1.Identification of the cisAB (c. 796A>C) allele and molecular docking of its transferase

Objective: To reveal the molecular basis of the cisAB (p. Met266Leu) glycosyltransferase by studying a proband with cisAB subtype and his family. Methods: A male newborn was selected as the research subject. Tube methods were used to identify ABO blood types of the proband and his family members. PCR-SSP detection, ABO gene sequencing, and cloning analysis were performed on the proband and some family members. The inheritance pattern of the subtype gene in the family was determined through pedigree analysis. Homology modeling was used to analyze the impact of amino acid variations on the structure of the transferase, and molecular docking was used to demonstrate the bifunctional activity of the transferase and the donor-receptor binding conformation. Results: Serological tests showed that the proband and his father had enhanced anti-H agglutination, and the grandmother had a forward and reverse discrepancy. Sequencing of the proband revealed heterozygous variations of c. 297A>G, c. 526C>G, c. 657C>T, c. 703G>A, c. 803G>C, and c. 930G>A compared with A1. 01 (compared with B. 01, lacking the c. 796C>A variation, namely harboring the c. 796A>C variation) and c. 261delG. Combined with cloning analysis, the proband's genotype was determined to be ABO

O. 01. 01, the father's genotype was ABO
cisAB (c. 796A>C)/ABO
O. 01. 02, and the grandmother's genotype was ABO
cisAB (c. 796A>C)/ABO
B102. Pedigree analysis indicated that the cisAB allele in this newborn was inherited from his father and grandmother rather than a natural mutation. Homology modeling showed that the side chain orientation and intermolecular forces of Leu266 in the cisAB (p. Met266Leu) transferase changed, and molecular docking demonstrated that the "binding pocket" of the active center of the variant enzyme could accommodate both UDP-GalNAc and UDP-Gal, indicating that the cisAB enzyme structure has bifunctional activity. Conclusion: The bifunctional activity of this cisAB (p. Met266Leu) enzyme is related to the nucleotide variation of c. 796A>C, and molecular docking indicates that the enzyme has dual affinity for A/B sugar donors.
2.Effect of reminiscence therapy of Montessori educational philosophy on the elderly with mild cognitive impairment

Objective To develop a reminiscence therapy program with Montessori educational philosophy in treatment of elderly people with mild cognitive impairment(MCI)in nursing homes and evaluate its effect.Methods On the basis of Montessori education principles,a reminiscence therapy program was drafted through literature review and had it finalised with expert consultations and a pilot study.Between February and May 2022,70 elderly with MCI were recruited from a nursing home in Liaoning Province using convenience sampling.The recruited elderly were randomly divided into a control group(n=35)and an intervention group(n=35)using a random number table.The elderly in control group received conventional cognitive training only while the elderly in the intervention group received reminiscence therapy in addition to the conventional cognitive training.Cognitive function and ability of daily living(ADL)were compared between the two groups after a 6-week intervention.Results The elderly in intervention group demonstrated statistically significant improvements in both cognitive function and dimensions(visuospatial/executive function,naming ability,delayed recall and orientation,as well as in ADL after a 6-week intervention(all P<0.05).Conclusion The Montessori philosophy based on reminiscence therapy improves cognitive function and activities of daily living in the elderly with MCI,particularly from the perspectives of visuospatial/executive function,naming ability,delayed recall and orientation.

The early non-invasive diagnosis,intervention and treatment of cardiac diseases are of great clinical significance,and the changes in myocardial microstructure play a key role in understanding alterations in cardiac function.Cardiac diffusion tensor imaging is a novel non-invasive technique,and its characteristic parameters have the capability to quantify the orientation and density of cardiomyocytes.In recent years,with the development and optimization of sequence design for cardiac diffusion imaging,this technique has been extensively investigated for exploring myocardial microstructure and assessing alterations in cardiac function.This review presents an overview of the technical development,characteristic parameters,preliminary clinical findings of cardiac diffusion tensor imaging,as well as other cardiac diffusion imaging models and applications.

Conventional 1.5T and 3.0T cardiac MRI(CMRI)had been widely used.The ultrahigh field MR behaved better in image resolution and signal-to-noise ratio.The domestic 5.0T whole-body ultrahigh field MRI had better balance between the field strength and quality in CMRI,which was expected to improve imaging quality and efficiency.The status challenges and future of 5.0T CMRI were reviewed in this paper.
7.Dosimetric effects of flatten filter on hippocampal-avoidance whole brain non-uniform intensity-modulated radiotherapy

Objective:To study the dosimetric differences of hippocampal-avoidance whole brain radiotherapy(HA-WBRT)between non-uniform 9-fields intensity-modulated radiotherapy-flatten filter(9F-IMRT-FF)and 9-fields intensity-modulated radiotherapy-flatten filter-free(9F-IMRT-FFF)mode.Methods:From January to December 2023,the simulated images of computed tomography(CT)location of 20 patients with intracranial metastases who had completed whole-brain radiotherapy at the Department of Radiotherapy of Chengde Central Hospital were selected.The radiotherapy plans of two kinds of modes of brand-new 9F-IMRT-FF and 9F-IMRT-FFF were redesigned in the planning system.The differences of dosimetric parameters of the two kinds of mode plans were compared,as well as the differences of irradiation efficiency of accelerator.Results:The exposure dose(D2%)of 2%volume of target region of planned target volume(PTV)of 9F-IMRT-FFF mode was significantly lower than that of 9F-IMRT-FF mode(t=-2.496,P<0.05).The exposure dose(D100%)of 100%volume of target region,the maximum dose(Dmax)and the mean dose(Dmean)of the left side of hippocampus of 9F-IMRT-FFF mode plan were significantly lower than those of 9F-IMRT-FF mode plan,with statistically significant(Z=-3.179,-3.018,-2.145,P<0.05),respectively.The D100%and Dmax of the right side of hippocampus of 9F-IMRT-FFF mode plan were significantly lower than those of 9F-IMRT-FF mode plan,with statistically significant(Z=-3.099,-3.260,P<0.05),respectively.The monitor unit(MU)value of 9F-IMRT-FFF mode plan increased by 25%than that of 9F-IMRT-FF mode plan,while the treatment time of 9F-IMRT-FFF mode plan shortened 38%than that of 9F-IMRT-FF mode plan,with statistically significant(t=-9.664,-13.312,P<0.05),respectively.Conclusion:Both the non-uniform 9F-IMRT-FF and the 9F-IMRT-FFF can meet the clinically requirement.The 9F-IMRT-FFF has better modulation capabilities in beams,which has more advantages in reducing the dose of radiotherapy for hippocampus,and reducing treatment time,and increasing treatment efficiency.
8.Theoretical Model Study on Morphology of Lipid Droplet on Inner Vascular Wall

Objective To establish the theoretical model for morphology of lipid droplets adhering to inner vascular wall, so as to provide the theoretical model for the study and analysis of the overall morphology of lipid droplets on inner vascular wall of patients with fat embolism. MethodsThe model of the droplet with variable radius on inner wall of the cylindrical tube was established to describe morphology of lipid droplets on inner vascular wall, and accuracy of the theoretical model was verified by Surface Evolver (SE) software simulation results. According to the theoretical model, the influencing patterns of lipid droplet volume and contact angle on dimensionless adhesion area Sb0 and blockage ratio κ of lipid droplets in blood vessels were analyzed. Results The theoretical model could predict contour parameters of adhesion morphology for lipid droplets on inner vascular wall, including the height and arc radius of lipid droplet at azimuth angle of 0 and π/2. The relative errors between contour parameters obtained from the theoretical model and corresponding parameters obtained from the SE simulation were smaller than 10%. For lipid droplets with the same dimensionless volume V0, Sb0 of inner vascular wall decreased with contact angle increasing, and blockage ratio κ increased with contact angle increasing. At the same contact angle, the smaller V0 , the smaller Sb0 and κ would be. Conclusions The established theoretical model with variable radius can well describe morphological characteristics of lipid droplets on inner vascular wall. The influening pattern of volume, contact angle and other parameters on height, adhesion area and cross-sectional area of lipid droplets can be accurately and quickly obtained through the theoretical model, indicating that the larger the contact angle of lipid droplets or the smaller the dimensionless volume, the lower the probability of embolism. The research findings provide theoretical support for the analysis on related diseases.
9.Analysis of clinical characteristics and prognosis of Epstein-Barr virus-related diseases in adults

Objective:To investigate the clinical characteristics and prognosis of Epstein-Barr virus-related diseases in adults.Methods:The clinical data of 59 patients with Epstein-Barr virus-related diseases in Huashan Hospital, Fudan University, Shanghai from January 2017 to August 2019 were analyzed retrospectively. The clinical manifestations of patients with infectious mononucleosis (IM), chronic active Epstein-Barr virus infection (CAEBV) and lymphoma in patients were compared. Patients were divided into acute course group (IM) and chronic course group (CAEBV+ lymphoma), and the results of labratory indications (blood rontine, liver function, imflammatory indications, Epstein-Barr virus DNA, Epstein-Barr virus antibody and T lymphocyte) were compared between two groups. Statistical analysis was performed by Mann-Whitney U test, chi-square test or Fisher exact probability test. Results:Among the 59 patients, 23 cases (39.0%) were diagnosed with IM, 23 cases (39.0%) were lymphoma and 13 cases (22.0%) were CAEBV. The clinical manifestations of patients with Epstein-Barr virus-related diseases were fever (57/59, 96.6%), lymphadenopathy (37/59, 62.7%) and splenomegaly (36/59, 61.0%). There were 17 patients in the chronic course group experienced hemophagocytic lymphohistiocytosis (HLH). The white blood cell counts, hemoglobin levels and platelet counts of patients in the chronic course group (4.07(1.94, 8.35)×10 9/L, 89.5(74.5, 108.0) g/L and 100(37, 161)×10 9/L, respectively) were all lower than those in the acute course group (9.91(6.75, 17.38)×10 9/L, 132.5(118.2, 152.0) g/L and 197(129, 233)×10 9/L, respectively), with statistically significant differences ( U=3.69, 5.22 and 3.61, respectively, all P<0.01). The levels of procalcitonin, C-reactive protein and serum ferritin in the chronic course group (0.45(0.15, 1.13) μg/L, 47.75(17.57, 84.67) mg/L and 2 000(682, 2 002) μg/L, respectively) were all higher than those in the acute course group (0.12(0.07, 0.28) μg/L, 6.39(3.13, 11.38) mg/L and 482(159, 1 271) μg/L, respectively), with statistically significant differences ( U=-2.95, -3.77 and -4.16, respectively, all P<0.01). The counts of CD4 + T lymphocytes, CD8 + T lymphocytes, CD19 + B lymphocytes and natural killer cells in the chronic course group (259.15(101.98, 509.26), 214.69(119.31, 529.47), 46.14(4.44, 135.87) and 81.09(41.53, 118.46)/μL, respectively) were all lower than those in the acute course group (738.88(592.20, 893.94), 1 609.17(920.88, 3 952.34), 144.52(83.65, 215.14) and 309.82(123.78, 590.68)/μL, respectively), with statistically significant differences ( U=3.66, 3.80, 2.90 and 3.40, respectively, all P<0.01), while the CD4 + /CD8 + T lymphocytes ratio in the chronic course group was higher (0.90(0.60, 1.70) vs 0.45(0.10, 1.28))( U=-2.29, P=0.02). Twenty-three patients with IM were all cured, while 10 patients with lymphoma died and 13 received chemotherapy. Seven patients with CAEBV died and six improved. Conclusions:The clinical characteristics of Epstein-Barr virus-related diseases in adults are fever, lymphadenectasis, splenomegaly.Chronic Epstein-Barr virus infection may be associated with HLH. The prognosis of adults with acute Epstein-Barr virus infection is good, while that of long-term chronic Epstein-Barr virus infection is poor.
10.A survey on the influence of parenting style and life events on emotional and behavioral problems in children and adolescents

Objective:To understand the influence of parenting style and life events on children and adolescents′ emotional behavior problems.Methods:The Achenbach′s Child Behavior Checklist (CBCL), Egma Minnen av Bardndosnauppforstran (EMBU), and Adolescent Self-Rating Life Events Check List (ASLEC) were used to investigate the children′s behavior problems, parenting style, and stressful life events among 420 children and adolescents (aged 6-16 years) in Beijing. Logistic regression analysis was conducted to examine the correlation among parenting style, stressful life events, and children′s behavior problems.Results:Among the 420 participants, boys mainly reported problems of social problems (9/44,20.5%), schizoid disorders (7/44,15.9%) and obsessive-compulsive problems (51/124,41.1%); while girls mainly reported depression (63/98,64.3%) and schizoid disorders (86/154,55.8%). Compared to those with normal CBCL scores, mothers of participants with abnormal scores were more likely to have refusal deny (13.1±5.2 vs. 11.8±3.7) and severe punishment (11.4±4.2 vs. 10.5±3.1) ( t=-3.00, -2.38 both P<0.05). Severe punishment ( OR=1.11 95% CI 1.02-1.22), overprotection by father ( OR=3.41,95% CI 1.34-15.96), and mother preference ( OR=1.74,95% CI 1.08-3.11) were the risk factors for boys′ behavior problems, including somatic complaints, immaturity, and social withdrawal behaviors (all P<0.05), while mother preference ( OR=1.31,95% CI 1.04-1.74), refusal deny ( OR=1.23,95% CI 1.06-1.47) and emotional warmth ( OR=1.04,95% CI 1.01-1.08) were the risk factors for girls′ behavior problems, including somatic complaints, cruelty, and immaturity (all P<0.05). Participants with abnormal CBCL total score tended to report greater problems in total stress, interpersonal relationship, learning pressure, loss, and health adaptation than those with normal CBCL total score, and the difference was statistically significant ( t=3.48, 2.87, 2.90, 3.29, 3.16, respectively, all P<0.05). Learning pressure and loss were risk factors of emotional behavior ( OR=1.10,1.21, respectively, both P<0.05). Conclusion:Negative parenting style and life events may affect the children′s emotional and behavior problems.
