Background:
High-mobility group box protein 1 (HMGB1) is a chromatin-binding protein involved in arthritis, ischemia, sepsis, atherosclerosis, neurodegenerative disorders, meningitis, and cancer. HMGB1 exhibits dual roles in cancer, acting as either a tumor suppressor or oncoprotein depending on context.
Objective:
This research aimed to elucidate HMGB1’s functional significance in cutaneous squamous cell carcinoma (cSCC).
Methods:
We overexpressed HMGB1 in cSCC cell lines using recombinant adenovirus and examined its effects on cell proliferation, colony formation, and cell migration.
Results:
Immunohistochemical analysis revealed elevated HMGB1 expression levels in cSCC tissue relative to normal epidermis. To assess the influence of HMGB1, we employed recombinant adenoviruses expressing HMGB1 to transduce SCC cell lines (SCC12 and SCC13). Enhanced HMGB1 expression significantly promoted cellular proliferation and colony formation capacity.Notably, HMGB1 overexpression elevated the levels of proliferation regulators, including P63, SOX2, CDK4 and CDK6. Furthermore, HMGB1 overexpression substantially enhanced tumor invasiveness, accompanied by upregulation of epithelial-mesenchymal transition (EMT) biomarkers. Mechanistically, overexpression of HMGB1 enhanced transforming growth factor-β signaling by increasing phosphorylation of SMAD2/3, the key mediators of EMT.
Conclusion
These data imply that HMGB1 acts as a tumor-promoting factor in cSCC.

Journal of Prevention and Treatment for Stomatological Diseases 2026;34(8):757-770
Objective:
To investigate the therapeutic effect of a dual-network hydrogel-microneedle (MN) on inhibiting oral squamous cell carcinoma (OSCC) recurrence and promoting tissue repair, thereby providing a foundation for the clinical treatment of OSCC.
Methods:
A Schiff base hydrogelformed between chitosan (CS) and poly(ethylene glycol) dibenzaldehyde (PEG-CHO), combined witha coordination-crosslinked hydrogel formed between sodium alginate (SA) and Cu²+, were jointly integrated to form the CS/PEG-CHO/SA/Cu²+ (CPSC) dual-network hydrogel base. The hydrogel formulation was optimized based on characterization by scanning electron microscopy, Fourier transform infrared spectroscopy, rheology, and swelling tests. Subsequently, 5-fluorouracil (5-FU) and gold nanorod (GNR) were incorporated into the CPSC hydrogel to fabricate an MN loaded with 5-FU/GNR (FG). The mechanical strength and dissolution behavior of FG MNs were characterized, and their photothermal performance was evaluated under near-infrared (NIR) irradiation using thermal imaging. The mouse fibroblast cell line L929 cells and the human oral mucosal epithelial-derived cell line GMSM-K cells were co-cultured with FG MNs for 2 and 4 days, respectively, and cell viability was assessed by the Cell Counting Kit-8 (CCK-8) assay and dead/live staining. Separately, the human tongue OSCC cell line Cal-27 cells were divided into four groups: a control group (cultured in complete medium), 5-FU MN group (co-cultured with 5-FU-loaded MNs), GNR MN+ NIR group (co-cultured with GNR-loaded MNs followed by NIR irradiation at 0.4 W/cm2 for 5 min), and FG MN+ NIR group (co-cultured with FG MNs followed by identical NIR irradiation). After 24 and 48 h of co-culture, Cal-27 cell viability was assessed using both CCK-8 assay and dead/live staining. All animal experiments were approved by the Institutional Animal Ethics Committee. A subcutaneous Cal-27-derived OSCC postoperative recurrence model was established in BALB/c nude mice. The mice were divided into four groups: a control group, 5-FU MN group, GNR MN+ NIR group, and FG MN+ NIR group. Mice in the control group received no intervention, while the other groups were treated with the corresponding MN patches. Additionally, the GNR MN+ NIR and FG MN+ NIR groups received NIR irradiation (0.4 W/cm2, 5 min). Tumors were excised and their volumes measured on day 7, and tumor recurrence and wound healing were assessed on day 14.
Results:
A dual-network hydrogel-MN was successfully constructed, demonstrating favorable mechanical and dissolution properties. Upon NIR irradiation for 5 min, the temperature rapidly increased to 48.4°C, reaching the threshold for effective tumor ablation. CCK-8 and dead/live staining results showed that FG MNs exhibited no cytotoxicity toward L929 and GMSM-K cells, indicating good cytocompatibility. However, Cal-27 cell viability in the chemotherapy-photothermal combination group was lower than that in the other three groups, with a statistical difference (P<0.0001). In the nude mouse model, subcutaneous Cal-27 tumors were resected 7 days post-implantation. No statistical difference in tumor volume was observed among groups at this timepoint, confirming successful model establishment. By postoperative day 14, the control group displayed complete tumor recurrence with unhealed wounds, while the 5-FU and GNR MN+ NIR groups showed mild recurrence and partial wound healing. The FG MN+ NIR group demonstrated superior efficacy in both inhibiting tumor recurrence and promoting tissue repair compared with all other groups, with a statistical difference (P<0.0001).
Conclusion
The dual-network hydrogel-MN system effectively suppresses postoperative recurrence of Cal-27-derived subcutaneous OSCC and promotes tissue repair through chemo-photothermal combined therapy.

Journal of Preventive Medicine 2026;38(2):196-200
Objective :
To investigate the status of electronic cigarette use and its influencing factors among residents aged ≥15 years in Chengdu City in 2021 and 2024, so as to provide the evidence for optimizing electronic cigarette control strategies.
Methods:
Two rounds of cross-sectional surveys were conducted in 2021 and 2024. Multi-stage cluster random sampling method was used to select permanent residents aged ≥15 years in Chengdu City as survey subjects. The Chinese Adult Tobacco Survey Questionnaire was used to collect information on gender, age, and electronic cigarette use. Complex sampling weighting was used to calculate the rate of electronic cigarette use and the proportion of reasons for electronic cigarette use. A complex sampling multivariable logistic regression model was used to analyze the influencing factors for electronic cigarette use.
Results:
A total of 7 950 and 6 920 people were surveyed in 2021 and 2024, respectively, including 3 868 and 3 653 males (48.65% and 52.79%), and 4 082 and 3 267 females (51.35% and 47.21%), respectively. The rate of electronic cigarette use in 2024 was lower than in 2021 (2.29% vs. 3.15%, P<0.05). In 2024, the rates of electronic cigarette use were lower in males, rural areas, people aged 15-<45 years, educational level of bachelor' s degree or above, occupation in government/public institutions or occupation in enterprises/commercial/service industries, and current smokers compared with 2021, while the rate of electronic cigarette use was higher in residents aged ≥65 years compared with 2021 (all P<0.05). In 2021 and 2024, the main reasons for electronic cigarette use were smoking cessation and the belief that electronic cigarettes are less harmful. Smoking cessation accounted for 27.78% and 23.28%, and the belief that electronic cigarettes are less harmful accounted for 24.26% and 22.10%, respectively. Complex sampling multivariable logistic regression analysis showed that age (15-<25 years, OR=15.682, 95%CI: 5.660-43.450; 25-<45 years, OR=7.463, 95%CI: 2.987-18.647; 45-<65 years, OR=3.425, 95%CI: 1.562-7.510), educational level (bachelor' s degree or above, OR=2.916, 95%CI: 1.444-5.891), and smoking (current, OR=23.992, 95%CI: 8.182-70.353; former, OR=20.393, 95%CI: 6.150-67.624) were influencing factors for electronic cigarette use in 2021. Gender (male, OR=2.279, 95%CI: 1.074-4.832), age (25-<45 years, OR=2.114, 95%CI: 1.148-3.892), educational level (high school/vocational high school/technical secondary school, OR=2.792, 95%CI: 1.414-5.514; bachelor' s degree or above, OR=3.321, 95%CI: 1.567-7.039), occupation (unemployed, retired, and retired, OR=0.449, 95%CI: 0.226-0.892), and smoking (current, OR=10.547, 95%CI: 5.563-19.994; former, OR=8.998, 95%CI: 3.833-21.125) were influencing factors for electronic cigarette use in 2024.
Conclusions
Compared with 2021, the rate of electronic cigarette use among residents in Chengdu City decreased in 2024. Young people, males, bachelor' s degree or above, and current smokers are the key populations for electronic cigarette control. The cognitive bias held by residents that electronic cigarette can aid in smoking cessation and pose minimal harm needs to be corrected.

Chinese Journal of Tissue Engineering Research 2025;29(8):1676-1683
BACKGROUND:In recent years, numerous studies have shown that autophagy and mitophagy play an important role in the regulation of bone metabolism.Under non-physiological conditions, mitophagy breaks the balance of bone metabolism and triggers metabolism disorders, which affect osteoblasts, osteoclasts, osteocytes, chondrocytes, bone marrow mesenchymal stem cells, etc. OBJECTIVE:To summarize the mechanism of mitophagy in regulating bone metabolic diseases and its application in clinical treatment. METHODS:PubMed,Web of Science,CNKI,WanFang and VIP databases were searched by computer using the keywords of"mitophagy, bone metabolism, osteoblasts, osteoclasts, osteocytes, chondrocytes, bone marrow mesenchymal stem cells"in English and Chinese. The search time was from 2008 to 2023.According to the inclusion criteria,90 articles were finally included for review and analysis. RESULTS AND CONCLUSION:Mitophagy promotes the generation of osteoblasts through SIRT1,PINK1/Parkin,FOXO3 and PI3K signaling pathways, while inhibiting osteoclast function through PINK1/Parkin and SIRT1 signaling pathways.Mitophagy leads to bone loss by increasing calcium phosphate particles and tissue protein kinase K in bone tissue.Mitophagy improves the function of chondrocytes through PINK1/Parkin,PI3K/AKT/mTOR and AMPK signaling pathways.Modulation of mitophagy shows great potential in the treatment of bone diseases, but there are still some issues to be further explored, such as different stages of drug-activated mitophagy, and the regulatory mechanisms of different signaling pathways.

A high-performance liquid chromatography method using pre-column derivatization with 6-aminoquinolyl-N-hydroxysuccinimidyl carbamate was developed to determine the content of 15 amino acids in the medicinal snakes Bungarus Parvus, Agkistrodon, and Zaocys. The results showed that the total amino acid(TAA) content ranged from 277.13 to 515.05 mg·g~(-1), with the top four amino acids in all three species being glutamic acid(Glu), glycine(Gly), aspartic acid(Asp), and lysine(Lys). The essential amino acid(EAA) content ranged from 74.56 to 203.94 mg·g~(-1), with Agkistrodon exhibiting the highest content. The non-essential amino acid(NEAA), semi-essential amino acid(semi-EAA), and medicinal amino acid(MAA) content ranged from 189.06 to 318.23, 12.89 to 33.53, and 179.83 to 342.33 mg·g~(-1), respectively, with Zaocys having the highest content in these categories. Amino acid nutritional value was evaluated using the amino acid ratio(RAA), amino acid ratio coefficient(RCAA), and amino acid ratio coefficient score(SRCAA), and the results indicated that all three medicinal snakes possessed good nutritional value. The amino acid composition was similar across the species, though significant differences in content were observed. Based on these differences, an orthogonal partial least squares-discriminant analysis(OPLS-DA) model was established, which could clearly distinguish between the three medicinal snake species. The key differences in amino acid content included Gly, tyrosine(Tyr), Glu, and serine(Ser), which may be related to the observed clinical application differences among the species. Further research into the mechanisms of these differential amino acids is expected to provide more insights into the clinical application disparities of these three medicinal snake species.

Chinese Journal of Experimental Traditional Medical Formulae 2025;31(21):90-101
ObjectiveTo establish a subcutaneous xenograft model of gastric cancer in nude mice and to identify differentially expressed genes (DEGs) affected by Jianpi Yangzheng Xiaozheng formula (JPYZXZ) in diffuse gastric cancer (DGC) using transcriptome sequencing. The study aims to identify potential key prognostic factors and preliminarily elucidate the therapeutic mechanism of JPYZXZ. MethodsSubcutaneous tumor tissues were collected from nude mice treated with JPYZXZ (6 g·kg-1)and from the untreated traditional Chinese medicine control group. High-throughput RNA sequencing was performed to extract and analyze total RNA and identify DEGs, followed by functional enrichment analysis. DEGs were intersected with cancer-associated fibroblast (CAF)-related genes identified from single-cell RNA sequencing (scRNA-seq) data. A Cox proportional hazards regression model (Cox model) was constructed to identify potential key targets, among which DAZ interacting protein 1 (DZIP1) was selected. The role of DZIP1 in DGC progression was further verified through in vitro and in vivo experiments. ResultsTranscriptome sequencing revealed that DEGs regulated by JPYZXZ were significantly enriched in biological processes such as focal adhesion, phosphatidylinositol 3-kinase/protein kinase B (PI3K/Akt) signaling pathway, and vascular development (P<0.05). Intersection analysis with CAF marker genes identified ten potential common target genes. Cox regression analysis combined with the Cancer Genome Atlas (TCGA) dataset for stomach adenocarcinoma (STAD) confirmed that DZIP1 is an independent prognostic factor significantly associated with poor outcomes. Transcriptome and immunohistochemical analyses showed that DZIP1 expression was significantly higher in gastric cancer tissues than in adjacent tissues, while JPYZXZ treatment downregulated DZIP1 expression in a dose-dependent manner. Clinical data further demonstrated that serum DZIP1 levels in patients treated with JPYZXZ were significantly lower than those in the control group. ConclusionJPYZXZ may inhibit the malignant progression of DGC by downregulating DZIP1 expression, thereby suppressing CAF activation and epithelial-mesenchymal transition (EMT). These findings provide experimental evidence and identify DZIP1 as a potential therapeutic target in DGC treatment.

Chinese Journal of Blood Transfusion 2025;38(8):1089-1093
Objective: To design HBV DNA proficiency testing and system comparison samples with different concentration gradients, analyze their detection results in PCR detection systems, evaluate the nucleic acid detection capabilities of laboratories and differences between detection systems, and put forward suggestions for continuous quality improvement to participating laboratories. Methods: Three groups of randomly numbered proficiency testing samples (with HBV DNA reference concentrations of <2, 7.5, and 30 IU/mL respectively) were taken as the detection objects. Using nucleic acid test data from 11 provincial blood station laboratories as the source, the samples were grouped by detection system and laboratory successively, and statistical analysis was conducted. Results: Statistical analysis of the detection data of the three groups of samples based on detection systems and laboratories showed that from low to high concentration, the coincidence rate between the detection results of different detection systems and laboratories and the expected results showed an increasing trend: 38.89%, 85.90%, and 100.00%; the same system exhibited certain differences in performance among different laboratories. Conclusion: Through this proficiency testing and system comparison, it is found that there are certain differences in the detection capabilities of different laboratories and different nucleic acid test systems. Blood station laboratories should standardize processes, strengthen quality management and data analysis on the basis of being familiar with the detection performance of their detection systems, and at the same time strengthen the control of laboratory interference factors to continuously improve the nucleic acid detection capabilities of blood station laboratories.
