1.Single-cell sequencing systematically analyzed the mechanism of Emdogain on the restoration of delayed replantation periodontal membrane.

The repair of the periodontal membrane is essential for the successful management of periodontal disease and dental trauma. Emdogain® (EMD) is widely used in periodontal therapy due to its ability to promote repair. Despite substantial research, the cellular and molecular mechanisms underlying EMD's effects, particularly at the single-cell resolution, remain incompletely understood. This study established a delayed tooth replantation model in rats to investigate these aspects. Tooth loss rate and degree of loosening were evaluated at 4 and 8 weeks. Micro-CT, HE staining, TRAP staining, and immunofluorescence staining were evaluated to assess EMD's efficacy. Single-cell sequencing analyses generated single-cell maps that explored enrichment pathways, cell communication, and potential repair mechanisms. Findings indicated that EMD could reduce the rate of tooth loss, promote periodontal membrane repair, and reduce root and bone resorption. Single-cell analysis revealed that EMD promotes the importance of Vtn+ fibroblasts, enhancing matrix and tissue regeneration functions. Additionally, EMD stimulated osteogenic pathways, reduced osteoclastic activity, and promoted angiogenesis-related pathways, particularly bone-related H-type vessel expression in endothelial cells. Gene modules associated with angiogenesis, osteogenesis, and odontoblast differentiation were identified, suggesting EMD might facilitate osteogenesis and odontoblast differentiation by upregulating endothelium-related genes. Immune cell analysis indicated that EMD did not elicit a significant immune response. Cell communication analysis suggested that EMD fostered pro-regenerative networks driven by interactions between mesenchymal stem cells, fibroblasts, and endothelial cells. In conclusion, EMD proves to be an effective root surface therapy agent that supports the restoration of delayed replantation teeth.

2.Jiaotai Pill Ameliorates Type 2 Diabetes in Rats Via Modulation of Neurotransmitters in HPA-Related Tissues

Objective To investigate the therapeutic effects of Jiaotai Pill(JTP)on type 2 diabetic(T2DM)rats,a systemic study was conducted by examining the changes in neurotransmitter levels related to the hypothalamic-pituitary-adrenal(HPA)axis.Methods The rats were divided into a normal group,a model group,a metformin group(183 mg·kg-1·d-1),and a JTP treatment group(3.3 g·kg-1·d-1).A T2DM model was established using a high-fat diet combined with streptozotocin.After 28 days of intragastric administration of JTP and metformin,metabolic indicators,Fasting blood glucose(FBG),insulin(INS),blood lipids,and pathological changes in the liver and kidneys were measured in each group of rats.Levels of eight neurotransmitters were quantified in serum,urine,and multiple tissues(brain,heart,liver,kidney,intestine,and adrenal gland)using ultra-high-performance liquid chromatography-tandem mass spectrometry.Results Compared to the T2DM model group,the JTP intervention groups showed varying degrees of reduction in water intake,food intake,FBG,INS,and blood lipids(P<0.05).Additionally,the fatty degeneration in the liver,glomerular lesions in the kidneys,and dyslipidemia were significantly improved.JTP intervention significantly modulated the disordered neurotransmitter levels in the blood,urine,and various tissues of T2DM rats(P<0.05),with the greatest number of neurotransmitters showing a notable recovery in blood,urine,brain,and adrenal gland.Conclusion JTP at a dosage of 3.3 g·kg-1·d-1 can improve the glucose and lipid metabolism in rats.JTP can regulate the HPA axis-related neurotransmitter system,mitigating metabolic disturbances and organ damage associated with T2DM.
3.Clinical characteristics and prognostic factors of 233 cases of Staphylococcus aureus bacteremia in adult patients

Objective To investigate the clinical characteristics and prognostic factors of Staphylococcus aureus bloodstream infections in adult patients for improving clinical treatment and identifying potential interventions.Methods Clinical data of inpatients diagnosed with S.aureus bloodstream infection confirmed by blood culture in a hospital from January 2016 to December 2023 were retrospectively reviewed.The data included patient age,gender,history of hospital admission,department of admission,underlying diseases,primary infection,quick Pitt bacteremia score(qPitt),invasive treatment,empirical anti-infective treatment,and treatment outcomes.Patients were assigned to case group or control group according to whether they died in hospital in order to identify the prognostic factors of patient outcomes.Binary logistic regression analysis was used to identify independent prognostic factors.Results A total of 233 cases of S.aureus bacteremia were identified.Multivariate logistic regression analysis showed that age ≥ 70 years old(OR=4.725,95%CI:1.228-18.173,P=0.024),diabetes mellitus(OR=8.161,95%CI:1.954-34.086,P=0.004),Charlson comorbidity index(CCI)≥ 5(OR=7.672,95%CI:1.901-30.963,P=0.004),hospital infection(OR=7.853,95%CI:1.588-38.832,P=0.012),and qPitt ≥ 2(OR=23.189,95%CI:4.461-120.552,P<0.001)were independent prognostic factors for poor outcome of patients with S.aureus bacteremia,while catheter-associated infection(OR=0.051,95%CI:0.005-0.579,P=0.016)was negatively correlated with mortality.Conclusions Advanced age,diabetes mellitus,high CCI,hospital infection,and high qPitt were independent prognostic factors for poor outcomes of patients with S.aureus bacteremia.The patients should be well managed by timely removal of eradicable lesions to improve patient outcomes.
4.Jiaotai pill and its main component enhance islet hormone secretion in type 2 diabetic rats by activating the THP1/TGase2/SERT/5-HT1FRpathway

ObjectiveTo investigate the relationship between Jiaotai pill (JTP), its main component berberine (BBR), and the serotonin (5-HT) system in regulating islet hormone secretion and alleviating pancreatic β-cell dysfunction during type 2 diabetes mellitus (T2DM) progression.MethodsT2DM rat model was established using a high-fat diet and streptozotocin injection. JTP, BBR, and Metformin were intragastrically administered for 35 days. The analyzed indices included blood glucose, blood lipids, islet hormones, and proteins related to 5-HT synthesis, secretion, and transport. Additionally, an in vitro model of glucose injury in islet cells was established to study the effects of JTP and BBR on islet hormone secretion following tryptophan hydroxylase 1 (TPH1) inhibition.ResultsJTP and BBR significantly improved blood glucose and lipid levels and islet morphology in T2DM rats. Both models exhibited reduced islet 5-HT levels and impaired islet hormone secretion. However, the administration of JTP and BBR reversed these effects. Furthermore, JTP and BBR upregulated the expression of TPH1(P = .0194, P = .0413) transglutaminase 2 (TGase2; P = .0492, P = .0349), serotonin transporter (SERT, P = .0090), and 5- hydroxytryptamine 1F receptor (5-HT1FR) in the islet 5-HT pathway (P = .0194). In the cell model, the regulatory effects of JTP and BBR on islet hormone levels were significantly weakened after TPH1 inhibition (P = .001), suggesting that JTP and BBR influence islet hormone secretion through the pancreatic 5-HT system.ConclusionThe islet 5-HT system is correlated with islet hormone secretion dysfunction in T2DM. JTP and BBR can improve islet hormone secretion by activating the TPH1/TGase2/SERT/5-HT1FR pathway in the islet 5-HT system in T2DM rats.
5.Current status of water management for terminal rinsing of digestive endoscope in 104 medical institutions in Fujian Province

OBJECTIVE To investigate the current status of water management for terminal rinsing of digestive en-doscope in medical institutions in Fujian Province,and to provide reference for improving regional quality control standards.METHODS An electronic questionnaire survey was conducted from Jul.2024 to Aug.2024 through convenient sampling in secondary and above hospitals of 9 prefecture-level cities in Fujian Province.The survey covered topics such as water treatment system configuration,maintenance and water quality monitoring.RESULTS A total of 108 questionnaires were distributed and 104 valid questionnaires were collected,with an effec-tive response rate of 96.30%.The survey revealed that 78.85%(82/104)of the hospitals adopted separate water supply for each department,and 82.69%used purified water for terminal rinsing.Only 19.23%installed the final filter membrane at the water outlet.In addition,66.35%of the hospitals did not specify the service life of the wa-ter supply pipeline,and the pipeline disinfection implementation rate was 60.58%,but 31.75%of them had ir-regular disinfection frequencies,with chlorine-based disinfectants(50.79%)and peracetic acid(34.92%)being the main disinfectants.The regular conductivity monitoring rate was 47.12%,and the microbial monitoring cov-erage rate reached 90.38%,with the monitoring frequency mainly being once every quarter(60.64%),but only 20.21%used R2A medium,and 12.77%adopted the membrane filtration method for inoculation.Tertiary hospi-tals were superior to secondary hospitals in terms of film membrane pore size pass rate(87.50%vs.56.86%),pipeline disinfection implementation rate(71.43%vs.50.91%)and advanced detection method application(P<0.05).CONCLUSIONS There are issues in the management of water used for terminal rinsing of digestive endo-scopes in Fujian Province,including non-standard equipment maintenance,inconsistent monitoring methods and insufficient awareness among management personnel.It is recommended to enhance management quality by optimizing the water treatment system setup,establishing standardized monitoring procedures and strengthening professional training for personnel.
6.Latent profile analysis of body image and its influencing factors in postoperative oral cancer patients

Objective:To analysis of the latent profiles and influencing factors of body image in patients with postoperative oral cancer.Methods:From July 2024 to March 2025, a total of 332 patients with primary oral cancer confirmed by pathology, aged ≥18 years, and undergoing oral cancer surgery at Hunan Cancer Hospital were selected using simple random sampling and cluster sampling. Among them, 25 were female and 307 were male. The body image scale and the Rosenberg self-esteem scale were used to investigate the patients. The main indicators included the total scale scores and scores on various dimensions of body image, such as appearance evaluation and health focus, with particular attention to satisfaction with facial appearance and oral function.The correlation between self-esteem and body image was analyzed, and differences in scores were compared based on gender, age, self-esteem level, and surgical procedure.Results:Among the 332 patients, 93.4% (310/332) were married, and 6.6% (22/332) were unmarried, divorced, or widowed. A total of 84.3% (280/332) underwent flap transplantation surgery, while 15.7% (52/332) did not. The body image distress in the 332 patients could be categorized into a body image adaptation group [80.12% (266/332)] and a body image disorder group [19.88% (66/332)]. Unmarried/divorced/widowed status ( P=0.020), undergoing flap transplantation ( P=0.006), and self-esteem level ( P<0.001) were identified as influencing factors for postoperative body image disorder in oral cancer patients. Conclusions:Given the varying levels of body image concerns among oral cancer patients, healthcare providers can implement targeted, personalized nursing interventions based on their distinct categories and influencing factors.
7.Transcriptome analysis of differentially expressed genes in the mice sciatic nerve in the early stage of Wallerian degeneration in vitro

Objective:To analyze the differentially expressed genes and related functional pathways of mouse sciatic nerves of Schwann cells(SCs)in early in vitro Wallerian degeneration(WD).Methods:The sciatic nerves of adult male C57BL/6J mice were Wallerian degeneration in vitro,and total RNA was extracted and transcriptome sequencing was performed at 3 h and 6 h after degeneration,respectively.The differentially expressed genes(DEGs),gene ontology(GO)and Kyoto encyclopedia of genes and genomes(KEGG)pathway enrichment were analyzed by bioinformatics.Results:Compared with the Control group,3961 and 5538 DEGs were screened in the WD 3 h group(WD3h)and the 6 h group(WD6h)of in vitro,respectively.The most significantly up-regulated genes mainly included molecules related to inflammation and immunity and neurotrophic factors.GO analysis showed that DEGs in both groups were enriched in positive transcriptional regulation and metabolic processes.KEGG pathway enrichment analysis revealed that DEGs were mainly concentrated in TNF signaling pathway,MAPK signaling pathway and ribosome production.Conclusion:At the early stage of WD,SCs up-regulates the genes related to inflammation and immunity to promote the progression of WD and secrete neurotrophic factors to support the survival of neurons,accompanied by the activation of TNF signaling path-way and MAPK signaling pathway.
8.Research advances in radiomics combined with multi-omics in hepatocellular carcinoma

Hepatocellular carcinoma(HCC)is the sixth most common malignant tumor in the world,accounting for 75%-85%.Due to the high incidence,strong heterogeneity and poor prognosis of HCC,early monitoring and fine assessment of high-risk populations are particularly important.In recent years,as a non-invasive method,radiomics has been widely used in the diagnosis,pathological characterization,selection of treatment methods and evaluation of curative effect of HCC.In addition,radiomics combined with next-generation sequencing technology and other multi-omics methods provide new ideas for HCC research.This paper reviews the research progress of radiomics combined with multi-omics in the field of HCC,in order to provide a new perspective for the diagnosis and treatment of HCC.
