1.Screening of initial processing methods for Ligusticum sinense slice based on differential metabolites

OBJECTIVE To screen the primary processing methods of Ligusticum sinense slice based on differential metabolites, and provide theoretical basis for the scientific processing of L. sinense. METHODS Using 13 groups of L. sinense slice processed by fresh-cutting or traditional methods as samples, UHPLC-QE-MS was employed for metabolite identification. Multivariate statistical analysis was applied to screen differential metabolites among the 13 sample groups, analyzing the effects of washing, soaking, drying methods, and drying cycles on both the relative expressions of differential metabolites and the contents of carboxylic acids and their derivatives in the samples (to reflect the total amino acid content). RESULTS Principal component analysis and partial least squares-discriminant analysis both showed significant intergroup differences among the 13 sample groups. A total of 688 differential metabolites were screened from the 13 sample groups, with carboxylic acids and their derivatives showing the highest proportion. The relative expression levels of phosphatidylcholine significantly increased after washing treatment, while tryptophan expression significantly decreased after soaking treatment. Samples dried at 50-60 ℃ showed significantly increased expression of psoralen, whereas those dried at 40 ℃ showed significantly decreased expression of methyl -p- methoxycinnamate. Both washing and soaking treatments significantly reduced the total amino acid content in samples, while secondary drying significantly increased it. The three controlled-temperature drying methods maintained relatively stable total content of amino acids in samples. CONCLUSIONS The optimal processing protocol for L. sinense slice is as follows: fresh L. sinense slice should be freshly cut at the production site, undergo quick washing after soil removal, and be dried twice at 40 ℃ (before and after slicing).
3.Diagnostic and prognostic value of combined detection of serum Lp-PLA2,NSE and S-100β in patients with carbon monoxide poisoning complicated with acute cerebral infarction

Objective To explore the value of combined detection of lipoprotein-associated phospholipase A2(Lp-PLA2),neuron specific enolase(NSE)and S-100 calcium binding protein β(S-100β)in the diagnosis and prognosis evaluation of acute cerebral infarction(ACI)in patients with carbon monoxide poisoning(CMP).Methods A total of 102 patients with CMP complicated with ACI admitted to the hospital from Jan-uary 2020 to November 2021 were selected as the study group,meanwhile,102 patients with simple CMP were enrolled as the control group.Patients in the study group were followed up for 6 months after discharge,ac-cording to the follow-up results,they were grouped into good prognosis group(60 cases)and poor prognosis group(42 cases).The serum levels of Lp-PLA2,NSE and S-100β were detected by enzyme-linked immunosor-bent assay(ELISA).The receiver operating characteristic(ROC)curve was applied to analyze the value of the combination of serum Lp-PLA2,NSE and S-100β in the early diagnosis and prognosis evaluation of patients with CMP and ACI.Results Compared with the control group,the levels of Lp-PLA2,NSE and S-100β in the study group were obviously higher(P<0.05).The ROC curve analysis results showed that the area under the curve(AUC)of the combined detection of serum Lp-PLA2、NSE、S-100β for the diagnosis of CMP complicat-ed with ACI was greater than the AUC of single detection of each indicator(P<0.001).Compared with the good prognosis group,the levels of Lp-PLA2,NSE and S-100β in the poor prognosis group were obviously higher(P<0.05).The results of ROC curve analysis showed that the AUC of the combined detection of ser-um Lp-PLA2、NSE、S-100β for the prognosis of patients with CMP complicated with ACI was greater than the AUC of single detection of each indicator(P<0.05).Conclusion The expression of Lp-PLA2,NSE and S-100β in serum of patients with CMP complicated with ACI is high,and the combined detection of the three has certain value in the diagnosis and prognosis evaluation for patients with CMP complicated with ACI.
4.GNAO1 gene-related dystonia

Dystonia is a movement disorder characterized by involuntary abnormal movements or postures due to sustained or intermittent muscle contractions. In recent years, rapid progress has been achieved in the genetics of this disease, leading to an in-depth understanding of common genes and phenotypes related to dystonia. GNAO1 is one of the genes closely associated with dystonia. GNAO1 gene, nevertheless, remains largely unknown to most clinicians. This article reviews the molecular genetic features, clinical characteristics, and therapeutic strategies of GNAO1 gene-associated dystonia, thereby increasing the knowledge of clinicians about GNAO1 gene-associated dystonia and preventing underdiagnosis and misdiagnosis in clinical practice.
5.Thrombus permeability predicts the outcome after endovascular therapy in patients with middle cerebral artery occlusive stroke

Objective:To investigate the predictive value of thrombus permeability for poor outcome and symptomatic intracranial hemorrhage (sICH) after endovascular therapy (EVT) in patients with middle cerebral artery occlusion stroke.Methods:Patients with middle cerebral artery occlusion stroke underwent EVT at the Cardiocerebrovascular Disease Hospital, the Affiliated Hospital of Yan'an University from January 2018 to January 2024 were included retrospectively. Thrombus attenuation increase (TAI) was used to evaluate thrombus permeability. The poor outcome was defined as a modified Rankin Scale score >2 at 90 days after procedure. sICH was defined as an increase of ≥4 of the National Institutes of Health Stroke Scale (NIHSS) score relative to baseline or lowest within 7 days after EVT, and CT scan showed cerebral hemorrhage. Multivariate logistic regression analysis was used to determine the independent influencing factors of poor outcome and sICH. Receiver operating characteristic (ROC) curves was used to evaluate the predictive value of TAI for poor outcome and sICH. Results:A total of 77 patients with middle cerebral artery occlusion stroke received EVT treatment were enrolled, including 44 males (57.1%), aged 62.1±12.4 years. Thirty-three patients (48.1%) had poor outcome, 35 (45.5%) experienced hemorrhagic transformation, of which 12 (15.6%) were sICH. Multivariate logistic regression analysis showed that TAI (odds ratio [ OR] 0.930, 95% confidence interval [ CI] 0.883-0.980; P=0.007) and sICH ( OR 0.868, 95% CI 0.784-0.961; P=0.006) were the independent influencing factors of poor outcome. ROC curve analysis showed that the area under the curve of TAI for predicting poor outcome at 90 days was 0.836 (95% CI 0.742-0.930). The cut-off value was 10.135 HU. The sensitivity and specificity were 73.0% and 92.5%, respectively. The area under the curve of TAI for predicting sICH was 0.750 (95% CI 0.637-0.902). The cut-off value was 18.200 HU. The sensitivity and specificity were 94.1% and 64.5%, respectively. Conclusions:TAI has certain predictive value for poor outcome and sICH after EVT in patients with middle cerebral artery occlusion stroke. Patients with higher thrombus permeability are less likely to develop sICH after EVT and have a higher likelihood of good outcome.
6.Effects of ingestion training based on food form modification on improving swallowing disorder after stroke

Objective To explore the effects of ingestion training based on food form modification on improving swallowing function among swallowing disorder patients after stroke. Methods From January 2018 and June 2019,this study selected 80 swallowing disorder patients after stroke of Department of Rehabilitation at Xuanwu Hospital Capital Medical University as subjects by convenience sampling.All of patients were divided into control group and observation group with the random number table,40 cases in each group.Control group carried out routine ingestion training. On the basis of that,observation group implemented ingestion training based on food form modification. The swallowing function was evaluated with the Modified Mann Assessment of Swallowing Ability(MMASA)before and after treatment. At the same,the rates of complication incidences and nutritional status were observed between two groups. Results After intervention,the score of MMASA of observation group were(144.10±16.56)higher than that(136.48±13.56)of control group with a statistical difference(P<0.05);the incidences of aspiration and pulmonary infection of observation group were lower and serum albumin was higher than those of control group with statistical differences(P<0.05). However,there was no statistical difference in the body mass index(BMI)between two groups(P>0.05). Conclusions Ingestion training based on food form modification and swallowing training can promote the recovery of swallowing function among swallowing disorder patients after stroke and reduce the incidences of complication which has a positive role in the prognosis of patients.
7.The Characteristics of the Working Memory in Patients with Chinese Aphasia

Objective To study the characteristics of the working memory in patients with Chinese aphasia in different ways of tasks and test materials .Methods Fifteen aphasia patients (aphasia group ,54~70 years old) and fifteen matched normal cases (control group) were requested to finish auditory retelling and auditory recognition tests of numerical materials ,content and function words .The maximum length of the string correctly completed was referred to as the working memory span .The working memory span was compared between the two groups .Results The working memory span of the aphasia group was significantly smaller than that of in the normal group (P<0 .01) .The working memory span of in patients with the aphasia group in the positive sequence retelling tasks were significantly greater than in reverse retelling ,positive recognition and reverse recognition (P<0 .01) .Reverse retell-ing was significantly greater than reverse recognition (P<0 .01) .The positive recognition was significantly greater than the reverse recognition(P<0 .01) .In retelling tasks ,the order of the working memory span was numbers , function words ,content words from high to low in the aphasia group ,but there was no significant difference be-tween them .In the positive recognition tasks ,the average working memory span for the numerical material was sig-nificantly greater than the content words(P< 0 .01) and the function words(P< 0 .01) .The content words were greater than the function words(P<0 .01) .In the reverse recognition tasks ,average working memory span for the numerical material was significantly greater than the function words (P<0 .01) .Conclusion The working memoryspan of in patients with aphasics significantly lags behind the normal people .Numbers are the easiest for aphasics . In different task modes ,the difficulties from high to low are as follow :positive retelling ,reverse retelling ,positive recognition and reverse recognition .
8.Correlation between CEUS parameters and angiogenesis and cell proliferation of gallbladder carcinoma transplant tumor in nude mice

Objective To explore the correlation between CEUS blood perfusion characteristics and angiogenesis and cell proliferation in nude mice models of gallbladder carcinoma transplant tumor.Methods Nude mice models of gallbladder carcinoma transplant tumor were established.The blood perfusion characteristics of the nude mice models were quantitatively analyzed,CEUS quantitative parameters and the expression levels of microvessel density (MVD),vascular endothelial growth factor (VEGF) and proliferating cell nuclear antigen (PCNA) immunohistochemically were obtained.The correlations were performed.Results Twenty tumors were successfully established.The CEUS of 16 tumors was successful.There was positive correlation between peak intensity (PI) and MVD,PCNA (r=0.635,0.514,P=0.008,0.042).The CEUS parameters were not correlated with expression of VEGF.MVD had positive correlation with expression of VEGF,PCNA (r=0.680,0.650,P=0.004,0.006);VEGF had positive correlation with expression of PCNA (r=0.611,P=0.012).Conclusion The PI of CEUS is positive correlation with MVD and PCNA in nude mice models of gallbladder carcinoma transplant tumor,which may be helpful to evaluate tumor angiogenesis and cell proliferation state.The MVD,VEGF and PCNA positively correlates to each other,which indicates there is the relationship of reciprocal promotion between tumor angiogenesis and cell proliferation,and is good for tumor growth.
9.Anti-angiogenic therapy for gallbladder carcinoma using contrast enhanced ultrasound

Objective To evaluate the feasibility of contrast enhanced ultrasound (CEUS) in gallbladder carcinoma anti-angiogenesis treatment.Methods Subcutaneous gallbladder carcinoma model was consturcted.The mice were divided into intervention-drug group and control group randomly.The mice of intervention-drug group were treated with Endostar (10 mg · kg-1 · d-1) by intraperitoneal injection for two weeks.Two groups of mice were detected by CEUS,then the time of arrival (AT),peak time (TTP),peak intensity (PI),area under the curve (AUC) were measured via time-intensity curve.Expression of MVD and VEGF both in the intervention and control groups were studied through immunohistochemistry.and the correlations between MVD,VEGF and CEUS parameteres were further analyzed.Results The mean values of PI in drug intervention group and control group were 10.8 ± 5.5 and 16.8 ± 5.8,respectively.The values of PI in intervention-drug group were lower than that in control group significantly (P < 0.05).There was no significant difference in AT,TTP,AUC between the two groups.The mean values of MVD on drug intervention group and control group were 8.5 ± 3.8 and 13.1 ± 3.5,respectively.The mean values of VEGF on drug intervention group and control group were 4.3 ± 0.5 and 4.7 ± 0.4,respectively.The values of MVD and VEGF in intervention-drug group were significant lower than that control group (P < 0.05).MVD and VEGF values of intervention-drug group were correlated with PI (r =0.712,P < 0.05;r =0.739,P < 0.05).Conclusion Endostar can inhibit the growth of gallbladder carcinoma and PI can be used as an effective marker to evaluatethe effect of anti-angiogenic therapy in gallbladder carcinoma.
10.Research Progresses in Single Molecule Enzymology

The single molecule imaging and technologies that developed in 1990 s have successfully probed the dynamics of single molecule enzyme catalysis in real time in vitro. Ever since then, single molecule enzymology has entered the golden age of rapid developing. Individual features of each enzyme hidden in the overall average have been discovered, and many new catalytic mechanisms have been proposed. Single molecule enzymology sheds light on the dynamic interactions between enzymes and substrates or products, deepening the understanding of biochemical reactions. This review described the recent research progresses of single molecule protease and ribozyme.
