1.Association between use of vibrating tools and low back pain in automobile manufacturing workers: A propensity score matching analysis

Background The prevalence of low back pain among workers using vibrating tools in the automobile manufacturing industry is high; however, research on associated risk factors remains limited. Objective To investigate the association between use of vibrating tools and low back pain among automobile manufacturing workers, and to estimate related population attributable risk percentage (PAR%). Methods A cross-sectional survey was conducted using the Chinese Musculoskeletal Disorders Questionnaire among workers with over one year of service in an automobile manufacturing enterprise. Workers were categorized into an exposed group (vibrating tool users) and a control group (non-users) based on their use of vibrating tools during work. The variables showing statistically significant differences between the two groups were selected as covariates for matching by 1∶2 nearest-neighbor propensity score. Chi-square test was used to compare the prevalence of low back pain between the matched exposed and control groups. Objective To evaluate the effect of dapagliflozin on myocardial function in early spontaneously hypertensive rats(SHR)with layer-specific global longitudinal strain(GLS).Methods A total of 45 male SHR aged 6 weeks were randomly divided into control group(normal saline),dapagliflozin group[1 mg/(kg·day)],and losartan group[10 mg/(kg·day)].Fifteen male Wistar-Kyoto(WKY)rats at same age with normal blood pressure were subjected and served as blank control group.During 8 weeks of intervention,systolic blood pressure(SBP)was measured,and conventional echocardiography and two-dimensional speckle tracking echocardiography(2DSTE)were performed and the results were collected to acquire the longitudinal strain of each layer of left ventricular(LV)myocardium.The parameters were compared among the groups.The pathological changes of myocardium were observed in each group of rats.Results Compared with the WKY group,LV ejection fraction(LVEF)and LV fraction shortening(LVFS)at week 8 were decreased in the control group(P<0.05),but no such decreases were observed in the dapagliflozin group and the losartan group.The GLS of endo-myocardium(GLSendo)at the 6th week was decreased,and GLSendo,GLSmid and GLSepi at the 8th week were all decreased in the control group than the WKY group(all P<0.05).But there were no statistical differences in the above 3 indicators in the dapagliflozin and losartan groups when compared with the WKY group(all P>0.05).The pathological results showed that myocardial interstitial fibrosis was observed in the control group at the 6th week.Conclusion Dapagliflozin can effectively improve myocardial function in early SHR.
3.Association between overtime work and work-related musculoskeletal disorders among male employees in the automobile manufacturing industry

ObjectiveTo explore the impact of overtime work on work-related musculoskeletal disorders in male employees in the automobile manufacturing industry. Methods A total of 1 731 male employees with more than one year of working experience from an automobile manufacturing industry were selected as the research subjects using judgment sampling method. The Musculoskeletal Disorder Questionnaire was used to investigate the prevalence of musculoskeletal disorder. Employees were divided into control group and overtime group, and a 1∶1 matching was performed using propensity score matching method, and 573 pairs were successfully matched. The prevalence of WMSDs in various body parts was compared between the two groups. Results The overtime working rate of the research subjects was 34.2%, and the prevalence of WMSDs was 57.1%. Overtime work increased the risk of WMSDs in the neck, shoulders, upper back, lower back, ankle/feet, and overall body of the workers (all P<0.05), with the odd ratio and 95% confidence interval of 1.43 (1.10-1.85), 1.38 (1.06-1.80), 1.42 (1.07-1.89), 1.28 (1.01-1.62), 1.37 (1.01-1.87), and 1.49 (1.17-1.89), respectively. However, there was no association between overtime work and the risk of WMSDs in the elbows, hands/wrists, hips, and knees of the subjects (all P>0.05). Conclusion Overtime work increases the risk of WMSDs in the neck, shoulders, upper back, lower back, ankles/feet, and overall body of male employees in the automobile manufacturing industry. Enterprises should improve labor organization, reduce overtime work, and protect the health of workers.

Since the approval of gemtuzumab ozogamicin,an antibody-drug conjugate(ADC)targeting CD33 in 2000,13 ADC drugs have been approved by the FDA.Although these drugs have clearly improved the survival of patients with various types of advanced cancers,their significant toxicity has compromised their therapeutic benefits.The adverse reactions of ADC drugs are complex and include on-target and off-target toxicities,where the payload drug is a determining factor.Antibody and linker may also affect the degree of toxicity.Combination therapy becomes an important strategy in anticancer treatment because of its increased efficiency,but treatment-related adverse reactions also increase accordingly.This review comprehensively analyzes the toxicity mechanisms of current ADC drugs and proposes various optimization strategies,including but not limited to optimizing linker molecules,upgrading antibody design,and changing drug administration strategies,to improve the overall safety profile of ADC drugs.
5.Clinical effects of minimally invasive negative pressure curettage combined with InMode diamond carving on axillary osmidrosis and axillary hyperhidrosis

Objective:To explore the efficacy of negative pressure suction curettage combined with InMode diamond carving in the treatment of axillary osmidrosis.Methods:A total of 200 patients with bromhidrosis, 89 males and 111 females, aged (27.85±7.98) years, were prospectively included in the study from February 2022 to February 2023 in Huamei Zixin Medical Cosmetology Hospital in Sichuan Province. According to different treatment methods, the patients were divided into 2 groups with 100 cases in each group by random number table method. The control group received negative pressure suction curettage, and the experimental group received combination with InMode diamond carving. After follow-up for one year, the efficacy, recurrence rate and incidence of adverse reactions were compared between the two groups.Results:In the experimental group, 94 cases were cured, 6 cases were effective, 0 cases were ineffective; the effective rate was 100% (100/100). In the control group, 90 cases were cured, 5 cases were effective and 5 cases were ineffective; the effective rate was 95% (95/100). The effective rate of experimental group was higher than that of control group, and the difference was statistically significant (χ 2=5.128, P=0.024). In the experimental group, no cases relapsed, and the recurrence rate was 0. In the control group, 5 cases relapsed, and the recurrence rate was 5.3% (5/95). The recurrence rate of experimental group was lower than that of control group, and the difference was statistically significant (χ 2=5.390, P=0.020) In control group, 5 cases of subcutaneous hematoma, 1 case of scar contracture, 7 cases of scar hyperplasia, and 1 case of skin necrosis occurred; the incidence of adverse reactions was 14.0% (14/100). In the experimental group, 2 cases of subcutaneous hematoma, 5 cases of scar contracture and 5 cases of scar hyperplasia occurred; the incidence of adverse reactions was 12.0% (12/100). There was no significant difference in the incidence of postoperative adverse reactions between the two groups (χ 2 = 0.177, P=0.674). Conclusions:Negative pressure suction curettage combined with InMode diamond carving has better curative effect and lower recurrence rate in the treatment of axillary osmidrosis.
6.Sanshentongmai Mixture Improves Oxidative Damage in Rat Cardiomyocytes H9C2 via Upregulation of microRNA-146a

Objective To investigate the effect of Sanshentongmai(SSTM)mixture on the regulation of oxidative damage to rat cardiomyocytes(H9C2)through microRNA-146a and its mechanism.Methods H9C2 were cultured in vitro,H2O2 was used as an oxidant to create an oxidative damage model in H9C2 cells.SSTM intervention was administered to the H9C2 cells.Then,the changes in H2O2-induced oxidative damage in H9C2 cells and the expression of microRNA-146a were observed to explore the protective effect of SSTM on H9C2 and its mechanism.H9C2 cells cultured in vitro were divided into 3 groups,including a control group,a model group of H2O2-induced oxidative damage(referred to hereafter as the model group),and a group given H2O2 modeling plus SSTM intervention at 500 μg/L for 72 h(referred to hereafter as the treatment group).The cell viability was measured by CCK8 assay.In addition,the levels of N-terminal pro-brain natriuretic peptide(Nt-proBNP),nitric oxide(NO),high-sensitivity C-reactive protein(Hs-CRP),and angiotensin were determined by enzyme-linked immunosorbent assay(ELISA).The expression level of microRNA-146a was determined by real-time PCR(RT-PCR).Result H9C2 cells were pretreated with SSTM at mass concentrations ranging from 200 to 1 500 μg/L.Then,CCK8 assay was performed to measure cell viability and the findings showed that the improvement in cell proliferation reached its peak when the mass concentration of SSTM was 500 μg/L,which was subsequently used as the intervention concentration.ELISA was performed to measure the indicators related to heart failure,including Nt-proBNP,NO,Hs-CRP,and angiotensin Ⅱ.Compared with those of the control group,the expressions of Nt-proBNP and angiotensin Ⅱ in the treatment group were up-regulated(P<0.05),while the expression of NO was down-regulated(P<0.05).There was no significant difference in the expression of Hs-CRP between the treatment group and the control group.These findings indicate that SSTM could effectively ameliorate oxidative damage in H9C2 rat cardiomyocytes.Finally,according to the RT-PCR findings for the expression of microRNA-146a in each group,H2O2 treatment at 15 μmol/L could significantly reduce the expression of microRNA-146a,and the expression of microRNA-146a in the treatment group was nearly doubled compared with that in the model group.There was no significant difference between the treatment group and the control group.Conclusion SSTM can significantly resist the H2O2-induced oxidative damage of H9C2 cells and may play a myocardial protective role by upregulating microRNA-146a.
7.Clinical value of 1, 3-β-D glucan and mannan IgM/IgG antibodies in the early diagnosis of invasive candidiasis

Objective:To evaluate the diagnostic value of 1, 3-β-D glucan (BDG), mannan IgM antibody (Mn-IgM) and mannan IgG antibody (Mn-IgG) in invasive candidiasis and to compare the differences in the diagnostic capability of serological markers used alone or in combination.Methods:Serum samples of 126 patients with invasive candidiasis and 104 healthy people who took physical examination during the same period were collected. BDG was detected by dynamic chromogenic method, and Mn-IgM and Mn-IgG were detected by ELISA. The sensitivity, specificity, positive predictive value, negative predictive value, Youden index, coincidence rate and Kappa value of the three serological markers used alone or in combination in the diagnosis of invasive candidiasis were analyzed and compared. The receiver operating characteristic (ROC) curves were drawn and the areas under the curves (AUCs) were calculated.Results:The levels of BDG, Mn-IgM and Mn-IgG in patients with invasive candidiasis were significantly higher than those in healthy people ( P<0.01). The sensitivity, specificity, Kappa value and AUC of BDG were 48.41%, 92.31%, 0.389 and 0.842. The sensitivity, specificity, Kappa value and AUC of Mn-IgM were 64.29%, 91.35%, 0.540 and 0.829. The sensitivity, specificity, Kappa value and AUC of Mn-IgG were 27.78%, 95.19%, 0.214 and 0.737. The sensitivity, specificity, Kappa value and AUC of BDG+ Mn-IgM were 76.19%, 88.46%, 0.637 and 0.921. The sensitivity, specificity, Kappa value and AUC of BDG+ Mn-IgG were 59.52%, 91.35%, 0.491 and 0.856. The sensitivity, specificity, Kappa value and AUC of Mn-IgM+ Mn-IgG were 69.84%, 90.38%, 0.588 and 0.891. The sensitivity, specificity, Kappa value and AUC of BDG+ Mn-IgM+ Mn-IgG were 80.16%, 88.46%, 0.679 and 0.922. Conclusions:The sensitivity of Mn-IgM was higher than that of BDG and Mn-IgG in the diagnosis of invasive candidiasis. When the serological biomarkers were used in combination, BDG+ Mn-IgM and BDG+ Mn-IgM+ Mn-IgG had relatively high Kappa value and AUC, showing high accuracy. The clinical diagnostic value of multiple serological biomarkers used in combination was significantly higher than that of any serological biomarkers used alone. Early combined detection and continuous monitoring of multiple serological biomarkers in patients with high risk of invasive candidiasis could be used clinically to adjust antifungal treatment strategies timely.
8. Immunogenicity and safety of co-immunization with 23-valent pneumococcal polysaccharide vaccine and influenza virus split vaccine for children aged 3-7 years

Objective:
To evaluate the immunogenicity and safety of concomitant administration of 23-valent pneumococcal polysaccharide vaccine (PPV23) and trivalent influenza vaccine (TIV) in preschoolers.
Methods:
A total of 1 035 children aged 3-7 years were enrolled in this study and randomly divided into three groups, inoculated PPV23, TIV and both, respectively. A one-year follow-up study was conducted for immunogenicity and safety analysis.
Results:
A total of 1 035 serological specimens were collected, including 327 in PPV23 group, 348 in TIV group and 360 in concomitant vaccination group. No significant differences in geometric mean concentrations (GMC) of seven pneumococcal serotypes were observed between the PPV23 group and the concomitant vaccination group. 9.Design, synthesis, and PPARalpha/gamma agonistic activity of novel tetrahydroisoquinoline derivatives.

A series of tetrahydroisoquinoline derivatives were prepared and their peroxisome proliferator-activated receptor (PPAR) alpha/gamma agonistic activities were evaluated to obtain more potent PPAR agonist. All of them were new compounds, and their structures were confirmed by 1H NMR and HR-MS. Three compounds exhibited higher agonistic activities of PPARgamma than that of the comparison, six compounds exhibited higher agonistic activities of PPARalpha than that of the comparison, and compound 8a was discovered as a highly potent PPARalpha/gamma agonist that is much more active than that of WY14643 and rosiglitazone. The development of potent PPAR agonists may offer a new choice for the treatment of diabetes.
10.Expression of bcl-2 mRNA and protein suppressed by antisense oligodexyonucleotide with phosphorothiote-modification in human melanoma A375 cells line

Objective To observe the effect of bcl-2 antisense oligodexyonucleotide (ASODN) by phosphorothiote-modification on the expression of bcl-2 mRNA and protein in human melanoma A375 cells line. Methods With the phosphorothiote-modification and liposome-encapsulation of ASODN, A375 cells were divided into ASODN group, nonsense oligodexyonucleotide (SODN) group and control group. Bcl-2 mRNA and protein were detected by RT-PCR and immunohistochemistry, respectively. Results The expression of bcl-2 protein was remarkably decreased in ASODN group than that in SODN group and control group (53.14 ±4.26 vs 138.22 ± 8.45, 53.14 ± 4.26 vs 141.08 ± 7.83, both P < 0.01 ). The level of bcl-2 mRNA was significantly lower than that in SODN group and control group (0.38 ± 0. 11 vs 0.96 ±0.13, 0. 38 ± 0.11 vs 0. 97 ± 0. 14, both P < 0. 01 ). Conclusion Bcl-2 antisense oligodexyonucleotide could down-regulate the bcl-2 level and block its protein expression.
