Objective:
To investigate the regulatory effect of corticosterone(CORT) pretreatment on the activation of microglia by lipopolysaccharide(LPS) and the inhibitory effect of Biochanin A(Bio A) on microglia activation.
Methods:
The MTT method was used to select the optimal concentrations for LPS,CORT and Bio A on BV2 cells;BV2 cells were divided into 5 groups: Control group,CORT(50 nmol/L) group,LPS(1 μg/mL) group,LPS(1μg/mL) + CORT(50 nmol/L) group, and LPS(1 μg/mL) + CORT(50 nmol/L) + Bio A(5 μmol/L) group;Except for the control group, each group was first incubated with CORT(50 nmol/L) for 2 h,and then each group was co-incubated with the corresponding concentrations of LPS(1 μg/mL) and Bio A(5 μmol/L) for 36 h;DCFH-DA probe method was used to detect reactive oxygen species(ROS) content; Western blot was used to detect the protein expression levels of inflammatory cytokines,5-hydroxytryptamine(5-HTT),glucocorticoid receptor(GR),mineralocorticoid receptor(MR),NOD-like receptor family pyrin domain containing 3(NLRP3),apoptosis-associated speck-like protein containing a CARD(ASC) and cysteine-aspartic protease 1(Caspase-1).
Results:
Compared with the CORT(50 nmol/L) and LPS(1 μg/mL) groups,cells in the CORT(50 nmol/L) + LPS(1 μg/mL) group showed increased cell viability,higher levels of ROS,and increased levels of inflammatory cytokines,NLRP3,ASC,and Caspase-1 protein expression( P<0. 05 ) .
Conclusion
A low dose of CORT pretreatment reinforces LPS- induced BV2 cell activation; Bio A inhibits CORT-pretreated LPS-induced BV2 cell activation. The mechanism of which may be related to the inhibition of NLRP3 inflammasome activation.

Objective:To quantify any correlation between the severity of spinal curvature of an adolescent with idiopathic scoliosis and their cardiopulmonary exercise endurance. Methods:The cardiopulmonary exercise test (CPET) results and the full-length spinal X-rays in a standing position of 64 adolescents with idiopathic scoliosis were reviewed retrospectively. Independent t-tests were used to compare the two datasets obtained from those with left or right thoracic scoliosis. The correlation between the Cobb angle and cardiopulmonary exercise endurance was analyzed using Pearson correlation coefficients, multiple factor linear regression and two-stage linear regression. Results:After adjusting for gender, age, height and weight, the multiple linear regression analysis showed that the Cobb angle was significantly negatively correlated with maximum tidal volume (β=-0.013) and significantly positively correlated with the rate of respiration (β=0.421). The relationship between the Cobb angle and cardiopulmonary exercise endurance was non-linear. With a Cobb angle > 34°, a 1° increase reduces cardiopulmonary exercise endurance by a factor of 1.4 on average. At smaller Cobb angles the corresponding increase is about 0.87 times. Conclusions:The Cobb angle is a negative predictor of ventilation during exercise among adolescents with idiopathic scoliosis. The more severe a patient′s spinal curvature, the lower the cardiopulmonary exercise endurance is likely to be.

Objectives:To investigate the diagnostic value of whole blood quantitative PCR for DNA load of Epstein-Barr virus (EBV) in post-transplant lymphoproliferative disease (PTLD) after allogeneic hematopoietic stem cell transplantation (allo-HSCT) .Methods:A total of 694 patients with hematologic diseases who underwent allo-HSCT at the Hematology Department of Peking University First Hospital from April 2004 to April 2019 were included, and their data were retrospectively analyzed.Results:①Among the 694 cases, 29 cases (22 males and 7 females, with a median age of 22 (1-52) years) developed PTLD after allo-HSCT with a cumulative incidence of 4.2% and a median onset time of 2.1 (0.8-20.6) months. ② Univariate analysis showed that age<30 years, diagnosis with aplastic anemia, human leukocyte antigen (HLA) mismatch, use of antithymocyte globulin (ATG) in preconditioning regimens, and EBV reactivation were the risk factors for the occurrence of PTLD. Multivariate analysis showed that EBV reactivation was an independent risk factor for the occurrence of PTLD. ③Further analysis of EBV reactivation cases showed that the peak value of EBV-DNA load was significantly higher in the PTLD group than that in the non-PTLD group ( P<0.001) and the incidence of PTLD increased with the increase of EBV-DNA load. Receiver operating characteristic (ROC) curve analysis indicated that PTLD was more likely to be diagnosed when the EBV-DNA load was >1.19×10 6 copies/ml (sensitivity 0.800 and specificity 0.768) . ④All patients with PTLD received rituximab-based treatment, with an overall response rate of 86.2% and an overall survival rate of 54.3%. Conclusion:The PTLD occurrence after allo-HSCT is highly correlated with EBV reactivation, and the higher the EBV-DNA load, the greater the risk of PTLD occurrence. The dynamic monitoring of EBV-DNA load plays an important role in predicting PTLD occurrence.
