1.Epidemiological investigation and analysis of a local dengue fever cluster outbreak in Qingpu District of Shanghai

ObjectiveTo analyze the epidemiological characteristics of a local dengue fever cluster outbreak in Qingpu District of Shanghai in 2024, and to provide a reference for subsequent dengue fever prevention and control. MethodsSeven confirmed local dengue fever cases reported through the National Notifiable Infectious Diseases Surveillance System in Qingpu District of Shanghai in 2024 were selected as the research subjects. Descriptive epidemiological methods were used to conduct investigation and analysis from the aspects of onset, medical treatment and reporting, clinical symptoms, travel and contact history within 15 days before onset, and activity trajectories. ResultsA total of 7 cases were identified in this outbreak. None of the cases had a travel history to dengue-endemic areas within 15 days prior to onset, while all had shared exposure environments and mosquito bite histories, indicating a local clustered transmission pattern. The main clinical manifestations included fever (100.00%) and myalgia (42.86%). All 7 cases were positive for dengue virus serotype 2 (DENV-2) by nucleic acid testing. Genetic sequencing showed that the virus strains belonged to the Cosmopolitan genotype and were most closely related to the epidemic DENV strains circulating in southern China in recent years. ConclusionThis outbreak might be a local secondary infection caused by the short-term stay of dengue fever-infected individuals, and the possible source of importation was dengue fever endemic areas in southern China.
2.Investigation of parasitic infection in food on market in Qingpu District of Shanghai during 2015‒2023

ObjectiveTo understand the status of parasitic infection in the food sold on market in Qingpu District of Shanghai, and to provide an evidence for the development of prevention and control strategies for parasitic infection applicable to Qingpu District. MethodsAquatic products, meat products and other foodstuffs sold on online shops, at farm product markets, supermarkets/foodstores and restaurants were sampled in Qingpu District, Shanghai, during 2015‒2023, based on the administrative division of Qingpu District. The parasitic infection in the food samples were examined using pressing method and digestion for detecting metacercariae in freshwater products and pickled products, using dissection microscopy for Anisakis larvae in seawater products, Taenia cysticercus and Trichinella encysted larvae in meat products. ResultsA total of 1 079 samples of food products were examined during 2015‒2023, with a total parasite infection rate of 13.44%. The total parasite infection rate of freshwater fish products was 3.40% (16/471), and the difference of parasite infection rates between different freshwater fish species was statistically significant (χ2=229.609, P=0.001). The total infection rate of Clonorchis sinensis was 3.18% (15/471), which had been detected in Pseudorasbora parva, Cyprinidae rhodeus, and Carassius auratus, with a positive rate of 77.78% (7/9), 50.00% (5/10) and 3.90% (3/77), respectively. Metorchis orientalis was detected in in Pseudorasbora parva, with a positive rate of 33.33% (3/9). The positive rate of Gnathostoma spinigerum (third-stage larvae) was 0.81%. Paragonimus metacercariae were not detected in the freshwater shrimps and crabs. The infection rate of seawater fish products was 26.46%. The difference of parasite infection rate in seawater fishes was statistically significant (χ2=109.181, P=0.001). A total of 53 pork and beef samples were tested, none was detected with Trichinella larva cysts, Taenia solium metacercariae, and Taenia saginata metacercariae. The total infection rate of pickled yellow mud snail products was 58.11% (43/74). Paragonimus metacercariae was not detected in any of the pickled aquatic product samples. ConclusionThere are different degrees of parasitic infection in freshwater products, seawater products and pickled aquatic products in Qingpu District of Shanghai. The risk of parasite infection from raw or undercooked foods is high. Health education on healthy dietary practices such as throughly cooked food should be strengthened for local residents.
3.Impact of curculigoside on neuronal pyroptosis in rats with acute cerebral infarction by regulating cGAS-STING signaling pathway

Objective:To explore effect of curculigoside on neuronal pyroptosis in rats with acute cerebral infarction(CI)by regulating cyclic guanosine-adenosine synthase(cGAS)-interferon gene stimulating factor(STING)signaling pathway.Methods:Fifteen rats were randomly selected as sham operation group,and remaining rats were constructed CI models by modified Longa suture method,CI rats successfully modeled were randomly divided into CI group,curculigoside group(5 mg/kg),SR-717 group(3 mg/kg cGAS-STING signaling pathway activator SR-717)and curculigoside+SR-717 group(5 mg/kg curculigoside+3 mg/kg SR-717),with 15 rats in each group,injected once a day for 7 consecutive days,sham operation group and CI group were given equal amounts of nor-mal saline.Neural function was evaluated by Zea-Longa score;inflammatory factors levels were detected by ELISA;TTC staining was used to evaluate volume of CI;TUNEL staining was used to detect neuronal apoptosis;immunofluorescence staining was used to detect GSDMD-N expression;Western blot was used to detect expression of pyroptosis and cGAS-STING pathway proteins.Results:Com-pared with sham operation group,Zea-Longa score,CI volume,IL-1β and IL-18 levels,apoptosis rate,number of GSDMD-N positive cells,NLRP3,cleaved-Caspase-1/Caspase-1,GSDMD-N,cGAS and STING protein expressions in CI group were significantly increased(P<0.05);compared with CI group,Zea-Longa score,CI volume,IL-1β and IL-18 levels,apoptosis rate,number of GSDMD-N positive cells,NLRP3,cleaved-Caspase-1/Caspase-1,GSDMD-N,cGAS and STING protein expressions in curculigoside group were significantly decreased(P<0.05),while the above indexes in SR-717 group had opposite trend(P<0.05);SR-717 reversed improvement effect of curculigoside on neuronal pyroptosis in CI rats.Conclusion:Curculigoside may improve neuronal pyroptosis in CI rats by down-regulating cGAS-STING signaling pathway.
4.Mechanistic study on circVAPA promoting the Hippo signaling pathway to inhibit liver regeneration via miR-101a-3p/TEAD3 axis

Objective To explore the molecular mechanism of circular RNA(circ)VAPA promoting the Hippo pathway to inhibit liver regeneration through the miR-101a-3p/TEAD3 axis.Methods A mouse model of 70%partial hepatectomy-induced liver regeneration was constructed,and the expressions of circVA-PA,miR-101a-3p,and TEAD3 were analyzed.Mouse embryonic hepatocyte BNL CL.2 cells were transfected with siRNA or overexpression plasmids and divided into the VAPA-NC group,the VAPA-NC+miR-101a-3p mimic group,the VAPA-NC+miR-101a-3p mimic-NC group,the VAPA-OE group,the VAPA-OE+miR-101a-3p mimic group and the VAPA-OE+miR-101a-3p mimic-NC group.CCK-8 method and flow cytometry were used to analyze the proliferation,apoptosis,and cell cycle changes of hepatocytes.Immunofluorescence staining was used to analyze the nuclear translocation of YAP1.Quantitative Reverse Transcription Real-Time PCR(qRT-PCR)was used to analyze the expressions of key genes in the Hippo pathway.The dual-lu-ciferase reporter gene assay was used to verify the targeting relationships between circVAPA and miR-101a-3p,as well as between miR-101a-3p and TEAD3.Results The level of circVAPA gradually increased during liver regeneration(P<0.05),while the level of miR-101a-3p first increased and then decreased(P<0.05).When miR-101a-3p was overexpressed alone,the cell proliferation rate was the highest(P<0.05),but it had no effect on the cell apoptosis rate(P>0.05).When circVAPA was overexpressed alone,it had no effect both on the cell proliferation rate and apoptosis(P>0.05).After co-overexpression of circVAPA and miR-101a-3p,the cell proliferation rate significantly decreased,and the cell apoptosis significantly increased(P<0.05).When miR-101a-3p was overexpressed alone,a large number of cells entered into the S phase.After co-overex-pression of circVAPA and miR-101a-3p,a large number of cells were blocked in the G2/M phase.The phos-phorylation level of the Hippo upstream gene YAP1 significantly increased at 6 hours after liver regeneration(P<0.05)and then rapidly decreased.However,co-overexpression of circVAPA and miR-101a-3p did not af-fect the level of p-YAP1 and the nuclear translocation of YAP1(P>0.05).The expression levels of the Hip-po downstream gene CTGF and the transcription factor TEAD3 first increased and then decreased during liver regeneration(P<0.05),and there was no significant change in CYR61(P>0.05).After co-overexpression of circVAPA and miR-101a-3p,the expression level of CTGF increased(P<0.05).Knocking down or overex-pressing circVAPA did not affect the expression of TEAD3(P>0.05),while overexpressing miR-101a-3p could significantly inhibit the expression level of TEAD3(P<0.05).The dual-luciferase reporter gene assay confirmed the targeting relationships between circVAPA and miR-101a-3p,as well as between miR-101a-3p and TEAD3.Conclusion circVAPA promotes the Hippo pathway to inhibit liver regeneration through the miR-101a-3p/TEAD3 axis.
5.Progress of clinical detection methodology of varicella vaccine serum antibody

Varicella is a common infectious disease caused by varicella-zoster virus (VZV), with high transmissibility and pathogenicity, and vaccination is a better preventive measure. The clinical diagnosis of varicella, the clinical immunogenicity evaluation of varicella vaccine, the continuous monitoring of the immune effect after marketing and the epidemiological investigation all require the detection of the antibody level of VZV in patients. Fluorescent antibody to membrane antigen method (FAMA) is the gold standard for detection of VZV antibody in serum, possessing advantages of high sensitivity and good specificity, playing an important role in detection of VZV antibody. However, the defects of FAMA operation complexity and low throughput are also troubling the clinical testing personnels. This article reviews research progress both at home and abroad on the technology of VZV serum antibody detection. The advantages and limitations of the current detection methods are discussed and the future development direction are highlighted on the dectection of VZV serum antibody IgG.
6.Impact of curculigoside on neuronal pyroptosis in rats with acute cerebral infarction by regulating cGAS-STING signaling pathway

Objective:To explore effect of curculigoside on neuronal pyroptosis in rats with acute cerebral infarction(CI)by regulating cyclic guanosine-adenosine synthase(cGAS)-interferon gene stimulating factor(STING)signaling pathway.Methods:Fifteen rats were randomly selected as sham operation group,and remaining rats were constructed CI models by modified Longa suture method,CI rats successfully modeled were randomly divided into CI group,curculigoside group(5 mg/kg),SR-717 group(3 mg/kg cGAS-STING signaling pathway activator SR-717)and curculigoside+SR-717 group(5 mg/kg curculigoside+3 mg/kg SR-717),with 15 rats in each group,injected once a day for 7 consecutive days,sham operation group and CI group were given equal amounts of nor-mal saline.Neural function was evaluated by Zea-Longa score;inflammatory factors levels were detected by ELISA;TTC staining was used to evaluate volume of CI;TUNEL staining was used to detect neuronal apoptosis;immunofluorescence staining was used to detect GSDMD-N expression;Western blot was used to detect expression of pyroptosis and cGAS-STING pathway proteins.Results:Com-pared with sham operation group,Zea-Longa score,CI volume,IL-1β and IL-18 levels,apoptosis rate,number of GSDMD-N positive cells,NLRP3,cleaved-Caspase-1/Caspase-1,GSDMD-N,cGAS and STING protein expressions in CI group were significantly increased(P<0.05);compared with CI group,Zea-Longa score,CI volume,IL-1β and IL-18 levels,apoptosis rate,number of GSDMD-N positive cells,NLRP3,cleaved-Caspase-1/Caspase-1,GSDMD-N,cGAS and STING protein expressions in curculigoside group were significantly decreased(P<0.05),while the above indexes in SR-717 group had opposite trend(P<0.05);SR-717 reversed improvement effect of curculigoside on neuronal pyroptosis in CI rats.Conclusion:Curculigoside may improve neuronal pyroptosis in CI rats by down-regulating cGAS-STING signaling pathway.

8.Progress in relationship between expression of solute carrier family 3 member 2,and disulfidptosis,ferroptosis and tumors

Ferroptosis and disulfidpthosis are associated with various diseases.Ferroptosis is distinguished by abnormal accumulation of iron ions and lipid reactive oxygen species,while disulfidptosis is marked by abnormal accumu-lation of disulfides.Solute carrier family 3 member 2(SLC3A2),a component of the solute carrier family,is implicated not only involved in the regulation of tumor cell proliferation,migration,invasion and apoptosis,but also in the processes of ferroptosis and disulfidptosis,playing a pivotal role in tumor growth and progression.This article provides an overview of the latest advancements in understanding the relationship between SLC3A2 and ferroptosis,disulfidptosis and tumors,aiming to establish a theoretical basis for targeted cancer therapies.
9.Study on neuroprotective effect of alpinetin on ischemic stroke rats by regulating Shh/Gli1 signaling pathway

Objective:To investigate neuroprotective effect of alpinetin on ischemic stroke(IS)rats by regulating Shh/Gli1 signaling pathway.Methods:Fifteen rats were randomly collected as control group,remaining rats were used to construct an IS model.Rats that successfully modeling were randomly grouped into model group,alpinetin group(5 mg/kg),Shh/Gli1 signaling pathway inhibitor cycloparamide group(15 mg/kg)and alpinetin+cycloparamide group(5 mg/kg alpinetin+15 mg/kg cycloparamide),with 15 rats in each group,and administered once a day for two consecutive weeks,control group and model group were given equal amounts of physiological saline.Zea-Longa scoring method was applied to evaluate neural function;ELISA was applied to detect inflammatory factors levels;mass of brain tissue was weighed and water content of brain tissue was calculated;TTC staining was applied to measure volume of cerebral infarction;HE staining was applied to observe nerve cell damage;TUNEL staining was applied to detect neuronal apoptosis;qRT-PCR and Western blot were used to detect mRNA and protein expressions of Shh and Gli1.Results:There were no abnormalities in hippocampal tissue of control group,while hippocampal tissue structure of model group rats was abnor-mal,with disordered cell arrangement and nuclear pyknosis of nerve cells,cell damage rate,Zea-Longa score,infarct volume,IL-1β,IL-6,TNF-α,brain tissue water content,and cell apoptosis rate in model group were obviously higher than control group(P<0.05),mRNA and protein levels of Shh and Gli1 were obviously decreased(P<0.05);compared with model group,cells in alpinetin group were arranged more neatly,and phenomenon of neuronal cell nucleus pyknosis was improved,cell damage rate,Zea-Longa score,infarct volume,IL-1β,IL-6,TNF-α,brain tissue water content,and cell apoptosis rate were obviously decreased(P<0.05),mRNA and protein levels of Shh and Gli1 were obviously increased(P<0.05),trend of above indicators in cyclophosphamide group was opposite,ciclopramide reversed neuroprotective effect of alpinetin on IS rats.Conclusion:Alpinetin may exert neuroprotective effects on IS rats by activating Shh/Gli1 signaling pathway.

The current situation of tumor radiotherapy teaching is far behind the development of radiotherapy technologies. The construction of a teaching system based on an artificial intelligence-powered automatic target delineation system and a standardized cancer radiotherapy case library is operable and practical for realizing the standardization and homogenization of clinical target volume delineation teaching, improving students' precision and speed of target volume delineation, and promoting students' learning interest, initiative, and efficiency, which can bring new vitality to the development of radiotherapy education and is worthy of further exploration and promotion.
