleviating depression-like behaviors.
METHODS:
The active compounds, key targets, and pathways of JWXYP were identified using TCMSP and TCMIP databases. Thirty-six SD rats were randomized equally into 6 groups including a control group and 5 chronic unpredictable mild stress (CUMS)-induced depression groups. After modeling, the 5 model groups were treated with daily gavage of normal saline, 1.8 mg/kg fluoxetine hydrochloride (positive control drug), or JWXYP at 1.44, 2.88, and 4.32 g/kg. The depression-like behaviors of the rats were evaluated using behavioral tests, and pathological changes in the liver and hippocampus were examined with HE staining. The biochemical indicators in the serum and brain tissues were detected using ELISA. Serum metabolomics analysis was performed to identify the differential metabolites using OPLS-DA, and gut microbiota changes were analyzed using 16S rDNA sequencing.
RESULTS:
Network pharmacology revealed that menthone and paeonol in JWXYP were capable of penetrating the blood-brain barrier to regulate inflammatory pathways and protect the nervous system. In the rat models subjected to CUMS, treatment with JWXYP significantly improved body weight loss, sucrose preference and open field activities, reduced liver inflammation, alleviated structural changes in the hippocampal neurons, decreased serum levels of TNF‑α, IL-1β, IL-6 and LBP, and increased 5-HT and VIP concentrations in the serum and brain tissue, and these effects were the most pronounced in the high-dose group. Metabolomics analysis showed changes in such metabolites as indole-3-acetamide and acetyl-L-carnitine in JWXYP-treated rats, involving the pathways for bile acid biosynthesis and amino acid metabolism. 16S rDNA analysis demonstrated increased gut microbiota diversity and increased abundance of Lactobacillus species in JWXYP-treated rats.
CONCLUSIONS
JWXYP alleviates depression-like symptoms in rats by regulating the neurotransmitters, inhibiting inflammation and oxidation, and modulating gut microbiota.

Objective To investigate the prevalence of influenza virus infections in infants and young children during the pandemic period of 2009 influenza A(H1N1)in Beijing. Methods Throat swabs were collected from children visited the affiliated Children's Hospital to Capital Institute of Pediatrics for influenza-like illness from June 1,2009 to February 28,2010.The specific gene segments of 2009 pandemic influenza H1N1 and seasonal influenza viruses were amplified from samples by real-time RT-PCR recommended by WHO and National Influenza Reference Center of China. Results Out of 4363 clinical samples tested by real-time RT-PCR,the total positive rate of influenza A viruses was 29.3%,including 623(14.3%)identified as 2009 pandemic influenza A(H1N1)and 657(15.1%)influenza A viruses without subtype identity.Among those pandemic influenza H1N1 positive,23 were severe cases with 5 deaths. The ages for 618 pandemic influenza H1N1 infected children with completed information were from 14 days to 16 years. The ratio of male to female wag 1.3:1.Among them,25.2% were patients in age group of 1 to 3 years old and distribution of children in age groups of 3 to 6 years old and 6 to 12 years old were similar(about 30.0%).During the survey period,it appeared only one prevalence wave of pandemic influenza H1N1.The positive rate of pandemic H1N1 increased in September and the peak(36.5%of positive rate)was in November and then declined to 2.7%in February 2010.The data from routine influenza virus surveillance from 20-30 clinical samples collected each week indicated an alternative prevalence of seasonal H3N2,pandemic H1N1 and influenza B during this study period.Respiratory syncytial virus(RSV)became predominant in children after the circulating of pandemic H1N1.Conclusion There was an epidemic of pandemic influenza H1N1 in children in Beijing from June 2009 to February 2010,especially in those of preschool and school aged children.Seasonal influenza viruses and pandemic influenza H1N1 were contributed alternatively.

Objective To explore the influence of pottery and weaving behavior therapy on rehabilitation of patients with schizophrenia. Methods 60 inpatients accorded with the diagnosis standard of CCMD-3 schizophrenia were selected as the research object, and were randomly divided into the intervention group and the control group. The antipsyehotie therapy wag adopted in the two groups. The intervention group received the pottery and weaving behavior therapy, and the control group only received the routine numing. Before and 12 weeks after the pottery and weaving behavior therapy, the patients were assessed and compared with NOSIE-30 (nurse observation scale for inpatient evaluation-30).Results Before the pottery and weaving behavior therapy.there was no statistical meaning in the scores of 7 factors(society ability, society interest, personal neatness, irritable bowel, representation of schizophrenia, depression and tardy factor)and the 3 total scores(P>0.05)in patients of the two groups.12 weeks after the pottery and weaving behavior therapy, the scores of NOSIE-30 in the intervention group were as follows.the total estimated score of disease station of (173.13±13.18),the total active score of(78.87±8.45),the society ability of (30.93±4.71),the society interest of (25.00±2.67) and the personal neatness factor of(22.93±3.27).These scores were obviously higher than those of the centrel group.with a statistical meaning(P<0.01).While the scores of irritable bowel(14.20±2.85),depression(6.27±3.23)and tardy factor(14.33±2.47)were obviously lower than those of the control group, with a statistical meaning(P<0.01).Conclusions The pottery and weaving behavior therapy improves the rehabilitation of society funetion of schizophrenia patients and ameliorates their part symptoms. So it has assistant function for rehabilitation of schizophrenia patients.
