In recent years, the incidence rate of tubal infertility has been rising. In China, most patients with tubal infertility prioritize conservative management or tubal reconstruction surgery in order to preserve fallopian tubes as far as possible. In order to meet the reproductive needs of tubal infertile patients, clinicians in the reproductive field need to evaluate various infertility risk factors comprehensively, weigh the pros and cons, and explore individualized reproductive programs which are more in line with the interests of patients. Correspondingly, higher requirements for the imaging evaluation efficiency of the fallopian tubes have been put forward. Hysterosalpingo-contrast sonography (HyCoSy), as a first-line imaging technique for evaluating tubal patency, still faces many controversies and dilemmas despite its value being widely recognized in clinical practice. This paper aims to provide viewpoints for the advancement of HyCoSy by reviewing literature and summarizing experiences.

To address the increasing need for detecting and validating protein biomarkers in clinical specimens, mass spectrometry (MS)-based targeted proteomic techniques, including the selected reaction monitoring (SRM), parallel reaction monitoring (PRM), and massively parallel data-independent acquisition (DIA), have been developed. For optimal performance, they require the fragment ion spectra of targeted peptides as prior knowledge. In this report, we describe a MS pipe-line and spectral resource to support targeted proteomics studies for human tissue samples. To build the spectral resource, we integrated common open-source MS computational tools to assemble a freely accessible computational workflow based on Docker. We then applied the workflow to gen-erate DPHL, a comprehensive DIA pan-human library, from 1096 data-dependent acquisition (DDA) MS raw files for 16 types of cancer samples. This extensive spectral resource was then applied to a proteomic study of 17 prostate cancer (PCa) patients. Thereafter, PRM validation was applied to a larger study of 57 PCa patients and the differential expression of three proteins in prostate tumor was validated. As a second application, the DPHL spectral resource was applied to a study consisting of plasma samples from 19 diffuse large B cell lymphoma (DLBCL) patients and 18 healthy control subjects. Differentially expressed proteins between DLBCL patients and healthy control subjects were detected by DIA-MS and confirmed by PRM. These data demonstrate that the DPHL supports DIA and PRM MS pipelines for robust protein biomarker discovery. DPHL is freely accessible at https://www.iprox.org/page/project.html?id=IPX0001400000.

Alcohol abuse leads to alcoholic liver disease and no effective therapy is currently available. Wuzhi Tablet (WZ), a preparation of extract fromthat is a traditional hepato-protective herb, exerted a significant protective effect against acetaminophen-induced liver injury in our recent studies, but whether WZ can alleviate alcohol-induced toxicity remains unclear. This study aimed to investigate the contribution of WZ to alcohol-induced liver injury by using chronic-binge and acute models of alcohol feeding. The activities of ALT and AST in serum were assessed as well as the level of GSH and the activity of SOD in the liver. The expression of CYP2E1 and proteins in the NRF2-ARE signaling pathway including NRF2, GCLC, GCLM, HO-1 were measured, and the effect of WZ on NRF2 transcriptional activity was determined. We found that both models resulted in liver steatosis accompanied by increased transaminase activities, but that liver injury was significantly attenuated by WZ. WZ administration also inhibited CYP2E1 expression induced by alcohol, and elevated the level of GSH and the activity of SOD in the liver. Moreover, the NRF2-ARE signaling pathway was activated by WZ and the target genes were all upregulated. Furthermore, WZ significantly activated NRF2 transcriptional activity. Collectively, our study demonstrates that WZ protected against alcohol-induced liver injury by reducing oxidative stress and improving antioxidant defense, possibly by activating the NRF2-ARE pathway.
7.On unified calculation of clinical nursing workload

Objective To explore a unified calculation method of clinical nursing workload in a general hospital,so as to compare objectively nursing workload among different departments.Methods As an attempt to figure out the nursing workload,direct nursing time of venous transfusion was used as the statistical basis and direct nursing time of nursing service items in different departments were standardized.Results Conversion coefficient among different nursing service items was determined,with the result of realization of reasonable calculation of nursing workload and objective comparison of nursing workload among different departments. Conclusions The unified calculation of clinical nursing workload,using direct nursing time of venous transfusion as the statistical basis,is easy in operation and reliable in results,which makes it operable and feasible in comprehensive performance evaluation system and bonus distribution with serving quality and nursing workload as main index.
8.Change of antithrombin Ⅲ in patients with atherosclerotic cerebral in-farction

AIM:To explore the change of antithrombin Ⅲ( AT-Ⅲ) in the patients with atherosclerotic cere-bral infarction .METHODS:Chromogenic substrate assay was used to measure the activity of AT-Ⅲ in 55 patients with atherosclerotic cerebral infarction and 55 healthy controls , and the correlation analysis was applied to determine the AT-Ⅲactivity with the severity of damage in central nervous system and general biochemical parameters .The levels of TNF-αand IL-6 in the plasma were detected by ELISA .Immunocomplex in the plasma was measured by enzyme immunoassay (EIA). The number and phenotype of the monocytes in peripheral blood were analyzed by flow cytometry .ELISA was also applied to determine the secretion of TNF-αand IL-6 from the monocytes after the stimulation of immunocomplex .The expression of AT-Ⅲin human brain vascular endothelial cells after the stimulation of TNF-αand IL-6 was observed by Western blotting . RESULTS:The activity of AT-Ⅲsignificantly decreased in the patients with atherosclerotic cerebral infarction , and nega-tively correlated with the damage degree of nervous system function , systolic pressure , diastolic pressure , glucose , choles-terol, triglyceride, low-density lipoprotein cholesterol and homocysteine , while positively correlated with high-density lipo-protein.In addition, the plasma levels of TNF-αand IL-6 increased significantly , accompanied with the enhancement of immunocomplex level .The numbers of CD14 + CD16 + and CD14 + CD32 + monocytes in peripheral blood were not changed , while CD14 +CD64 +monocytes increased obviously .The secretion of TNF-αand IL-6 by monocytes were signifi-cantly enhanced after stimulated with immunocomplex , while the protein expression of AT-Ⅲ in the human brain vascular endothelial cells was down-regulated after co-incubated with TNF-αor IL-6.CONCLUSION:Decreased AT-Ⅲactivity in the patients with atherosclerotic cerebral infarction is one of the risk factors of cerebral infarction , and related with the dis-ease severity .The production of pro-inflammatory cytokines through immunocomplex from CD 14 +CD64 +monocytes may be involved in the mechanism .Improvement of AT-Ⅲactivity may protect against cerebral ischemia .
9.Association of miR-199a expression with clinicopathologic characteristics and prognosis of renal cell carcinoma.

OBJECTIVETo investigate the association of miR-199a expression with the clinicopathologic characteristics and the survival of patients with renal cell carcinoma (RCC).
METHODSReal-time quantitative RT-PCR was used to detect the expression of miR-199a in the tumor tissues and paired adjacent normal tissues from 67 patients with RCC. The correlations of miR-199a expression with the clinicopathologic characteristics and survival of RCC patients were analyzed.
RESULTSPositive miR-199a expression was detected in both the RCC and adjacent normal tissues, but the tumor tissues showed a significantly lower expression level by a mean of 13.7 folds (P=0.003). The expression of miR-199a was negatively correlated with tumor recurrence in RCC patients in T stage (P<0.05), but showed no significant correlations with the patients age, histological type of the tumor, lymph node metastasis, distal metastasis, or Fuhrman grade (P>0.05). The patients with lowered miR-199a expression in the tumor tissue had a significantly shorter mean survival time than those without miR-199a down-regulation (P=0.017 by Log-rank test).
CONCLUSIONA decreased expression of miR-199a is significantly correlated with a higher tumor stage, a greater likeliness of tumor recurrence, and a poorer prognosis in RCC patients. miR-199a can serve as a promising prognostic factor of renal cell carcinoma.

10.Invention of precise positioning instrument regulating puncture in percutaneous nephrolithotripsy and its clinical application

ObjectiveTo summarize the research and manufacturing ideas of precise positioning instrument and its feasibility and safety in the minimally invasive percutaneous nephrolithotripsy.Methods The precise positioning instrument was preliminarily made by integrating ruler with protractor.Fifty cases of kidney stones or upper ureteral stones were recruited in the study.There were 32 males and 18 females with mean age of 47 years (21 -75 years).Of them,6 cases had open surgery history and 31 cases had different degrees of hydronephrosis.The distance from the mid-point of renal pelvis to the plane of the dorsal surface and the distance from the incident point of the mid-point of pelvis on the dorsal surface to spinous process were measured by ruler on the preoperative CT images.According to the measured results,the point,angle,direction and depth of needle were calculated by geometric formula,and then fix position puncture and percutaneous puncture channel expansion were completed by precise positioning instrument and calibrated by Carm X-ray machine.ResultsThe puncture channels were established successfully in all of the 50 cases.Forty-six cases were punctured successfully by just one try,and the success rate of one puncture only was 92.0%.C-arm was used in none of the cases during the channel establishing process.Each patient was Xrayed 1 -3 times,and the average was 1.4 times.Targeting deflection or renal pelvis injury occurred in none of these cases.ConclusionThe application of the homemade precise positioning instrument is a safe and quick method for the establishment of puncture channel during percutaneous nephrolithotripsy,meanwhile the surgeons can reduce the exposure to radiation.
