1.Genetic characteristics of VP1 region of Coxsackievirus A10 in Yunnan Province

Objective To investigate the genetic characteristics of the VP1 region of Coxsackievirus A10 (CVA10) in Yunnan Province. Methods Fecal samples of suspected hand, foot and mouth disease (HFMD) were subjected to real-time fluorescent quantitative PCR for detection of enterovirus CVA10. Positive samples were subjected to VP1 gene sequence amplification and Sanger sequencing. Sequence splicing was performed with DNAstar7.1 Seqman software, and nucleotide sequence and amino acid site analysis were performed using Mega 6.0 software. Results The sequencing of VP1 gene of CVA10 obtained a sequence of 894 nucleotides, encoding 298 amino acids. Compared with the original strain, there were mainly three active amino acid mutation regions, 13-33, 141-142, and 283-285. The nucleotide difference rate between the Yunnan isolates and the reference strain ranged from 16.92% to 30.90%, and the amino acid difference rate ranged from 2.58% to 4.00%. C1 and C2 group nucleotide difference was 10.58%, and the amino acid difference rate was 1.80%. The VP1 150-176 region exhibited highly conserved characteristics. Six CVA10 strains and Sichuan strain MW178898 belonged to the C1 group of the C genotype. The other 14 CVA10 strains belonged to the C2 group. Conclusion VP1 gene mutation is active and CVA10 is an important pathogen of HFMD in Yunnan. C2 genotype of CVA10 is dominant in this study, and C1 and C2 have co-circulated in Yunnan. It is necessary to strengthen monitoring and develop multivalent vaccines containing CVA10 epidemic genotype.
2.Analysis of research topics on child and adolescent healthcare in community nursing based on LDA model

Objective:To explore the current status and thematic content of child and adolescent healthcare in community nursing, so as to provide reference for research on child and adolescent healthcare in community nursing.Methods:Literature on child and adolescent healthcare in community nursing published from January 2004 to November 2024 in the Web of Science core collection database was systematically searched. Bibliometric methods were used to analyze the number of publications, journal distribution, and country distribution. The themes were analyzed using the latent Dirichlet allocation (LDA) model.Results:A total of 17 240 articles were included. The overall number of articles issued showed an upward trend year by year. Out of a total of 200 source publications, PLoS One ranked highest in terms of the number of articles issued (674) . A total of nine themes were obtained based on the LDA model, which further resulted in three merge topics, namely, the family environment of children and adolescents in community nursing and risk factors, the prevention and treatment of diseases and public health of children and adolescents in community nursing, and the social issues and mental health concerns of child and adolescent healthcare in community nursing. Conclusions:The research on child and adolescent healthcare in community nursing is generally on the rise, and the thematic analysis and mining of related studies provides a reference for child and adolescent healthcare services and related research in community nursing in China.
3.Cognition and influencing factors of rural child caregivers in the context of rural revitalization: a qualitative study

Objective:To explore in depth the nurturing cognition of caregivers of children aged 1 to 7 years in rural areas in the context of rural revitalization, and to reveal the facilitating and hindering factors from the perspective of positive psychology, so as to provide scientific basis for improving the "family-centered" child nurturing care services in rural revitalization areas.Methods:This study utilized a descriptive qualitative research methodology. Semi-structured in-depth interviews were conducted in July 2024 with 20 visiting patients at the Outpatient Clinic of Shuangbai Maternal and Child Health Hospital in Chuxiong Yi Autonomous Prefecture of Yunnan Province. The data was organized and analyzed through the content analysis method.Results:Caregivers' nurturing cognition in the area were centered around three core themes, including perceptions and learning about nurturing, factors facilitating positive nurturing, and factors hindering positive nurturing.Conclusions:Caregivers in rural revitalization areas continue to face multidimensional challenges in their nurturing cognition and lack positive traits. The family factor is a significant barrier to the establishment of positive nurturing cognition. Healthcare professionals should make use of the perspective of positive psychology to analyze in-depth the cognition of caregivers and their influencing factors, and improve the "family-centered" child nurturing care services in rural revitalization areas.

Objective:To do genetic analysis on the VP1 gene of 2 Coxsackievirus A12 (CV-A12) isolated from hand, foot and mouth disease (HFMD) surveillance in Yunnan province.Methods:Coxsackievirus isolation was carried out in RD cells from the clinical samples. CV-A12 strains were identified by realtime RT-PCR and sequencing technology from the positive cultures. The VP1 region of the CV-A12 strain was further sequenced. The VP1 gene sequences were initially aligned and then used to construct the phylogenetic tree with GenBank reference strains.Results:The two CV-A12 strains had the highest homology with the Yunnan reference strains in 2018 and the amino acids in VP1 region had specific mutations with other cluster reference strains at multiple sites.Conclusions:The CV-A12 in the HFMD cases in Yunnan province has occured regional specific mutation in VP1 gene.
9.Determination of Polysaccharide Content in Dried Peel of Seeding Watermelon and Its Mechanism of Lowering Blood Glucose

Objective Determination of polysaccharide content in dried peel seeding watermelon, and its mechanism of lowering blood glucose.Methods The content of polysaccharides in dried peel of seeding watermelon was determined by the method of phenol-sulfuric acid.Mice were given starch and sucrose load,and the mouse blood glucose was examined.The inhibitory activities of seeding watermelon against α-glucosidase were tested by the colorimetry of pNPG.Results The calibration of polysaccharide was A=0.066 4 C+0.022 6, R2=0.999 5, the content of polysaccharide in dried peel of seeding watermelon was 4.45% (n=10,RSD=1.80%);50,100 and 150 mg·kg-1 of polysaccharide could significantly reduce the starch load in mice blood glucose(P<0.01);100 and 150 mg·kg-1 of polysaccharide significantly reduced sucrose load blood glucose(P<0.01);polysaccharide concentration in 18 mg·mL-1 could significantly inhibit the activity of α-glucosidase as the inhibitory rate was (73.19±3.45)% (n=10).Conclusion Seeding watermelon polysaccharide has effect on lowering blood glucose in starch and sucrose load mice, and inhibits α-glucosidase significantly.
10.Role of nuclear factor-E2 related factor 2-antioxidant response element pathway in cardio-protection by ischemic or pinacidil postconditioning against ischemia-reperfusion injury in isolated rat hearts

Objective To evaluate the role of nuclear factor-E2 related factor 2 (Nrf2)-antioxidant response element (ARE) pathway in cardio-protection by ischemic or pinacidil postconditioning ( IP,PP) against ischemia-reperfusion (I/R) injury in isolated rat hearts.Methods Fifty-six male SD rats of both sexes weighing 200-250 g were anesthetized with intraperitoneal amobarbital sodium.The isolated rat hearts were perfused in a Langendorff apparatus with Krebs-Hensleit buffer (K-H).Fifty-six isolated rat hearts with I/R injury were randomly divided into 7 groups ( n =8 each):normal control group (group C) ; group I/R; group IP and group PP1-4 postconditioning with 4 different concentrations of pinacidil.After 20 min of equilibration,the perfusion was suspended for 40 min (global ischemia) followed by 60 min of reperfusion in group I/R.In group IP after 40 min of global ischemia,the isolated hearts underwent 6 cycles of 10 s reperfusion and 10 s ischemia followed by 58 min of reperfusion.In group PP1-4 at the end of 40 min of global ischemia,the isolated hearts were perfused with K-H containing pinacidil 5,10,30 and 50μmol/L for 5 min respectively followed by 55 min reperfusion with regular K-H.Left ventricular developed pressure (LVDP) and LVEDP were measured immediately before global ischemia and at the end of 60 min reperfusion.Myocardial specimens were obtained at the end of reperfusion for detection of Nrf2,quinopeoxidoreductase (NQO1),HO-1 and SOD1 mRNA (by RT-PCR) and protein (by Western blot) expression.Results I/R significantly up-regulated Nrf2,NQO1,HO-1 and SODI mRNA and protein expression,decreased LVDP and increased LVEDP in group I/R as compared with group C.IP and 30,50 μmol/L pinacidil postconditioning further significantly increased Nrf2,NQO1,HO-1 and SOD1 mRNA and protein expression and IP,5,10,30,50 μmol/L pinacidil postconditioning significantly increased LVDP and decreased LVEDP as compared with group I/R.Conclusion Ischemic or pinacidil postconditioning can attenuate I/R injury by activating Nrf2-ARE pathway in isolated rat hearts.
